Changes in the Moisture Stability of $CaS:Eu^{2+}$ Phosphors with Surface Coating Methods

  • To improve the moisture stability of the $CaS:Eu^{2+}$ red phosphor, surface coatings with silica nanoparticles were performed using five different methods, i.e., $P_1$, $P_2$, $P_3$, $P_4$, and $P_5$. The phosphors were coated with silica nanoparticles using a dip coating method ($P_1$) and sol-gel method ($P_2$). The phosphors were coated using a solution containing silica nanoparticles and poly(1-vinyl-2-pyrrolidone), PVP, $(P_3$). The phosphors were also coated with silica nanoparticles by reacting with the 1-vinyl-2-pyrrolidone (VP) monomer ($P_4$) or by reacting with mixtures containing VP and tetraethylorthosilicate ($P_5$). A decrease in the photoluminescence (PL) intensity was observed regardless of the coating methods. However, the moisture stability of the phosphors was enhanced by the coating when aged in a temperature-controlled humidity chamber. Among these methods, the $P_4$ (or $P_5$) method exhibited the greatest increase in moisture stability of the phosphors. The coated phosphors showed a relatively constant intensity with aging time, whereas the uncoated phosphor showed a decrease.

Expression of a $\beta$-1,3-Glucanase Gene from Bacillus circulans in B. subtilis and B. megaterium (Bacillus subtilis와 Bacillus megaterium에서의 $\beta$-1,3-glucanase 유전자의 발현)

  • A Bacillus circulans KCTC3004 $\beta$-1,3-glucanase gene contained in a recombinant plasmid pLM460 derived from subcloning the original recombinant plasmid pLM530 was trasferred into a new shuttle vector plasmid pLMS1180 by ligating linearized DNAs of pLM460 and pUB110. B. subtilis RM125 and B. megaterium ATCC14945 transformed with pLMS1180 produced the $\beta$-1,3-glucanase substantially. Most of the enzyme was produced during the exponential growth period. The maxium activities of the $\beta$-1,3-glucanase produced by the Bacillus transformants were compared with that of the B. circulans gene donor strain. The B. subtilis RM125 (pLM1180) enzyme showed the activity 14 times higher than that of the gene donor cells, followed by the B. megaterium ATCC14945 (pLMS 1180) enzyme with activity 5 times higher than that of the gene donor cells. While E. coli secreted about 7% of the produced enzyme, B. subtilis excreted the enzyme into the medium wholly and B. megaterium about 97% of the total product. The SDS-PAGE of this enzyme produced in E. coli (pLMS1180), B subtilis (pLMS1180) or B. megaterium (pLMS1180) indicated a molecular weight of 38,000. The enzymes overproduced in three different host cells hydrolyzed laminarin to produce mainly laminaribiose, laminaritriose, and laminarioligosaccharides. The plasmid pLMS1180 was stable in B. megaterium, E. coli, but was unstable in B. subtilis.

  • The addition of L-cysteine without blocking amino and carboxyl groups to${\beta},{\beta}$-dinitrostyrene derivatives(11a-e) were investigated. ${\beta},{\beta}$ -Dinitrostyrene derivatives(11a-e) easily undergo addition reactions with L-cysteine to from s-(2,2-dinitro-1-phenylethyl)-L-cysteine(12a), s-[2,2-dinitro-1-(p-methyl)phenylethyl]-L-cysteine (12b), s-[2,2-dinitro-1-(p-methoxy)phenylethyl]-L-cystein e(12c), s-[2,2-dinitro-1-(p-chloro)phenylethyl]-L-cysteine (12d) and s-[2,2-dinitro-1-(p-nitro)phenylethyl]-L-cysteine( 12a), respectively. The structure of adducts were confirmed by means of spectral data, molecular weight measurement and elemental analysis.

  • The liver is an important organ in the regulation of glucose and lipid metabolism. It is responsible for systemic energy homeostasis. When energy need exceeds the storage capacity in the liver, fatty acids are shunted into nonoxidative sphingolipid biosynthesis, which increases the level of cellular ceramides. Accumulation of ceramides alters substrate utilization from glucose to lipids, activates triglyceride storage, and results in the development of both insulin resistance and hepatosteatosis, increasing the likelihood of major metabolic diseases. Another sphingolipid metabolite, sphingosine 1-phosphate (S1P) is a bioactive signaling molecule that acts via S1P-specific G protein coupled receptors. It regulates many cellular and physiological events. Since an increase in plasma S1P is associated with obesity, it seems reasonable that recent studies have provided evidence that S1P is linked to lipid pathophysiology, including hepatosteatosis and fibrosis. Herein, we review recent findings on ceramides and S1P in obesity-mediated liver diseases and the therapeutic potential of these sphingolipid metabolites.

Comparison between the Balance of Skilled and Less-Skilled Players during Successful and Failed Front Kick and Turning Side Kick Motions (태권도 품새 우수·비 우수선수 간 앞차고 몸돌아 옆차기의 성·패에 따른 균형성 비교)

  • The aim of this study is to identify the appropriate movement for maintaining postural balance during Front Kick and Turning Side Kick motions. To do so, ten Taekwondo athletes: five skilled players(S, body mass: $65.0{\pm}5.8kg$, height: $172.3{\pm}3.7cm$, age: $20.0{\pm}1.2yrs$, career: $9.0{\pm}1.9yrs$) and five less-skilled players(LS, body mass: $67.1{\pm}5.5kg$, height: $173.2{\pm}5.1cm$, age: $19.4{\pm}1.7yrs$, career: $9.6{\pm}1.7yrs$) participated in this study. A three-dimensional motion analysis was performed on the participants using eight infrared cameras and two force plate(sampling frequency of 200 Hz and 2000 Hz for S and LS players, respectively). The participants' motions were divided into: a front-kick phase(P1) and a turning-side-kick phase(P2). For P2(p<.05), the range and root mean square(RMS) of the ground reaction torque and the M-L mean velocity of COP were greater for LS than for S; similarly, for P2(p<.05), the M-L range, A-P range, and velocity of the COP were greater for LS than for S. Further, the M-L range and maximum velocity of the COP was greater for failure than for success(p<.05). The femoral biceps muscle for bending the knee joint was significantly stronger in S than in LS(p<.05). It is expected that these results will be useful in developing a training program for improving the balance and stability of Taekwondo poomsae athletes and improve their front-kick and turning-side-kick motions.

Prevalence and Risk Factors for the Weight Loss during Hospitalization in Children: A Single Korean Children's Hospital Experience

  • Purpose: Undernutrition during hospitalization increases the risk of nosocomial infection and lengthens the disease courses. The aim of this study was to evaluate the risk factors of weight loss during hospitalization in children. Methods: All the patients who were admitted in general wards between April and May 2014 were enrolled. Patients aged >18 years and discharged within 2 days were excluded. Weight loss during hospitalization was defined as a decrease in body weight of >2% in 8 hospital days or on the day of discharge. Patients who lost body weight during hospitalization were compared with patients who maintained their body weights. Significant parameters were evaluated by using the multivariate logistic regression analysis. Results: We enrolled 602 patients, of whom 149 (24.8%) lost >2% of their body weight. Complaint of pain (p=0.004), admission to the surgical department (p=0.001), undergoing surgery (p=0.044), undergoing abdominal surgery (p=0.034), and nil per os (NPO) durations (p=0.003) were related to weight loss during hospitalization. The patients who had high weight-for-age tended to lose more body weight (p=0.001). Admission to the surgical department (odds ratio [OR], 1.668; 95% confidence interval [CI], 1.054-2.637; p=0.029) and long NPO durations (OR, 1.496; 95% CI, 1.102-2.031; p=0.010) were independent risk factors of weight loss during hospitalization. The patients with high weight-for-age tended to lose more weight during hospitalization (OR, 1.188; 95% CI, 1.029-1.371; p=0.019). Conclusion: Greater care in terms of nutrition should be taken for patients who are admitted in the surgical department and have prolonged duration of nothing by mouth.

Relationship between maximum bite force and facial skeletal pattern (최대 교합력과 안면 골격 형태에 관한 연구)

  • The purpose of this study was to measure maximum bite force and to investigate its relationship with anteroposterior, vertical, and transverse facial skeletal measurements. From among the dental students at the College of Dentistry, forty subjects (26 male and 14 female) were selected. With two sets of strain gauge, maximum bite force at the right and left first molars and anterior teeth was measured in the morning and afternoon. After taking lateral and posteroanterior cephalograms, fifty and nineteen variables were evaluated, respectively Paired t-tests and an independent t-test were done and correlation coefficients were obtained. 1. The maximum bite force at the first molars was $68.0\pm13.9kg$. in males and $55.6\pm10.5kg$ in females (p<0.05) while the force at the anterior teeth was $8.4\pm4.9kg\;and\;1.1\pm3.4kg$ respectively (p<0.05). 2. Some tendency for a greater value of maximum bite force at the preferred side was observed but not statistically significant (p>0.05). 3. Significant difference was observed between the strong bite force group and the weak bite force group in some cephalometric and other measurements (p<0.05). N-S-Ar, S-Ar-Go, FH-Hl, IMPA and MMO showed a significant difference in posterior maximum bite force (P). N-S-Ar and FH-H1 also showed a significant difference in anterior maximum bite force (A). 4. Several cephalometric variables showed some correlation with maximum bite force (p<0.05). N-S-Ar, S-Ar-Go, UGA, FH-H6, FH-H1, body weight and MMO were significantly correlated with posterior maximum bite force (P). Go-Me, P-1 and IMPA were significantly correlated with anterior maximum bite force (A).

Effects of Antiseptics on the Protein Change of Plasma Membrane in various Fungal Cells (진균류의 원형질막 단백질 변화에 미치는 식품보존제의 영향)

  • Plasma membrane from the fungal cells (Aspergillus phoenicis, Rhizopus acidus, Candida albicans) treated with sodium benzoate (S.B), potassium sorbate (P.S) and calcium propionate (C.P) during the cultivation were separated. The contents and patterns of plasma membrane proteins compared with those of the control. The growth of A. phoenicis was decreased by the average 64.0% in the S.B treatment. That of R. acidus was inhibited by the average 69.0% in the P.S treatment. Also, That of C. albicans was showed the deminution of the average 59.5% in the S.B treatment. The contents of protein involved in the plasma membrane of the each fungal cells were inhibited the average 41.0%, 41.7% and 59.5% in the S.B treatment, respectively. In case of A. phoenicis, the changes in the protein pattern involved in the plasma membraneshowed the aspect similar to the control on the 1st day and 2nd day of cultivation in the treatment group, but $116\;KD{\sim}97\;KD$ band almost disappeared in the 5th day of cultivation, and $45\;KD{\sim}29\;KD$ band was uncleared through the cultivation. In S.B treatment group R. acidus was showed the loss of $116\;KD{\sim}97\;KD$ band from the middle stage of cultivation and P.S, and C.P treatment group were started the loss at the early stage and completely lost at the 36 hours of cultivation. In C. albicans, $116\;KD{\sim}97\;KD$ band were started the loss at the early stage to compare with the control and $66\;KD{\sim}45\;KD$ band were dimmed at the 96 hours of cultivation. Especially, the C.P treatment group were perfectly lost at the 96 hours of cultivation.

Roles of Mitogen-Activated Protein Kinases (MAPKinases) in H-ras-induced Invasiveness and Motility of MCF10A Cells

  • Ras는 세포의 성장과 분화 등 여러 필수적인 세포기능에 없어서는 안될 중요한 역할을 담당하며 Ras가 mutation되면 암 등의 치명적인 결과를 초래한다. Ras 발현은 유방암에서 tumor aggressiveness의 지표로 간주되고 있으며 유방세포의 침습성과 연관이 있다고 알려져 있으므로 ras가 전이과정에 미치는 영향에 관한 연구는 중요한 의미를 갖는다. 본 연구의 선행연구결과, H-ras와 N-ras 모두 transformed phenotype을 나타내지만 H-ras 만이 암전이에 있어서 중요한 침윤성을 유도하는 것을 밝혔다. 이 결과는 MCF10A 세포에서 H-ras와 N-ras에 의한 신호전달경로가 각각 다른 생물학적 전이활성을 나타냄을 시사한다. 세포의 이동성은 침습성에 있어서 결정적인 역할을 하므로, 본 연구에서 H-ras와 N-ras로 형질전환된 MCF10A세포에서 이동성을 시험한 결과, 세포의 이동성이 N-ras가 아닌 H-ras MCF10A 세포에서만 크게 증가된다는 것을 보았다. 이는 침습성을 나타내는 H-ras가 세포의 이동성을 증가시키는데 작용한다는 것을 말한다. H-ras에 의해 유도된 침습성과 이동성에 대한 분자적 기전에 관하여 연구하기 위하여 H-ras MCF10A와 N-ras MCF10A 세포에서 Ras의 downstream effector들, 특히 mitogen-activated protein kinases(MAPKinases)들인 JNK1, ERK, p38의 활성화를 살펴본 결과 p38 MAPKinase가 H-ras MCF10A 세포에서 현저하게 활성화됨을 보았다. p38 MAPKinase 저해제인 SB203580를 처리하던지 dominant negative p38 (DN p38) transfectant로 p38을 불활성화시켰을 때 세포침습성 및 이동성이 저해되는 결과를 얻었다. SB203580 처리한 H-ras MCF10A 세포에서 전이에 관여하는 효소인 MMP-2 분비가 감소되었다. H-ras에 의해 유도된 침습성과 이동성은 DN JNK1 transfectant에서는 변화가 없었으나 DN MEK transfectants에서는 유의성있게 감소되었다. 이상의 결과를 종합하면, MCF10A 세포의 침윤성과 이동성에는 p38 MAPKinase 활성이 중심적인 역할을 하며, JNK 활성은 영향을 미치지 않고, ERK-1/2 활성은 충분하지는 않으나 필요하다는 것을 알 수 있었다.
