Scientific Analysis of Formulation Theory of Chungpesagan-tang; The purgative Action of Chungpesagan-tang

  • To analyze scientifically the fundamental formulation theory and drug interaction on the purgative action of Chungpesagan-tang, which is one of the traditional Korean medical prescriptions that has been most frequently used for stroke, water extracts of herbal medicines with Rhei Rhizoma were administered into mice and the purgative action was measured. Water extracts of Raphani Semen or Cimicifugae Rhizoma with Rhei Rhizoma had more potent purgative activity than Rhei Rhizoma alone, although Raphani Semen alone displayed no purgative action. However, Platycodi Radix and Puerariae Radix inhibited the purgative action of Rhei Rhizoma. When Rhei Rhizoma with Raphani Semen or Cimicifugae Radix was extracted with water, sennoside A content in the water extract was increased. However, Puerariae Radix and Platycodi Radix decreased the content of sennoside A in the extracted water. The transforming rate of sennoside A of Rhei Rhizoma was inhibited by Puerariae Radix and Platycodi Radix. The transforming rate of sennoside A of Chungpesagan-tang to rheinan-throne by human intestinal microflora and its purgative activity were similar to those of Rhei Rhizoma. Finally we thought that the purgative action of Chungpesagan-tang could be controlled by the addition and subtraction of Chungpesagan-tang-composing herbal medicines.

  • In vivo antioxidant activities were screened over 30 kinds of crude drugs which are most frequently prescribed in oriental medicine. Of these, only Ginseng Radix, Cimicifugae Rhizoma, Zingiberis Rhizoma(steam dried), Alismatis Rhizoma, and Liriopes Tuber were shown to be positive in the activity.

  • Quantitative methods for the marker compounds of Cimicifugae Rhizoma, Achyranthis Radix, Artemisiae Capillaris Herba, Moutan Cortex Radicis and Arecae Semen, respectively, were developed using HPLC-DAD. Using the established methods, each extract of the natural medicines were evaluated. In addition, long term and accelerated stability test in the extracts were examined for six months. No significant change in content of the marker compounds of each extract observed during the time of investigation.

In Silico Molecular Docking Comparison of Tubocurarine and the Active Ingredients of Cimicifugae rhizoma on Acetylcholine Binding Proteins (In Silico 분자결합 분석방법을 활용한 tubocurarine과 승마 추출성분 actein의 아세틸콜린 결합 단백질 활성 부위에 대한 결합 친화도 비교 분석)

  • Actein is the well-known active ingredient of Cimicifugae rhizoma (Black cohosh). In this study, we investigated and compared the binding affinity of tubocurarine, actein, and actein derivatives on the B&C domain of the acetylcholine binding protein through in silico computational docking studies. The three-dimensional crystallographic structure of the acetylcholine binding protein B&C domain was obtained from the PDB database (PDB ID: 2XYT). An in silico computational autodocking analysis was performed using PyRx, Autodock Vina, Discovery Studio Version 4.5, and NX-QuickPharm based on scoring functions. The actein showed an optimum binding affinity (docking energy), with the acetylcholine binding protein at -10.50 kcal/mol as compared to the tubocurarine (-9.80 kcal/mol). The interacting amino acids tryptophan 84 and tryptophan 147, in the B domain of the acetylcholine binding protein active site, significantly interacted with the actein and 27-deoxyactein, and (26R)-actein. The centroid XYZ grid position of the tubocurarine was X=38.300689, Y=112.053467, and Z=51.991022, but the actein and its derivatives showed values around X=26.4, Y=127.3, Z=43.7. These results clearly indicated that actein and its derivatives could be a more potent antagonist to the acetylcholine binding protein than tubocurarine. Therefore, the extract of Cimicifugae rhizoma or actein containing biomaterials can substitute for the botulinum toxin-mediated acetylcholine receptor regulation, and be applied to the anti-wrinkle cosmetics industry.

Scientific Analysis of the Formulation Theory of Chungpesagan-tang; In Vitro Cytotoxicity of Chungpesagan-tang

  • To analyse scientifically the fundamental formulation theory and drug interaction of Chungpegagan-tang, the extraction level of puerarin and daidzin, the transforming activity of puerarin and daidzin to daidzein by human intestinal bacteria and in vitro cytotoxicity against tumor cell lines of Chungpesagan-tang were investigated. When Puerariae Radix was extracted with Chungpesagan-tang composing herbal medicines, the puerarin extraction level from these polyprescriptions was decreased by the extraction with Raphani Semen or Cimicifugae Rhizoma, but the other herbal medicines increased it. The activity transforming puerarin and daidzin to daidzein by human intestinal bacteria was increased by Raphani Semen, Cimicifugae Rhizoma and Angelicae Tenuissimae Radix, but decreased by Scutellariae Radix and Rhei Rhizoma. Puerariae Radix did not showed in vitro cytotoxicity against tumor cell lines. However, by its anaerobic incubation with human intestinal bacteria, it showed a potent cytotoxicity. When the main components, puerarin and daidzin, of Puerariae Radix were incubated with human intestinal bacteria, the main metabolites were daidzein and calycosin. These metabolites had the most potent cytotoxicity, compared to those of puerarin and daidzin. Raphani Semen, Rhei Rhizoma and Chungpesagan-tang had also the potent cytotoxicity against tumor cell lines by the anaerobic incubation with human intestinal bacteria.

  • 'Bojungikgi-tang' consists of Astragali Radix, Ginseng Radix, Atractylodis Rhizoma alba, Glycyrrhizae Radix, Angelicae gigantis Radix, Aurantii nobilis Pericarpium, Cimicifugae Rhizoma and Bupleuri Radix. It has been widely prescribed in oriental medicine for controlling digestive functions and for health. Experimental studies were implemented on analgesic, sedative and antipyretic actions. The results showed significant analgesic and antipyretic actions were recognized. Sedative actions were significantly noted.

Genotoxicological Safety of Hot Water Extracts of the γ-Irradiated Astragali Radix, Atractylodes Rhizoma, and Cimicifugae Rhizoma in Vitro (감마선 조사 황기, 백출 및 승마 열수 추출물의 in vitro 유전독성학적 안전성 평가)

  • As the utilization of medicinal herbs in food and bio-industry increases, safe hygienic technologies for them are demanded. To consider the possibility of application of radiation technology for this purpose, the genotoxi-cological safety of three r -irradiated medicinal herbs were studied. Astragali Radix, Atractylodes Rhizoma and Cimicifugae Rhizoma were irradiated at 10 kGy, and then were extracted with hot water. The genotoxicity of the extracts was examined in two short-term in vitro tests: (1) Salmonella reversion assay (Ames test) in strains of TA98 and TA100; (2) Micronucleus test in cultured Chinese hamster ovary (CHO) cells. The extract was treated at maximum doses of 5 mg/plate in Salmonella reversion assay, and 1 mg/mL in micronucleus test where growth of CHO cells was inhibited by 50%. In Salmonella reversion assay with or without metabolic activation, both ex-tracts of irradiated and non-irradiated herbs showed no significant differences in formation of revertant colonies compared with the negative control. And also in micronucleus test, the incidences of micronucleus in CHO cells cultured with extracts of irradiated herbs were almost same as negative control in less than 3%. These results of two in vitro tests suggest that ${\gamma}$-irradiated herbs do not show mutagenicity and cytogenetic toxicity. Further tests of in vivo genotoxicity and chronic toxicity are needed to ascertain the safety of ${\gamma}$-irradiated herbs.

  • In this dissertation, I will focus on the channel entry, the effect and the treatment throughout books of oriental medicine from ancient to modern in order to classify the medicines of the Spleen as main or supplementary organ. The results are as follows: 1. Proofreading of one Ryang based on the Ratio of Maximum and Minimum Dose in the Decoction of ≪Treatise on Cold Damage Diseases≫ (≪상한론≫ 탕제에서 한약 하루 먹는 량 최대와 최소 비율에 근거한 복용량 1 량(兩) 교정)

  • Objectives : The purpose of this study was to proofread 'one ryang' in the Decoction of ${\ll}$Treatise on Cold Damage Diseases${\gg}$. Methods : I found out the ratio of maximum dose and minimum dose in this book. On the basis of the ratio, I corrected 'one ryang' in diverse decoctions. Results : In any decoction, maximum dose of medicinal medica in one decoction could not exceed four times minimum dose. Specifically, in the case that maximum dose in one decoction is sixteen ryang, it could not exceed eight times minimum dose in the same decoction. Any medicinal medica used in two decoctions or more, its maximum dose could not exceed four times minimum dose in other decoctions. On the basis of these results, it should be changed into three ryangs that are one ryang dose of 'Haematitum' of Seonbokdaeja Tang, 'Ginger' of Bujageongang Tang, Baektong Tang, Baektonggajeodamjep Tang and Senggangsasim Tang. Furthermore it should be changed into two ryangs that are one ryang dose of 'Coptidis Rhizoma' of Sohamhyung Tang, 'Ginger' of Dowha Tang, 'Ginseng Radix' of Whubaksenggangbanhagamchoinsam Tang, 'Polyporus, Poria Sclerotium, Alismatis Rhizoma, Talcum and Asini Corii Colla' of Jeoryeong Tang, 'Cimicifugae Rhizoma, Atractylodis Rhizoma Alba and Anemarrhenae Rhizoma' of Mahuangshengma Tang and 'Cassiae Cortex Interior' of Gyejigamchoryonggolmoryeo Tang. Conclusions : These results suggest that one ryang of thirteen medicinal medica such as Haematitum or Ginger of eleven decoctions such as Seonbokdaeja Tang or Bujageongang Tang should be changed into two or three ryangs.
