수종 근관충전세멘트의 Streptococcus Sanguis에 대한 항균효과에 관한 실험적 연구 (ANTIMICROBIAL EFFECT OF ROOT CANAL CEMENTS ON STREPTOCOCCUS SANGUIS)

  • This study was designed to compare the antimicrobial effect of the several root canal cements and to determine the duration of their activity. After Strep. sanguis 9811 was streaked on the surface of BHI agar and Plain agar, PVC tubes filled with root canal cements were applied and cultured for 48 hours, aerobically. Following results were obtained, 1. All of the examined root canal cements had antimicrobial activity with varying degree. 2. The antimicrobial activity of N2 was larger than any other root canal cements. Nogenol and Tubliseal showed the lowest activity. ZOE, AH26, Procosol and PCA cement showed the moderate antimicrobial activity. (P>0.05) 3. At one day after mixing the root canal cements, all of the root canal cements except N2 and PCA cement showed greatly reduced antimicrobial activity. 4. At three days after mixing, PCA cement showed the reduced antimicrobial activity. N2 showed the reduced activity at 7 days after mixing.

한국독활(韓國獨活)과 중국독활(中國獨活)의 항산화효능 비교 연구 (Comparative Studies on the Anti-oxidation Activities of Aralia continentalis Root and Angelica pubescens Root)

  • Objectives : The present study compared Aralia continentalis Root and Angelica pubescens Root used in Korea and China respectively concerning their anti-oxidant effect Methods : We tested the anti-oxidant effect through in vitro experiment and in vivo experiment that induced oxidative stress using ethanol. Results : 1. DPPH scavenging activity was stronger in Aralia continentalis Root than in Angelica pubescens Root 2. Superoxide anion radical scavenging activity was similar between Aralia continentalis Root and Angelica pubescens Root 3. The linoleic acid peroxidation inhibition effect was stronger in Aralia continentalis Root than in Angelica pubescens Root. 4. The phenolic component was higher in Aralia continentalis Root than in Angelica pubescens Root. 5. Both Aralia continentalis Root and Angelica pubescens Root increased the concentration of GSH and decreased SOD activity in mice, in which oxidative stress was induced, and the effect was stronger in Aralia continentalis Root. 6. Aralia continentalis Root increased GSH peroxidase activity but Angelica pubescens Root did not have such an effect. 7. Neither Aralia continentalis Root nor Angelica pubescens Root had a significant effect on catalase, ADH and ALDH in mice, in which oxidative stress was induced. Conclusions : Aralia continentalis Root has a stronger anti-oxidant effect than Angelica pubescens Root. Thus, although Aralia continentalis Root is not an original plant recorded in botanical literature, it may be usable based on the data about its effects.

Increasement of Eleutherosides and Antioxidant Activity in Eleutherococcus senticosus Adventitious Root by Jasmonic acid

  • This study was carried out to investigate the impacts of jasmonic acid (JA) on adventitious root culture of Eleutherococcus senticosus. Adventitious root of E. senticosus were treated with jasmonic acid (JA) and cultured for 30 days. JA inhibited the root growth but increased eleutherosides accumulation, total phenolic contents and antioxidant activity. Among various concentrations of JA, 1.0 mg/L JA increased the total phenolic contents in E. senticosus adventitious root to $39.81{\mu}g/g$, about 2.6 times higher than that of the control. Consequently, high accumulation of total phenolic contents led to increase the antioxidant activity to 82.41%. The antioxidant activity of control was 37.89% at $2500{\mu}g/mL$. A linear correlation ($R^2$ = 0.9937, 0.9648 and 0.9883) was also shown between antioxidant activity (at 1250, 1875, and $2500{\mu}g/mL$) and total phenolic contents of adventitious root of E. senticosus.

당근 Hairy root의 성장 및 Anthocyanin 함량에 미치는 Polyamime의 영향 (Effects of Polyamine on Growth and Anthocyanin Contents of Carrot Hairy Root)

  • The effects or polyamines on growth, anthocyanin contents and $\beta$-glucan synthetase(GSII) activity in carrot hairy root were studied. Growth of hairy root was stimulated somewhat when each polyamine concentration was treated, especially addition of 1mM spermidine resulted in about 20% increase. On the whole, the axial diameter of hairy root was increased in response to increase in concentration of polyamine. On the other hand, GSII activity was stimulated in response to increase in concentration of polyamine, especially addition of 1mM spermine resulted in about 100% increase of activity. Therefore increased activity of GSII stimulated growth and thickness of hairy root. Anthocyanin contents were not affected by the polyammine.

마치현 부위별 에탄올 추출물의 항산화 활성 (Antioxidant Effects of Ethanol Extracts from Different Parts of Portulacae Herba)

  • Objectives : This study reviewed the antioxidant activity of each part of Portulacae Herba (PH) in order to better analyze the possibility of PH being used as a natural material. Methods : The root, stem, and leaves of the PH were separated, dried at $50^{\circ}C$, put in a 70% ethanol and then extracted three times every six hours. Antioxidant activities was performed by measuring the total polyphenol and total flavonoid contents, DPPH, ABTS, nitrite, hydroxyl radical scavenging activity, and $Fe^{2+}$ chelating. Results : The total polyphenol and total flavonoid contents was highest in the root, then the leaves, and lastly the stem. DPPH and ABTS radical scavenging activities for the root extract indicated the highest activity followed by the leaf and stem extracts. It also showed similar activity to that of the ascorbic acid. Hydroxyl radical scavenging activity and $Fe^{2+}$ chelating activity resulted to be highest in the root extract and then the leaf and stem extracts. Nitrite scavenging activity decreased with higher pH levels, and activity was highest in the order of root extract, leaf extract, and stem extract. Also, in reaction to a solution with a pH 1.2 level, the root extract showed similar scavenging activity to that of BHT. Conclusions : Therefore, based on the above results, it was verified that the roots of PH have a high antioxidant level, and they can be considered to have potential to be applied to functional food, cosmetics, and medicine with antioxidant efficacy.

Phytotoxicity and DPPH Radical Scavenging Activity of Barley Seedling Extracts

  • A series of methanol extracts from leaf and root parts in spring- and winter-barley plants were assayed to determine their allelopathy and antioxidant activity. The methanol extracts applied on filter paper in a Petri-dish significantly inhibited root growth of Chinese milkvetch (Astragalus sinicus L.) seedlings. Leaf extracts at 25 and 50 g $L^{-1}$ inhibited root growth of Chinese milkvetch seedlings more than root extracts. No difference in phytotoxic effects of spring- and winter-barley seedlings extracts on root growth of Chinese milkvetch was observed. Methanol extracts dose-dependently increased DPPH free radical scavenging activity in vitro. DPPH free radical scavenging activity was higher in the methanol extracts from winter-barley seedlings than in those from spring-barley seedlings, and from leaf extracts than from root extracts. The antioxidant potential of the individual fraction from the methanol extracts of spring-barley seedlings was in order of n-butanol>ethyl acetate>water>chloroform>n-hexane fraction. By means of HPLC analysis, spring-barley (200.62 mg $100g^{-1}$) had more amount of total phenol acid than winter-barley (114.08 mg $100g^{-1}$). Especially, ferulic acid was detected in spring-barley extract (183.46 mg $100g^{-1}$) as the greatest amount. These results suggest that early seedlings of barley plants had potent allelopathy and antioxidant activity, and their activities were differently exhibited depending on plant parts and growing condition.

Antioxidant Effect of Tea Tree Root Extracts using Various Extraction Methods

  • To investigate antioxidant effects of tea tree root extracts using various extraction methods, cytotoxicity, DPPH and ABTS radical scavenging, SOD, nitrite scavenging activity and inhibitory activity of lipid peroxidation, reducing power, ferrous ion chelating activity were measured. Cytotoxicity for RAW 264.7 cells was not observed at concentrations treated with below 90 ㎍/mL in all extracts. The maximum DPPH radical, nitrite scavenging, SOD activity and inhibitory activity of lipid peroxidation were obtained at the ethylacetate and 70% ethanol extract. The maximum ABTS radical scavenging activity was obtained at the ethylacetate and hot water extract. However, in the case of reducing power and ferrous ion chelating activity, they were obtained at 70% ethanol and hexane extract, respectively. Nitrate scavenging activity showed the most excellent scavenging ability of 59.6% at 90 ㎍/mL of ethylacetate. The hexane extract had the highest ferrous ion chelating activity, showing 61.05% at 50 ㎍/mL, 66.07% at 70 ㎍/mL and 76.81% at 90 ㎍/mL, respectively. The results of this research show that the ethylacetate and 70% ethanol extracts of tea tree root can be used as a natural material for scavenging the radicals. However, future study is necessary to understand the mechanism of antioxidant activity by identification of substances.

6년생 인삼(연풍)의 뿌리부위별 항산화 활성 (Antioxidant Activity from Different Root Parts of 6-year-old Panax ginseng C.A. Meyer (Yun-poong))

  • 6년생 인삼(Panax ginseng C.A. meyer, 연풍)의 뿌리를 주근, 지근, 세근으로 나누고 각각의 중앙부위의 직경에 따라, 직경이 큰 것에서부터 1에서 4까지 등급을 나누었다(MR 1~4, LR 1~4, FR 1~3). 각각의 시료는 동결건조한 뒤 환원당 함량, 페놀함량 및 항산화 효과를 비교하였다. 환원당 함량은 직경에 따른 영향은 없었으나, 주근, 지근, 세근으로 부위가 세분화됨에 따라 환원당 함량이 감소하는 경향을 나타내었다. 총 페놀화합물의 함량은 FR-3이 8.19 mg/g으로 다른 부위보다 2배 정도 높은 페놀함량을 나타내었다. 수소 공여능의 $IC_{50}$값을 비교한 결과 FR-3의 $IC_{50}$ 값이 7.03 mg/mL로 가장 높았고, MR-2에서 37.02 mg/mL로 가장 낮았다. 전체적으로 세근의 수소공여능이 주근과 지근에 비해 높았다. ABTS 라디칼 소거능 역시 FR-3에서 24.87%의 가장 높은 라디칼 소거능을 나타내, 다른 부위보다 세근의 활성이 유의적으로 높음을 알 수 있었다. FRAP value의 측정 결과 역시 FR-3이 $0.23{\mu}M$로 가장 높게 나타났다. 본 연구를 통해 인삼 연풍의 항산화 활성은 주근과 지근보다는 세근에서 유의적으로 높게 나타났으며, 세근 중에서도 직경이 작은 부위에서 높은 페놀함량 및 항산화 활성을 나타내는 것을 알 수 있었다.

추출조건에 따른 도깨비부채 뿌리의 멜라닌 억제효과 (Extraction Conditions Affect Melanin Inhibitory Activity in Rodgersia podophylla Root)

  • Background: The inhibitory effect of Rodgersia podophylla root extract on melanin production has been reported, however, the study on the optimal extraction conditions that increase melanin inhibitory activity has not yet been performed. Methods and Results: In this study, we compared the melanin inhibitory activity of R. podophylla root extract obtained through different preprocessing and extraction methods. The melanin inhibitory activity was examined using Melan-A melanocytes. The results indicated that the inhibitory activity of R. podophylla roots collected in August was higher than that of the roots collected in May and November. Additionally, non-dried R. podophylla roots exhibited higher activity than dried roots, and the stirring extract was more active than the ultrasonic extract. Conclusions: Collection of R. podophylla root when the temperature is high, without drying, and stirring extraction are considered to be the optimal extraction conditions for increased melanin inhibitory effect.

Antioxidant activity of ethanol extracts from the root and bark of Ulmus davidiana

  • Antioxidant activities of Ulmus davidiana root and bark extracts were evaluated by various antioxidant tests, including DPPH radical-scavenging, nitric oxide-scavenging, superoxide anion radical-scavenging, and ABTS radical-scavenging assays, and superoxide dismutase (SOD)-like activity and reducing power analysis, along with the determination of total phenolic and flavonoid contents. Both the extracts showed strong antioxidant activities by these testing methods. Ulmus davidiana root extract possessed strong reducing power and nitric oxide-scavenging activity, and high scavenging activities against free radicals including the superoxide anion, and the ABTS and DPPH radicals, but a weaker scavenging activity of SOD. In contrast, the Ulmus davidiana bark extract exhibited a strong SOD-like activity, but all the other activities were weak. It was observed that the total phenolic and flavonoid contents of the Ulmus davidiana root extract were higher than those of the Ulmus davidiana bark extract.
