연화부를 포함한 판재의 항복거동과 항복강도 (Yielding behavior and yield strength of plate structure containing softened region)

  • Welded joint often contains soft or softened regions such as the HAZ of TMCP steel welded with high heat input. In this study, the equivalent yield strength of plate structure containing softened region was predicted by FEM analysis, and its incremental behavior was explained with the results of the analysis. The calculated results of yield strength indicated the following for the plate structures. 1) As the softened region starts to yield, shear stress begins to build up along the boundary between base metal and softened region. This results in multi-axial stress condition which gives restraint on the softened region. 2) Restraint effect has a significant influence on the distribution of the shear stress, the nominal stress, and the strain. 3) The yielding behavior of softened region becomes the same as that of base metal when both ratios of length to width and thickness to width of softened region are larger than 30 and 13 respectively.

An experimental investigation of the effects of chronic stress on bone-to-implant contact

  • Objectives: This study aimed to investigate the effects of chronic restraint stress on the osseointegration of titanium implants. Materials and Methods: Twenty adult male Wistar albino rats were used in the study. After surgical insertion of titanium implants into the metaphyseal part of the tibial bone, rats were randomly divided into two groups: a control group (CNT group) and an experimental restraint stress group (RS group). In the CNT group, titanium implants were inserted surgically, and rats received no further treatment during the 47-day experimental period. In the RS group, restraint stress was applied for 3 hours per day for 45 days, beginning 2 days after implant surgery. Weight of the rats was measured prior to surgery and at the end of the study to analyze the effects of stress. At the end of the experimental period, rats were euthanized, and implants and surrounding bone tissues were used for undecalcified histological analysis. Serum cortisol levels were assessed in cardiac blood samples from the rats following centrifugation. Results: Average weight of rats in the RS group was lower than that of rats in the CNT group after the experimental protocol had been completed (P<0.05). Further, serum cortisol levels were higher in the RS group than in the CNT group (P<0.05). There were no significant differences in boneimplant connection levels between the two groups (P>0.05). Conclusion: The data analyzed in this study suggest that chronic restraint stress did not adversely affect rats during a 45-day osseointegration period.

The Anti-apoptotic Effect of Ghrelin on Restraint Stress-Induced Thymus Atrophy in Mice

  • Thymic atrophy is a complication that results from exposure to many environmental stressors, disease treatments, and microbial challenges. Such acute stress-associated thymic loss can have a dramatic impact on the host's ability to replenish the necessary naïve T cell output to reconstitute the peripheral T cell numbers and repertoire to respond to new antigenic challenges. We have previously reported that treatment with the orexigenic hormone ghrelin results in an increase in the number and proliferation of thymocytes after dexamethasone challenge, suggesting a role for ghrelin in restraint stress-induced thymic involution and cell apoptosis and its potential use as a thymostimulatory agent. In an effort to understand how ghrelin suppresses thymic T cell apoptosis, we have examined the various signaling pathways induced by receptor-specific ghrelin stimulation using a restraint stress mouse model. In this model, stress-induced apoptosis in thymocytes was effectively blocked by ghrelin. Western blot analysis demonstrated that ghrelin prevents the cleavage of pro-apoptotic proteins such as Bim, Caspase-3, and PARP. In addition, ghrelin stimulation activates the Akt and Mitogen-activated protein kinases (MAPK) signaling pathways in a time/dose-dependent manner. Moreover, we also revealed the involvement of the FoxO3a pathway in the phosphorylation of Akt and ERK1/2. Together, these findings suggest that ghrelin inhibits apoptosis by modulating the stress-induced apoptotic signal pathway in the restraint-induced thymic apoptosis.

Stress-induced Decrease of Granule Cell Proliferation in Adult Rat Hippocampus: Assessment of Granule Cell Proliferation Using High Doses of Bromodeoxyuridine Before and After Restraint Stress

  • Stress is known to inhibit granule cell proliferation in the hippocampus. However, recent studies suggest that the commonly used dose of bromodeoxyuridine (BrdU) is insufficient to label all fractions of granule cells. Furthermore, stress-induced changes in BrdU availability may influence the labeling of newly born cells. To investigate whether changes in BrdU availability affect measurements of stress-induced granule cell proliferation, granule cell proliferation was assessed using injection of high doses of BrdU before and after restraint stress lasting 1 h. In addition, to determine whether stress-induced changes in plasma corticosterone levels were influenced by the BrdU, time-dependent changes in plasma corticosterone levels over 2 h after BrdU injection were compared with total accumulated plasma corticosterone levels [as determined by areas under the curve (AUC)]. Restraint stress significantly reduced the numbers of BrdU-labeled cells and clusters in the granule cell layer (GCL) of rats that received BrdU after stress, and decreases of similar magnitude were observed when the rats were given BrdU before stress. BrdU injection enhanced the stress-induced plasma corticosterone response, but there was no difference between the mean AUCs of plasma corticosterone levels of animals injected with BrdU before or after stress. These observations suggest that restraint stress decreases granule cell proliferation, and that this may be influenced by the extent and duration of plasma corticosterone increases rather than by changes in the availability of BrdU.

튜브 용접부의 용접변형 및 잔류응력에 관한 연구 (A Study on Welding Distortion and Residual Stress for Tubular Welded Joint)

  • The purpose of this study is to evaluate the distortion and the residual stress of GTA tubular welds between tube and head. In order to do it, the heat input model for GTA welding process was first developed by experiment and FE analyses. The welding distortion and the residual stress distribution of the tubular welds according to welding pass and various restraint degrees were evaluated by using FEA with the heat input model. From FEA results, it was found that the residual stress and the radial distortion at the weld toe of tube part decrease with a decrease in the number of welding pass. However, the maximum residual stresses in each direction of tubular welds are almost constant regardless of the external restraint degree. It was mainly due to the high internal restraint of the welds.

구속 스트레스에 의한 타액선 조직내의 Clusterin 발현 (Expression of Clusterin in the Salivary Gland under Restraint Stress)

  • 일상생활에서 우리는 항상 스트레스에 노출되어 있으며, 스트레스에 대한 반응은 우리 삶의 질과 건강에 중요한 영향을 미친다. 특히 정서적으로 중요한 구강안면영역에도 다양한 스트레스성 질환이 존재한다. 그러나 스트레스와 질병에 관한 오랜 연구에도 불구하고 스트레스성 구강안면질환의 병인은 아직 명확하지 않다. 이에 저자는 최근 임상의학에서 관심이 집중되어지고 있는 스트레스와 질병에 대한 분자생물학적 접근의 일환으로 스트레스성 구강안면질환의 병인을 규명하고자 본 연구에 임하였다. 즉, 스트레스 단백질로 알려진 clusterin이 스트레스의 생리적 반응으로서 세포보호작용 결과 발현된다는 사실에 기초하여, 스트레스 부여 후 타액선 조직 내에서의 그 발현변화를 추적하였다. Sprague-Dawley계 웅성 백서(200-230g/bw)를 구속 스트레스 부여군(구속장치에 구속한 후 0, 3, 5일에 희생) 및 정상군으로 나누고, 각각 악하선을 적출하였으며, 면역조직화학법 및 Northern Blot을 이용하여 Clusterin의 변화를 관찰하였다. 그 결과는 다음과 같다. 1. 구속 스트레스 부여군의 clusterin 단백질은 실험 즉일군에서 증가되었고, 그 이후에는 감소되었다. 2. 구속 스트레스 부여 3일군과 5일군에서 핵붕괴 및 핵농축 등의 핵변화를 동반한 apoptosis가 유도되었다. 3. 구속 스트레스 부여 결과, 선포세포가 도관세포보다 일찍 세포사하였다. 4. 구속 스트레스 부여군의 clusterin mRNA는 실험 즉일군에서만 미약하게 관찰되었다. 즉, 타액선 조직은 스트레스 단백질인 clusterin을 생산하여 세포를 보호함으로써 스트레스 상황에 적응하지만, 생리적 적응한계를 넘는 스트레스에 노출될 때에는 조직이 apoptosis됨이 확인되었다. 따라서, 본 연구결과는 구강건조증등 스트레스성 타액선관련 증상 및 질환의 병리적 기전에 대한 규명에 도움이 되리라고 생각하며, 또한 clusterin은 향후 생체에 가해진 스트레스에 대한 표식자(marker)로 사용될 수 있을 것으로 사료된다.

구속 스트레스 쥐 모델에서 스트레스 반응 감소에 대한 사카린 섭취의 효과 (Effect of Saccharin Intake in Restraint-induced Stress Response Reduction in Rats)

  • Purpose: Stress activates the sympathetic nervous system and hypothalamic-pituitary-adrenal (HPA) axis and induces the release of glucocorticoids. Saccharin is 300 times sweeter than sucrose, but does not increase blood insulin levels. Thus, this study was designed to evaluate the effect of saccharin intake in restraint-induced stress response reduction in rats. Methods: Adult male Sprague-Dawley (SD) rats had stress induced by restraint for 2 hours/day for 1 week. Saccharin was provided in sufficient amounts to allow them to intake it voluntarily at 0.1% diluted in water. The Y-maze test and forced swim test (FST) were performed to evaluate cognitive function and the depressive behavior of the rats. The protein expression of the glucocorticoid receptor (GR) in hippocampal cornu ammonis (CA) 1 was investigated by using immunohistochemistry. Results: It was found that, the percentage of alternation in the Y-maze test was significantly (p<.01) higher in the Stress + saccharin group than in the Stress group. Immobility time in the FST was significantly (p<.01) lower in the Stress + saccharin group than in the Stress group. Also, the positive cells of GR in hippocampus CA1 were significantly (p<.05) lower in the Stress + saccharin group than in the Stress group. Conclusion: This study showed that there was an effect of saccharin intake in restraint-induced stress response reduction in rats.

원주방향 균열의 균열열림에 미치는 압력유기굽힘의 구속 효과 (Effect of Restraint of Pressure Induced Bending on Crack Opening for Circumferential Crack)

  • This study evaluated the effect of restraint of pressure induced bending(PIB) on crack mouth opening displacement(CMOD) for circumferential through-wall crack in pipe by using both elastic and elastic-plastic finite element analyses. The analyses results showed the restraint of PIB was decreased crack opening for a given crack length and tensile stress, and the effect was considerable for large crack and short restraint length. Also, the restraint effect on CMOD was independent on the variation in pipe diameter and decreased with increasing pipe thickness, and it depended on not total restraint length but short restraint length for non-symmetrically restrained. Additionally, the effect of restraint of PIB was more significant in the elastic-plastic analysis results compared with in the elastic analysis results.

고빈도 120 Hz 전침이 Cold-Restraint 스트레스 유발 흰쥐의 위점막에 미치는 영향(影響) (The Effects of 120 Hz High Frequency Electroacupuncture on the Gastric Mucosa in Cold-Restraint Stressed Rats)

  • Objective : The present studies investigated the effects of 120 Hz high frequency electroacupuncture (EA) on the stress-induced stomach dysfunction in relation to its effect on the level of stress hormones and gastric mucosal damages. The gastric mucosal injury was induced by cold-restraint stress and two acupoints corresponding to Zusanli and Sanyinjiao in man were used. Methods : Cold-restraint stress produced typical gastric lesions in all rats of the stressed groups, but the number of ulcers as well as the mean ulcer diameter were reduced by 120 Hz EA pre-treatment. Results : The degranulation value of gastric mast cell was significantly higher in cold-restrained rats than in control ones. However, with the significant reduction of degranulation values of gastric mast cells in EA pre-treated rats compared with cold-restrained rats. Cold-restarint stress induced an elevated mRNA expression of pro-inflammatory gene such as cyclooxygenases-2 and tumor necrosis factor $(TNF)-{\alpha}$, but these expression were down-regulated in EA pre-treated rats. Immunohistochemical analysis showed that while the $inhibitory-{\kappa}B{\alpha}$ and $TNF-{\alpha}$ immunorection in the surface epithelium of the stomach tended to increase, both reactions in the EA pre-treated rats showed similar pattern as observed in controls. Conclusion : These results suggest that 120 Hz EA may act as a therapeutical means for gastric mucosal damages through an activation of pituitary adrenal system. It could be concluded that 120 Hz high frequency electroaucupuncture affords a good protective potential against stress-induced gastrointestinal dysfunction.

  • In this report, stress analysis of restraint specimen was done by numerical method (finite element method). Calculations were done by elastic-plastic analysis and thermo-elastic-plastic analysis. The results showed similarities for both cases, and by thermo-elastic-plastic analysis transient characteristics of welding could be found.
