  • The purpose of this study was to investigate the extraction method of phenolic compounds from Rubus coreanum and antioxidative activity. antioxidative activities of Rubus coreanum were tested with ability of donating hydrogen to DPPH, and HPLC, fluorometry which measure the amount of MDA after reacting linoleic acid with $H_2O$$_2$, and LDL with $H_2O$$_2$ and FeCl$_2$. The most suitable extraction conditions of the phenolic compounds from Rubus coreanum was 3 times with 60% ethanol, and the yield of extract containing 35% moisture was 15.28%. In extraction efficacy of phenolic compounds, 60% ethanol was superior to water as extraction solvent, and extraction efficacy with 60% ethanol did not differ from disolving by water after evaporation of 60% ethanol extract. 60% ethanol extract of Rubus coreanum had an ability of hydrogen donating to DPPH, MDA determination showed the antioxidative effect with inhibition ratio of 77.91% on linoleic acid oxidation by addition of Rubus coreanum extract with the concentration of 1.500 ppm. and about 65.74% of LDL oxidation was inhibited by addition of 1,000 ppm.

  • This study analyzed the antibacterial activity of a Rubus coreanum (Bokbunja) ethanol extract. The antimicrobial activity was determined by disc diffusion, minimum inhibitory concentration (MIC), and growth inhibition methods with seven kinds of bacteria related to foodborne illness (Bacillus cereus, Escherichia coli, Escherichia coli O157:H7, Listeria monocytogenes, Pseudomonas aeruginosa, Staphylococcus aureus, and Salmonella typhimurium). In the results, disc diffusions of the ethanol extract from R. coreanum (9.8-17.5 mm at $4,000{\mu}g/disc$) clearly showed the antimicrobial activity of the extract against all tested microorganisms. Rubus coreanum promoted an inhibitory effect as follows: E. coli O157:H7 > P. aeruginosa > L. monocytogenes > E. coli > S. aureus > B. cereus ${\geq}$ S. typhimurium. In the MIC test, R. coreanum showed high antimicrobial effect against L. monocytogenes at 500 ppm. Moreover, the R. coreanum ethanol extract showed strong growth inhibition against microorganisms, similar to the MIC results. These results show that a R. coreanum ethanol extract has powerful antimicrobial activity against all tested microorganisms, suggesting that R. coreanum will be useful as a potential natural preservative.

  • The antimutagenic and cytotoxic activities of ethanol and water extracts from Rubus coreanum were investigated in this study. Their antimutagenic activities were measured by the Ames test and their cytotoxic activities were evaluated by the growth inhibition of cancer cells via the 3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyl tetrazolium bromide (MTT) assay and the sulforhodamine B (SRB) assay. In the results, the inhibition rates of the ethanol and water extracts toward mutagenicity induced by 4-NQO were 95.0% and 93.6% at 5 mg/plate, respectively, while their inhibition rates against mutagenicity induced by sodium azide were 27.2% and 40.8%, respectively. According to MTT assay, the cytotoxicity values of the ethanol extract against Hep3B and HeLa cells were 67.2% and 68.5%, respectively, and the values for the water extract were 65.8% and 66.4%, respectively. In the SRB assay, the ethanol and water extracts inhibited over 60% of cancer cell growth. In conclusion, both the ethanol and water extracts of Rubus coreanum offer potentially good antimutagenic and anticancer effects.

  • Two ellagitannins, pedunculagin and 2,3-(S)-Hexahydroxydiphenoyl (HHDP)-D -glucose and three condensed tannins, (+)-catechin, (-)-epicatechin and procyanidin B-4 which were isolated from Rubus coreanum were evaluated for their antioxidative effects with 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical and lipid peroxidation generation system mediated by addition of $H_2O$$_2$to rat liver homogenate (TBARS). The ellagitannins, 2,3-(S)-HHDP-D-glucose and pedunculagin, showed more potent antioxidative activities by DPPH and TBARS than condensed tannins.

  • To estimate the inhibitory effect of Rubus coreanum extract on melanin biosynthesis, we tested its inhibitory effects on tyrosinase promoter in B16 mouse melanoma cells. Rubus coreanum extract by methanol had inhibitory effect approximately 10% on tyrosinase promoter at $100{\mu}g/mL$. In the MTT assay, the same extract exhibited very low cytotoxicity under $100{\mu}g/mL$. The fractions of methylene chloride, butyl alcohol and water did not showed the inhibitory effect on tyrosinase promoter, but the fraction of ethyl acetate exhibited inhibitory effect approximately 11% at $100{\mu}g/mL$.
