Phytophthora infestans에 의해 발생하는 토마토 잎마름 역병은 전 세계적으로 발생하여 토마토 생산에 막대한 피해를 준다. 이 병의 방제를 위하여 저항성 품종의 이용은 매우 중요하다. 저항성 품종육종의 초기 및 본 육종에서 토마토 유전자원의 저항성 평가는 매우 중요하다. 본 연구는 2009년에 유묘 및 성체식물을 대상으로 78종의 토마토 시판 품종과 계통에 대한 잎마름역병 저항성을 검정하였다. Legend를 포함한 모든 시판 품종은 감수성이지만, California 대학에서 분양 받은 KNU-2, KNU-6-1, KNU-11, KNU-13, KNU-14-1 계통과 저항성 유전자 Ph-3를 가지고 있는 L3708, $AV107-4{\times}L3708$, $07-15{\times}L3708$, $BS67{\times}L3708$ 및 $06-9-62A{\times}06-9-62A$ 계통은 고도의 저항성인 것으로 나타났다. 본 연구에서 선발된 토마토 잎마름역병에 고도의 저항성을 가지는 10계통은 저항성 품종육종을 위한 유용한 자원으로 활용될 수 있다.
Using the abundant available information about the tomato genome, we developed DNA markers that are linked to disease resistant loci and performed marker-assisted selection (MAS) to construct multi-disease resistant lines and varieties. Resistance markers of Ty-1, T2, and I2, which are linked to disease resistance to Tomato yellow leaf curl virus (TYLCV), Tomato mosaic virus (ToMV), and Fusarium wilt, respectively, were developed in a co-dominant fashion. DNA sequences near the resistance loci of TYLCV, ToMV, and Fusarium wilt were used for primer design. Reported candidate markers for powdery mildew-resistance were screened and the 32.5Cla marker was selected. All four markers (Ty-1, T2, I2, and 32.5Cla) were converted to cleavage amplification polymorphisms (CAPS) markers. Then, the CAPS markers were applied to 96 tomato lines to determine the phenetic relationships among the lines. This information yielded clusters of breeding lines illustrating the distribution of resistant and susceptible characters among lines. These data were utilized further in a MAS program for several generations, and a total of ten varieties and ten inbred lines were constructed. Among four traits, three were introduced to develop varieties and breeding lines through the MAS program; several cultivars possessed up to seven disease resistant traits. These resistant trait-related markers that were developed for the tomato MAS program could be used to select early stage seedlings, saving time and cost, and to construct multi-disease resistant lines and varieties.

In order to develop chili pepper bacterial spot resistant cultivars and near-isogenic lines (NILs) to prompt the molecular mapping of the resistance gene, we have run backcross breeding program since 1994. Two resistance genes against Xanthomonas axonopodis pv. vesicatoria Bs2 from Fla. XVR 3-25 and Bs3 from our breeding line 25-11-3-2, were introduced into a land race, Chilseongcho (abbreviated to Chilseong hereafter) with good fruit guality. We report here the testing of $BC_4F_3\;to\;BC_4F_5$. We found that $BC_4F_5$ lines of the crosses were homozygous with respect to the respective genes of introduction. The lines, in which Bs2 gene was introduced, were hypersensitively resistant to both race 1 and race 3 of X. axonopodis pv. vesicatoria, whereas, those in which Bs3 was introduced were resistant to race 1.
This review describes the stratagies of development of virus-resistant Alstroemeria plants using the genetic modification system. Despite of increasing of its importance in cut flower market, improvements of some horticultuirally important traits such as fragrance, long vase-life, virus resistance and tolerance against abiotic stresses are lack of the breeding program in Alstroemeria. Of these traits, virus-resistance is quite difficult to develop in Alstroemeria plants due to the limitations of genetic variation in the existed germplasm. To extend the genetic variation, plant biotechnological techniques such as genetic transformation and tissue culture should be combined to develop virus-resistant line in Alstroemeria. In this review, several strategies for the generation of virus-resistance by using natural resistance genes, pathogen-derived genes and other sources including pathogen-derived proteins, virus-specific antibodies and ribosome-inactivating proteins are presented. Also, brief histories of breeding, tissue culture, and transformation system in Alstroemeria plants are described to inderstand of the application of transgenic approach for the development of virus-resistance in Alstroemeria species.
Insects are among the most important abiotic and biotic constraints to rice production. National rice research programs are in various stages in the development and implementation of integrated pest management (IPM) stratagies for rice insect control. Among the various control tactics, insect resistant cultivars are sought as the major tactic in rice IPM. Through the activities of interdisciplinary teams of scientists significant progress has been made in the development and release of insect resistant cultivars to farmers. Because of its compatibility with other control tactics insect resistance has proven to fit well into the IPM approach to rice insect control agents and minimize the need for insecticide applications. The development of biotypes which overcome the resistance in rice plants has been a significant constraint in the breeding of rice for resistance to insects. Most notable examples in Asia are the green leafhopper, Nephotettix virescens, brown planthopper, Nilaparvata lygens and the Asian rice gall midge, Orseolia oryzae. The current breeding stratege is to develop rice cultivars with durable resistance on which virulent biotypes cannot adapt. In spite of the significant progress made in the breeding of insect resistant cultivars there are still numerous important rice insect species for which host plant resistance as a control tactic has not been fully utilized. Advances in biotechnology provide promise of solving some of the problems that have limited the use of host plant resistance as a major tactic in the integrated management of rice insect pests.

Bud rot (BR) is the most devastating disease affecting oil palm (Elaeis guineensis) crops in Colombia. Its causal agent, Phytophthora palmivora, initiates the infection in immature oil palm leaflets producing necrotic lesions, followed by colonization of opportunistic necrotrophs, which increases disease damage. To improve the characterization of the disease, we transformed P. palmivora using Agrobacterium tumefaciens-mediated transformation (ATMT) to include the fluorescent proteins CFP-SKL (peroxisomal localization), eGFP and mRFP1 (cytoplasmic localization). The stability of some transformants was confirmed by Southern blot analysis and single zoospore cultures; additionally, virulence and in vitro growth were compared to the wild-type isolate to select transformants with the greatest resemblance to the WT isolate. GFP-tagged P. palmivora was useful to identify all of the infective structures that are commonly formed by hemibiotrophic oomycetes, including apoplastic colonization and haustorium formation. Finally, we detected cell death responses associated with immature oil palm tissues that showed reduced susceptibility to P. palmivora infection, indicating that these tissues could exhibit age-related resistance. The aim of this research is to improve the characterization of the initial disease stages and generate cell biology tools that may be useful for developing methodologies for early identification of oil palm materials resistant or susceptible to BR.
Although zoysia grass (Zoysia spp.) has a lot of excellent chracteristics as warm-season grass, it have been limited in use due to slow establishment, low seed production, poor shade tolerance and other factors. Breeding trials have been continued from 1900's, much attentions have been paid especially in U.S.A., Korea and Japan recently. In U.S.A., more than 24 varieties had been evaluated at National Turfgrass Evaluation Program(NTEP) from 1991 to 1995 and some were regsistered as commercial. After the 6th International Turfgrass Research Conference at Japan in 1989, Japan Turfgrass In-corporation (JTI) sponsored by private companies and government carried out breeding programs for pest, salt, and shade tolerant and herbicide resistant varieties. JTI also has been trying to im-prove vigor and breed evergreen zoysia Korean breeders collected germplasms since 1960's. After USDA breeders came to Korean penesula in 1982, Korean breeders joined with USDA zoysia breeding project for several years. Many interspecific hybrids and natural selected varieties were breeded that period both in U.S.A and Korea. Breeding objectives were to extend green color period, improve leaf quality and density, and better leaf color at dormant stage. Since 1990's, zoysia grass breeding trials are getting more diverse in many points such as random amplified polymorphic DNA (RAPD) assay for ecotype identification. The objectives of this study are to evaluate germplasms in Korea, and also review the present status and future prospect in zoysia grass breeding in the world.
본 시험은 고추의 육종을 위한 병 저항성 고추를 선발하고자 하였다. 국내에서 시판중인 역병 저항성 품종인 PR계통 21품종, 미국에서 도입해온 36품종과 역병감수성 품종인 '슈퍼마니따'를 사용하여 역병, 탄저병, 흰가루병의 병 저항성 검정을 수행하였다. $Phytophthora$$capsici$에 의해 발병하는 고추역병의 유묘검정은 서로 다른 교배형(A1, A2, Sterile)을 가지는 3 균주를 사용하여 접종하였다. 그 결과 대부분의 PR계통 고추들은 각각의 교배형에 대하여 저항성 또는 중도저항성을 나타내는 것으로 조사되었다. '역강홍장군'은 서로 다른 교배형의 세 가지 균주에 대하여 저항성을 나타냈으며, 'PR-다따'와 'PR-마니따'는 각각의 교배형에 저항성 또는 중도저항성을 보였다. 외래종의 경우, 'NuMex J.E.Parker', 'Omni Color'과 'SCM-334' 품종은 세 가지 균주에 저항성을 나타냈고, 'Sweet Banana'와 'Tabasco'를 포함한 몇몇 품종들은 각각의 균주에 중도저항성을 나타냈다. 흰가루병에 대하여 'Orange Habanero', 'black Cuban'은 저항성을 나타냈으며, '슈퍼마니따'와 'PR 금동'은 중도저항성을 나타냈다. 'Santa Fe Grnade', 'NuMex Pinata' 품종은 흰가루병에 매우 심한 감수성인 것으로 조사되었다. 탄저병의 경우, 'Aji Limon'과 'C. baccatum var. pendelum 3-4' 품종은 저항성인 것으로 조사되었다. 'Pobalno', 'Omni Color', 'Negro', 'Mesilla', 'Mulato', 'Bhut Jolokia', 'Big Dipper', 'Black Cuban', 'NuMex Pinata'와 'NuMex Big Jim'은 중도저항성을 나타내는 것으로 조사되었다. '태산'을 제외한 대부분의 PR계통들은 탄저병에 대하여 감수성 내지 심한 감수성인 것으로 확인되었다. 본 연구에서 확인된 저항성 개체는 이후에 고추 병저항성 육종에 유용한 유전자원으로 이용될 수 있다.
고추 역병은 그 병원균의 토양전염성 때문에 약제에 의한 방제효과가 낮아 저항성 품종의 개발이 기대되어 왔다. 고추 역병에는 CM334, AC2258, PI201234 등 다수의 저항성 유전자원이 보고되었으며, 이들의 저항성의 유전에 관한 연구로 진행되었다. 그러나 저항성의 유전양식은 실험에 사용한 재료와 연구자에 따라 1개, 2개, 혹은 3개 이상의 유전자, 다수의 유전자에 의한 양적 유전 등 다양하였다. 최근에는 분자적 방법으로 양적형질유전자좌(QTL)를 구명하는 연구가 보고되고 있으며, 분자표지를 이용한 선발 기술도 이미 육종 현장에서 활용되고 있다. 최근 저항성 품종이 다수 출시됨에 따라 새로운 병원형(pathotype), 즉, 레이스(race)의 출현에 관심이 높아지면서 이에 대한 연구가 보고되고 있다. 모두 품종과 병원균주간에 특이적 변이가 있으며, 이를 토대로 몇 개의 병원형(race)으로 분류할 수 있었다. 그러나 판별품종이 통일되지 않았고 시험에 사용한 품종들의 저항성 유전자의 조성도 달라 세계적으로 통일된 레이스분류체계는 아직 없는 실정이다. 이러한 배경에서 보다 안정된 저항성 품종의 육성을 위해서는 한 가지 저항성 재료보다는 다수의 저항성 유전자원에서 저항성을 도입하고, 육성과정에 육성품종의 보급 대상지역의 여러 균주를 사용하여 선발하는 것이 필요할 것이다.
벼 도열병은 안정적인 쌀 생산을 위협하는 대표적인 병으로, 도열병 방제에는 저항성 품종의 육종이 가장 효과적으로 이용되었다. 병원균의 꾸준한 수집과 이들을 이용한 저항성 검정 테스트는 신품종 육성 프로그램에서 중요한 과정이다. 1985년 이래 우리나라의 도열병균은 8개의 판별품종을 이용한 레이스 판별시스템을 구축하여 분류하였다. 그러나 기존의 판별품종은 유전자형이 연구된 바 없어 새로운 레이스 출현이나 병 저항성 붕괴 등에 대하여 과학적으로 분석하여 신속하게 대처하기가 어려웠다. 따라서 이 연구에서는 유전자형이 밝혀진 단인자 저항성 계통과 우리나라에서 수집된 200개 균주의 병원성 반응을 분석하여, 새로운 판별 품종 시스템의 필요성을 제시하였다. 또한, 24개의 단인자 저항성 계통의 병원성 반응을 통해 9개의 대표 저항성 유전자를 선발하였으며, 이에 따른 10개의 대표균주 그룹과 그 저항성 반응을 인덱스화 하고 30개의 한국형 대표균주를 선발하였다. 이 연구는 향후 도열병 저항성 검정 및 벼 신품종 육종에 기초자료로서 활용될 수 있을 것이다.
