Quality Characteristics of Sulgidduk (a Traditional Korean Rice Cake) Admixed with Cheese Powder (치즈가루 첨가량을 달리한 설기떡의 품질특성)

  • We explored the quality characteristics of Sulgidduk prepared using different amounts of cheese powder (0%, 6%, 12%, 18%, 24%; all w/w); physico-chemical properties were determined. Proximate composition analysis showed that the moisture content of Sulgidduk decreased with a rise in the level of added cheese powder. Lightness and yellowness values were lowest in control Sulgidduk and highest in Sulgidduk with 24% added cheese powder. Texture profile analysis showed that all of hardness, cohesiveness, springiness, and brittleness increased with a rise in cheese powder level. Scanning election microscopy indicated that the number of air cells fell as the ratio of cheese powder to rice powder increased. Sensory evaluation tests indicated that color, cheese smell, and greasiness rose as the cheese powder level increased, whereas softness and moistness fell. Consumer acceptance testing showed no significant difference in overall acceptability scores among samples. In conclusion, we suggest that cheese powder may be incorporated into Sulgidduk without affecting sensory qualities.

Improvement of Takju Quality by a Ripening-Fermentation Process Using Honey and Extension of Shelf Life by Control of Takju Mash Sediment (벌꿀을 이용한 고액분리 숙성 탁주의 주질 향상과 고형분의 조절에 따른 품질유지기간 증대)

  • This study was conducted to improve quality of takju where natural honey is used to control the ripening fermentation and the amount of sediment derived from takju mash. A koji was prepared using rice starch and Aspergillus awamori var. kawachii. Takju mash was prepared by alcohol fermentation with Saccharomyces cerevisiae and a 3-step addition of steamed rice. The clean part of the mash (CPM) was separated from the sediment at $5^{\circ}C$ and 5% (w/v) of natural honey was added and then ripened for 23 days at 5, 10, or $15^{\circ}C$. Temperature, pH, acidity, and total sugar content showed no significant differences, but a 0.2 percent reduction in alcohol content occurred during storage. However, CPM ripened with honey had a comparatively higher score on sensory evaluation than did immature CPM with added honey added. Takju with 8% alcohol content was prepared by mixing the water from the mixed CPM ripened with honey together with 100, 50, and 25% of the frozen sediment. Several quality characteristics of the takju were checked over 37 days of fermentation at $10^{\circ}C$. The pH was sustained between 4.1 and 4.3, and changes in the number of viable yeast cells, acidity, total sugar amounts, and alcohol content showed similar patterns but differences in scale. Smaller amounts of sediment affected the stability of the takju. Mixing the CPM ripened with natural honey at low temperature moderately reduced the amount of sediment in the mash and resulted in a highly flavorful takju with an extended shelf life.

Anatomical Difference Between Two Rice Cultivars Selected to Bensulfuron (Bensulfuron에 내성(耐性) 및 감수성(感受性)인 수도품종간(水稻品種間)의 해부학적(解剖學的) 비교(比較) 연구(硏究))

  • Bensulfuron concentrations of $10^{-7}$, $10^{-6}$, $10^{-5}$ and $10^{-4}M$ were applied to agar medium on susceptible (cv. KH 17854 and cv. IR 1846) and tolerant (cv. Chinsurah Boro II and IR 14252) rice cultivars were grown for microscopic inspection. Susceptible cultivars showed the decrease in shoot and root growth at the concentration of $10^{-5}M$ while ones showed no difference. Such a tendency was also observed from microscopic inspection in the elongation zone of shoot meristematic tissue. Seedlings grown in soil for 10 days were transfered to distilled water containing only bensulfuron solutions. There were significant differences between cultivars in terms of supression of shoot meristematic activity and swelling of cell volume. Observations of those cells made it clear that especially susceptible cultivars showed the irregular cell layering, vacuolation, cell swelling and partial damage in membrane of shoot tissue. The major response of root tips of susceptible cultivars showed the disorganization of cortex, rupture and contraction of membrane, inhibition of cell division, swelling and emergence of lateral root while tolerant ones showed no such responses.

  • Effects of butachlor [N-(butoxymethyl)-2-chloro-2', 6'-diethyl acetanilide] and 1,8-naphthalic anhydride (NA) on post-germination growth, mesocotyl and root anatomy and root-cell membrane permeability of rice (Orvza saliva L.) were investigated. Lengths of mesocotyl and radicle were markedly decreased as the application rates of butachlor increased from 0.1 to 100 ppmW and NA from 1 to 100 ppmW, but there was no effect on coleoptile elongation. Application of butachlor-NA resulted in increase in coleoptile elongation, but decrease in mesocotyl elongation. Partial breakdown of cortical cells in root and mesocotyl was caused by either trutachlor or NA treatments, resulting in increase in intercellular air space. Further increase in the intercellular air space of root and mesocotyl was obtained when butachlor was applied in combination with NA. Increase in root-cell membrane permeability occurred when either butachlor or NA was applied. However, butachlor-NA treatments resulted in reduction in the permeability.

Effects of rrhGM-CSF on Morphology and Expression of PCNA in Regenerating Rat Liver (재생 중인 흰쥐 간의 형태학적 변화 및 PCNA 발현에 미치는 rrhGM-CSF의 영향)

  • Liver regeneration is a result of highly coordinated proliferation of hepatocytes and nonparenchymal liver cells. Partial hepatectomy (PH) is the most often used stimulus to study liver regeneration because, compared with other methods that use hepatic toxins, it is not associated with the tissue injury and inflammation, and the initiation of the regenerative stimulus is precisely defined. Granulocyte macrophage-colony stimulating factor (GM-CSF), which is a cytokine able to regulate the proliferation and differentiation of epithelial cells, was first identified as the most potent mitogen for bone marrow. Particularly, rrhGM-CSF, which is highly glycosylated and sustained longer than any other types of GM-CSF in the blood circulation, was specifically produced from rice cell culture. In this experiment, effects of rrhGM-CSF administration were evaluated in the regenerating liver after 78% PH of rats. Morphological changes induced by PH were characterized by destroyed hepatocyte plate around the central vein and enlarged nuclear cytoplasmic ratio and increased hepatocytes with two nuclei. And then, proliferation of liver cells (parenchymal and nonparenchymal) and rearrangement of plates and lobules seemed to be carried out during liver regeneration. These alterations in the experimental group preceded those of the control. Since proliferating cell nuclear antigen (PCNA) is known to be a nuclear protein maximally elevated in the S phase of proliferating cells, the protein was used as a marker of liver regeneration after PH in rats. PCNA levels by western blot analysis and immunohistology were compared between the two groups. PCNA protein expression of two groups at 12 hr and 24 hr after injury showed similar pattern. The protein expression showed the peak at 3 days in both groups, however, the protein level of the experimental group was higher than that of the control. On immunohistochemical observations, the reaction product of PCNA was localized at the nuclei of proliferating cells and the positive reaction in experimental group at 3 days was clearly stronger than that in control group. The results by Western blotting and immunohistology for PCNA showed similar pattern in terms of the protein levels. In conclusion, rrhGM-CSF administration during liver regeneration after 78% PH accelerated breakdown and restoration of the hepatic plate and expression of PCNA. These results suggest that rrhGM-CSF might play an important role during liver regeneration in rats.

Induction of ${\beta}$-carotene by Ozone and Hydrogen Peroxide and Extraction Using Vegetable Oil from Microalga Dunaliella bardawil (미세조류 Dunaliella bardawil에서 오존과 과산화수소에 의한 ${\beta}$-carotene의 축적과 식용기름을 이용한 추출)

  • Halotolerant microalga Dunaliella bardawil was reported to massively accumulate the ${\beta}$-carotene, which protects cells from excess light intensity. Maximum specific growth rate of 0.168/hr was achieved when cells were cultivated at 1 N NaCl, pH 8.0, light intensity 80 ${\mu}E/m^{2}/s$, agitation 70rpm. For the effectiv accumulation of ${\beta}$-carotene, ozone ro hydrogen peroxide was added to media which was irradiated with white fuorescent lamps with moderate light intensity of 250 ${\mu}E/m^{2}/s$. As a result, maximum volumetric content of ${\beta}$-carotene was 324 ${\mu}$g/㎖. The ${\beta}$-carotene extraction efficiency of vegetable oils was in the order of olive oil, sesame oil, rice brain oil, corn oil, and soy bean oil. Sonication and warming was effective in ${\mu}$-carotene extraction and finally 96.9% of ${\beta}$ could be extracted using olive oil.

Development of GIS for the Food Chain Assessment around Kori Nuclear Power Plant Using ArcView (ArcView를 이용한 고리 원전 주변 육상생태계 평가를 위한 GIS 구축)

  • Geographical Information System(GIS) was established to display the calculation results which show the concentration change with time and regions in case of an accidental release of radionuclides from Kori Nuclear Power Plants. GIS included the commercial program, ArcView(ESRI), and a basic digital map of 1:5000 scale lot 20km by 20km around Kori area. The object for the presentation was $^{131}I$ concentration in rice which is one of staple foodstuffs. Provided by deposited $^{131}I$ concentrations, ECOREA-II code computed the $^{131}I$ concentration of the soil and the plant in the area divided by In unit cells in total, in which the concentrations also varied with time. The results were introduced into the attributed data of previously designed polygon cells in ArcView. In order to display the concentration change with time by monotonic color, the RGB value for ArcView color lamp was controlled. This display definitely helped the concentration change around Kori area be acceptable to public.

Fungal Development, Respiration and Activity of Oxidative Enzymes in Rice Plants Inoculated with Pyricularia oryzae in Both Compatible and Incompatible Combinations (벼 도열병균에 감염된 친화 및 비친화 조합 벼에서의 균생장, 호흡 및 산화효소 활성)

  • Appressorial formation of Pyricularia oryzae on leaves showed no marked difference between highly resistant Tongil and susceptible Norm No. 6. Race N-2 of the blast fungus penetrated directly into motor cells of susceptible cultivar Norm No. 6, later extensively spreading hyphae were developed, while in the cultivar Tongil, after penetration, no further hyphal extension resulted. In discoloration of infected tissues, the highly resistant cultivar Tongil not only discolored rapidly, but also the percentage of discolored cells was higher than the susceptible cultivars, Jinheung and Norm No. 6. The respiratory rate, was generally higher in infected tissue than in healthy tissue. No significant difference in the respiration rate of resistant Suwon No. 180 was not found between the infected and healthy leaf tissue, whereas, in susceptible Jinheung, a marked increase in respiratory rate was caused by blast infection. The respiratory rate increased at the appearance of the first visible symptom in all cultivars resistant or susceptible. Higher peroxidase activity was found in infected tissues as compared with healthy tissue. Peroxidase activity increased in resistant and susceptible reactions. Particularly, in resistant reaction, the increase of the activity was more pronounced. In highly resistant reaction, there was no difference in peroxidase activity between healthy and infected tissues. Ascorbic acid oxidase, hydroquinone oxidase and catechol oxidase had the same trend as the peroxidase. In contrast, activity of catalase rather decreased in leaf tissues infected with compatible races of the fungus.

Anti-proliferative and Pro-apoptotic Effects by Lees Extracts of Ehwa Makgeolli Containing Oriental Herbs (한방이화주 주박 추출물에 의한 암세포 항성장 및 세포사멸 기전 연구)

  • In the present study, ethanol extracts and their subsequent organic solvent fractions were extracted from the lees of Ehwa Makgeolli containing oriental herbs, a commercialized traditional Korean rice wine, and the prepared lees samples were designated as from KSD-E3-1 to KSD-E3-5. First, their effects on cell viability and on the expression of pro-apoptotic ATF3 and NAG-1 genes in human colorectal HCT116 cells were investigated. Among the treated lees samples, the hexane fraction (KSD-E3-2) and the ethyl acetate fraction (KSD-E3-3) of lees extracts from Ehwa Makgeolli significantly reduced cell viabilities, in a dose dependent manner. The treatment with KSD-E3-2 and KSD-E3-3 also increased the expression of pro-apoptotic NAG-1 and ATF-3 genes and their proteins, which were detected with RT-PCR and Western blot analysis, respectively. In addition, poly-(ADP-ribose) polymerase (PARP) cleavage was detected by treatment with the fraction KSD-E3-3, indicating that KSD-E3-3 could induce apoptosis in HCT116 cells. Interestingly, this PARP cleavage was recovered by transfection of NAG-1 small interfering RNA. The results indicate that NAG-1 is one of the genes responsible for apoptosis induced by the fraction KSD-E3-3 from Ehwa Makgeolli. Overall, the findings may help in understanding the molecular mechanisms of the anti-proliferative and pro-apoptotic activities mediated by the lees of Ehwa Makgeolli.

Dietary supplementation with L-glutamine enhances immunity and reduces heat stress in Hanwoo steers under heat stress conditions

  • This study investigated the effects of L-glutamine (Gln) supplementation on growth performance, physiological traits, heat shock proteins (HSPs), and gene expression related to muscle and adipose tissue development in Hanwoo steers under heat stress (HS) conditions. Eight Hanwoo steers (initial body weight [BW] 570.7 ± 43.6 kg, months of age 22.3 ± 0.88) were randomly separated into two groups, control and treatment, and supplied with the concentration (1.5% of BW kg/day/head) and rice straw (1.5 kg/day/head). The treatment group were fed the Gln supplementation (0.5% of concentration, as-fed basis) once a day at 08:00 h. Blood samples for the assessment of haematological and biochemical parameters and the separation of peripheral blood mononuclear cells (PBMCs) were collected four times, at 0, 3, 6, and 10 weeks of the experiment. Feed intake was measured daily. BW to analyze growth performance and hair follicle collection to analyze the expression of HSPs were executed four times at 0, 3, 6, and 10 weeks. To analyze gene expression, longissimus dorsi muscle samples were collected by biopsy at the end of the study. As a result, growing performance, including final BW, average daily gain, and gain-to-feed ratio, were not different between the two groups. Leukocytes including lymphocytes and granulocytes, tended to increase in the Gln supplementation group (p = 0.058). There were also no differences in biochemical parameters shown between the two groups, except total protein and albumin, both of which were lower in the Gln supplementation group (p < 0.05). Gene expressions related to muscle and adipose tissue development were not different between the two groups. As temperature-humidity index (THI) increased, HSP70 and HSP90 expression in the hair follicle showed a high correlation. HSP90 in the hair follicle was decreased in the treatment group compared with the control group at 10 weeks (p < 0.05). Collectively, dietary Gln supplementation (0.5% of concentration, as-fed basis) may not be influential enough to affect growth performance and gene expression related to muscle and adipose tissue development in steers. However, Gln supplementation increased the number of immune cells and decreased HSP90 in the hair follicle implying HS reduction in the corresponding group.
