Chrysanthemum boreale M. is an important medicinal plant that has been historically used in herbal medicine and in the health food throughout East Asia. This study was conducted to investigate the influence of abscisic acid (ABA) and salicylic acid (SA) on plant growth, mineral content and effective components, such as essential oil, amino acid and cumambrin A, by means in order to increase the productivity and the quality of flowerheads in the plant. Yields of flowerheads were increased by 12.7%, 21.7% and 15.5% by ABA, SA and both treatments, respectively, as compared with the control. Inorganic nutrient content was changed by PGRs; SA treatment was increased by nitrogen, phosphorus and magnesium content but decreased by potassium of C. boreale M. flowerheads. Total content of amino acid was increased by SA but decreased by ABA treatment. Essential oil content and yields were increased to 9.7% and 33.8% by SA treatment. Moreover, the content of terpene, monoterpenoids and sesquiterpenoids, were improved by ABA treatment, especially, germacrene-D content was increased by 39.1%, as compared to control. In addition, yields of cumambrin A, sesquiterpene compound exhibiting blood-pressure activity, increased in all PGRs treatments, but its concentration in the C. boreale M. flowerheads only increased by ABA and both treatment. The experiment suggests that PGRs using ABA and SA could increase the yields and quality of C. boreale M. flowerheads.
Purpose: The Helicobacter pylori stool antigen (HpSA) enzyme immunoassay is a non-invasive test for the diagnosis and monitoring of H. pylori infection. But, there are few validation studies on the HpSA test after eradication in children. The aim of this study was to assess the diagnostic accuracy of HpSA enzyme immunoassay for the detection of H. pylori to confirm eradication in children. Methods: From January 2001 to October 2003, 164 tests were performed in 146 children aged 1 to 17.5 years (mean $9.3{\pm}4.3$ years). H. pylori infection was confirmed by endoscopy-based tests (rapid urease test, histology, and culture). All H. pylori infected children were treated with quadruple regimens (Omeprazole, amoxicillin, metronidazole and bismuth subcitrate for 7 days). Stool specimens were collected from all patients for the HpSA enzyme immunoassay (Primier platinum HpSA). The results of HpSA tests were interpreted as positive for $OD{\geq}0.160$, unresolved for $$0.140{\leq_-}OD$$<0.160, and negative for OD<0.140 at 450 nm on spectrophotometer. Results: 1) One hundred thirty-one HpSA tests were performed before treatment. The result of HpSA enzyme immunoassay showed three false positive cases and one false negative case. The sensitivity, specificity, positive predictive value, and negative predictive value of HpSA enzyme immunoassay before treatment were 96.4%, 97.1%, 90%, and 99%, respectively. 2) Thirty-three HpSA enzyme immunoassay were performed at least 4 weeks after eradication therapy. The results of HpSA enzyme immunoassay showed two false positive cases and one false negative case. The sensitivity, specificity, positive predictive value, and negative predictive value after treatment were 88.9%, 91.7%, 80%, and 95.7%, respectively. Conclusion: Diagnostic accuracy of the HpSA enzyme immunoassay after eradication therapy was as high as that of the HpSA test before eradication therapy. The HpSA enzyme immunoassay was found to be a useful non-invasive method to confirm H. pylori eradication in children.
To decrease stress in eel (Anguilla japonica) during its culture or transportation, aspirin (ASA) known as analgesic, antiinflammatory and antithrombic agent was administrated by dipping or oral routes. Concentrations of aspirin (ASA) and salicylic acid (SA) in eel plasma were simultaneously measured by a high performance liquid chromatography (HPLC). The plasma was acidified with 0.2 M HCl and 0.2 M orthophosphoric acid, and mixed with acetonitrile. ASA and SA extracted with acetonitrile were analyzed by the HPLC equipped with reversed phase Novapak C18 column (4 ㎛ silica, 150×4 mm) and UV detector(237 nm). The mobile phase consisted of 740 ㎖ water, 900 ㎕ orthophosphoric acid (85%) and 180 ㎖ acetonitrile. The retention times of ASA, SA and 2-methylbenzoic acid(MBA) were 4.8 min, 8.4 min and 11.5 min, respectively. The limit of quantification was 0.01 ㎍/㎖ for SA and 0.05 ㎍/㎖ for ASA. The mean recovery from eel plasma was 70.8~99.6% for ASA and 95.2~100.3% for SA. This HPLC method was applied to analyze ASA and SA of eel plasma after either dipping in a concentration of 20 ppm or feeding the feed supplemented with 50 ㎎/kg BW. Only SA was detected in eel plasma after the administration of ASA by dipping or oral routes because the drug was quickly decomposed into SA in eel plasma. The amount of SA in eel plasma reached the highest value at 3hr in dipping and 7 days in oral administration. When the ASA-administrated eel were kept in ASA free aquaria, 0.02-0.03 ㎍/㎖ of SA were detected 48 hr after the administration in both routes.
A search of the Streptomyces avermitilis genome reveals that its closest homologs are several O-methyltransferases. Among them, one gene (viz., saomt5) was cloned into the pET-15b expression vector by polymerase chain reaction using sequence-specific oligonucleotide primers. Biochemical characterization with the recombinant protein showed that SaOMT5 was S-adenosyl-L-methionine-dependent Omethyltransferase. Several compounds were tested as substrates of SaOMT5. As a result, SaOMT5 catalyzed O-methylation of flavonoids such as 6,7-dihydroxyflavone, 2',3'-dihydroxyflavone, 3',4'-dihydroxyflavone, quercetin, and 7,8-dihydroxyflavone, and phenolic compounds such as caffeic acid and caffeoyl Co-A. These reaction products were analyzed by TLC, HPLC, LC/MS, and NMR spectroscopy. In addition, SaOMT5 could convert phenolic compounds containing ortho-dihydroxy groups into O-methylated compounds, and 6,7-dihydroxyflavone was known to be the best substrate. SaOMT5 converted 6,7-dihydroxyflavone into 6-hydroxy-7-methoxyflavone and 7-hydroxy-6-methoxyflavone, and caffeic acid into ferulic acid and isoferulic acid, respectively. Moreover, SaOMT5 turned out to be a $Mg^{2+}$-dependent OMT, and the effect of $Mg^{2+}$ ion on its activity was five times greater than those of $Ca^{2+}$, $Fe^{2+}$, and $Cu^{2+}$ ions, EDTA, and metal-free medium.
Characteristics of natural radioactivity were investigated for soils collected from seven sites of different bed rocks distributed in the Keum River area of Korea by the use of a Gamma-ray spectrometry. Specific activity (SA) and SA ratio (SAR) of typical naturally occurring radioactive nuclide such as $^{226}$Ra, $^{228}$Ac and $^{40}$K were determined for the soil samples. The SA values of $^{226}$Ra, $^{228}$Ac and $^{40}$K in 41 soils of 7 sites are 26.7-485 (74.2 ${\pm}$ 72.2), 30.9-157 (90.7 ${\pm}$ 32.7) and 203-1558 (990 ${\pm}$ 203) Bq/kg, respectively. The SA of $^{226}$Ra has very different values by the soils and the sites. Especially the SA of $^{226}$Ra in a soil sample of Ogcheon site is 485 Bq/kg while most SA of 41 soil samples are < 100 Bq/kg. SA of $^{228}$Ac has a little different values with the soils and sites, however the SA of $^{40}$K has almost constant values in all soil samples. The SAR values of $^{26}$Ra/$^{228}$Ac, $^{226}$Ra/$^{40}$K and $^{228}$Ac/$^{40}$K in 41 soils of 7 sites are 0.343-6.11 (0.865 ${\pm}$ 0.883), 0.0258-0.759 (0.0814 ${\pm}$ 0.1l17) and 0.0373-0.178 (0.0945 ${\pm}$ 0.0373), respectively. The SARs of $^{226}$Ra/$^{228}$Ac and $^{226}$Ra/$^{40}$K have very different values by the soils and the sites, however the SAR of $^{228}$Ac/$^{40}$K has a little difference by the soil and sites.
Gold sodium thiomalate (GST, gold compound) is a widely used anti-arthritic, anti-rheumatic and anti-inflammatory drug that is considered a good alternative to sodium salicylate (NaSA) for individuals who cannot tolerate salicylates. Nitric oxide (NO) synthesized by inducible nitric oxide synthase (iNOS) has been implicated as a mediator of inflammation. Recent evidence suggests that anti-inflammatory effect of NaSA lies in the inhibition of iNOS, but nothing has been reported about the direct effect of iNOS expression by GST. The present study was designed to elucidate sequentially the action mechanisms of GST and NaSA on lipopolysaccharide (LPS) plus interferon-gamma (IFN-$\gamma$) induced iNOS expression in RAW 264.7 macrophages. Both GST and NaSA inhibited NO production and iNOS protein expression in a dose dependent manner. GST inhibited iNOS mRNA expression induced by LPS plus IFN-$\gamma$, whereas NaSA did not. These findings suggest that GST may exert anti-arthritic, anti-rheumatic and anti-inflammatory effect by inhibiting iNOS expression induced by LPS plus IFN-$\gamma$ at transcriptional level, whereas NaSA exert its effect by inhibiting iNOS expression at the translational or posttranslational level.
Rice varieties with suitable flour-making quality are required to promote rice processed-food industry and boost rice consumption in Korea. 'Namil (SA)-flo1' is an advanced mutant line with floury endosperm which shows good flour-making quality under dry-milling process. Genetic analysis was carried out to localize the chromosomal region responsible for the floury endosperm of 'Namil (SA)- flo1'. By using 94 F2 progenies, which were derived from 'Namil (SA)-flo1' ${\times}$ 'Milyang 23', floury grains percentage was investigated as phenotypic data, and genotyping was conducted with 54 SSR markers. Association analysis showed that the target genetic region for floury endosperm is on middle-low region of chromosome 5. Through further association analysis with increased number of SSR markers on chromosome 5, we found that genotypic variation in RM164 explains 79.7% of the variation in floury grains percentage of F2:3 seeds. The floury endosperm locus was localized on 17.7-20.7 Mbp region of chromosome 5 and will be further analyzed for fine mapping and gene identification.

Twenty strains of Salmonella paratyphi A, 55 of S. typhi, 7 of Shigella flexneri, and 14 of Sh. sonnei which were isolated in Taegu area in 1977, were tested for the susceptibility to antimicrobial drugs. All strains of S. paratyphi A were resistant to sulfisomidine(Sa), but none was resistant to chloramphenical(Cm), tetracycline(Tc), streptomycin(Sm), ampicillin, nalidixic acid, kanamycin, gentamicin, amikacin, 1:20 mixture of trimethoprim and sulfamethoxazole, carbenicillin, cephaloridine, and rifampicin. Only one strain. of S. typhi was multiply resistant to Cm, Tc, Sm, and Sa, but all strains were susceptible to the other drugs tested. The resistant strain carried R plasmid; R(Cm Tc Sm Sa). All strains except one were highly resistant to Cm, Tc, Sm, and Sa, and all except one of multiply resistant strains carried R plasmid; R(Cm Tc Sm Sa).
The effects of salicylic acid(SA) and water deficit on growth and proline accumulation were investigated in cucumber(Cucurmis sativus L.) seedlings. Exogenous application of SA(100 $\mu$M-1 mM) led to a noticeable decrease in root and shoot growth, and dry weight of seedlings. Anatomical observation on leaf of cucumber revealed that the thickness of all leaf tissue components decreased in SA-treated plants. The effect was most pronounced on the width of the adaxial epidermis. In the separate effects of SA(0, 100, 500 and 1000 $\mu$M) and water deficit induced by PEG(0, 4.4, 7.0 and 9.6 %) on growth, the water deficit treatments had greater effects on growth traits than SA. Combinations of SA and PEG(SA+PEG) decreased shoot and root dry matter, and root length. Proline increased slightly in SA-treated seedlings, but exhibited a marked increase in water deficit application. Combinations of SA+PEG induced higher proline in both shoots and roots than SA stress alone. Shoots had higher proline than roots. Our data support a role of SA potentiating the osmotic stress response of germinating cucumber seedling.
The effect of salicylic acid (SA) on C $d^{2+}$ - induced physiological toxicity in Commelina communis was investigated. 3- weeks old Commelina communis was transferred to and grown in Hoagland solution in the presence or absence of 100 $\mu$M C $d^{2+}$ and SA for 3 weeks. In the treatment of C $d^{2+}$ + SA, the length of stem was increased to 0.7 cm for 3 weeks (C $d^{2+}$, 2.1cm; control, 7.2 cm). C $d^{2+}$ + SA reduced total chlorophyll content up to 86%, and changed chlorophyll a/b ratio below 1.6. C $d^{2+}$ + SA also reduced about 40-78% of water potential, but C $d^{2+}$ increased 16-39% from 1 week to 3 weeks. C $d^{2+}$ + SA also inhibited 27% of Fv/Fm, but in case of C $d^{2+}$, Fv/Fm was not changed. The treatment of C $d^{2+}$ + SA showed about 37-58% inhibition of photosynthetic activity when measured at various light intensity (500-1000 $\mu$mol $m^{-2}$$s^{-1}$ ). In the case of C $d^{2+}$ treatment, photosynthetic activity was inhibited to 12-15%. Similar effect was found in terms of stomatal conductance. Therefore, it could be concluded that the treatment of C $d^{2+}$ + SA into plant decrease or block various physiological activities and lend to die by double effects of both chemicals.cts of both chemicals..
