In the present study, we investigated the plant growth characteristics and biological activity of four asparagus cultivars grown using two cultural methods and tested the possibility of domestic open field. The number of shoots, buds, roots, shoot and root fresh and dry weights, and total dry weight of the four asparagus cultivars grown in a plastic house were higher than those of the same cultivars grown in an open field. Of the cultivars grown in the open field, Jersey Giant had greater shoot number than the other cultivars. In plastic house cultivation, the number of buds in Jersey Supreme was greater than the other cultivars. The total flavonoid content of the Jersey Giant was greater than the other cultivars, but cultural method was unaffected. The total polyphenol contents in asparagus cultivars grown in the plastic house were higher than those of cultivars grown in the open field. The total polyphenol content of the Jersey Giant grown the plastic house was significantly higher than those of other cultivars. Antioxidant activity such as catalase (CAT) and peroxidase (POX) did not differ significantly with cultural methods and among the cultivars. Ascorbate peroxidase (APX) activity of asparagus cultivars grown in the open field was higher than that of cultivars grown in the greenhouse; the highest APX activity was detected in UC157. Thus, greenhouse cultivation is expected to improve plant growth characteristics and biological activities of asparagus cultivars; each cultural method should be considered when selecting a suitable cultivar for high yield and high bioactive compound content.
Salinity is one of the major abiotic stressors that inhibits the growth, yield, and productivity of crop plants. Therefore, it is necessary to develop crops with increased salt tolerance for cultivation in saline soils such as is found in reclaimed land. The objective of this study was to develop a salt-tolerant silage rice line that grows on reclaimed land. In order to develop this salt-tolerant silage rice, we transferred Saltol, a major QTL associated with salt tolerance, from IR64-Saltol, a salt-tolerant indica variety, into Mogyang, a susceptible elite japonica variety. To determine the effect of salt stress, Mogyang and IR64-Saltol cultivars were grown on a medium containing various concentrations of NaCl in in vitro conditions. Shoot length was found to decrease with increasing salt concentrations, and root growth was almost arrested at NaCl concentrations over 50 mM in the Mogyang cultivar. Based on these preliminary results, we screened five salt-tolerant lines showing superior growth under salt stress conditions. Polymerase chain reaction and sequencing results showed that the introgression types of Saltol QTL were derived from the IR64-Saltol cultivar in almost all selected lines. Based on the observed growth and physiological characteristics, the new Saltol introgression lines showed higher salt tolerance compared to the Mogyang parental cultivar. The salt-tolerant lines identified in this study could be used as a genetic resource to improve rice salt tolerance.

This study was performed to establish light intensity conditions for producing stem cuttings for aeroponic systems suitable for seed potato production using a closed-type plant production system. Shoot tip cultured plantlets of 'Sumi' and 'Chubaek' potato (Solanum tuberosum L.) were acclimatized, cuttings were collected, and stem cuttings were planted. The seedlings were raised for 40 days at different LED light intensities (60, 120, 180, and 240 μmol·m-2·s-1), and were cultivated in an aeroponic system for 80 days. When stem cuttings were raised at 60 μmol·m-2·s-1 LED light intensity, the plant height was the longest, at 17.3 cm for 'Sumi' and 16.1 cm for 'Chubaek', and the number of nodes was the highest in both cultivars. The higher light intensities, produced smaller plants with fewer nodes. The leaf areas, SPAD values, and Fv/Fm values differed slightly between cultivars. The fresh weight of stem cuttings, and the production rate of healthy stem cuttings were the highest at 60 μmol·m-2·s-1. In the aeroponic system, seedlings raised at 60 μmol·m-2·s-1 with LED light intensity showed a difference between the cultivars, but the fresh weight of stems and leaves above the planting plate was the heaviest. In addition, below the planting plate the stem cuttings were longest and the root weight was heaviest at 60 μmol·m-2·s-1 LED light intensity. The number of stolons also differed between cultivars, but was greatest for seedlings raised at 60 μmol·m-2·s-1 LED light intensity, at 4.2/plant for 'Sumi' and 7.7/plant for 'Chubaek'. At 60 μmol·m-2·s-1 LED light intensity, the tuber number and total tuber weight were the best, but the higher the light intensity, the smaller the total tuber number and total tuber weight for both cultivars. In conclusion, when producing potato stem cuttings for aeroponic systems using a closed-type plant production system, the most suitable LED light intensity for raising seedlings was found to be 60 μmol·m-2·s-1.

Melon leaves showing yellowing symptoms were analyzed using electron microscopy and RT-PCR for major cucurbit-infecting-viruses (CMV, MNSV, CGMMV, SqMV, WMV, KGMMV, PRSV and ZYMV) reported in Korea, but these viruses were not detected. As the result of further analysis by next-generation sequencing (NGS), the virus was identified as Cucurbit aphid-borne yellows virus (CABYV), and then confirmed by RT-PCR using CABYV-specific primers. When photosynthetic capacity was measured based on chlorophyll fluorescence yield (ChlFY), the leaves of the diseased plants showed $4.09{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$, which was one-third of the readings observed for unaffected normal plants ($12.36{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$). The root functions of plants affected by leaf yellowing symptoms (LYS) was $0.28mg{\cdot}g^{-1}$, about half that measured for the normal unaffected plants ($0.48mg{\cdot}g^{-1}$). Cytological observations revealed that there were no morphological differences in the palisade parenchyma and mesophyll spongy cells of the leaves between the diseased and the normal plants. However, the same leaf cells of the affected plants contained more starch granules compared to those of the normal, unaffected plants. We conclude that the LYS of muskmelon is not merely a physiological disorder but a viral disease caused by CABYV and spread by aphids.
The following informations are known from physical and chemical characteristics of orchard soils and nutritional diagnosis of orange leaves in Jeju. 1. Most orange orchards are located on terrace and cindercone thus soil moisture and microclimate of an orchard will greatly be affected by its topography. 2. Excessive well drainage, shallow soil depth, high content of gravels, low solid phase ratio and strong wind will give severe problem of soil moisture and wind errosion, thus the exte- nsion of soil depth is necessary for maintain nutrients, water and sufficient root volume. 3. Available soil water was significantly and positively correlated with organic matter content and clay content also contributes to available soil water. Vinyl mulching was greatly helpful for soil water conservation, wind errosion prevention, soil temperature increases during winter. 4. Abundant amphoteric amorphous allophane take a key role to fix phosphorus and also rations and thus it is the major factor to determine fertilizer efficiency. Lime and phosphorus must be applied in deeper soil layer. Release of filed phosphorus must be reevluated for availability. 5. Organic matter such as see weeds will greatly increase fertilizer efficiency and low fertilizer efficiency during spring may be related to available soil water. 6. Nitrogen was in superoptimum and Mg was enough but P and Ca were somewhat deficient according to leaf analysies while K was deficient according to fruit analysis. Phosphorus application increased sugar/acid ratio and potassium decreases rind percentage. 7. Manganese deficiency and toxicity appeared in a few places. Iron and boron were enough. Most places showed tendency of copper excess but some places showed copper deficiency. 8. Soiling after elimination of rock base, application of slow release fertilizer and abundant organic matter, vinyl mulching and drip irrigation will increase fertilizer efficiency greatly and fruit yield drastically.
An experiment was conducted in 1990~1991 to study the effects of various soil amelioration on the soil productivity and machine workability at tidal land paddy field of Kyewhado Substation, Homam Crop Experiment Station. The soil, Munpo Series(fine sandy loam, Typic Fluvaquents) was treated with gipsum, rice straw, rice straw compost and foreign soil(at 20cm depth) after deep ploughing. The results are surmerized as follows. 1. Sand and clay were slightly increased, while silt was slightly decreased in the rice straw and compost plots. The soil texture was changed from loam to sand loam by the addition of foreign soil 2. Soil bulk density and porosity was decreased in the rice straw, compost and foreign soil addition plots. 3. Cone penetration resistance was $12.5kg/cm^2$ at 10cm of soil depth before experiment and $12.5kg/cm^2$ at 20cm of soil depth after experiment except control, and the root zone was expended down to 20cm. 4. Soil salt content before experiment was 0.46 and 0.48% for surface soil(10cm) and subsoil(20cm), respectively ; The salt content of ameliorated plot was 0.26~0.32% and 0.16~0.31%, respectively, indicating good leaching of soil salt by the soil improvements. 5. The yields of rice in different treatments were in the order of the foreign soil addition > compost > gypsum > rice straw > control.
Tomatoes are flooded differently 0, 5, 10 and 15cm, according to the developing stages such as flowering stage under the condition of greenhouse. Along with this, they are treated according to the time condition such as 6, 12, 24, 48 and 120 hours. The results obtained are summarized as follows. Plant height decreased in the depth of $0{\sim}10cm$ for over 48 hours, in the depth of 15cm for over 24 hours. Number of leaves was the same as in control, and it decreased over. Number of flowers and fruit setting of individuals decreased conspicuously according as the depth and the hours got greater and longer. Adventitious root occurred remarkably in the depth of $0{\sim}10cm$, for over 24 hours and in the depth of 15cm, 12 hours. Epinastic curvature increased greatly as the depth and the hours got greater and longer. Diffusion resistance of stomata cell increased as the depth and the hours got greater and longer. Diseases occurred conspicuously as the hours of flooding got longer rather than as the depth greater. The preventing of diseases caused by insecticide was observed, but it was not greater than in the seedling and transplanting stage. Fertilization was effective in the case of increasing the weight of shoot. Number of fruits per plant did not decrease in the depth of 0cm up to 24 hours, but decreased on the deeper level of flooding and increased as the hours got longer. Moreover with the exception of 120 hours per respective depth of the treatment, average weight of a fruit got greater as the depth and the hours got greater and longer. In the case of epinastic curvature and diffusion resistance, there was negative correlation between all the other investigated characters and positive correlation between weight of a fruits and average weight of a fruit.
Soybean is the most promising crop for substituting rice on the paddy field. Excessive water stress is a common limiting factor in soybean yield under paddy soil condition. This study was carried out to identify changes in leaf total nitrogen and petiole ureide content under excess water conditions for establishing a screening system related to waterlogging tolerance. Waterlogging treatment was conducted by maintaining the water level on the soil surface for 10 days at the early vegetative growth stage ($V_5$) and the flowering stage ($R_2$). Leaf total nitrogen content, SPAD value and ureide content in petiole decreased in all soybean varieties in response to waterlogging, but the degree of decrease was much lesser in Pungsannamulkong and Muhankong than in Jangyeobkong and Myungjunamulkong, at 21 days after waterlogging treatment. This result means that root and nodule recovery rates were much higher in Pungsannamulkong and Muhankong than in Jangyeobkong and Myungjunamulkong after waterlogging treatment. The ureide and leaf nitrogen content showed high positive correlation with SPAD value, regardless of waterlogged stages. In conclusion, leaf nitrogen content, ureide content in petiole and leaf greenness were identified as promising indicator for screening soybeans which are tolerant of excess water.

This study was conducted to investigate the effect of some kinds of tree litters covered the soil surface on the germination, establishment, sward composition, and herbage production of the grasses sown by the oversowing method. The litters were leaves of Pinus rigida Miller, Quercus variabilis Blume, Larix leptolepis Gord, and Alnus hirsuta Rupr. The results obtained were as follows; 1. In the Petri dish, germination was affected by grass species and kinds of litters, especially the percolate from Pinus litter seriously suppressed the early germination of small size seeds such as ladino clover and timothy. 2. In the pot, establishment was slightly suppressed in ladino and timothy by the percolate from Pinus litter, while in others it was decreased relatively. 3. Among the 6 species investigated, the small size seeds of ladino clover and timothy and the large size seeds of red clover, they could be early put into the litter spaces and established well. 4. In Larix litter, the establishment percentage of all species were decreased seriously owing to low moisture retention capacity. 5. Grass height and root length were different in kinds of litters and species. 6. The percentages of establishment on each surface treatments of burning, treading, raking, and the control under Quercus tree were 36, 46, 37, and 31%, respectively. 7. The sward percentage of oversown grasses and legumes was 57.8% in burning, 70.9% in treading, 59.6% in raking, and 54.0% in the control. 8. Treading treatment was most effective to destroy existing vegetation and improve soil-seed contact when oversowing and showed the best result. 9. Yield of dry matter per unit area in treading treatment was higher (p<0.05) than those of the other treatments. The above experimental results suggest the importance of direct oversowing on the litters in the mountainous land and forest land.
The physical properties of seven perlites different in particle size distribution were investigated to develop perlite bag culture in Korea. Particle sizes of 1.0-2.8mm and larger than 2.8 mm were rather evenly distributed in S-1 (1.2-5 mm), S-2 (0.15-5 mm) and S-5 (parat No.1). Larger particles were less in S-3 (1-3 mm), S-4 (Parat No.2), S-6 (OTAVI) and S-7 (Agroperl B-3). S-4, S-6 and S-7 contained lots of particles less than 1 mm in size. Total porosity was similar among substrates with the value of $59{\sim}62%$. Container capacity was between 35-40% regardless of substrates except in S-2 with 27.7%. Water content, which was about 60% at 0 kPa, was decreased sharply at 4.90 kPa regardless of substrates, which meant the easily available water was plenty in any kind of perlite tested. Substrates, S-1, S-2 and S-3 with different particle size distribution, were investigated to evaluate for perlite bag culture. Six tomatoes (Licopersicon esculentum Mill. cv. Rokkusanmaru) were planted in a perlite bag of 40 liters with the dimension of 120cm in length and 34cm in width. The amount of nutrient solution supplied and its drainage dependent on daily integrated radiation didn't show any regular trend during the growth. Roots in the bag were distributed evenly in S-1 and S-2 than in S-3. Plant grown in S-1 showed the highest total and marketable yield of 8,628 and 7,759 kg/10a, respectively. The number of small size fruits and malformed fruits were more in S-3. Consequently, S-1 with the particle size distribution of 1.2-5 mm is suggested as desirable substrate for perlite bag culture.
