DNA uptake in dry embryos of rice by DNA imbibition was detected by monitoring the expression of chimeric vectors. The selective markers of expression vectors used were ${\beta}-glucuronidase$ ronidase (GUS) and hygromycin phosphotransferase (HPT) genes under the control of CaMV35 S promoter. Frequency of transient expression of the foreign gene was generally 30-50% varying according to the types of vectors and rice cultivars. Dot blot analysis and DNA sequence analysis of inverse polymerase chain reaction products showed that selected rice in hygromycin B (HmB) medium had HPT gene and CaMV35S promoter DNA sequence in genomic DNA of rice. To investigate what ratio of rice having two marker genes simultaneously as rice embryos imbibed the vector DNA having two HPT and GUS gene, transform ants selected in lImB medium were subjected to PCR for GUS gene. It was shown that about 90 percentage of surviving ones in HmB medium had GUS gene.S gene.

Crop status monitoring and yield prediction at higher spatial resolution is a valuable tool in various decision making processes including agricultural policy making by the national and local governments. A prototype crop forecasting system was developed to project the size of rice crop across geographic areas nationwide, based on daily weather pattern. The system consists of crop models and the input data for 1,455 cultivation zone units (the smallest administrative unit of local government in South Korea called "Myun") making up the coterminous South Korea. CERES-rice, a rice crop growth simulation model, was tuned to have genetic characteristics pertinent to domestic cultivars. Daily maximum/minimum temperature, solar radiation, and precipitation surface on 1km by 1km grid spacing were prepared by a spatial interpolation of 63 point observations from the Korea Meteorological Administration network. Spatial mean weather data were derived for each Myun and transformed to the model input format. Soil characteristics and management information at each Myun were available from the Rural Development Administration. The system was applied to the forecasting of national rice production for the recent 3 years (1997 to 1999). The model was run with the past weather data as of September 15 each year, which is about a month earlier than the actual harvest date. Simulated yields of 1,455 Myuns were grouped into 162 counties by acreage-weighted summation to enable the validation, since the official production statistics from the Ministry of Agriculture and Forestry is on the county basis. Forecast yields were less sensitive to the changes in annual climate than the reported yields and there was a relatively weak correlation between the forecast and the reported yields. However, the projected size of rice crop at each county, which was obtained by multiplication of the mean yield with the acreage, was close to the reported production with the $r^2$ values higher than 0.97 in all three years.
The objective of this study was to determine the molecular/crystalline structures and phase transition properties of starches isolated from six rice cultivars grown in Korea. Apparent amylose content was highest in starch obtained from the Saemimyeon cultivar (30.8%) and lowest in that obtained from the Sheonhyangheukmi cultivar (20.3%). Starch from the Saemimyeon cultivar had a lower proportion of short chains (DP 6-12) and a the higher proportion of long chains (DP${\geq}37$) than that seen in other rice starches. Saemimyeon had relatively higher pasting temperature ($86.5^{\circ}C$), gelatinization temperature ($72.1^{\circ}C$) and gelatinization enthalpy (14.2 J/g) than these values found for other rice starches. The onset temperature and enthalpy for ice crystallization of rice starch ranged from $-27.1{\sim}-20.2^{\circ}C$ and 241.1~264.8 J/g, respectively. The ice melting enthalpy measured in excess water (67% water content) of rice starches was 282.4~310.1 J/g. Among the rice starches examined, starch obtained from Sheonhyangheukmi, with the lowest amylose content, showed the lowest glass transition temperature (${T_g}^{\prime}$).
We conducted a QTL analysis of grain quality traits using 117 $BC_3F_4$ and $BC_3F_5$ lines developed from a cross between Ilpumbyeo and Moroberekan. Genotypes of 117 $BC_3F_5$ lines were determined using 134 simple sequence repeat (SSR) markers. A linkage map constructed using 134 SSR markers was employed to characterize quantitative trait loci (QTL). The 117 $BC_3F_4$ and $BC_3F_5$ lines were evaluated for eleven grain quality traits in 2005 and 2006. A total of 18 QTLs were identified for eleven traits, and the phenotypic variance explained by each QTL ranged from 9.9% to 35.2%. Moroberekan alleles contributed positive effects in the Ilpumbyeo background at two QTL loci for 1,000 grain weight. Four QTLs, two for chalky rice and one each for 1,000 grain weight and head rice were consistently detected in two consecutive years indicating that these QTLs are stable. Clusters of QTLs were observed in three chromosome regions. One cluster harboring five QTLs including head rice and brown rice ratio near SSR markers RM190 and RM314 was detected on chromosome 6. Another cluster harboring grain weight and white belly was detected on chromosome 2. Increase in white belly at this locus might be due to the increase in grain weight due to the presence of the Moroberekan allele. The Moroberekan alleles at two QTL loci, gw3 and gw4 associated with increased grain weight might be useful in breeding programs to develop high-yielding cultivars.

Gel consistency data collected from various experiments in same location during 1988 to 1992 were analysed to clarify the degree of varietal and environmental variation in rice. Most of japonica and Tongil-type rice varieties were belonged to soft gel consistency, and average gel length of japonica rice was longer than that of Tongil-type varieties. Gel length of glutinous rice cultivars was extremely long, near 100mm. Year variation of gel consistency was recognized, but did not affect on varietal difference. Gel consistency of most rice varieties and pedigree lines tested varied slightly between different seeding dates or nitrogen levels, but gel length of two indica rice varieties were decreased with the later seeding dates or the higher nitrogen levels. Rice varieties belonged to extremely hard or extremely soft gel consistency showed less variation of gel length between different years, seeding dates or nitrogen levels compared with intermediate ones.

The transplanting date of rice affects grain yield and quality, and it is changed by the environment during cultivation. Thus, it is important to provide the optimal transplanting dates for rice growers under global warming environment. In this study, transplanting date experiments with thirty-day-old seedlings of three cultivars (early, mid, and mid-late maturity) were conducted at the National Institute of Crop Science in Suwon from 2018 to 2021 to determine the optimal transplanting dates for and quantify the effect of planting dates on yield and quality. Transplanting date was strongly associated with culitvar across every year. Clear relationships between transplanting date and head rice yield in early and mid-late maturing rice varieties were observed, and the highest head rice yields were observed during transplantings in mid and late June for early maturing cultivar and mid June for mid-late maturing cultivar. It is obvious that the optimal rice transplanting dates have been shifted and are better later than the optimal transplanting dates in 2002-2004. Days to heading was also strongly associated with the transplanting date and cultivar with 89% of the variation explained. Days to heading was reduced in the later transplanting dates. Grain yield was strongly associate with biomass production during ripening(R2=0.85), however translocated biomass from leaf and stem showed little association with grain yield. The results from this study reconfirmed the importance of shifting optimal transplanting dates to maximize head rice yield for the Mid-plain area in South Korea.
To evaluate cold tolerance of rice cultivars on the basis of a relationship between cold tolerance in the field test and a biochemical character, transition-temperature of respiratory activity of mitochondria(TRM) which were isolated from different parts and at different growth stages and from etiolated rice seedling was investigated, and its relationship with degree of cold tolerance(DCT) in the field experiment was studied. The results obtained were summarized as follows. 1. The transition-temperature of respiratory activity of mitochodria (TRM) isolated from etiolated seedling ($25^{\circ}C$, two week-grown in the dark) of cold tolerant cultivars tended to be low, while that of cold susceptible cultivars to be high. 2. The correlation between TRM of etiolated seedling and the degree of cold tolerance(DCT) was $r=0.8935^{**}$ in 1988 and $r=0.8236^{**}$ in 1989. The correlation coefficient between TRM of 4-leaf seedling and DCT was $0.6239^{**}$ 3. TRM of young panicle was correlated with DCT except a few varieties. 4. TRM of seedling increased with growth as $12.5^{\circ}C$, $14.5^{\circ}C$, $15.5^{\circ}C$ and $16.5^{\circ}C$ at 1, 2, 3~4 and 5weeks respectively. 5. The transition temperature of mitochondrial respiratory activity was not significantly influenced by growth stages as $15.0{\sim}16.0^{\circ}C$ in Pungsanbyeo and $17.0{\sim}18.0^{\circ}C$ in Satbyelbyeo, while it was significantly different depending upon the degree of cold tolerance(tolerant, medium, susceptible).
This study was conducted to obtain the basic information on salt-tolerance of rice cultivars in relations with growth retardation and cation content. As the salt-level increased, less retardation in plant height and dry weight were shown in salt-tolerant than salt-susceptible rice cultivars. Salt tolerant cultivars showed lower N $a^{+}$ and higher $K^{+}$ content and lower N $a^{+}$ / $K^{+}$ ratio at each salt-levels than salt-susceptible ones, while there were no significant differences in $Ca^{++}$ and $Mg^{++}$ content. At the tillering stage, the plant height and dry weight of the salt-treated plots were significantly correlated with N $a^{+}$ and $K^{+}$ content and N $a^{+}$ / $K^{+}$ ratio, implying that N $a^{+}$ and $K^{+}$ content could be an indicator of salt-tolerance of a rice cultivar. There were no tiller-depending differences in cation content in all cultivars. N $a^{+}$ content and N $a^{+}$ / $K^{+}$ ratio in leaves were lower at the top and higher at the bottom. In three $F_1$ hybrids between salt tolerant parent Pokkali and three salt susceptible parents, plant height, dry weight, $K^{+}$ content and N $a^{+}$ / $K^{+}$ ratio were similar to those of Pokkali parent, while N $a^{+}$ content was intermediate of the parents. So, it seemed that salt tolerance is dominant over salt susceptibility. In $F_2$ of Pokkali/wx817 cross, genetic segregation of plant height, dry weight, $K^{+}$ content and N $a^{+}$ / $K^{+}$ ratio varied continuously and was biased onto Pokkali side, while that of N $a^{+}$ content showed normal distribution with intermediate mode. Rice is the world's three major crops that is the staple food of Asian. Japonica type rice is consumed in Korea. Rice cultivation area decreases steadily. That is affected by decrease of the farm income with the inventory of 1.7 million tons, which is 2 to 3 times more than the optimal amount due to the decrease in the consumption of rice per person and cultivate high yield varieties. In recent years, as income level has improved, interest in health has been high and consumption for health food has been steadily increasing. For health food, rice is added by adding grains such as millet, sorghum, oats, beans and colored rice rather than white rice. In 1997, RDA(Rural Development Administration) developed black rice 'Heuknambyeo', and then 20 varieties were bred until 2017. In CBARES(Chungbuk Agricultural Research and Extension Services), we have developed new rice varieties 'Cheongpungheukchal' in 2010, 'Cheongpungheukhyangchal' in 2014, 'Cheongpungheukchal' is high in farming preference because of high yield. Black rice has high content of GABA and water-soluble phenol, DPPH radical scavenging activity, and consumers are interested in the function in the body. Therefore, functional and antioxidant activities(anthocyanin, total polyphenol, ${\alpha}-glucosidase$ inhibition) and antioxidant activities(ABTs, DPPH) were analyzed by comparing white and black rice. Testing varieties are 'Chucheongbyeo', best quality cultivars 'Daebo' and 'Samgwang' as white rice which are cultivated much in Chungbuk area, and black rice are 10 varieties including 'Cheongpungheukchal', 'Cheongpungheukhyangchal', 'Josaengheukchal' and so on. It has transplanted on 25th May, at CBARES research paddy by 100% fertilizer recommendation rate. Harvesting time was 50 days after heading by varieties, and has researched growth properties, yield and yield components, functional and antioxidant activities. Anthocyanin content was not measured because there was no pigmented in white rice, the highest value of anthocyanin content was 'Shintoheukmi' and in the range of 125.6~249.6mg/100g by black rice varieties. Total polyphenol content was high 'Cheongpungheukchal' and 'Shintoheukmi' and in the range of 96.68~244.34 mg/100g in black rice, white rice lower than blackish rice at 19.84~22.51mg/100g. ${\alpha}-glucosidase$ inhibition was high 'Cheongpungheukchal', 'Cheongpungheukhyangchal' and 'Shintoheukmi' in the rage of 75.87~98.85% by black rice varieties, especially 'Samgwang' was 80.75% and the other white rice was higher than 58~68%.
Soybean cultivars for bean sprouting, and for cooking with rice are related closely with Korean dietary tradition, but their yields have been kept in low level. The yielding responses of Danyup, a cultivar for bean sprouting and Gumjung, a cultivar for cooking with rice were compared with Paldal as standard cultivar in planting density within 10 to 80 plants per ㎡. Yield of Paldal increased as planting density increased from 10 plants up to 80 plant/㎡. But, Danyup reached at threshold as planting density reached up to 40 plants/㎡. Gumjung did not have significant difference in yield within planting density. Danyup had the highest yield, 445 kg/l0a at 80 plants/㎡. The highest yield of Paldal was 406kg/10a at 40 plants /㎡ while Gumjung had 373kg/ l0a at 40 plants. The yield responses of cultivars were influenced by canopy structure and light utilization. Paldal had short and straight canopy with determinate growth habit responded well at the heavy density, and had 6.2 in critical LA!. But Gumjung, tall and dense canopy with indeterminate growth habit had limited utilization of light within canopy with 5.4 in critical LAI and high rate of lodging. Gumjung was concluded as a low yielding cultivar, since it did not have good response to increasing source size and sink number.
