Isolation of Novel Non-Toxic Bacillus thuringiensis from Soil Samples in Korea (한국 토양으로부터 새로운 무독성 Bacillus thuringiensis 균주의 분리)

  • Four Bacillus thuringiensis isolates obtained from soil samples in Korea produce parasporal inclusions non-toxic to 10 insect species of three orders, Lepidopera, Diptera and Coleoptera. These four isolates are named NTB-1, NTB-2, NTB-3 and NTB-4, respectively. The morphology of parasporal inclusions of four isolates observed by phase contrast- and scanning electron microscope was all ovoid. Characterization of four non-toxic B. thuringiensis isolates was determined by sodium dodecyl sulfate-polyacrylamide gel electrophoresis and restriction endonuclease analysis. The results showed that parasporal inclusion proteins and total plasmid DNA profiles of four isolates are different from other known non-toxic B. thuringiensis strains', suggesting that four isolates are novel.

  • In this paper, the topology optimization of inner-wall stiffener of cylindrical containers for the use as a rocket fuel tank is presented. Such structures for space mission should have high stiffness against the buck]ins while their weight should be maintained low from the viewpoint of cost and performance. Therefore, in the present work the reciprocal of critical buckling load is adopted as an objective function and the total mass of stiffener is constrained to a prescribed value. Due to the restriction of computational resources a section of cylindrical container is topologically optimized and this result is repeated to obtain the full design. Also, for manufacturability the concept of periodic topology pattern in design domain is newly introduced. In the numerical examples, the results by the proposed approach are investigated and compared with those of isogrid design.

  • The deletion of the distal long arm of chromosome 1 is associated with a characteristic facial appearance and a pattern of associated malformations. Characteristic manifestations include a round face with prominent 'cupid's bow' and downturned corners of the mouth, thin vermilion borders of lips, a long upper lip with a smooth philtrum, a short and broad nose, epicanthal folds, apparently low-set ears, micrognathia, microcephaly, abnormal hands and feet, variable cardiac or genital anomalies, moderate to severe mental retardation, and growth retardation. Using fluorescent in situ hybridization (FISH) analysis to map precisely the deletion, we present a case of chromosome 1q44 deletion with craniofacial characteristics, multiple congenital anomalies, and growth and psychomotor retardation. In comparison with other reported cases of 1q43-44 deletion, the subject does not show hydrocephalus, seizure, syn- or polydactyly of hands, and a urogenital anomaly. However, an arachnoid cyst, pinpoint dimple on the midline of the forehead, a right-sided supernumerary nipple and auricular pit, polydactyly of the right foot, adducted thumb, and flexion restriction of the proximal interphalangeal joint with a simian line in both hands were observed additionally.

  • To gain a better understanding of the methylation imprinting changes associated with heat stress in early development, we used bisulfite sequencing and bisulfite restriction analysis to examine the DNA methylation status of imprinted genes in early embryos (blastocysts). The paternal imprinted genes, H19 and Igf-2r, had lower methylation levels in heat-stressed embryos than in control embryos, whereas the maternal imprinted genes, Peg3 and Peg1, had similar methylation pattern in heat-stressed embryos and in control embryos. Our results indicate that heat stress may induce aberrant methylation imprinting, which results in developmental failure of mouse embryos, and that the effects of heat shock on methylation imprinting may be gene-specific.

  • Along with the rapid emergence of the Internet and e-commerce, online auctions are hitting the spotlights. The inconveniences found in off-line auctions, such as time and place restriction and limited number of items, are solved in the online auction. However, not so many auction sites have integrated auction information systems, which monitor the present status of auctions, resulting in greater inconvenience for the online auction users. Moreover, there is no auction site that suggests the appropriate starting or closing price that is useful for users when they make, their bids, What the online auction users need is an auction system that can solve such problems. This study is purported for solving the problems by designing and implementing a real time auction system that applies the comparison search functions and the agent functions. In other words, an integrated database system using a bidder-oriented agent for providing information is built so that the users can search and compare the information on the item they are interested in and make a faster and more accurate purchase. Also the appropriate starting and closing prices are offered to the sellers and bidders through the integrated system for a closer and more accurate comparison and analysis of the prices. For future work, the product recommendation service, which accurately reflects the bidding patterns, and the methods for studying the multi bidding pattern will be applied to the suggested system to realize a real time auction information system that supports CRM(Customer Relationship Management) .

Laser Head Design and Heat Transfer Analysis for 3D Patterning (3차원 패터닝을 위한 레이저 헤드설계 및 열해석)

  • A laser head was designed for micro-scale patterning and joining applications. The target feature size of the pattern was $100{\mu}m$, and optics were designed to perform the target. Two singlet lenses were combined to minimize the chromatic aberration, and the geometry of the lenses was calculated by using the raytracing method with a commercial software program. As a restriction of lens design, the focal length was set at 100mm, and the maximum diameter of the lens or beam size was limited to 10mm for the assembly in the limited cage size. The maximum temperatures were calculated to be $1367^{\circ}C$, $1508^{\circ}C$, and $1905^{\circ}C$ for 10, 12, and 15 Watts of power, respectively. A specially designed laser head was used to compensate for the distance between the object and the lens. The detailed design mechanism and 3D data were presented. The optics design and detailed performance of the lens were analyzed by using MTF and spot diagram calculation.

Discussion on the Late Miocene Biogenic Opal Crash in the Andaman Sea (마이오세 후기 안다만해에서 생물기원 오팔 함량의 폭감에 대한 논의)

  • Biogenic opal crash at about 6.7 Ma was identified at both IODP Site U1447 and NGHP Site 17 in the Andaman Sea. The different biogenic opal content and general variation pattern between two sites may be attributed to the different concentration of analytical reagent and sedimentation rate estimated by the different chronological approaches. Nevertheless, this study suggests that the biogenic opal crash in the Andaman Sea is closely related to the restriction of Indonesian Throughflow and to the decreasing strength of Indian summer monsoon during the late Miocene, both of which resulted in the reduction of nutrient supply.

  • BamHI restriction pattern and genomic library of Herpes simplex virus type 2 (HSV-2) strain G were constructed, and locations of the glycoproteins gB and gD, and tk genes on the fragments were detected by Southern blot analysis. HSV-2 genomic DNAs were cleaved into twenty-seven fragments by BamHI enzyme in the range of 0.72 to 15.08 (total 150.44 kb), which were cloned into the BamHI site of pBluescript SK(+) to construct genome library of the HSV-2. The library was named by the order of the fragment size from smallest one to largest one. HSV-2 glycoprotein gD gene was located in pHLA2-21 and pHLA2-22 recombinant plasmids, gB gene in pHLA2-24 plasmid, and tk gene in pHLA2-11 clone by Southern blot analysis.

  • The ascomycetous fungus Taphrina wiesneri, the pathogen of cherry witches' broom, is highly pathogenic to Prunus yedoensis, the most widely planted cherry trees in Korea as park and roadside trees. A collection of 13 strains of the pathogen in Korea and Japan was characterized by 18S rDNA gene sequence and restriction fragment length polymorphism (RFLP) analysis. In cluster analysis based on 18S rDNA gene sequence the strains were divided into 2 clusters. In RFLP analysis of the rDNA-IGS region using HhaI, the strains were separated into four patterns, B, C, D and G, of which pattern G was new.

  • The plant pathogenic bacterial genus Pectobacteirum consists of heterogeneous strains. The P. carotovorum species is a complex strain showing divergent characteristics, and a new subspecies named P. carotovorum subsp. brasiliensis has been identified recently. In this paper, we re-identified the P. carotovorum subsp. brasiliensis isolates from those classified under the subspecies carotovorum and newly isolated P. carotovorum subsp. brasiliensis strains. All isolates were able to produce plant cell-wall degrading enzymes such as pectate lyase, polygalacturonase, cellulase and protease. We used genetic and biochemical methods to examine the diversity of P. carotovorum subsp. brasiliensis isolates, and found genetic diversity within the brasiliensis subsp. isolates in Korea. The restriction fragment length polymorphism analysis based on the recA gene revealed a unique pattern for the brasiliensis subspecies. The Korean brasiliensis subsp. isolates were divided into four clades based on pulsed-field gel electrophoresis. However, correlations between clades and isolated hosts or year could not be found, suggesting that diverse brasiliensis subsp. isolates existed.
