BCG 접종 후 발생한 화농성 림프절염 환자의 단핵구에서 Toll-like receptor 2의 발현 (Toll-like receptor 2 expression on monocytes from patients with BCG vaccine-associated suppurative lymphadenitis)

  • 목 적 : TLR2는 숙주의 항결핵 방어면역의 중요한 역할을 하는 것으로 알려져 있다. 본 연구는 BCG 접종 후 발생한 화농성 림프절염 환자 단핵구에서 TLR2의 발현과 TLR2 리간드 자극에 의한 $TNF-{\alpha}$와 IL-6의 생성을 조사하여 화농성 림프절염 발병과 TLR2의 연관성을 알아보고자 하였다. 방 법 : BCG 접종 후 발생한 화농성 림프절염 환자 16명과 건강 대조군 10명의 말초 혈액에서 단핵구를 분리하고, TLR2 리간드인 Pam3CSK4로 자극한 후 유세포분석과 역전사중합효소반응을 이용하여 TLR2의 발현을 측정하였고, 자극 후 $TNF-{\alpha}$와 IL-6의 생성을 측정하여 TLR2의 발현 정도를 간접적으로 조사하였다. 결 과 : BCG 접종 후 발생한 화농성 림프절염 환자 단핵구의 TLR2 발현 정도($3.39{\pm}1.2%$)는 대조군($4.64{\pm}2.6%$)에 비하여 유의하게 감소하였고, 단핵구 자극에 의한 $TNF-{\alpha}$와 IL-6의 생성도 대조군($TNF-{\alpha}$, $1,098.5{\pm}94.3pg/mL$; IL-6, $6,696.3{\pm}544.3pg/mL$)에 비하여 환자군($TNF-{\alpha}$, $775.5{\pm}60.8pg/mL$; IL-6, $4,645.8{\pm}583.9pg/mL$)에서 유의하게 감소하였다. 그리고 자극 시간에 따른 TLR2 발현 정도와 $TNF-{\alpha}$와 IL-6의 생성 증가가 유사한 양상을 나타내었다. 결 론 : 본 연구의 결과에서 BCG 접종 후 발생한 화농성 림프절염 환자군 단핵구의 TLR2 발현 감소가 연관되어 있고, M. bovis BCG의 리간드 인식에 TLR2가 관여함을 추정할 수 있다.

안구내 악성종양에 대한 저준위 방사선요법에 관한 연구 (A study of Brachytherapy for Intraocular Tumor)

  • 연구의 목적 및 중요성 안구에 발생하는 악성종양의 치료에 일반적으로 사용되는 적출술이나 외부 방사선치료법은 시력상실 및 충분한 종양선량을 얻을 수 없는 단점이 있다. 최근 들어 기존치료법의 단점들을 줄이면서 치료 효과를 높이기 위한 치료법으로서 방사성동위원소가 삽입된 기구를 이용한 근접 방사선요법이 외국에서 개발되어 시행되고 있다. 본 연구에서는 안구내 악성종양의 새로운 치료법인 근접 방사성요법을 시행하기 위하여 접합한 형태의 기구를 개발하고, 기구에서 방출되는 방사선의 선량 및 선량분포측정을 통하여 종양 및 주변조직에 분포되는 선량을 보다 정확하게 계산해 보고자 하였다. 연구의 내용 및 범위 안구내 악성종양의 치료를 위한 근접 방사선요법의 연구를 위하여, 1. 안구의 모델을 결정하고, 적당한 방사성동위원소를 선택하였으며, 2. 적합한 형태의 기구를 설계 및 제작하고, 3. 제작된 기구의 방사선 투과계수 및 기구에서 방출되는 방사선의 선량분포를 측정하였으며, 4. 이들 측정 결과와 기존의 컴퓨터 선량계산모델들의 계산 결과와 비교하였다. 연구 결과 및 활용에 대한 건의 본 연구를 통해 다음과 같은 결과를 얻었다. 1. 안구의 모델은 지름 25mm인 구형으로 결정하였으며, 근접 치료에 사용될 방사성동위원소는 크기, 반감기, 발생 방사선의 에너지 및 구입의 용이성 등을 고려하여 Ir-192가 가장 적합한 것으로 판단되었다. 2. 기구는 인체 조직과의 생물학적 반응 및 피폭 선량 차폐를 고려하여 순금으로 제작하였으며, 기구의 크기는 지름이 15mm, 17mm 및 20mm인 3종류로 하였고, 두께는 모두 1.5mm로 하였다. 3. 기구의 방사선 투과계수를 TLD 및 Film으로 측정한 결과 표면에서 모두 0.71, 표면의 1.5mm거리에서 각각 0.45, 0.49이었다. 4. Ir-192가 삽입된 기구의 선량분포 측정 결과를 Gary Luxton 등의 컴퓨터 선량계산모델 및 원자력병원에서 보유하고 있는 방사선 치료계획장치인 CAP-PLAN의 선량계산모델의 계산 결과와 비교하여, 흡수선량은 ${\pm}10\%$ 이내에서 거리로는 0.4mm 이내에서 대부분 일치하였다. 최대 오차는 각각 $11.3\%$ 및 0.8mm로 나타났다. 이상의 결과들로부터 새로 개발한 금으로 된 기구와 Ir-192 seed를 이용한 근접 방사선 치료법으로 안구의 악성종양에 대한 치료를 보다 효과적으로 시행할 수 있을 것으로 기대된다.

수막종에 대한 선형가속기형 정위방사선수술 (LINAC-based Stereotactic Radiosurgery for Meningiomas)

  • 목적 : 수막종 환자에 대한 선형가속기를 이용한 정위방사선수술의 임상 경과, 영상의학적 반응, 그리고 신경학적 후유증을 분석하여 수막종에서 정위방사선수술의 역할을 알아보고자 하였다. 대상 및 방법 : 1995년 2월부터 1999년 12월까지 26명의 수막종 환자에 대해 선형가속기를 이용한 정위방사선수술을 시행하였다. 9명은 외과적 절제술이 선행되었으며 나머지 17명은 정위방사선수술만 시행되었다. 대상환자들의 남녀 비는 7:19이었고, 연령 분포는 $14\~67$세(중앙값 51세)이었다. 정위방사선수술 당시 17명에서 병변으로 인한 신경학적 증상을 보였다. 종양 용적의 범위는 $0.7\~16.5\;cm^3$ (중앙값 $4.7\;cm^3$)이었고, 정위방사선수술 시 처방선량은 최대선량 기준으로 $46\~90\%$ (중앙값 $80\%$) 등선량곡면에 $10\~20\;Gy$ (중앙값 15 Gy)이었다. 임상 추적관찰 기간은 $1\~71$개월(중앙값 27개월)이었고 영상 추적관찰 기간은 $1\~52$개월(중앙값 25개월)이었다. 결과 : 정위방사선수술 당시 임상 증상이 있었고 임상 추적관찰 기간이 1년 이상인 14명 중 13명$(93\%)$에서 증상의 소멸 및 완화를 보였으며, 나머지 1명은 치료 후 4개월부터 종괴효과로 증상이 악화되어 7개월 후에 외과적 절제술을 시행 받았다. 영상 추적관찰 기간이 1년 이상인 14명의 환자들 중 7명$(50\%)$은 정위방사선수술 후 $6\~25$개월 (중앙값 11개월)에 종양 크기의 감소를 보였고, 6명$(43\%)$은 종양 크기의 변화가 없었다. 종양 크기의 증가를 보인 1명$(7\%)$은 외과적 절제술을 시행 받았다. 6명$(23\%)$의 환자에서 정위방사선수술 후 새로운 신경학적 후유증이 발생하였으며, 이 중 5명은 일시적 후유증으로 스테로이드 투여 등의 보존적 치료 후에 증상이 소멸되었고, 1명은 정위방사선수술로 인한 조직 괴사로 외과적 절제술을 시행 받았다. 결론 : 정위방사선수술을 이용한 수막종의 치료는 외과적 절제술이 불가능한 경우, 불완전한 절제술을 시행한 경우, 그리고 수술 후 재발한 경우에 안전하고 매우 효과적인 치료 방법으로 판단된다. 하지만 더 오랜 기간의 추적관찰이 필요하며 종양용적이 크고 주위에 결정장기가 있는 경우에는 신경학적 후유증을 줄이기 위해 좀 더 신중한 치료 계획 및 선량 결정이 필요하다.

  • The wall shear stress in the vicinity of end-to end anastomoses under steady flow conditions was measured using a flush-mounted hot-film anemometer(FMHFA) probe. The experimental measurements were in good agreement with numerical results except in flow with low Reynolds numbers. The wall shear stress increased proximal to the anastomosis in flow from the Penrose tubing (simulating an artery) to the PTFE: graft. In flow from the PTFE graft to the Penrose tubing, low wall shear stress was observed distal to the anastomosis. Abnormal distributions of wall shear stress in the vicinity of the anastomosis, resulting from the compliance mismatch between the graft and the host artery, might be an important factor of ANFH formation and the graft failure. The present study suggests a correlation between regions of the low wall shear stress and the development of anastomotic neointimal fibrous hyperplasia(ANPH) in end-to-end anastomoses. 30523 T00401030523 ^x Air pressure decay(APD) rate and ultrafiltration rate(UFR) tests were performed on new and saline rinsed dialyzers as well as those roused in patients several times. C-DAK 4000 (Cordis Dow) and CF IS-11 (Baxter Travenol) reused dialyzers obtained from the dialysis clinic were used in the present study. The new dialyzers exhibited a relatively flat APD, whereas saline rinsed and reused dialyzers showed considerable amount of decay. C-DAH dialyzers had a larger APD(11.70$\pm$1.32mmHg/min)compared to CF dialyzers(4.32$\pm$0.55mmHg/min)(p<0.05). However, there was no observable difference in the UFR between the two dialyzers. Neither APD nor UFR showed any significant increase with an increasing number of reuses for up to more than 20reuses. A substantial number of failures observed in APD(larger than 20mmHe/min)on the reused dialyzers(2 out of 40 CP and S out 26 C-DAK) were attributed to the Possible damage on the fibers. The CF 15-11 HFDs which failed APD test did not show changes in the UFR compared to normal dialyzers indicating that APD is a more sensitive test than UFR test to evaluate the integrity of the fibers. 30527 T00401030527 ^x For quantitative measurement of reflected light from a clinical diagnostic strip, a prototype old reflectance photometer was designed. The strip loader and cassette were made to obtain more accurate reflectance parameters. The strip was illuminated at 45˚c through optical fiber and the intensity of reflected light was determined at rectanguLat angle using a photodiode. The kubelka-munk coefficient and reflection optical density were determined ar four different wavelengths(500, 550, 570 and 610nm) for blood glucose strip. For higher concentration than 300mg/41 about glucose, a saturation state of abforbance was observed at 500, 550 and 570nm. The mice were divided into two groups. One group was injected with Photofrin II, intravenously (2.5mg/kg body weight). The other group received no Photofrin II. Mice from both groups were irradialed for approximately 15 minutes at 100, 300, or 500 mW/cm2 with the argon (488nm/514.5 nm), dye(628nm) and gold vapor (pulsed 628 nm) laser light. A photosensitizer behaved as an added absorber. Under our experimental conditions, the presence of Photolfrin II increased surface temperature by at least 40% and the temperature rise due to 300 mW/cm2 irradiation exceeded values for hyperthermia. Light and temperature distributions with depth were estimated by a computer model. The model demonstrated the influence of wavelength on the thermal process and proved to be a valuable tool to investigate internal temperature rise. 30536 T00401030536 ^x We investigated the structural geometry of thirty-eight Korean femurs. The purpose of this study is to identify major geometrical differences between Korean femurs 3nd others that we believe belong to Caucasians so that we would be able to get insights into the femoral component design that fits Asians including Koreans. We utilized computerized tomography (CT) images of femurs extracted from cadavers. The CT images were transformed into bitmap data by using a film scanner, and then analyzed by using a commercially available software called Image v.1.0 and a Macintosh IIci computer.The resulting data were compared with already published data. The major results show that the geometry of the Korean femurs is significantly different from that of Caucasians: (1) the anteversion angle and the canal flare index are greater by the amount of approximately 8˚ and 0.5, respectively, (2) the shape of the isthmus cross section is more round, and (3) the distance between the teaser trochanter and the proximal border of the isthmus is shelter by about 15 mm. The results suggested that the femoral component suitable for Asians should be different from the currently-used components designed and manufactured mostly by European or American companies. 30537 T00401030537 ^x It is well known that nonlinear propagation characteristics of the wave in the tissue may give very useful information for the medical diagnoisis. In this paper, a new method to detect nonlinear propagation characteristics of the internal vibration in the tissue for the low frequency mechanical vibration by using bispectral analysis is proposed. In the method, low frequency vibration of f0( = 100Hz) is applied on the surface of the object, and the waveform of the internal vibration x (t) is measured from Doppler frequency modulation of silmultaneously transmitted probing ultrasonic waves. Then, the bispectra of the signal x (t) at the frequencies (f0, f0) and (f0, 2f0) are calculated to estimate the nonlinear propagation characteristics as their magnitude ratio, w here since bispectrum is free from the gaussian additive noise we can get the value with high S/N. Basic experimental system is constructed by using 3.0 MHz probing ultrasonic waves and the several experiments are carried out for some phantoms. Our department has developed an information system for routine hematology using down-sized computer system. The operating system and database management system were SCO UNIX and SCO foxbase, respectively. For program development, we used Xbase language provided by SCO foxbase. The C language was used for interface purpose. To make the system use friendly, pull-down menu was used. The system connected to our hospital information system via application program interface (API), so the information related to patient and request details is automatically transmitted to our computer. Our system interfaced with fwd complete blood count analyzers(Sysmex NE-8000 and Coulter STKS) for unidirectional data tansmission from analyzer to computer. The polymer valve showed smaller regurgitation volume and transvalvular pressure drop compared to the mechanical valve at higher heart rate. Because the point of maximun temperature of laser-irradiated gelatin exists below the surface due to surface cooling, evaporation at the boiling temperature is made explosively from below the surface. The important parameters of this process are the conduction loss to laser power absorption (defined as the conduction-to-laser power parameter, Nk), the convection heat transfer at the surface to conduction loss (defined as Bi), dimensionless extinction coefficient (defined as Br.), and dimensionless irradiation time (defined as Fo). Dependence of Fo on Nk and Bi has been observed by experiment, and the results have been compared with the numerical results obtained by solving a 2-dimensional conduction equation. Fo and explosion depth (from the surface to the point of maximun temperature) are increased when Nk and Bi are increased.To find out the minimum laser power for explosive evaporative removal process, steady state analysis has been also made. We obtained a two-dimensional current density distribution within a phantom by processing the real and imaginary MR images from a 4.77 NMR machine. We implemented a resistivity mage reconstruction algorithm using the finite element method and sensitivity matrix. Optimal stimulus parameters, which can minimize muscle fatigue and can achieve effective muscle contraction were determined in slow and fast muscles in Sprague-Dawley rats. Stimulus patterns of our stimulator were designed to simulate electromyographic activity monitored during locomotion of normal subjects. Muscle types of the lower extremity were classified according to their mechanical property of contraction, which are slow muscle (msoleus m.) and fast muscle (medial gastrocneminus m., rectus femoris m., vastus lateralis m.). Optimal parameters of electrical stimulation for slow muscles were 20 Hz, 0.2 ms square pulse. For fast muscle, 40 Hz, 0.3 ms square pulse was optimal to produce repeated contraction. Higher stimulus intensity was required when synergistic muscles were stimulated simultaneously than when they were stimulated individually. Electrical stimulation for each muscle was designed to generate bipedal locomotion, so that individual muscles alternate contraction and relaxation to simulate stance and swing phases.   • The wall shear stress in the vicinity of end-to end anastomoses under steady flow conditions was measured using a flush-mounted hot-film anemometer(FMHFA) probe. The experimental measurements were in good agreement with numerical results except in flow with low Reynolds numbers. The wall shear stress increased proximal to the anastomosis in flow from the Penrose tubing (simulating an artery) to the PTFE: graft. In flow from the PTFE graft to the Penrose tubing, low wall shear stress was observed distal to the anastomosis. Abnormal distributions of wall shear stress in the vicinity of the anastomosis, resulting from the compliance mismatch between the graft and the host artery, might be an important factor of ANFH formation and the graft failure. The present study suggests a correlation between regions of the low wall shear stress and the development of anastomotic neointimal fibrous hyperplasia(ANPH) in end-to-end anastomoses. 30523 T00401030523 ^x Air pressure decay(APD) rate and ultrafiltration rate(UFR) tests were performed on new and saline rinsed dialyzers as well as those roused in patients several times. C-DAK 4000 (Cordis Dow) and CF IS-11 (Baxter Travenol) reused dialyzers obtained from the dialysis clinic were used in the present study. The new dialyzers exhibited a relatively flat APD, whereas saline rinsed and reused dialyzers showed considerable amount of decay. C-DAH dialyzers had a larger APD(11.70${\pm}$1.32mmHg/min)compared to CF dialyzers(4.32${\pm}$0.55mmHg/min)(p<0.05). However, there was no observable difference in the UFR between the two dialyzers. Neither APD nor UFR showed any significant increase with an increasing number of reuses for up to more than 20reuses. A substantial number of failures observed in APD(larger than 20mmHe/min)on the reused dialyzers(2 out of 40 CP and S out 26 C-DAK) were attributed to the Possible damage on the fibers. The purpose of this study is to identify major geometrical differences between Korean femurs 3nd others that we believe belong to Caucasians so that we would be able to get insights into the femoral component design that fits Asians including Koreans. We utilized computerized tomography (CT) images of femurs extracted from cadavers. The CT images were transformed into bitmap data by using a film scanner, and then analyzed by using a commercially available software called Image v.1.0 and a Macintosh IIci computer.The resulting data were compared with already published data. The major results show that the geometry of the Korean femurs is significantly different from that of Caucasians: (1) the anteversion angle and the canal flare index are greater by the amount of approximately 8˚ and 0.5, respectively, (2) the shape of the isthmus cross section is more round, and (3) the distance between the teaser trochanter and the proximal border of the isthmus is shelter by about 15 mm. In the method, low frequency vibration of f0( = 100Hz) is applied on the surface of the object, and the waveform of the internal vibration x (t) is measured from Doppler frequency modulation of silmultaneously transmitted probing ultrasonic waves. Then, the bispectra of the signal x (t) at the frequencies (f0, f0) and (f0, 2f0) are calculated to estimate the nonlinear propagation characteristics as their magnitude ratio, w here since bispectrum is free from the gaussian additive noise we can get the value with high S/N. Basic experimental system is constructed by using 3.0 MHz probing ultrasonic waves and the several experiments are carried out for some phantoms. The system connected to our hospital information system via application program interface (API), so the information related to patient and request details is automatically transmitted to our computer. Our system interfaced with fwd complete blood count analyzers(Sysmex NE-8000 and Coulter STKS) for unidirectional data tansmission from analyzer to computer. The important parameters of this process are the conduction loss to laser power absorption (defined as the conduction-to-laser power parameter, Nk), the convection heat transfer at the surface to conduction loss (defined as Bi), dimensionless extinction coefficient (defined as Br.), and dimensionless irradiation time (defined as Fo). Dependence of Fo on Nk and Bi has been observed by experiment, and the results have been compared with the numerical results obtained by solving a 2-dimensional conduction equation. Fo and explosion depth (from the surface to the point of maximun temperature) are increased when Nk and Bi are increased.To find out the minimum laser power for explosive evaporative removal process, steady state analysis has been also made. Stimulus patterns of our stimulator were designed to simulate electromyographic activity monitored during locomotion of normal subjects. Muscle types of the lower extremity were classified according to their mechanical property of contraction, which are slow muscle (msoleus m.) and fast muscle (medial gastrocneminus m., rectus femoris m., vastus lateralis m.). Optimal parameters of electrical stimulation for slow muscles were 20 Hz, 0.2 ms square pulse. For fast muscle, 40 Hz, 0.3 ms square pulse was optimal to produce repeated contraction. Higher stimulus intensity was required when synergistic muscles were stimulated simultaneously than when they were stimulated individually. Electrical stimulation for each muscle was designed to generate bipedal locomotion, so that individual muscles alternate contraction and relaxation to simulate stance and swing phases.   • 대부분의 공익성을 띈 콘텐츠들은 진부한 내용으로 메시지가 표면에 드러나는 경우가 많은데, 관객들은 이러한 콘텐츠를 흥미롭게 받아들이지 않는다. 애니메이션 또한 공익적 메시지를 전달하려다 보면 내용이 전반적으로 진부해질 수밖에 없는 것이 사실이다. <북극곰 파오>(PAO The Polar Bear, 2013)는 진부하고 교과서적인 내용인 지구 온난화 문제를 이야기함에 있어서 관객에게 흥미로운 경험이 되도록 하는 것에 초점을 두고 제작하였다. 또한 라이브 윈도우(Live windows)의 특성을 활용하여 애니메이션을 보는 것에 그치지 않고 관객의 참여를 통해 서사를 이루어낼 수 있도록 다중 형식 스토리(The Multiform Story)로 구성하였다. 본 논문에서는 <북극곰 파오>의 제작 사례를 통하여 극장과 라이브 윈도우에서의 차이점에 대하여 알아보고 이를 토대로 관객 참여를 위한 인터랙션 활용 사례를 살펴본다. 극장과 라이브 윈도우의 차이에 대해서는 장소의 특성에 따른 환경적 측면과 관객의 차이에 대해 분석해 보고, 인터랙션의 활용 사례는 불특정 다수를 위한 사용자 환경 디자인과 인터랙션에 대한 반응으로 서사가 바뀌는 과정을 중심으로 살펴보고 밀라노 광장에서의 상영 결과를 통해 공공예술로서의 애니메이션이 나아가야할 방향에 대하여 제시해본다. 라이브 윈도우의 특성상 <북극곰 파오>의 관객은 애니메이션을 보러 온 관객이 아니라 각기 다른 목적지를 향해 가고 있는 거리의 사람들이다. 이들을 대상으로 서사를 가진 애니메이션을 상영한다는 것은 새로운 시도였다. 밀라노 광장에서 상영이 시작되었을 때, 관객들은 본인의 참여로 이야기의 흐름이 바뀌는 경험을 통해 상당한 만족감을 느꼈으며, 흥미로운 경험의 끝에 지구 온난화 문제를 자연스럽게 떠올릴 수 있었을 것이다. <북극곰 파오>의 메시지를 통해 사람들의 습관과 생각을 얼마나 변화시켰는지 당장은 알 수 없지만 이러한 시도를 통해서 애니메이션의 사회적 역할의 가능성을 엿볼 수 있는 계기가 되었다. 세계적으로 수많은 애니메이션이 제작되고 있다. 대부분 극장과 TV, 영화제를 목표로 제작되고 있는데, 조금 더 다양한 방법을 통해서 관객과 만날 수 있어야 한다. 그 중 하나가 바로 공공예술로서의 애니메이션이고, <북극곰 파오>는 그러한 의미에서 새로운 시도였다고 할 수 있다. 또한 앞으로 공공예술로서의 애니메이션은 우리가 함께 더불어 살 수 있는 생각을 공유하고 나눌 수 있는 흥미로운 경험을 선사하기 위해 노력해야 한다. 다양한 매체의 예술이 공익을 생각하는 예술로 바뀌어가고 있는 만큼 애니메이션도 다양한 테크놀로지와 융합하여 새로운 형태의 공공예술로 나아갈 수 있을 것이다.

적정 구성 배양 HCT-8 기반 대장암 스페로이드의 암 줄기세포능 및 항암제 내성 평가의 비교 평가 연구 (Comparative Evaluation of Colon Cancer Stemness and Chemoresistance in Optimally Constituted HCT-8 cell-based Spheroids)

  • 암은 비균질적으로 구성된 세포집합체로 간질세포 및 세포 외 기질로 구성된 미세환경과 상호작용에 의해 발병, 전이, 심화되는 복잡한 질병이다. 하지만, 기존의 2차원 배양 세포 기반 플랫폼이 3차원적 생체 환경과 암의 비균질성을 대표하기 힘든 한계를 극복하기 위해 스페로이드 배양 세포를 비롯한 다양한 플랫폼 개발이 활발해지고 있다. 본 연구에서는 특히 감염, 염증 및 식이적 환경성 영향력에 민감한 HCT-8 대장암 세포주를 기반으로 하여 3차원 스페로이드 배양법을 보다 효과적인 방법으로 개선하고, 대장암 스페로이드 세포를 기반으로 암의 비균질적인 특질과 항암내성 연구의 간단하고 개선된 플랫폼을 제시고자 하였다. 3차원 배양법 최적화를 위해 물리적 배양환경 조성과 배양배지 구성에 따른 스페로이드 형태형성을 비교 분석하고 암 줄기세포군의 증가 양상을 확인한 결과, 필수요소로 구성된 제한 배지와 균일한 형태의 비부착성 표면 배양접시에서 배양된 스페로이드가 균일한 형태의 구형을 형성하고 암 줄기세포군이 증가함을 확인하였다. 대장암 스페로이드 세포를 기반으로 대장암 치료제인 5-Fluorouracil (5-FU)에 대한 화학적 감응성 변화를 측정한 결과, 암 줄기세포가 5-FU에 대한 화학적 감수성 저해의 원인이 되며, 최적배양 조건에서 암 줄기세포의 약제 내성의 표현이 증대되었다. 이는 암줄기세포의 항암제 내성에 대한 잠재적 위험성을 내포하는 것으로, 이 방법론은 감염, 염증 및 식이적 요인과 연관된 대장암 스페로이드 세포 기반 항암제 약물반응을 검증하기 위해 효과적이면서 간소한 시험법으로 활용될 수 있을 것이다.

  • 목 적 : B형 간염 백신은 접종 후 시간이 경과하면서 항체가는 감소하나, 면역학적 기억은 약 10년 이상 유지되며, 항체가가 감소하더라도 임상적으로 의미있는 감염에 대한 예방효과가 있어 현재 정상 면역 기능을 가진 경우 3회 접종 후 예방 가능한 항체가 형성된 경우는 추가접종을 권장하고 있지 않다. 저자들은 최근 우리나라 중부지역 소아의 B형 간염 항체 보유율과 B형 간염 백신의 면역학적 기억에 대해 연구하고자 하였다. 방 법 : 2003년 3월부터 2005년 5월까지 단국대학교병원(병동, 외래)에서 간염 항원, 항체 검사(RIA, 혹은 EIA)를 받은 소아(6개월-18세)를 대상으로 하였고, 측정한 B형 간염 항체가가 10 IU/mL 이상인 경우를 양성으로 하였다. B형 간염 항체가가 음성인 소아 중 B형 간염 백신을 1회($Euvax^{(R)}$ 혹은 $Hepavaxgene^{(R)}$) 추가 접종한 후 1개월 후에 측정한 항체가가 10 mIU/mL 이상의 양전을 보인 경우를 면역기억반응이 있다고 정의하였다. HBsAg, anti-HBs를 검사받은 대상 중에서 $-20^{\circ}C$에 보관된 169명의 혈청에서 EIA로 IgG anti-HBc를 측정하였다. 결 과 : 총 3,448명이 RIA 또는 EIA로 HBsAg/anti-HBs 검사를 받았으며 그 중 3,277명(남아 1,923명, 여아 1,354명)을 연구 대상으로 하였다. 미숙아, 면역기능 저하자, 검사 전 수혈을 받았거나 면역글로불린을 투여 받은 소아를 제외한 3,277명 중에서 1,913명(58.4%)이 anti-HBs가 양성이었고, 그 중에서 남아는 1,115명(58.3%)이었다. HBsAg 양성은 29명(0.9%)였다. 나이에 따른 anti-HBs 양성률은 6-12개월까지는 78.6%(88/112명), 1-3세는 62.7%(455/726명), 4-6세는 51.9%(407/784명), 7-12세는 49.5%(441/890명), 13-15세는 63.4%(220/347명) 그리고 16-18세까지는 72.2%(302/418명)였다. 과거에 기본접종을 받았으나 HBsAg/anti-HBs 모두 음성을 보여 추가 접종을 받은 소아는 80명이었고, 1개월 뒤 측정한 간염 항체 검사 결과 양성률은 88.8%(71/80명), 무반응자는 9명이었다. IgG anti-HBc를 검사한 169명 중에서 5명(3.0%)이 양성을 보였고, 5명 모두 HBsAg이 음성이었다. 5명 중 anti-HBs가 음성인 소아가 4명으로 모두 1회 접종 후 항체 양전을 보였다. 결 론 : 우리나라 중부지역 소아의 B형 간염 항체 양성률이 나이가 들면서 감소를 보이고 있으나 1회 접종으로 10 IU/mL 이상으로 항체가 증가를 보여 이는 B형 간염 백신의 장기간의 면역학적 기억을 보여 주는 것으로 생각된다.

Protein Kinase C Inhibitor (PKCI)에 의한 방사선 민감도 변화와 c-fos Proto-oncogene의 전사 조절 (Effect of Protein Kinase C Inhibitor (PKCI) on Radiation Sensitivity and c-fos Transcription Activity)

  • 목 적 : Ataxia-Telangiectasia (AT) 증은 여러 가지 유전적 결함을 갖는 질병으로 방사선 민감도가 비정상적으로 상승되어 있는 것이 특징이다 AT 환자에서 공통적으로 존재하는 ATM 유전자는 현재까지 방사선 신호전달에 관여하는 것으로 알려진 Pl-3 kinase와 유사한 구조임이 알려져 ATM이 방사선 신호전달경로에 중요한 작용을 할 것으로 추정하게 되었다. 본 연구에서는 AT 세포와 정상세포에 PKCI를 과발현 시킴으로써 방사선 신호전달에 관여하는 PKC를 억제하여 이것이 방사선 민감도에 미치는 영향을 관찰하고, 방사선에 의해 유도되는 early response gene인 c-fos transcription의 차이를 측정하여 ATM과 PKCI에 의한 신호전달이 c-fos 유전자 전사에 미치는 영향을 분석하고자 하였다. 대상 및 방법 : PKCI expression vector를 작제한 후 정상세포인 LM217과 AT세포인 AT5BIVA에 transfection 시킨 후 plasmid의 genomic DNA에 결합된 것은 polymerase chain reaction (PCR) 방법으로 확인하였고 PKCI의 mRNA 발현 여부는 northern blotting으로 확인하였다. 방사선 민감도는 아포토시스로 측정하였으며 PKCI가 과발현된 각 세포주에 5 Gy의 방사선을 조사한 후 48시간에 세포를 모아 TUNEL방법으로 아포토시스 세포의 수를 측정하였다. c-fos 유전자의 전사는 reporter 유전자로 c-fos CAT plsmid를 $\beta$-gal expression vector와 같이 각 세포주에 transfection 시키고 36시간이 지난 후 CAT assay를 하여 activity를 측정하고 동시에 $\beta$-gal assay를 시행하여 transfection 효율을 보정해 주었다. PKCI, Ras의 영향을 보기 위하여는 PKCI, Ras expression vector와 c-fos CAT plasmid를 cotransfection하고 CAT activity로 측정 하였다. 결 과 : 이 실험의 결과 LM과 AT 세포에서 PKCI가 방사선 민감도에 미치는 영향과 c-fos 전사에 미치는 영향을 처음으로 보여주었다. PKCI의 과발현이 LM 세포에서는 방사선 민감도를 증가시켰지만 AT세포에서는 오히려 약간 감소시키는 작용을 나타내었다. c-fos 전사는 AT 세포에서 LM 세포에 비하여 70배 낮게 나타났는데 PKCI가 과발현 됨으로써 LM 에서는 c-fos의 전사가 감소되었지만 AT 세포에서는 영향이 없었다. Ras 단백으로 c-fos를 유도시키고 여기에 PKCI 발현 백터를 contransfection 하면 LM세포에서는 induction 이 감소되었지만 AT 세포에서는 영향이 없었다. 즉 LM과 AT 세포에서의 PKCI에 의한 반응의 차이는 Ras와 관련된 signal transduction pathway라는 것을 알 수 있었다. 결 론 : PKCI는 정상세포에서는 방사선에 의한 세포 손상을 증가시키지만 AT 세포에서는 별 영향을 보이지 않는 것을 알 수 있었으며, 두 세포간의 이러한 차이는 c-fos proto-oncogene의 전사차이로 설명할 수 있겠다. 이러한 차이가 AT 세포의 방사선 민감도의 한 원인일 것으로 생각된다.

As a result, accelerated senescence and enzymatic browning OCCUI', Recent l'esearch on minimally processed fl'uits and vegetables has mainly focused on methods to inhibit browning, due to ban on use of sulfur dioxide, In order to retard or prevent these physiological changes, val'ious al ternatives, reducing agents. acidulants, chelating agents and inol'ganic sal ts have been evaluated for use on fresh cut fl'ui ts. Al though some agents were effective replacement for sulfur dioxide. consum$\textregistered$I'S demandless use of chemical on such products. Shel~ life of minimally processed products has been extended by inhibition of metabolic reactions associated with loss of quality and by inhibition of aerobic spoilage caused by wide variety of microorganisms. Appl ication of ~I.-\ packaging, including vacuum packaging, retards the rate of respiration, prevents growth of aerobic spoilage organisms, inhibits oxidation and color deterioration. Tissue softening is another major problem in minimally processed products because enzymes re 1 a ted to ce 11 wa 11 degrada t i on are not inactivated. Various treatments have been investigated for retardation of the softening of sliced products. Some studies have concentrated on the application of an active packaging system with ~I, l. packaging and calcium infi 1 tration as possible measures to retain firmness of processed products. In my opinion, one important step for production of minimally processed frui ts wi th favorabl e color of cut surface and firm texture is the selection of better cultivar. As the view, changing tendency of fresh color by apple cultivars and relationship between the tendency and PPO activity will be discussed in the seminar. In addition to the topic, research result on quality enhancement of fresh apple slices by heat shock treatment will be introduced.
