Recombinant human granulocyte macrophage colony-stimulating factor (rhGM-CSF) could accelerate burn wound healing in hamster skin

  • Burns are one of the most devastating forms of trauma and wound healing is a complex and multicellular process, which is executed and regulated by signaling networks involving numerous growth factors, cytokines, and chemokines. Recombinant human granulocyte macrophage colony-stimulating factor (rhGM-CSF) was specifically produced from rice cell culture through use of a recombinant technique in our laboratory. The effect of rhGM-CSF on promotion of deep second-degree burn wound healing on the back skin of a hamster model was evaluated through a randomized and double-blind trial. As macroscopic results, hamster skins of the experimental groups showed earlier recovery by new epidermis than the control groups. Immunohistochemical reactions of proliferating cell nuclear antigen and transforming growth factor-b1, which are indicators of cell proliferation, were more active in the experimental group, compared with the control group. On electron microscopy, basal cells in the epidermis of the experimental group showed oval nuclei, prominent nucleoli, numerous mitochondria and abundant free ribosomes. In addition, fibroblasts contained well-developed rough endoplasmic reticulum with dilated cisternae. Bundles of collagen fibrils filled the extracellular spaces. Particularly, ultrastructural features indicating active metabolism for regeneration of injured skin at 15 days after burn injury, including abundant euchromatin, plentiful free ribosomes, and numerous mitochondria, were observed. These findings suggest that use of rhGM-CSF could result in accelerated deep second-degree burn wound healing in animal models.

예측미생물학을 활용한 미강 식이섬유 함유 떡갈비의 유통기한 설정 (Application of Predictive Microbiology for Shelf-life Estimation of Tteokgalbi Containing Dietary Fiber from Rice Bran)

  • 본 연구는 미강 추출 식이섬유 혼합물을 첨가한 떡갈비의 미생물학적 안전성 확보와 유통기한 산출을 위하여 예측미생물학을 이용하였다. 이를 위해 미강 추출 식이섬유 혼합물을 0, 1, 2, 3%의 비율로 첨가하였으며 냉장($4{\pm}1^{\circ}C$) 조건에서 15일간 저장하며 일반세균수, 혐기성균, 저온균, 내열성균, 대장균군의 미생물학적 변화를 관찰하였다. 일반세균수의 초기 균수(저장 0일)의 경우 3.23-3.85 log CFU/g을 나타내었으며 혐기성균, 저온균의 경우 비슷한 초기균수를 나타내었다. 일반세균수와 혐기성균의 경우 저장 3-5일차에서 균수의 급격한 증가를 나타내었다. 저장기간 동안 측정된 균수를 Baranyi function을 바탕으로 성장예측곡선과 생육 지표를 예측하였으며, 성장예측곡선의 적합성을 검증한 결과 일반세균수, 혐기성균, 저온균의 경우 처리구간 모두 0.923 이상의 높은 $R^2$값을 나타내었으며 $B_f$, $A_f$의 경우 역시 이상적인 값인 1에 가까운 값은 나타내었다. RSME 값 역시 모두 0.65 이하를 나타내어 실측치와 예측치 간의 높은 정확성을 나타내었다. Baranyi function 식 (1), (2)을 이용하여 계산된 균수가 5 log CFU/g이 되는 시점인 predicted shelf-life의 경우 control, T1, T2, T3의 경우 각각 2.5, 3.5, 3.5, 3.6일로 예측되었으며 안전계수(1/1.5)를 고려한 estimated shelf-life의 경우 1.7, 2.3, 2.3, 2.4일로 측정되었다 미강 식이섬유 혼합물이 첨가된 처리구가 control보다 약 0.6-0.7일 긴 유통기한을 가지는 것으로 측정되었고 식이섬유가 3% 함유된 떡갈비가 2.4일로 가장 긴 유통기한을 갖는 것으로 나타났다. 본 연구를 통해, 미강 추출 식이섬유 혼합물이 첨가된 떡갈비의 유통기한을 예측미생물학을 적용하여 측정할 수 있었으며, 식품산업에 있어서 이러한 예측미생물학은 식품의 제조 가공 판매 등을 결정할 때 미생물의 정량적 위험성을 판단하는 도구로서 사용될 수 있을 것으로 기대된다.

고속도로 휴게소에서 판매되는 식품의 세균학적 품질 (Bacteriological Quality of Foods on Sale at Resting Places of the Highways in Korea)

  • 1995년(4월과 5월), 1997년(5월과 7월) 및 1999년(1월)에 경부 .호남 고속도로 및 중부 . 영동 고속도로의 20개 ,휴게소의 김밥, 햄버거, 호도과자 및 자장면에 대한 계절별 위생 실태와 조리 형태에 의한 미생물적 안전성 시험을 실시한 결과 식중독균은 검출되지 않았으나 대장균과 많은 수의 대장균군 세균이 검출되어, 고속도로 휴게소에서 판매되고 있는 식품의 위생 실태가 매우 불량함을 알수 있었다. '95년 김밥 시료에서는 전 제품에서 대장균군(8.8$\times$10 4~6.6$\times$10 5 cells/g)과 대장균이 검출되었고,'95년 햄버거 또한 전 제품에서 대장균군(1.8$\times$10 2~4.7$\times$10 4 cells/g)이 검출되었다. '97년 시료에서는 김밥은 22곳 제품 중 16곳 제품에서 대장균이 양성이었고, 대장균군은 21곳 제품에서 검출되었다. '97년 햄버거는 14곳 제품 중 7곳 제품에서 1.7$\times$10 2~1.9$\times$10 7 cells/g의 대장균군이, 2곳 제품에서 대장균이 양성으로 나타나서 김밥과 햄버거의 위생 상태는 매우 불량하였다. 호도과자는 조사 대상 6곳 제품 중 대장균군과 대장균이 모두 검출되지 않아 위생 상태가 양호하였다. 반면 자장면은 6곳 제품 중 3곳 제품에서 7.1$\times$10 2~2.0$\times$10 3 cells/g수준의 대장균군이 검출되고 대장균은 검출되지 않았다. '95년과 '97년 김밥 시험 결과와 '99년 결과를 비교할 때 기온이 높은 계절의 위생 관리에 더욱 주의해야 함을 알 수 있었다. 김밥과 기타 식품의 세균학적 품질을 비교한 결과, 김밥 >햄버거 >자장면 >호도과자의 순으로 나타나 김밥과 햄버거와 같은 복합재료를 사용하는 식품의 경우에 더 많은 오염이 일어날 수 있으므로 조리 .가공 및 유통 과정의 엄격한 미생물절-안전성 관리가 요구된다.

Insertional Transposon Mutagenesis of Xanthomonas oryzae pv. oryzae KXO85 by Electroporation

  • The bacterial leaf blight, which is caused by Xantho-monas oryzae pv. oryzae, is the most damaging and intractable disease of rice. To identify the genes involved in the virulence mechanism of transposon TnS complex, which possesses a linearized transposon and transposase, was successfully introduced into X. oryzae pv. oryzae by electroporation. The transposon mutants were selected and confirm the presence of transposition in X. oryzae pv. oryzae by the PCR amplification of transposon fragments and the Southern hybridization using these mutants. Furthermore, transposon insertion sites in the mutant bacterial chromosome were deter-mined by direct genomic DNA sequencing using transposon-specific primers with ABI 3100 Genetic Analyzer. Efficiency of transposition was influenced mostly by the competence status of X. oryzae pv. oryzae cells and the conditions of electroporation. These results indicated that the insertion mutagenesis strategy could be applied to define function of uncharacterized genes in X. oryzae pv. oryzae.

천년초 열매 분말을 첨가한 설기떡의 품질 특성 (Quality Characteristics of Sulgidduk Added with Cheonnyuncho Fruit Powder)

  • This study is performed to investigate the quality characteristics of sulgidduk added with cheonnyuncho fruit powder. Sulgidduk was prepared with freeze-dried cheonnyuncho, rice flour, sugar syrup and salt. The sulgidduk was made with various amounts (0, 3, 6, 9, 12%) of added cheonnyuncho fruit powder. The proximate composition, pH, Hunter's color value, texture profile analysis, sensory characteristics and SEM of cheonnyuncho sulgidduk were being examined. The addition of cheonnyuncho fruit powder has a tendency to decrease the moisture contents of sulgidduk while the crude ash, crude lipid and crude protein increased. The pH of sulgidduk decreased when the amounts of cheonnyuncho fruit powder increased. As the amount of cheonnyuncho fruit powder increased, the lightness (L) decreased, while the redness (a) and yellowness (b) increased. In texture profile analysis, the hardness decreased with increasing cheonnyuncho fruit powder contents, whereas cohesiveness, springiness, gumminess and brittleness increased. The results of sensory evaluation showed that the sulgidduk with 3%, 6% of cheonnyuncho fruit powder got the highest scores on the color, taste, springiness and acceptability. The air cells of sulgidduk observed by SEM were big and uniform with the amount of cheonnyuncho fruit powder increased. In conclusion, these results show that the quality and preference increased when 3%, 6% of cheonnyuncho fruit powder was added to the sulgidduk.

Acetoxycycloheximide의 생리활성 및 과 생산균주 (Biological Activity of Acetoxycycloheximide and Its Producing Microoganism)

  • 77-AG-567 균주는 Pyricularia oryzae와 Sphasrotheca fuliginea에 대한 항균활성을 나타냈다. 활성 물질을 정제해 가는 과정에서 이 균주는 blasticidin S와 또 다른 항생물질을 동시에 생산한다는 사실을 알았다. 이 화합물은 UV 및 $^1$H NMR 스펙트럼에 의해 E-73이라고도 일컬어지는 acetoxycycloheximide로 밝혀졌다. 균주를 동정해본 결과 77-AG-567균주는 Streptomyces atbutus로 판단되며 지금까지 blasticidin S와 acetoxycycloheximide를 동시에 생산하는 것으로 알려져 온 S. albulus와는 다른 특성을 나타냈다. 특히 주목할만한 차이점은 포자의 표면이였다. 또한 acetoxycycloheximide는 지금까지 효모와 종양세포에만 생리활성을 나타내는 것으로 알려져 왔으나 본 실험결과 Pyricularia oryzae와 Sphasrotheca fuliginea에도 항균활성을 가진다는 사실을 알았다.

저온 캡슐화에 의한 Weissella cibaria CMU의 생존율 증진 효과 (Enhancement of Viability of Weissella cibaria CMU by Low-Temperature Encapsulation)

  • In this study, we aimed to investigate the effects of low-temperature encapsulation on the viability of Weissella cibaria CMU under harsh conditions and in freeze-dried foods during storage. The capsules were prepared by gelation of sodium caseinate at different concentrations (5%, 6%, 7%, and 8%) with a combination of 0.5% gellan gum and 2% $glucono-{\delta}-lactone$. The size distribution of the capsules was determined using a Mastersizer 3000 laser diffraction particle size analyzer. Scanning and transmission electron microscopy revealed that the capsule with 6% sodium caseinate had a smooth and rounded external surface, with reproducibility. The acid, bile, and heat tolerances of the encapsulated cells were significantly higher than those of the control under prolonged acid (5 h), bile (12 and 24 h), and heat (2 h) exposure, respectively. During storage for up to 6 months at $4^{\circ}C$ or $25^{\circ}C$, the viability of encapsulated Weissella cibaria CMU in beef and vegetable rice porridge was effectively improved.

VARIOUS NIR SAMPLE PRESENTATIONS FOR AGRICULTURAL PRODUCTS SUCH AS INTACT FRUITS, SINGLE GRAINS, VEGETABLE JUICE, MILK AND THE OTHERS

  • Sample presentation, which means how to set samples to an NIR instrument, is very important in Near Infrared (NIR) Spectroscopy. When sample presentation is not suitable for the samples that you use, very good spectra can not be obtained even if you use a sophisticated NIR instrument. In my presentation, various NIR sample presentations for agricultural products such as intact fruits, single grains, vegetable juice and the others will be explained. In case of peaches with thin peel, the fiber optics of Interactance can be used. However, the fiber optics are not suitable for oranges with relatively thick peel. In this case, transmittance method is useful. As for a small sample such as single grains, a specially designed cell is needed. The cell in transmittance mode has been developed and then applied to single kernels of rice and soybean. In this case we also used the fiber optics. As regards liquid type of sample, a cuvette cell made of quartz in transmittance mode is popular. However, it is time-consuming to wash and dry it. In order to compensate this disadvantage the sample presentation using normal test tubes as sample cells have been developed and applied to milk, rumen juice and urine of a milking cow. An individual test tube can be used for each sample if you use the calibration equation with sample cell compensation. The test tube cell has also been applied to spinach juice for determination of undesirable constituents. It is concluded that sample presentation is most important for NIR Spectroscopy.

Involvement of Extracellular Matrix and Integrin-like Proteins on Conidial Adhesion and Appressorium Differentiation in Magnaporthe oryzae

  • Conidial adhesion and appressorium formation of Magnaporthe oryzae on the rice surface are important early events in the infection process. As an initiative step to understand the mechanisms underlying these cellular processes at a biochemical level, the effect of a human fibronectin antibody (HFA) and RGD peptides on conidial adhesion and appressorium formation was evaluated. HFA inhibited conidial adhesion and appressorium formation in a dosage-dependent manner. RGD peptides also inhibited these cellular events. Conidial adhesion and appressorium formation inhibited by RGD peptides were restored by chemicals involved in the cyclic AMP-dependent signaling pathway. These results suggest that extracellular matrix proteins might be involved in conidial adhesion and appressorium formation through integrin-like receptor mediation and modulation of cAMP-dependent signaling in the cells.

Effects of Wood Vinegar Mixted with Insecticides on the Mortalities of Nilaparvata lugens and Laodelphax striatellus(Homoptera: Delphacidae)

  • Effects of wood vinegar on the activity of various insecticides were determined by measuring the mortality of two species of rice planthoppers, Nilaparvata lugens and Laodelphax striatellus. Wood vinegar itself did not show insecticidal activity on planthoppers. When the planthoppers were treated with wood vinegar mixed with one of insecticides such as BPMC, dinotefuran, imidacloprid, carbosulfan or insect growth regulators, the planthopper mortality induced by carbosulfan was greatly increased by the wood vinegar in comparison with a single carbosulfan treatment. Wood vinegar showed no effect on other insecticides. In addition, the wood vinegar-carbosulfan mixture significantly reduced AChE activity of planthoppers, which is a target molecule of carbosulfan. This result suggests that wood vinegar has a synergistic effect on the insecticidal activity of carbosulfan. Our study provides information on a potential role of wood vinegar in facilitation of activity of specific insecticides.
