Variations of pH Values and EC of Stemflow of 5 Tree Species in Chunchon Districts, Kangwon-do (강원도(江原道) 춘천시(春川市) 지역(地域)에 있어서 5개(個) 수종(樹種) 수간류(樹幹流)의 pH 및 전기전도도(電氣傳導度) 변화(變化))

  • This study investigated pH values and EC changes of rainfall and stemflow were investigated for 5 tree species such as Pinus koraiensis, P. densiflora, Larix leptolepis, Quercus variabilis and Q. mongolica in the Experiment Forests, College of Forest Science, Kangwon National University. The results were as follows; 1. Mean month pH values of stemflow were much lower than those of rainfall. Among tree species, pH values of the coniferous such as P. densiflora, P. koraiensis and L. leptolepis were lower those of the deciduous such as Q. mongolica and Q. variabilis. Mean month pH values of stemflow ranged from 3.23 to 5.03 (average : 4.23) for P. koraiensis, 3.57 to 5.16 (average : 4.36) for P. densiflora, 3.41 to 6.69 (average : 4.45) for L. leptolepis, 3.86 to 6.11 (average : 4.70) for Q. variabilis and 4.39 to 6.13 (average : 5.18) for Q. mongolica. 2. Mean month EC of the coniferous species was 1.5 to 2 times higher than that of the deciduous species. Mean month EC of stemflow for P. koraiensis, P. densiflora, L. leptolepis, Q. variabilis and Q. mongolica was ranged from 29.0 to $946.5{\mu}S/cm$(average : $258.5{\mu}S/cm$), 18.6 to $907.5{\mu}S/cm$(average : $220.3{\mu}S/cm$), 27.0 to $918.0{\mu}S/cm$(average : $216.2{\mu}S/cm$), 16.5 to $301.6{\mu}S/cm$(average : $125.4{\mu}S/cm$) and $25.2{\sim}225.7{\mu}S/cm$(average : $101.5{\mu}S/cm$), respectively. 3. Average pH values and EC were highly variable according to season. Average pH values appeared lower in spring than those in summer and autumn, but EC appeared higher in spring than those lower in summer and autumn. 4. According to precipitation, there was little difference in and pH values of stemflow the coniferous and the deciduous showed low pH valuess in the case of less than 50mm precipitation. However, EC of stemflow showed a difference depending on the precipitation, and EC of the coniferous and the deciduous was high more than $100{\mu}S/cm$ at the precipitation of less than 50mm.

  • We have developed a plasmid vector, pKCH1, for the purpose of higher plant transformation. It contains the promoter region of cauliflower mosaic virus 35S transcript (P35s) and the terminator region of nopaline synthase gene (Tnos) with unique cloning sites, Bam HI and Xba I, between them. After inserting a foreing gene at the cloning sites, P35s-foreign gene-Tnos cassette can be recovered by using a restriction enzyme Hind III.

  • At pH 7.4 (physiological pH), skin is permselective to cations, due to the net negative charge of the current passing channels (pores) in skin. This causes the convective solvent flow (electroosmotic flow) from anode to cathodal direction.   • To understand the properties of the cauliflower mosaic virus (CaMV) 35S promoter and the effect of the deleted maize alcohol dehydrogenase I-S (Adhl-S) intron 1 on the expression of the CaMV $35S{\beta}-glucuronidase$ (GUS) gene in potato (Solanum tuberosum L. cv. Superior), we constructed a chimeric gene and transferred it into potato with Agrobacterium tumefaciens mediated method. The pLS201, a gene transfer vector of 17.7 kilobase pairs, was composed of the CaMV 35S promoter, the 249 base pairs of deleted maize Adhl-S intron 1, the GUS reporter gene, and the kanamycin resistance gene as a selectable marker for transformation. The GUS activity was examined by histochemical and spectrophotometric assay in transformed potato plants. The GUS activity was found primarily around the vascular tissue cells in stem and root. In the spectorophotometric assay, the level of GUS activity of transgenic potato transformed with CaMV 35S/249 bp of intron 1 fragment-GUS (pLS201) was compared with that of potato transformed with CaMV 35S-GUS (pBI121). The quantitative spectrophotometric assay showed that the level of GUS activity in potato transformed with pLS201 was higher in leaf, stem and root by 30-, 34- and 42-fold, respectively than those in potato transformed with pBI121. This results indicate that the inclusion of the deleted maize Adhl-S intron 1 resulted in increament of the GUS gene expression in transgenic potato.potato.

EPIDEMIOLOGICAL EVALUATION OF PERIODONTAL STATUS IN KOREAN ADULTS (한국인의 치주조직상태에 관한 역학조사)

  • To evaluate the periodontal status of Korean adults, 3,988 adults(20-69 years old) who lived in Kwangju, Chonnam, Taegu, and Kyungbuk districts were periodontally examined by plaque index(PI), calculus index(CI), gingival index(GI), periodontal disease index (PDI), and papillary bleeding index(PBI). The periodontal examination charts were grouped according to age and sex. And subsequently 200 periodontal examination charts in each age group were randomly selected according to sex. The differences of the indices among age groups and those between male and female in each age group were statistically analyzed by Chi-Square test. In PI, CI, GI, PDI, and PBI, there was no statistical significance in the difference between male and female in each age group(p>0.05). In male, PI of 60s age group($1.44{\pm}0.68$) was significantly higher than that of any age group(p<0.05), and in female PI of 20s age group($0.86{\pm}0.39$) was significantly lower than that of any other age group(p<0.05). CI of 60s age group in both male($1.67{\pm}0.70$) and female($1.63{\pm}0.91$) was significantly higher than that of any age group(p<0.05). GI of 20s age group in both male($0.85{\pm}0.66$) and female($0.67{\pm}0.60$) was significantly lower and GI of 60s age group in both male($1.37{\pm}0.60$) and female($1.44{\pm}0.84$) was higher than that of any age group(p<0.05). PDI in male was the lowest in 20s age group($0.70{\pm}0.73$), and was the highest in 60s age group($4.93{\pm}1.96$) (p<0.05). In female PDI of 20s age group($0.71{\pm}0.74$) was significantly lower than that of any age group(p<0.05). In male PBI of 20s age group($0.38{\pm}0.31$) was significantly lower than that of any age group(p<0.001). PBI in female was the lowest in 20s age group($0.35{\pm}0.30$) (p<0.001), and was the highest in 60s age group($1.09{\pm}0.76$) (p<0.05). The data insist that all the young populations of 20s age group should be interested in routine check for early diagnosis and prevention of periodontal disease, and a special education program of periodontal health care for young generation must be prepared, and the periodic recall check system for periodontal examination of middle age group must be supported by government.

  • Defects in the mitotic spindle or in the attachment of chromosomes to the spindle are believed to release an activated form of spindle checkpoint complex that inhibits APC-dependent ubiquitination and subsequently arrests the cell cycle at metaphase. When the spindle assembly is disrupted, the fission yeast mitotic arrest deficient (mad) mutants fail to arrest and rapidly lose viability. To enhance our understanding of the molecular mechanisms for the pathway of checkpoint function, the functional characterizations of Mad 1 p from Schizosaccharomyces pombe involved in this process have been carried out. Yeast two-hybrid and various deletion analyses of S. pombe Mad1 p reveal that the C terminus of Mad1p is critical for the binding of Mad2p and maintenance of Mad 1 p-Mad2p interaction. In addition, it was found. that the Mad1p region (residues 206-356) is essential for Mad1p-other checkpoint components. Mad1p truncating this region is sufficient to bind Mad2p but abolishes the checkpoint function, indicating that the checkpoint function is necessary for interaction of Mad 1 p-other checkpoint components. The possible functions of S. pombe Mad1p at the cell cycle checkpoint are discussed.

  • The quantitative separations of a mixture containing equal amount of each anion such as Si(IV), As(V), P(V), S(VI), W(VI) and Cr(VI) are carried out by the elution through 20${\times}3.14cm^2$ column of anion exchange resin, Dowex 1${\times}$8. The eluents are a mixture of 0.07 M hydrochloric acid and 0.03 M sodium chloride (pH = 1.30) for Si(IV), As(V) and P(V) species, a mixture of 0.6 M sodium chloride and 0.3 M sodium hydroxide for S(VI), W(VI) and Cr(VI) species, and 0.1 N sodium sulfite (pH = 3.48) for P(V) and As(V) species. The subsidiary anions in a standard mixture such as Si(IV), As(V), S(VI), P(V) and W(VI) are separated together from large amount of Fe(III) by the elution through 30cm${\times}3.14cm^2$ column of the resin, Dowex${\times}$50w${\times}$12, using a mixture of 0.1 M sodium nitrate and 2 percent dimethylsulfoxide aqueous solution as an eluent. Si(IV), As(V), S(VI), P(V) and W(VI) eluted together are separated quantitatively under the same conditions as in the separations of the anion mixture. By the conditions obtained in the separations of the standard mixture, Fe(III) and all of the subsidiary anions in steel are quantitatively separated.

Phenotypic and Molecular Characteristics of Children with Progressive Familial Intrahepatic Cholestasis in South China

  • Purpose: Progressive familial intrahepatic cholestasis (PFIC) is a rare genetic autosomal recessive disease caused by mutations in ATP8B1, ABCB11 or ABCB4. Mutational analysis of these genes is a reliable approach to identify the disorder. Methods: We collected and analyzed relevant data related to clinical diagnosis, biological investigation, and molecular determination in nine children carrying these gene mutations, who were from unrelated families in South China. Results: Of the nine patients (five males, four females) with PFIC, one case of PFIC1, four cases of PFIC2, and four cases of PFIC3 were diagnosed. Except in patient no. 8, jaundice and severe pruritus were the major clinical signs in all forms. γ-glutamyl transpeptidase was low in patients with PFIC1/PFIC2, and remained mildly elevated in patients with PFIC3. We identified 15 different mutations, including nine novel mutations (p.R470HfsX8, p.Q794X and p.I1170T of ABCB11 gene mutations, p.G319R, p.A1047P, p.G1074R, p.T830NfsX11, p.A1047PfsX8 and p.N1048TfsX of ABCB4 gene mutations) and six known mutations (p.G446R and p.F529del of ATP8B1 gene mutations, p.A588V, p.G1004D and p.R1057X of ABCB11 gene mutations, p.P479L of ABCB4 gene mutations). The results showed that compared with other regions, these three types of PFIC genes had different mutational spectrum in China. Conclusion: The study expands the genotypic spectrum of PFIC. We identified nine novel mutations of PFIC and our findings could help in the diagnosis and treatment of this disease.

Proteolysis Analysis and Sensory Evaluation of Fermented Sausages using Strains Isolated from Korean Fermented Foods

  • We studied the proteolysis and conducted a sensory evaluation of fermented sausages using strains derived from Kimchi [Pediococcus pentosaceus-SMFM2021-GK1 (GK1); P. pentosaceus-SMFM2021-NK3 (NK3)], Doenjang [Debaryomyces hansenii-SMFM2021-D1 (D1)], and spontaneous fermented sausage [Penicillium nalgiovense-SMFM2021-S6 (S6)]. Fermented sausages were classified as commercial starter culture (CST), mixed with GK1, D1, and S6 (GKDS), and mixed with NK3, D1, and S6 (NKDS). The protein content and pH of GKDS and NKDS were significantly higher than those of CST on days 3 and 31, respectively (p<0.05). Sodium dodecyl sulfate-polyacrylamide gel electrophoresis showed that the NKDS had higher molecular weight proteins than the GKDS and CST. The myofibrillar protein solubility of the GKDS and NKDS was significantly higher than that of the CST on day 31 (p<0.05). The GKDS displayed significantly higher pepsin and trypsin digestion than the NKDS on day 31 (p<0.05). The hardness, chewiness, gumminess, and cohesiveness of the GKDS were not significantly different from those of the CST. The GKDS exhibited the highest values for flavor, tenderness, texture, and overall acceptability. According to this study, sausages fermented using lactic acid bacteria (GK1), yeast (D1), and mold (S6) derived from Korean fermented foods displayed high proteolysis and excellent sensory evaluation results.

Desmutagenic Effect of Water Extract from Artemisia capillaris THUNB on the Mutagenicity of Benzo[a]pyrene (Benzo[a]pyrene의 변이원성에대한 인진쑥 물 추출물의 항돌연변이 효과)

  • The antimutagenic activity of the extract of Artemisia capillaris THUNB on the mutagenicity induced by benzo(a)pyrene [B(a)P] in the presense of S9 mixture was studied using bacterial mutagenic assay system. Samples harvested in summer and autumn were extracted using ethanol and hot water. Among these extracts the water extract of summer sample had the strongest inhibitory effect against the mutagenenicity of B(a)P, The water extract of Artemisia capillaris THUNB was separated again into ethanol soluble and insoluble parts. The ethanol insoluble part(El) of water extract exhibited higher inhibition effects than the ethanol soluble part against the mutagenic activity of B(a)P. El showed dose-dependent activity on the mutagenicity of B(a)P in SOS Chromotest and Ames test. The 50% inbibition concentraction $(IC_{50}$ of El were $200{\mu}g/assay$ $600{\mu}g/plate$ and $800{\mu}b/plate$ in E. coil PQ37 S. typhimurium TA100 and TA98 respectively. El were showed desmutagenic effect but had no effect on the DNA repair system for B(a)P-induced mutagenesis. HPLC analysis showed that the formation of aflatoxin M1 by cytochrome P-450 1A1 known as playing an impotant role on B(a) P-induced mutagenicity was highly inhibited by El. Therefore we encluded that B(a)P-induced mutagencity can be reduced possible due to the interference of el with cytochrome P-450 1A1-dependent bioactivation.
