Zoogloea Ramigera 115SLR로부터 다당류 생합성에 관여하는 유전자의 분리 및 염기서열 결정 (Cloning and Sequencing of a Gene Involved in the Biosynthesis of Exopolysaccharide in Zoogloea Ramigera 115SLR)

  • Zoogloea ramigera 115SLR로부터 다당류 생합성에 관여하는 유전자를 분리하기 위해서 균주의 genomic DNA로부터 제조된 gene bank로부터 plasmid pLEX3이 얻어졌다. 이로부터 재조합된 5.0 kb DNA fragment를 포함하는 plasmid pLEX10은 다당류의 형태를 변환시키는 유전자를 포함하고 있으며, 이중에서 upstream 영역에 해당하는 1.7 kb DNA fragment가 분리되었다. 1.7 kb DNA 염기서열의 결과로부터 단백질을 인지 할 수 있는 2개의 ORF가 존재하였으며, 50 kDa 단백질을 인지 할 수 있는 ORF3은 X. campestris의 다당류 생합성 유전자들인 gumC와 R. meliloti의 exoP와 아미노산의 동질성을 나타내었다. ORF4는 N-terminal 영역이 결여된 단백질을 인지하며, Thermotoga maritime의 다당류 export에 관여하는 단백질과 동질성을 보였다. Z. ramigera 115SLR and Z. ramigera 115SLR/pLEX10은 각각 slime또는 capsule 형태의 다당류를 생합성하며 이들로부터 생합성된 다당류양은 각각 0.26% (w/v) and 0.16% (w/v)였다.

Design and Development of High-Repetition-Rate Satellite Laser Ranging System

  • The Accurate Ranging System for Geodetic Observation - Mobile (ARGO-M) was successfully developed as the first Korean mobile Satellite Laser Ranging (SLR) system in 2012, and has joined in the International Laser Ranging Service (ILRS) tracking network, DAEdeoK (DAEK) station. The DAEK SLR station was approved as a validated station in April 2014, through the ILRS station "data validation" process. The ARGO-M system is designed to enable 2 kHz laser ranging with millimeter-level precision for geodetic, remote sensing, navigation, and experimental satellites equipped with Laser Retro-reflector Arrays (LRAs). In this paper, we present the design and development of a next generation high-repetition-rate SLR system for ARGO-M. The laser ranging rate up to 10 kHz is becoming an important issue in the SLR community to improve ranging precision. To implement high-repetition-rate SLR system, the High-repetition-rate SLR operation system (HSLR-10) was designed and developed using ARGO-M Range Gate Generator (A-RGG), so as to enable laser ranging from 50 Hz to 10 kHz. HSLR-10 includes both hardware controlling software and data post-processing software. This paper shows the design and development of key technologies of high-repetition-rate SLR system. The developed system was tested successfully at DAEK station and then moved to Sejong station, a new Korean SLR station, on July 1, 2015. HSLR-10 will begin normal operations at Sejong station in the near future.

요추부 추간판 탈출 정도와 SLR, valsalva test의 관계 (A Study of the Relationship between Straight Leg Raising, Valsalva Test and Size, Position of Lumbar Disc Herniation)

  • Objectives : This study investigated the relationship between straight leg raising(SLR), valsalva test and size, position of lumbar disc herniation. Methods : We took SLR and valsalva test on 105 patients with lumbar disc herniation. According to the result of MRI findings, this study classified three groups of 105 patients with lumbar disc herniation, bulging, protrusion and extrusion. According to the position of lumbar disc herniation, 72 patients that were diagnosed protrusion and extrsuion were sorted 4 groups, lateral, lateral postero-lateral, central postero-lateral, central. The association size, position of lumbar disc herniation and SLR, valsalva test were analysed. Results : The bigger size of disc herniation, the more positive result of SLR and valsalva test, the lower angle of SLR test. There was not significant association between the position of lumbar disc herniation and the angle of SLR test. Conclusions : The SLR and valsalva test is an useful physical examination to speculate about the degree of lumbar disc herniation.

거창 인공위성 레이저 추적을 위한 운영 시스템 개발 (Development of Operation System for Satellite Laser Ranging on Geochang Station)

  • 한국천문연구원은 인공위성 정밀 궤도 결정, 우주 감시, 우주 측지 등 과학 연구 및 국가적 우주 미션을 수행하기 위해 거창 SLR(satellite laser ranging) 시스템을 개발하였다. 시스템을 구성하는 여러 서브시스템 중 하나인 운영 시스템은 다른 서브시스템을 제어하고 관측 알고리즘을 기반으로 수동 및 자동 관측 모드를 제공하여, 지상에서 인공위성까지의 거리를 계산하는 소프트웨어로써 네트워크 기반의 서버와 클라이언트 방식으로 개발되었다. 본 연구에서는 운영 시스템의 요구사항을 분석하고, 서버 및 클라이언트 통신을 위한 개발환경, 소프트웨어 구조 및 관측 알고리즘을 기술한다. 그리고 개발된 운영 시스템을 이용하여 지상보정 및 측지 전용 인공위성 STARLETTE에 대한 레이저 추적을 통해 취득한 관측 데이터를 처리하여 ILRS(international laser ranging service) 국제기구에서 배포한 전 세계 SLR 관측소와 거리측정 정밀도를 비교 분석하였다.

  • YLPODS (Yonsei Laser-ranging Precise Orbit Determination System) is POD system using SLR (Satellite Laser Ranging) data. YLPODS is developed for two main purposes. The first purpose is to verify the result of POD using GPS data. The second purpose is to perform POD using only SLR data. In this study, YLPODS performance test is presented for checking the reliability of POD using only SLR data. To perform POD, the information of CHAMP and TOPEX mission is applied and SLR NP (Normal Point) data is used. The test is performed by checking both range precision and 3D accuracy (radial, along, cross direction). To confirm of 3D accuracy, CHAMP GENESIS orbit and TOPEX JPL orbit of NASA are used.

  • Several selective excitation pulses are used in MRI. Because of the nonlinearity of the Block equation, the pulse problem is nonlinear generally. Recently, Shinnar & Le Roux have proposed a direct solution of this problem. In this paper, we introduce the SLR algorithm and design pulses using SLR algorithm. This SLR pulse produces a specified slice profile. For example, we demonstrate the sinc function pulse with piece wise constant duration ${\Delta}t$. Further, we will design $\pi/2$ pulse and slice profile.

Localization of Genes Involved in Exopolysaccharide Biosynthesis in Zoogloea ramigera 115SLR

  • Mutants having altered levels and/or types of EPS in exopolysaccharide biosynthesis were isolated after NTG mutagenesis of Zoogloea ramigera 115SLR. Mutant candidates were classfied with five groups based on the observed characteristics for EPS biosynthesis pattern. The recombinant plasmids pLEX3BS and pLEX3BM were constructed from pEX3B which was previously isolated from genomic DNA of Z. ramigera 115SLR. Plasmid pLEX3BM with a 7.8 kb insert DNA contains an additional 1.8kb DNA fragment which is not present in pLEX3BS containing 13 kb insert DNA. Plasmid pLEX3BM was able to complement the mutation responsible for the changes in morphology of Z. ramigera 115SLR. However, the complementation of EPS negative mutant strains was not successful with pLEX3BM. Plasmid pLEX3BS on the other hand complemented the mutation responsible for the loss of EPS biosynthesis, resulting in the restoration of Z. ramigera 115SLR phenotype. But this plasmid was not able to complement the morphological mutant strain, Z. ramigera 115SLR.

Risk Prediction Using Genome-Wide Association Studies on Type 2 Diabetes

  • The success of genome-wide association studies (GWASs) has enabled us to improve risk assessment and provide novel genetic variants for diagnosis, prevention, and treatment. However, most variants discovered by GWASs have been reported to have very small effect sizes on complex human diseases, which has been a big hurdle in building risk prediction models. Recently, many statistical approaches based on penalized regression have been developed to solve the "large p and small n" problem. In this report, we evaluated the performance of several statistical methods for predicting a binary trait: stepwise logistic regression (SLR), least absolute shrinkage and selection operator (LASSO), and Elastic-Net (EN). We first built a prediction model by combining variable selection and prediction methods for type 2 diabetes using Affymetrix Genome-Wide Human SNP Array 5.0 from the Korean Association Resource project. We assessed the risk prediction performance using area under the receiver operating characteristic curve (AUC) for the internal and external validation datasets. In the internal validation, SLR-LASSO and SLR-EN tended to yield more accurate predictions than other combinations. During the external validation, the SLR-SLR and SLR-EN combinations achieved the highest AUC of 0.726. We propose these combinations as a potentially powerful risk prediction model for type 2 diabetes.

  • Satellite laser ranging (SLR) system which measures the round trip time of laser to satellites is one of the important techniques in space geodesy. SLR system gives a powerful tool to determine the precise orbit of satellites, the center of mass of the Earth, and etc because it provides instantaneous range measurements of millimeter level precision. China Transportable Ranging Observation System (TROS) was built in 1999 and other four SLR stations were founded in China. TROS has been upgraded to the new electronic system capable of KHz ranging since last year, and succeeded in KHz SLR technology. TROS has been operated in KASI headquarter for research of space geodesy since August 2008, which will be operated for 12 months by August 2009. Now ISCEA and KASI keep strong relationship in SLR field.
