Characteristics of Vibrio anguillarum Isolated from Seawater Cultured Rainbow Trout Oncorhynchus mykiss in Korea (해수 사육 무지개송어(Oncorhynchus mykiss)에서 분리된 Vibrio anguillarum의 특성 분석)

  • From 2014 to 2017, mortalities of seawater-cultured rainbow trout Oncorhynchus mykiss, were observed in the Goheung and Jeju areas of Korea, with Vibrio anguillarum (seven strains: RT1, 2, 3, 4, 5, 6, and 7) identified as the etiological agent. The phenotypic (based on API 20NE, API ZYM, and E-test kits), serotypic (slide agglutination tests with O1, O2, O3, O4, and O7 antisera), and genotypic (16S rRNA and ompU sequencing) characteristics of the seven RT strains were analyzed and compared to those of seven additional V. anguillarum stains (SF, isolated from sweet fish; FM, isolated from flathead mullet; ATCC43305; ATCC43311; ATCC43307; ATCC43308; and KCTC2711). The phenotypes of the RT strains showed variance, while the slide agglutination tests of the RT1-7, SF, and FM strains all showed positive reactions with serotype O1 antiserum. The 16S rRNA and ompU sequences of the RT1-7, SF, and FM strains were affiliated with V. anguillarum ATCC43305 (Serotype O1), but the ompU sequence of the SF strain differed from those of the RT1-7, FM, and ATCC43311 strains, including one amino acid substitution. We thus confirmed that serotype O1 V. anguillarum, with multiple phenotypes, continues to infect seawater-cultured rainbow trout in Korea.

Prevention Strategies for Viral Nervous Necrosis (VNN) in Sevenband Grouper Epinephelus septemfasciatus Aquaculture Farms (능성어(Epinephelus septemfasciatus) 양식장에서의 바이러스성신경괴사증(VNN) 예방대책)

  • Viral nervous necrosis (VNN) is a serious disease of sevenband grouper Epinephelus septemfasciatus in Korean aquaculture farms. However, we suggest the following preventative methods for hatcheries: 1) disinfecting rearing water, 2) selecting spawners via ELISA and PCR, 3) selecting eggs via PCR, 4) disinfecting fertilized eggs, and 5) proper facilities management. When these methods are implemented, nervous necrosis virus (NNV)-free fish are produced because vertical and horizontal transmission is prevented. However, horizontal transmission of NNV through rearing seawater sourced from the environment during grow-out stages in sea cages can still occur. Live NNV vaccines with a low rearing temperature or Poly(I:C) immunization are very effective at preventing horizontal transmission of NNV in rearing farms. Furthermore, even after VNN is contracted, fish mortality can be reduced by administering Poly(I:C).

Eicosapentaenoic Acid Production of Allteromonas putrefacients KS-90 Isolated from Fish Intestines (생선 내장에서 분리한 Alteromonas putrefaciens KS-90에 대한 Eicosapentaenoic Acid 생산)

  • Marine baceteria of fish interstines were screened for high eicosapentaenoic acid(EPA) productivity. An isolated bacteria, KS-90, identified and designated as Allteromonus putrefaciens KS-90. A. putrefaciens KS-90 was found to be a rich source of EPA production and it was observed on incuibation at 4~12.5$^{\circ}C$ and pH 7.0. The production of EPA reached 18mg/g of dry cell weight when A. putrefaciens KS-90 was grown in the medium containing 1.0% pepton, 0.5% yeast extract, 0.025% meat extract and 2.0% glucose in 1/2 concentration of an artificial seawater, pH for 48 hr at $25^{\circ}C$. This value accounted for 24.7% of the total fatty acid in the extractable lipids.

  • Rockfish Sebastes schlegeli was exposed to fluoranthene, a ubiquitous polycyclic aromatic hydrocarbon, at 1 and 10 $\mu$g/L for 4 weeks followed by depuration period of 8 weeks. Although the fluoranthene in the p]asma reached only 1.8$\~$1.9 times seawater concentration, it was 6.5 $\~$ 15.7 times higher in the liver, spleen and bile indicating efficient accumulation in the lipid -containing body tissues. When the exposed fish were then maintained in clean water, rapid fluoranthene decline occurred in the initial 2 weeks followed by a rather slow phase. This result suggests that fluoranthene accumulates efficiently provided the existence in the culture medium, but the contaminant disappears rapidly once the chemical source is removed. The fluoranthne residue in fish tissues my be a good indifator for relent PAHs exposure.

On the Distribution of PCBs and Organochlorine Pesticides in Fish and Sediment of the Asan Bay (아산만해역 어류체내와 퇴적물중의 PCB와 유기염소계농약 분포)

  • Distribution of Polychlorinated biphenyls (PCBs) and organochlorine pesticides in sediments, seawater and fish tissues were examined. Concentrations of ${\Sigma}PCBs$ and ${\Sigma}DDTs$ in the intertidal sediments of Asan Bay ranged from $1.90{\sim}3.27\;ng\;g^{-1}$ dw (dry weight) and $0.51{\sim}1.26\;ng\;g^{-1}$ dw, respectively. ${\Sigma}PCBs$ concentrations in the tissue of greenling (Hexagrammos otakii), olive flounder (Paralichthys olivaceus) and finespotted flounder (Pleuronichthys cornutus) were $1.27{\pm}0.55$, $1.30{\pm}0.71$ and $1.19{\pm}0.43\;ng\;g^{-1}$ ww (wet weight), respectively. The profile of PCBs in the sediments and the 3 species of fish differed as witnessed by the dominance of tetra- to penta-chlorinated congeners in sediments while penta- to hexa-chlorinated congeners dominated in fish. ${\Sigma}DDTs$ concentrations were $0.67{\pm}0.52$, $0.79{\pm}0.61$, $1.58{\pm}1.05\;ng\;g^{-1}$ ww respectively in the tissue of greenling, olive flounder and finespotted flounder. The p,p'-DDE appeared to be the most important metabolite of DDT in studied fishes. The concentrations of PCBs and organochlorine pesticides measured in this study for fish were generally lower than those of previous studies. Compared to the pollutant concentration quality guidelines in marine fish, the environment of Asan Bay appears to be in a healthy state.

  • Currently, land-based fish farms utilizing seawater have introduced and are utilizing various equipment such as real-time water quality monitoring systems, facility automation systems, and automated dissolved oxygen supply devices. Furthermore, data collected from various equipment in these fish farms produce structured and unstructured big data related to water quality environment, facility operations, and workplace visual information. The big data generated in the operational environment of fish farms aims to improve operational and production efficiency through the development and application of various methods. This study aims to develop a system for effectively analyzing and visualizing big data produced from land-based fish farms. It proposes a data visualization process suitable for use in a fish farm big data analysis system, develops big data visualization tools, and compares the results. Additionally, it presents intuitive visualization models for exploring and comparing big data with time-series characteristics.

Optimum Feeding Rate in Growing Olive Flounder Paralichthys olivaceus Fed Practical Expanded Pellet at Optimum Water Temperature (19-21℃) (적수온(19-21℃)에서 배합사료를 공급한 육성기 넙치(Paralichthys olivaceus)의 적정 공급률)

  • We investigated the effects of feeding rate on the growth performance, blood components, and histology of growing olive flounder Paralichthys olivaceus. We determined the optimum feeding rate (initial fish mean weight of $240{\pm}10.9$ g) at the optimum water temperature. Two replicated groups of fish were fed a commercial diet at rates of 0%, 0.5%, 0.75%, and 1.0% body weight (BW) per day, and to satiation. Feeding trial was conducted using a flow-through system with 10 1.2-metric ton aquaria receiving filtered seawater at $19-21^{\circ}C$ for three weeks. Weight gain (WG) for fish fed to satiation was significantly higher than that of unfed fish and fish fed at 0.5% and 0.75% BW per day. The WG of fish fed at 1.0% BW per day was significantly higher than that of unfed fish and of fish fed at 0.5% BW per day. However, there were no significant differences in WG between fish fed at 0.5% BW per day and those fed at 0.75% BW per day, between fish fed at 0.75% BW per day and those fed at 1.0% BW per day, and between fish fed at 1.0% BW per day and those fed to satiation. The specific growth rates of fish fed at 1.0% BW per day and those fed to satiation were significantly higher than those of unfed fish and of fish fed at 0.5% BW per day. Broken-line regression analysis of weight gain showed that the optimum feeding rate of olive flounder weighing 240 g was 1.09% BW per day at the optimum water temperature.

Trimethoprim Resistance of Vibrio parahaemolyticus Isolated from the Fish Farm (어류 양식장에서 분리한 Vibrio parahaemolyticus의 trimethoprim 내성)

  • Vibrio parahaemolyticus species, which cause acute gastroenteritis in humans, were isolated from farmed fish and seawater and their antimicrobial-resistance pattern and factor were investigated. They exhibited the highest resistance to ampicillin (88.9%), followed by trimethoprim (51.9%) and rifampin (22.2%). The relatively high resistance to trimethoprim was unexpected because trimethoprim was not commonly used in fish farming in Korea. R plasmid related resistance was identified by the treatment of novobiocin (7 ug/mL) and it was named as pVPBW1. A putative trimethoprim resistance gene in 2.0 kb fragment of pVPBW1 was also confirmed.

Genotoxic and Neurotoxic Potential in Marine Fishes Exposed to Sewage Effluent from a Wastewater Treatment Plant

  • Concentrations of industrial, agricultural and natural chemicals have been increasing in secondary effluents without their combined sub-lethal effects having been elucidated. In this study, two assays (the comet and acetylcholinesterase assays) were combined to evaluate the genotoxic and neurotoxic effects of effluent from the Noksan wastewater treatment plant (WWTP) on two local marine fish species (flounder and sea eel). The fish were exposed to WWTP secondary effluent that had been diluted with filtered seawater to final concentrations of 1%, 10% and 50%. Analysis of fish samples collected 3 and 5 days after exposure showed that DNA damage occurred in flounder exposed to 50% effluent and in sea eels exposed to 10% or 50% effluent. Furthermore, it was found that acetylcholinesterase (EC:3.1.1.7, AChE) activity decreased in both species when exposed to 10% effluent, indicating the presence of large amounts of genotoxic and neurotoxic chemicals in the effluent. Our results indicate that the comet and AChE assays are promising tools for biomonitoring of secondary effluents.

Species-Specific Duplex PCR for Detecting the Important Fish Pathogens Vibrio anguillarum and Edwardsiella tarda

  • Vibriosis caused by Vibrio anguillarum and edwardsiellosis caused by Edwardsiella tarda are septicemic diseases of many commercially important freshwater and marine fishes, and threaten the aquaculture industry in Korea. Early diagnosis and accurate identification of these two bacterial species could help to prevent these diseases and minimize the damage to cultured marine species. This study designed a duplex polymerase chain reaction (PCR) method for the simultaneous detection of two major fish pathogens: V. anguillarum and E. tarda. Each pair of oligonucleotide primers exclusively amplified the target groEL gene of the specific microorganism. Twenty-two Vibrio and ten non-Vibrio enteric species were used to check the specificity of the primers, which were found to be highly specific for the target species, even among closely related species. The detection limit was 400 pg for V. anguillarum and 4 ng for E. tarda when mixed purified DNA was used as the template. This assay showed high specificity and sensitivity in the simultaneous detection of V. anguillarum and E. tarda from artificially inoculated seawater and fish.
