The purpose of this study was to locate the quantitative trait loci (QTL) associated with quality properties in the recombinant inbred lines derived from the 'Milyang 23' and 'Gihobyeo' cross. Four quality-related traits; protein content, amylose content, fat acid content and sensory value were measured. Eight QTLs for protein content were detected on chromosomes 1 (two loci), 3, 6, 7 and 8 (three loci), each accounting for $6.0\%\~15.2\%$ of the phenotypic variation. Three QTLs for amylose content were detected on chromosomes 6 and 7 (two loci), each explaining from $7.3\%\;to\;24.4\%$ of the phenotypic variation. Six QTLs for fat acid content were detected on chromosomes 2 (two loci), 3, 6 (two loci) and 7, each explaining form $5.5\%\;to\;14.0\%$ of the phenotypic variation. Six QTLs for sensory value were detected on chromosomes 2, 6, 7(two loci) and 8 (two loci), each accounting for $5.5\%\~10.3\%$ of the phenotypic variation.
Streptomyces coeUc%r (Muller) cells were treated with $100 \mu$M hydrogen peroxide for I hour and proteins synthesized during hydrogen peroxide stress were labeled with L-[$^{35}S$]-methionine. Total cellular proteins were extracted and analyzed by two-dimensional polyacrylamide gel electrophoresis. In exponential growth phase, synthesis of about 100 proteins was increased by hydrogen peroxide treatment. These proteins were named as Pin (£eroxide-inducib]e) proteins and classified into 4 subgroups according to their induction time after hydrogen peroxide treatment. About 60 of them were found to be induced within 20 minutes and maintained throughout I hour of treatment. In stationary growth phase. synthesis of 62 proteins was increased by hydrogen peroxide and 21 of them were the same Pins found in exponential growth phase. Proteins from the mutants which are resistant to hydrogen peroxide were obtained in exponential growth phase and compared with those from the wild type on two-dimensional gel. The three mutants, N7, N9. and N24, were found to have higher constitutive leve]s of ]5, 17, and 15 Pin proteins respectively, than the wild type. 9 of these Pin proteins (D74.7a, E76.0c, E23.3. F50.7, F47.2a. F25.5, G39.6b, G24.0, H39.6a) increased in two of the three mutants and 3 proteins (F39.7, H6I.7. 120.8) increased in all of the three mutants. These proteins might play important roles in the response of S. coelic%r to hydrogen peroxide.
Yam, Prunella was stepwise extracted with hexane, chloroform, ethyl acetate, butanol, and water. Anti-microbial activity of each extract was investigated. Hexane extract was tested for anti-microbial effect on Streptocaccus mutans, one of causative factor of dental caries. Methanol extracts of 7 plants were investigated to anti-microbial effects on S. mutans KCTC 5316, P. gingivalis KCTC 5352, S. aureus KCTC 1927 by means of agar diffusion method. Methanol extract of Yam and Prunella revealed anti-microbial activity against S. mutans, P. gingivalis, and S. aureus. Also, hexane fraction of Yam revealed anti-microbial activity against S. mutans. In sequence of hexane, chloroform, ethylacetate, butanol fraction by Prunelia acted as potent anti-microbial agent on P. gingivalis. The measured MIC of hexane fraction of Yam and Prunella on S. mutans KCTC 5316 strain was 0.25 mg/ml and 0.5 mg/ml and the MIC of hexane fraction of Prunella on S. aureus was 0.5 mg/ml. The hexane fraction of Yam and Prunella suppressed viable ceil counts(VCC) of S. mutans, especially after 24 hrs. The Prunella hexane fraction suppressed VCC of S. aureus, after 12 and 24 hrs. Tested concentrations were 0.1, 0.25 and 0.5 mg/ml. the results were compared with control (0 mg/ml). The pH of S. mutans media and GTase activity were determined to evaluate the anticariogenic activity of Yam, Prunella hexane fraction. The pH were increased from 5.6 to 7.0-7.2 in concentration of 2.0 mg/ml. Yam hexane extraction revealed 35% inhibition to GTase activity and Punella inhibited 25% of GTase. These results suggest that the hexane extracts of Yam and prunella have Antibacterial activities against S. mutans, P. gingivalis, S. aureus and have preventive effect on dental caries.

In this paper, S-type transformed MSA and transformed MSA load by a Capacitor not to limit the electric force on the MSA is proposed. Bandwidth of the designed and fabricated S-type transformed MSA is 5.75% at the resonated frequency of 2.24GHz and the resonant frequency and bandwidth versus change of any arbitrary feed point is observed. also, E-plane and H-plane in the Measured radiation pattern characteristic of S-type transformed MSA is compared and analyzed.
The antimicrobial effects of ethanol and organic acids(acetic, citric, lactic. propionic, tartaric acid), either alone or in combination against four foodborne microorganisms (Listeria monocytogenes, Salmonella typhimurium, Staphylococcus aureus and Escherichia coli O157:H7) in tryptic soy broth were determined. Area under the growth curve, minimum generation time, maximum growth rate, and detection time were measured by using automated turbidometer Bioscreen(Labsystem, Finland), for 24 hr at $30^{\circ}C$. All microorganisms were not grown at 7% ethanol in the media. The 0.1% propionic acid showed the strongest inhibitory effects against S. aureus, L. monocytogenes and E. coli O157 : H7 compared with other organic acids, whereas 0.01% organic acids did not show significant inhibitory effect against microorganisms tested (p > 0.01) except S. aureus. The combination of 1% ethanol and 0.01% organic acids were significantly more effective than alone on growth of S. aureus and L. monocytogenes(p < 0.01).
Purpose: Following up the cases of cow's milk-sensitive enteropathy (CMSE), We observed the development of clinical tolerance with cow's milk and other foods. We investigated the clinical outcome of CMSE. Methods: We reviewed the clinical records of patients who had been admitted and diagnosed as CMSE by responses to cow's milk challenge and elimination test and the findings of small intestinal biopsy at Department of Pediatrics, Taegu Catholic University Hospital from March 1992 to March 1997. All of them were being fed with protein hydrolysate before 6 months old, and tried cow's milk and other foods challenge test at following each two month. Twenty-one cases of them returned to be followed. The age at admission was $30.7{\pm}8.8$ (18~47) days old and at survey was $43.4{\pm}23.7$ (16~84) months old. Results: 1) Although the body weight at birth of the patients was 25~75 percentile, all on admission was below 3 percentile. The body weight on interview was 25~75 percentile. 2) The development of clinical tolerance in cow's milk was observed at 16~24 months of age and the tolerance rate was 61% at 12 months of age, 90% at 16 months of age. The development of clinical tolerance in other foods was observed at 10~24 months of age and the tolerance rate was 33% at 12 months of age, 80% at 18 months of age. 3) Adverse reactions after challenge test with cow's milk were observed at 19 cases, manifestated as vomiting (31%), diarrhea (31%), irritability or lethargy (21%), skin rash (10%), and abdominal distention (5%). 4) Comparing serum IgE and milk RAST positive group on admission (5 cases) and negative group on admission (16 cases), there was no significant difference at the age of tolerance in cow's milk (p>0.05), the age of tolerance in other foods (p>0.05), allergy history in family, and the incidence of other allergic diseases. 5) The history of family allergy was observed in 3 cases (14%) in 21 patients and 3 cases (14%) showed rhinitis, urticaria or asthma through a follow-up interview. Conclusion: The development of clinical tolerance in cow's and other foods was sharply increased at 12 months of age and most of all tolerated within 24 months of age. CMSE is a temporary disorder of infancy.
To improve the stability of recombinant pBR322-trip$^+$ plasmid (pLTW24) in E. coli culture, a positive selection system was devised. A DNA fragment containing pheW$^+$ gene (a structural gene for tRNA$^{phe}$ was isolated and inserted into the pBR322-trip$^+$ plasmid(pLTP24). A temperature sensitive host strain. LC901-pheS$^{-ts}$, was constructed for this plasmid by transducing pheS$^{-ts}$ allele (phenylalanyl-tRNA synthetase) to E. coli LC901 using P1kc bacteriophage. The LC901-pheS$^{-ts}$ cells were unable to grow at a restrictive temperature when they had lost the pBR322 :: pheW$^+$ (pLTP24) plasmid. The effects of pheW$^+$ gene on the plasmid stability and the expression level of trip$^+$ gene in LC901-pheS$^{-ts}$ strain were investigated. The proportion of Trip$^+$ colonies among LC901-pheS$^{-ts}$ strain carrying plasmid pLTP24 was 99%, whereas that of LC901 strain carrying plasmid pLTW24 was 7% at the end of 20 generations. After 100 generations of growth, the strain LC901-pheS$^{-ts}$ carrying plasmid pLTP24 showed little loss of plasmids. While the majority of plasmid pLTW24 in LC901 strain were lost in the same period. The activities of tryptophan synthetase (T. Sase) and anthranilate synthetase (A. Sase) in LC901 strain carrying pLTW24 were about 1.2 times and 1.8 times respectively of those in LC901-pheS$^{-ts}$ strain carrying pLTP24.
Ergosta-4,6,8(14),22-tetraen-3-one (1), ergosta-7,24(28)-dien-3-ol (2), and 5,8-epidioxyergosta-6,22-dien-3-ol(3) were isolated from the fruit body of Phellinus pini. Their structures were based on spectroscopic methods including IR, MS, and NMR (1D and 2D). These compounds were screened for their ability to inhibit nitric oxide (NO) production in LPS-activated RAW 264.7 cells. Compounds 1, 2, and 3 reduced NO production in the assay with $IC_50$ values of 29.7 ${\mu}M$ (1), 15.1 ${\mu}M$ (2), and 18.4 ${\mu}M$ (3) respectively. They also suppressed the expression of protein and m-RNA of iNOS and COX-2 in a dose dependent manner by western blot analysis and RT-PCR experiment in LPS-activated microglial cells.
This study was accomplished to illuminate factors of contamination of microbes in the culture medium and effect of antibiotics on prevention of contamination in the medium when bovine follicular oocyte was matured, fertilized and developed in vitro. 1. When washed or unwashed semen diluted with TCM 199 was incubated for 24∼72hr, contamination was come out. 2. When diluted semen with TCM 199 which has penicillin, streptomycin, gentamycin, kanamycin or nystatin was incubated for 24∼72hr, contamination was not come out only in kanamycin. 3. When imported semen which was diluted in TCM 199 with penicillin, streptomycin, gentamycin, kanamycin or nystatin was incubated for 24∼72hr, contamination was not come out in all treatments. 4. When semen which was diluted in BO, CZB, Ham's F10 or TCM 199 was incubated for 24∼72hr, kanamycin showed no contamination in all treatments, but gentamycin showed contamination in CZB, Ham's F10 and TCM 199. 5. When the semen diluted in BO was moved at 24hr after incubation into BO and incubated for 72hr, contamination was not come out, but when it was moved into the TCM 199 and incubated for 72hr, contamination was come out at 48 to 72hr of incubation. 6. When the semen diluted in BO, BO+BSA or BO+FBS containing gentamycin, kanamycin or nystatin was incubated for 24∼72hr, the diluted semen in BO or BO+BSA showed no contamination in all antibiotics but the diluted semen in BO+FBS showed no contamination only in kanamycin. 7.The Pseudomonas cepacia, Serratia liquefaciens, Klebsiella pneumaniae was respectively isolated in the semen of A, B, and C bull and the microbes are highly affected by amikacin, tobramycin and kanamycin. 8. When bovine folicular oocyte was in vitro matured, fertilized and developed in the simple medium with kanamycin, 26.6% was developed to over 32cell stage embryo.
