In Vitro Chemosensitivity Test of SK-302B on Human Colon Carcinoma Cell Lines

  • SK-302B, an antibiotic purified from soil Streptomyces sp. 302, was structurally identified as echinomycin (C/sub 50/H/sub 66/N/sub 11/S/sub 2/). In the present experiment, the possibility of SK-302B as an anticolon cancer agent was investigated by using chemosensitivity system (MTT assay, clonogenic assay). Treatment of SK-302B on various colon cancer cell lines resulted in a significant cytotoxicity and tumor colony formation inhibition. These studies showed that SK-302B had a potent inhibition on colon cancer cells.

  • 광대역 통합망(BcN, Broadband convergence Network)은 유무선 통합, 통신과 방송 융합, 음성 및 데이터의 통합을 통하여 사용자가 원하는 품질과 보안의 정도에 따라 차별화된 광대역 멀티미디어 서비스 제공 및 관리가 가능한 네트워크이다. 본 고는 광대역 통합망의 개념 및 특징을 간략히 설명하고 SK Telecom 주관 UbiNet 컨소시엄의 1단계 (04년${\sim}$05년) 시범 사업을 통해 개발된 BcN 서비스의 내용, 인프라 구조 및 시스템 요구사항 등을 제시한다. 그리고 향후 전개될 2단계 BcN 서비스 및 SKT의 BcN 추진 방향을 제시한다.

The Clinical Effects of an Herbal Antimicrobial Feed Additive in Growing Chickens (천연물 유래 항생제 대체 사료첨가제의 육계 임상 효능에 대한 연구)

  • The natural herbal antimicrobial additive, Flavo-$SK^{TM}$, was developed by Zoonosis Research Center of Wonkwang University. The purpose of this study was to evaluate the effects of Flavo-$SK^{TM}$ on the health status and performance of growing chickens. This study was conducted on the growing chickens (n=20,000) for 31 days in a growing chickens husbandry. The animals were divided with two groups; Flavo-$SK^{TM}$ treated group (n=10,000) and commercial diet feeding group (n=10,000). The Flavo-$SK^{TM}$ treated animals had provided with commercial diet adding the Flavo-$SK^{TM}$ as 0.29%. During the study period, we compared clinical signs, weight increase rate, diet consumption amount, gross finding, necropsy findings and histopathological findings between the treated group and non treated group. As the results of this clinical trial, the natural herbal antimicrobial additive, Flavo-$SK^{TM}$, showed the effects on disease reduction. It is suggested that Flavo-$SK^{TM}$ has the antimicrobial effects.

Viscum Album Var Hot Water Extract Mediates Anti-cancer Effects through G1 Phase Cell Cycle Arrest in SK-Hep1 Human Hepatocarcinoma cells

  • Viscum album var (VAV) also known as mistletoe, has long been categorized as a traditional herbal medicine in Asia. In addition to its immunomodulating activities, mistletoe has also been used in the treatment of chronic hepatic disorders in China and Korea. There are numerous reports showing that VAV possesses anti-cancer effects, however influence on human hepatocarcinoma has never been elucidated. In the present study, hot water extracts of VAV was evaluated for its potential anti-cancer effect in vitro. SK-Hep1 cells were treated with VAV (50-400ug/ml) for both 24 and 48 hours then cell viability was measured by cell counting kit-8 (CCK-8). Flow cytometry analysis was used to measure the proportion of SK-Hep1 in the different stages of cell cycle. RT-PCR and Western blot analysis were conducted to measure expression of cell cycle arrest related genes and proteins respectively. VAV dose dependently inhibited the proliferation of SK-Hep1 cells without any cytotoxicity with normal Chang liver cell (CCL-13). Flow cytometry analysis showed that VAV extract inhibited the cell cycle of SK-Hep1 cells via G1 phase arrest. RT-PCR and Western blot analysis both revealed that cyclin dependent kinase 2 (Cdk2) and cyclin D1 gene expression were significantly down regulated while p21 was upregulated dose dependently by VAV treatment. Combined down regulation of Cdk2, Cyclin D1 and up regulation of p21 can result in cell death. These results indicate that VAV showed evidence of anti-cancer activity through G1 phase cell cycle arrest in SK-Hep1 cells.

Sensitization of TRAIL-resistant SK-Hep1 Human Hepatocellular Carcinoma Cells by Luteolin (SK-Hep1 인체 간암 세포에서 Luteolin에 의한 TRAIL 저항성 감소 효과)

  • In this study, we examined the effect of luteolin to enhance TRAIL-induced anticancer effect in SK-Hep1 cells. We found that combined use of TRAIL with luteolin markedly enhanced the cytotoxicity compared to either agent alone by inducing apoptosis. Furthermore, combined treatment of TRAIL with luteolin significantly induced activation of death receptor pathway-related proteins as well as PARP-cleavage and activation of effector caspases. Also, our result indicated that upregulation of DR4 and DR5 by luteolin combination may contribute to enhanced susceptibility of SK-Hep1 cells to TRAIL.

Molecular Mechanism of the Antiproliferative Effect by Ginseng Panaxynol on a Human Malignant Melanoma Cell Line, SK-MEL-1 (인체 흑색종 세포주 SK-MEL-1에 대한 인삼 panaxynol의 항증식 효과 기전)

  • In this study, the molecular mechanism of the growth inhibitory effect of panaxynol was investigated in a human malignant melanoma cell line, SK-MEL-l. In the cell cycle analysis, panaxynol arrested cell cycle progression of SK-MEL-I at the G1 phase. Immunoblot analysis demonstrated that panaxynol increased $p21^{WAF1}$ and decreased cdc2 expression. Protein levels of pl6, p27, E2F-1, Rb, and p53 were not changed. Thus, the changes in expression levels of $p21^{WAF1}$ and cdc2 apparently mediate the cell cycle arrest caused by panaxynol. In addition, cycloheximide (CHX) partially reversed the growth inhibition by panaxynol, which suggested that new protein synthesis was required. On the other hand, LLnL, a proteasome inhibitor, increased antiproliferative effect of panaxynol. This may be due to stabilization of the protein(s) responsible for the growth inhibition such as $p21^{WAF1}$. In summary, these results demonstrate that panaxynol inhibits proliferation of SK-MEL-I by inducing cell cycle arrest at the G1 phase and the inhibitory effect is mediated by the increased level of $p21^{WAF1}$ as well as decreased cdc2 expression.

Potential prevention effects of Rubus occidentalis seed on UVB-induced MMP-1 production and procollagen degradation in CCD-986sk cells

  • UV exposure induces matrix metalloproteinases (MMPs) and extracellular matrix-degrading enzymes expression. We studied the protective effect of Rubus occidentalis seed against UVB-generated skin photoaging using human fibroblast cells (CCD-986sk). We used an ELISA kit to measure the supernatents of procollagen type I and MMP-1 in CCD-986sk cells after they were exposed to UVB irradiation. The CCD-986sk cells that were used with RC-E/E after the UVB irradiation caused higher levels of type I procollagen and lesser levels of MMP-1 compared with the control group. Furthermore, the RC-E/E treated group showed lesser MMP-1 levels and higher procollagen type I levels than the untreated counterpart. Therefore, it can be concluded that Rubus occidentalis seed can prevent from skin photoaging.
