Isolation and Characterization of Pathogen-Inducible Putative Zinc Finger DNA Binding Protein from Hot Pepper Capsicum annuum L.

  • To better understand plant defense responses against pathogen attack, we identified the transcription factor-encoding genes in the hot pepper Capsicum annuum that show altered expression patterns during the hypersensitive response raised by challenge with bacterial pathogens. One of these genes, Ca1244, was characterized further. This gene encodes a plant-specific Type IIIA - zinc finger protein that contains two Cys$_2$His$_2$zinc fingers. Ca1244 expression is rapidly and specifically induced when pepper plants are challenged with bacterial pathogens to which they are resistant. In contrast, challenge with a pathogen to which the plants are susceptible only generates weak Ca1244 expression. Ca1244 expression is also strongly induced in pepper leaves by the exogenous application of ethephon, an ethylene releasing compound. Whereas, salicylic acid and methyl jasmonate had moderate effects. Pepper protoplasts expressing a Ca1244-smGFP fusion protein showed Ca1244 localizes in the nucleus. Transgenic tobacco plants overexpressing Ca1244 driven by the CaMV 355 promoter show increased resistance to challenge with a tobacco-specific bacterial pathogen. These plants also showed constitutive upregulation of the expression of multiple defense-related genes. These observations provide the first evidence that an Type IIIA - zinc finger protein, Ca1244, plays a crucial role in the activation of the pathogen defense response in plants.

The Cortical Activation by Functional Electrical Stimulation, Active and Passive Movement (능동 및 수동 운동과 기능적 전기자극에 의한 대뇌 피질의 활성화)

  • We investigated the activation of the cerebral cortex during active movement, passive movement, and functional electrical stimulation (FES), which was provided on wrist extensor muscles. A functional magnetic resonance imaging study was performed on 5 healthy volunteers. Tasks were the extension of right wrist by active movement, passive movement, and FES at the rate of .5 Hz. The regions of interest were measured in primary motor cortex (M1), primary somatosensory cortex (SI), secondary somatosensory cortex (SII), and supplementary motor area (SMA). We found that the contralateral SI and SII were significantly activated by all of three tasks. The additional activation was shown in the areas of ipsilateral S1 (n=2), and contralateral (n=1) or ipsilateral (n=2) SII, and bilateral SMA (n=3) by FES. Ipsilateral M1 (n=1), and contralateral (n=1) or ipsilateral SII (n=1), and contralateral SMA (n=1) were activated by active movement. Also, Contralateral SMA (n=3) was activated by passive movement. The number of activated pixels on SM1 by FES ($12{\pm}4$ pixels) was smaller than that by active movement ($18{\pm}4$ pixels) and nearly the same as that by passive movement ($13{\pm}4$ pixels). Findings reveal that active movement, passive movement, and FES had a direct effect on cerebral cortex. It suggests that above modalities may have the potential to facilitate brain plasticity, if applied with the refined-specific therapeutic intervention for brain-injured patients.

SOUND PRODUCTION AND BEHAVIOUR OF CRABS, PORTUNUS TRITUBERCULATUS(MIERS) (II) (꽃게의 소리와 행동 (II))

  • For the development of acoustic fishing method, we had been researched a fundamental study which concerned on the sound production and behaviour of crabs was conducted. For specimen crabs such as Portunus trituberculatus and Charybdis japonica were selected. Croaking noise were recorded by the Cassette-recorder (Sony model CF-1600) through the under water monitor microphone, and analyzed in frequencies by Octave band analyzer, Rion SA-55, and sound pressure level of source by sound level meter, SM-5844. The following are the results obtained from the present investigations : When sound production of crabs (Portunus trituberculatus( female ) : carapace width $12.6\~15$cm) were attracted to another crabs in the water of anechoic aquarium, efficacy of phonotaxis was $84\~100\%$ and velocity by phonotaxis was $6.5\~7.2cm/sec$. The time required for copulation ranged from 90 minute to 95 minutes by Charybdis japonica, at that time there was no sound production with their copulation.

  • Electric pump system is new technology for next generation propulsion unit. The system has simple structure which dose not need gas generator, injector and turbine and might better pump for low cost and low payload rocket. Therefore, this paper suggests conceptual design of electric-pump Permanent-Magnet Synchronous Motor (PMSM) which has 50 kW & 50,000 RPM for rocket. To satisfy the system's requirement, electromagnetic analysis is conducted for suitable inner and outer diameter of stator and rotor which uses 4000 Gauss cylinder magnet and Inconel 718 can to fix whole rotor. Futhermore, to confirm rotational vibration, rotordynamics analysis is conducted. By this analysis, Campbell diagram is printed. From the diagram, natural frequency could be determined for the only motor and dynamo meter test bench.

Assembling Disjoint Korean Syllables Using Two-Step Rules (2단계 규칙을 이용한 해체된 한글 음절의 결합)

  • With increasing usages of a messenger and a SMS, many young people are habitually using a new-style of sentences with intentionally disjoint Korean syllables. To develop a natural language interface system in these environments, we should first develop a technique that converts a sequence of disjoint Korean syllables to a correct sentence. Therefore, we propose a method to assemble a sequence of disjoint Korean syllables into a correct sentence by using two-step rules. In the first step, the proposed method assembles CVC (consonant-vowel-consonant) forms of simple-disjoint Korean syllables by using manual heuristic rules. In the second step, the proposed method assembles CCVCC forms of double-disjoint Korean syllables by using a mapping table and a transformation-based learning technique. In the experiment, the proposed method showed the perfect precision of 100% in assembling simple-disjoint Korean syllables and the high precision of 99.98% in assembling double-disjoint Korean syllables.

In Vitro Study of Anti-inflammatory and Analgesic Effects of Salvia Miltiorrhiza (SM) Extracts Using Luciferase Reporter Gene Assay (Luciferase reporter gene assay를 이용한 단삼(丹蔘)추출물의 소염, 진통작용에 대한 in vitro 연구)

  • Objectives: In order to identify the anti-inflammatory and analgesic properties of Salvia miltiorrhiza (Dan-Sam), widely used in Korean traditional medicine, an in vitro screening system was designed using pGL3, a luciferase reporter vector, and the tumor necrosis factor (TNF)-${\alpha}$ and cyclooxygenase (COX)-2 as target genes. Methods: The promoter regions of each gene were generated by PCR using the human chromosome as template DNA, and inserted into pGL3 vector with Kpn I and Hind III. The final construct was transfected into human myelomonocytic leukemia cells (U-937) that could be differentiated and activated by phorbol 12-myristate 13-acetate (PMA) or lipopolysaccharide (LPS). Using this system, the anti-inflammatory and analgesic effects of several herbal extracts regarded to have the medicinal effects of diminishing body heat and complementing Qi were tested. The chemicals PD98059 and berberine chloride were used as controls of the transcriptional inhibitors of TNF-${\alpha}$ and COX-2, respectively. Results: Salvia miltiorrhiza (Dan-Sam) demonstrated significant decrease of TNF-${\alpha}$ and COX-2 mRNA in the in vitro assay system. In MTT assay, Salvia miltiorrhiza (Dan-Sam) did not significantly inhibit the survival and proliferation of human myelomonocytic leukemia cells (U-937). Conclusions: Salvia miltiorrhiza (Dan-Sam) was found to exhibit the significant medicinal properties of anti-inflammatory and analgesic effects.

Changes of the Characteristics of ′Kurakatawase′ and ′Mibaek′ Peaches during Storage Period (′창방조생′과 ′미백′ 복숭아의 저장중 특성 변화)

  • This study was conducted to investigate the change of qualities of peaches by different packing types and humidity during storage period. The weight loss ratios of 'Kulakatawase' and 'Mibaek' by non-packing were about 6.8% and 4.9% for 4 days storage at room temperature, respectively whereas, the peaches by packing with 30㎛ LDPE were less than 1% for 25 days storage at low temperature and high humidity(95 $\pm$3%). The firmness values of the peaches were not decreased during storage at low temperature compared to the firmness values of the peaches during storage at room temperature. Little difference of total acidity and soluble solids of the peaches was during storage at low temperature. The contents of fructose and glucose in peaches were increased slightly after storage for 25 days but that of sucrose was decreased slightly. When peaches were stored at low temperature(0∼2$\^{C}$) and high humidity(95$\pm$ 3%) after packing with 30㎛ LDPE or 25㎛ perforated polyolefin film 'Kurakatawase ' and 'Mibaek' were able to storage for 20 and 25days, respectively.

Comparison of Two Vitrification Methods of In Vitro Development Oocytes Collected from Porcine Antral Follicles Using Open Pulled Straw (OPS) Techniques

  • The advantages of the OPS techniques(Vajta G et al, Mol Reprod Dev 51: 53-58,1998) give 1) high survival rates of various types of eggs, 2) quick and simple process, 3) inexpensive equipment and reduced chilling injury. The efficiency of IVM/IVF technique in the porcine species is relatively lower than that obtained in other species such as ruminants. Two experiments were designed to investigate the effects of in-vitro fertilization of porcine oocytes matures using different OPS protocol for chilling and warming of vitrification. Porcine oocytes from ovaries collected at abattoir were matured for 44 hours in TCM199 Earle's salt supplemental with pyruvate, pff, L-cysteine, hormones and gentamycin. Oocytes were denuded and fertilized with frozen boar semen by common method. Porcine embryos produced routinely by in-vitro culture system of NCSU23 medium. The vitrification and the warming were conducted by OPS method with the glass micropipette instead of straw vessels and modified the protocol of G.Vajta(1999). In Exp 1, Chilling/Warming:Holding Medium(HM)+EG+DMSO/HM +sucrose Medium(SM) at 39$^{\circ}C$ warm stage. In Exp 2, : PBS+CS+EG+Ficoll+ Trehalose/PBS+Trehalose at 25$^{\circ}C$ stage. Filling, freezing, packing, thawing out and further culturing were performed to follow the basic protocol of G Vajta. During IVM-lVC and post-warming, fertilization parameter and developmental potential were compared to and statistically analysed. It was not significantly different from Exp 1 and Exp 2 but 25$^{\circ}C$ of stage was slightly higher on the morula/blastocyst forming rate and better atmosphere for worker than that at 39$^{\circ}C$ stage.

SEROTYPES AND ANTIBIOTIC SUSCEPTIBILITIES OF STREPTOCOCCUS MUTANS ISOLATED FROM DENTAL PLAQUES OF CHILDREN (치아우식아동과 그의 모친(母親)의 치태(齒苔)에서 분리(分離)한 Streptococcus mutans의 혈청형(血淸型) 분포(分布) 및 약제내성(藥劑耐性)에 관(關)한 연구(硏究))

  • A total of 141 strains of Streptococcus mutans were isolated from dental plaques of 153 subjects. Eighty-two children with caries-experience, 18 caries-free children and their mothers were participated. All isolates were examined for their serotypes by immunodiffusion method with 7 reference sera and their antibiotic susceptibilities to 7 antibiotics by agar dilution method using 7 kinds of antibiotics, and were compared their results by caries experience (DMFT ratio) and by intrafamilial levels. 1. Isolation rate of Streptococcus mutans were greater in samples of carious teeth than those of sound teeth, and in mothers than in children. 2. Multiple serotypes of Streptococcus mutans were occasionally found in a single samples. 3. Of the total 141 isolates (83 isolates from children, and 58 from their mothers), type c isolates were most prevalent (63.8%). Type d,e and f were found, comprizing 14.9%, 10.6% and 6.4% respectively. Serotype g, a and untypable strain were also found but far lower frequencies (2.8-0.7%), and type b was detected. 4. These results suggest that there are no significant correlation among the distribution of serotypes, antibiotic susceptibilities, caries experience and intrafamilial relationships. 5. Most of isolates were susceptible to chloramphenicol (100%), penicillin (95.7%), ampicillin (94.3%), and gentamicin (92.2%), but about one-third isolates were resistant to cephaloridine (27%), streptomycin (33.3%) and kanamycin (47.6%), resulting that 91 strains (64.5%) among 141 isolates were resistant to one or more drugs used. 6. Of the 91 resistant strains, 20 different resistant patterns were observed, and the most frequently encountered patterns were KM, SM and CE.

Solidification Crackin in Root Pass for One-side Welding of 590MPa Class Steel for Pressure Vessels by FCAW (FCAW에 의한 590MPa급 고장력압력용기강의 초층편면용접부에서 발생하는 고온균열)

  • It is well known that solidification cracking often occurs in welds of root pass for one-side welding under the conditions of high welding currents and speeds. In this study, the solidification in 590MPa class steel for pressure vessels SPPV490 was investigated by using flux-cored arc welding(FCAW) with 4 types of welding wires and welding conditions of 200∼280A and 2.8∼ 4.2mm/sec. In order to compared the result of cracking in SPPV490, 0.2%C steel for welded structure of SWS400 and 0.45%C steel for machine structural SM45C were also used as base metals. As the results, all the cracks formed in some welding conditions were observed near the center of weld bead. The solidification cracks were generally initiated near the upper surface of bead and propagated toward the inner part. The solidification cracking generally increased with welding current and welding speed in the same base metal and welding material. In cracking susceptibility, SPPV490 showed higher cracking susceptibility than SWS400 in all welding conditions and welding materials. It was considered that cracking susceptibility could not be evaluated with the hardness of weld metals. The cracking ratio increased with decreasing of a/b(a and b; the width of the upper surface and the back surface of the bead) as shape factor of bead. The cracking tendency with shape factor of bead was extended under the condition of higher welding currents.
