Cloning of cDNA Encoding Putative Cellular Receptor Interacting with E2 protein of Hepatitis C Virus (C형 간염바이러스 E2 단백질에 결합하는 추정 세포수용체 cDNA의 클로닝)

  • E2 glycoprotein of hepatitis C virus (HCV) comprises a surface of viral particle together with E1 glycoprotein, and is thought to be involved in the attachment of HCV viral particle to receptor (s) on the permissible cells including hepatocytes, B cells, T cells, and monocytes. We constructed a phage library expressing cellular proteins of hepatocytes on the phage surface, which turned out to be 8.8${\times}$$10^5$ cfu of diversity and carried inserts in 95% of library. We screened both cDNA phage library and 12-mer peptide library to identify the cellular proteins binding to E2 protein. Some intracellular proteins including tensin and membrane band 4.1 which are involved in signal transduction of survival and cytoskeleton organization, were selected from cDNA phage library through several rounds of panning and screening. On the contrary, membrane proteins such as CCR7, CKR-L2, and insulin-like growth factor-1 receptor were identified through screening of peptide library. Phages expressing peptides corresponding to those membrane proteins were bound to E2 protein specifically as determined by neutralization of binding assay. Since it is well known that HCV can infect T cells as well as hepatocytes, we examined to see if E2 protein can bind to CCR7, a member of C-protein coupled receptor family expressed on T cells, using CCR7 transfected tells. Human CCR7 cDNA was cloned into pcDNA3.1(-) vector and transfected into human embryonic kidney cell, 293T, and expressed on the surface of the cell as shown by flow cytometer. Binding assay of E2 protein using CCR7 transfected cells indicated that E2 protein bound to CCR7 by dose-dependent mode, giving rise to the possibility that CCR7 might be a putative cellular receptor for HCV.

An Analysis on Behavior Characteristics between Gifted Students and Talented Students in Open-end Mathematical Problem Solving (개방형 문제 해결과정에서 수학 영재아와 수학 우수아의 행동특성 분석)

  • This study is intended to reconsider the meaning of the education for gifted/talented children, the foundation object of science high school by examining the behavior characteristics between gifted students and talented students in open-end mathematical problem solving and to provide the basis for realization of 'meaningful teaming' tailored to the learner's level, the essential of school education. For the study, 8 students (4 gifted students and 4 talented students) were selected out of the 1 st grade students in science high school through the distinction procedure of 3 steps and the behavior characteristics between these two groups were analyzed according to the basis established through the literature survey. As the results of this study, the following were founded. (1) It must be recognized that the constituent members of science high school were not the same excellent group and divided into the two groups, gifted students who showed excellence in overall field of mathematical behavior characteristics and talented students who had excellence in learning ability of mathematics. (2) The behavior characteristics between gifted students and talented students, members of science high school is understood and a curriculum of science high school must include a lesson for improving the creativity as the educational institutions for gifted/talented students, unlike general high school. Based on these results, it is necessary to try to find a support plan that it reduces the case which gifted students are generalized with common talented students by the same curriculum and induces the meaningful loaming to learners, the essential of school education.

  • Recent technical developments have transformed how neuroscientists can probe brain function. What was once thought to be difficult and perhaps impossible, stimulating a single set of long range inputs among many, is now relatively straight-forward using optogenetic approaches. This has provided an avalanche of data demonstrating causal roles for circuits in a variety of behaviors. However, despite the critical role that neuropeptide signaling plays in the regulation of behavior and physiology of the brain, there have been remarkably few studies demonstrating how peptide release is causally linked to behaviors. This is likely due to both the different time scale by which peptides act on and the modulatory nature of their actions. For example, while glutamate release can effectively transmit information between synapses in milliseconds, peptide release is potentially slower [See the excellent review by Van Den Pol on the time scales and mechanisms of release (van den Pol, 2012)] and it can only tune the existing signals via modulation. And while there have been some studies exploring mechanisms of release, it is still not as clearly known what is required for efficient peptide release. Furthermore, this analysis could be complicated by the fact that there are multiple peptides released, some of which may act in contrast. Despite these limitations, there are a number of groups making progress in this area. The goal of this review is to explore the role of peptide signaling in one specific structure, the bed nucleus of the stria terminalis, that has proven to be a fertile ground for peptide action.

A Case analysis on the treatment of mathematics anxiety utilizing a program to change students' thought of mathematics ('생각 바꾸기 프로그램'을 적용한 수학불안 치유 사례분석)

  • This case study examined mathematics anxiety of a public high school sophomore who was unable to perform well in mathematics but later overcame his fear of mathematics. In this study, he showed high levels of mathematics anxiety in the assessment tools that evaluate mathematical anxiety factors. Cognitive and behavior treatments were carried out to alleviate his anxiety. First, cognitive treatments that were implemented include: understanding his own problems, writing down his thoughts on a record sheet, and changing intermediate and core beliefs. This paper explored cognitive and affective changes and reactions during the treatment process. Second, behavioral treatments that were conducted include: the divided-page method and peer tutoring. The divided-page technique involves the test subject to write down and solve his problems on a note to see what kind of cognitive and affective changes occur during the process. This paper also explored how Su-chul, an overly competitive student, changed and reacted cognitively and affectively through peer tutoring. The results revealed that Su-chul's exam anxiety, as well as other factors, has decreased. Moreover, he regained his self-confidence by solving math problems that he had felt difficult. His competitive attitude also has turned into a cooperative and thoughtful one.

Improvement Plans for Increasing Satisfaction among Other Departments Based on The Survey (설문조사를 바탕으로 부서 간 만족도 향상을 위한 개선 안)

  • Purpose Recently, there has been increasing demands for clinical departments at Asan Medical center in Seoul. They want to see the results of lab tests about blood samples as soon as possible and to be reported to them on the day. As the main contents of the survey, we explained the goodness of Asan medical center's in vitro and the points to be improved. Based on this, it became an opportunity to create a positive image of In vitro laboratory. In addition, we could identify their specific requirements through the surveys. Materials and Methods In September 2016, a total of 14 questionnaire surveys were conducted for 49 clinical departments and outpatient nurses at Asan Medical center in Seoul. The survey consists of description questions to be able to express the intention of the individual and the questions made on the Likert 4 point scale. The main contents of 14 questions are composed of goodness of In Vitro laboratory and points to be improved. Results 62% answered that the best service in In Vitro laboratory was "good accuracy and reproducibility". On the other hand, as an inconvenience when requesting blood tests, 73% pointed out that "the result report time was long", which was recognized as a part to be improved. There are many contents that "The result of all tests is reported within 2 hours" on the day of the examination. In the question - 'Are there some examination results which do not coincide well with clinical observations?', 55 of 56 people answered "no". Above all, the majority answered that waiting for re-examination results is too long. This problem must be causing discomfort to the patients. Conclusion In order to improve these problems, the first thing is to increase the number of blood tests by using the current personnel, equipments and reagents to the fullest by item in Asan Medical Center's Nuclear medicine in vitro part. Secondly, in case of re-examinations, we use the "AMIS message" to show other clinical departments the reporting time. This methode improves the efficiency of work with nurses and increases satisfaction of custom service. Thirdly, the correlation was evaluated by selecting the test species that can be carried out by the shortening method. Currently, C-peptide and insulin are implemented in the reaction process to shorten 2 hours into 1 hour. Finally, we are considering purchasing new equipments for quick test results.

Comparative Evaluation of 18F-FDG Brain PET/CT AI Images Obtained Using Generative Adversarial Network (생성적 적대 신경망(Generative Adversarial Network)을 이용하여 획득한 18F-FDG Brain PET/CT 인공지능 영상의 비교평가)

  • Purpose Generative Adversarial Network(GAN) is one of deep learning technologies. This is a way to create a real fake image after learning the real image. In this study, after acquiring artificial intelligence images through GAN, We were compared and evaluated with real scan time images. We want to see if these technologies are potentially useful. Materials and Methods 30 patients who underwent 18F-FDG Brain PET/CT scanning at Severance Hospital, were acquired in 15-minute List mode and reconstructed into 1,2,3,4,5 and 15minute images, respectively. 25 out of 30 patients were used as learning images for learning of GAN and 5 patients used as verification images for confirming the learning model. The program was implemented using the Python and Tensorflow frameworks. After learning using the Pix2Pix model of GAN technology, this learning model generated artificial intelligence images. The artificial intelligence image generated in this way were evaluated as Mean Square Error(MSE), Peak Signal to Noise Ratio(PSNR), and Structural Similarity Index(SSIM) with real scan time image. Results The trained model was evaluated with the verification image. As a result, The 15-minute image created by the 5-minute image rather than 1-minute after the start of the scan showed a smaller MSE, and the PSNR and SSIM increased. Conclusion Through this study, it was confirmed that AI imaging technology is applicable. In the future, if these artificial intelligence imaging technologies are applied to nuclear medicine imaging, it will be possible to acquire images even with a short scan time, which can be expected to reduce artifacts caused by patient movement and increase the efficiency of the scanning room.

Community Structures of Macrozoobenthos in the Subtidal Area of the Nakdong River estuary, Korea (낙동강 하구 조하대 대형저서동물의 군집구조)

  • In order to see the spatio-temporal distribution patterns of macrobenthic fauna in subtidal areas of Nakdong River estuary, benthic fauna samples were collected seasonally at 8 sites in subtidal areas of Nakdong River estuary from February, 2013 to November, 2015. Sandy sediment facies was found in the southern part of the barrier islands in Nakdong River estuary while muddy sediment facies occupied at sites in offshore areas. The bottom salinity of the study area was measured greater than 30 psu during the study period. The mean species richness and density were estimated as 266 species and $859ind\;m^{-2}$, respectively. Polychaete worms were the most abundant fauna group, and four polychaete species such as Goniada maculata, Sternaspis chinensis, Magelona japonica, and Heteromastus filiformis occurred as dominant species in this study area. Pseudopolydora kempi known as an opportunistic species occurred dominantly only in August, 2013. From the multivariate analyses, the macrobenthic fauna in the subtidal area of the Nakdong River estuary could be divided into 2 assemblages that each occupied sites near barrier islands and offshore areas by sediment facies.

Examining the Influence of Fit between Project Radicalness and Organizational Innovativeness on the Supply Chain Integration (프로젝트 급진성과 조직 혁신성 간의 조화가 공급망 통합에 미치는 영향)

  • This study was conducted to empirically examine the impact of fit between project radicalness and innovativeness of the supply chain on the degree of supply chain integration. In order to test the research hypotheses, a survey was employed. The results show that the degree of supply chain integration varies on the fit between project radicalness and innovativeness of the supply chain. Specifically, when a supply chain consists of two morphogenetic organizations and the supply chain integration project is radical, we see very highly integrated supply chain. We also find high-level integration of supply chain when project is radical even though both a morphogenetic and a homeostatic organization compose a supply chain. However, we find opposite result, which is low integration, if a project is incremental rather than radical and a supply chain is composed of same types of the organizations mentioned above. Interestingly, degree of integration seems to be always low when a supply chain is composed of two homeostatic organizations, regardless of project radicalness. With these findings, this study concludes that a project whose radicalness is match with the supply chain's innovativeness should be chosen for successful supply chain integration.

  • This study is progressed function ratio, it's trued taste by an experiment to present data for human work light weight aggregate development that use clink ash progressed liquid limit, small success limit, wear loss in quantity, sand equivalent, sieve cutting examination. 80 : 20's match of function rain examination is 1.4, and that use rubble Goljae as ckink ash lightweight aggregate's capacity ratio increases by 1.0 increase of function rain many. Also, examination multiplied delicate flavor gradually according to increase of the mixing rate, and absorption coefficient increased. This is judged by phenomenon that appear by special quality upper clink ash of polystyrene bid and porosity's increase between lightweight aggregate. It is case that use aggregate of wear loss in quantity is 13.5 in sand equivalent and a wear loss in quantity experiment and although case that mix 20% increases by 14.4, this phenomenon by weak tissue of lightweight aggergate be judge. When it's as a these experiment, the statue prevention floor of a street improvement specifications is prescribing so that satisfy by sand equivalent 20, CBR 10. This is showed result that this satisfies in quality standard all in match experiment ago that see.

Fashion-cultural Products Design Development Based on the Lian Pu of Chinese Beijing Opera: Focused on Chinese Four Major Novels of Wonder (중국 경극 검보를 활용한 패션문화상품 디자인개발: 중국의 사대기서를 중심으로)

  • The Beijing Opera is one of the leading representatives of Chinese culture, which includes literature, music, dance, martial arts, and a type of performance that stems from the Chinese cultural history that is still relevant today. The purpose of this study is to develop fashion-cultural products from the Lian Pu of the Beijing Opera, a Chinese cultural tradition that receives abundant positive feedback from around the world, showing its value in both academic and practical fields. This study was carried out first as a theoretical study of the literature, definition and types of facial make up used in the Opera, as a way of examining the formative aspect. Secondly, an analysis was conducted on the main characters, 'Guan Yu' and 'Zhang Fei' of "The Romance of the Three Kingdoms", 'Li Kiu' and 'Lu Zhishen' of "All Men are Brothers" and 'Monkey King' of "Journey to West", employing the collection belonging to 'Yongqi Zhao' who is an expert on the Chinese Beijing Opera. Thirdly, two concepts were categorized, based on the analytic results of the abovementioned characters, each of which were then further categorized into three sub concepts. In regard to cultural development designs, the results of an analysis on the facial make-up color, form, and texture of the four main characters were utilized to construct the themes, "Modern Chic" and "Traditional Splendor". The simplest form that has been represented in the four figures has been applied to "Modern Chic" to show a modern image in which black, white and light blue has been used alongside the vivid red, which is a Chinese favorite, to highlight the characters. In "Traditional Splendor", which is focused on the stage art of the Opera, we see more artistic traditions and colors, to further appeal to our emotions. Traditional motifs have been applied using traditional Chinese arts, in order to develop strong and brilliant colors. The two styles of cultural products were developed in the form of women's scarves and men's ties; a total of 24 designs were expressed, using Illustrator CS6. In the final step, 4 scarves and 6 ties were produced as a sample, using high quality silk. The development of these cultural fashion products will bring an opportunity to show how Chinese traditional culture can be widely utilized in commercial market design.
