The purpose of this study was to develop a system capable of restoring the seismic performance of a precast concrete (PC) connection damaged by an earthquake. The developed PC connection consists of a top-and-seat angle, post-tensioning (PT) tendons, and U-shaped steel. The PC beam can be replaced by cutting the PT tendons in the event of damage. In addition, the seismic performance of the developed PC beam-column connection was evaluated experimentally. A PC beam-column connection specimen was fabricated, and a quasistatic cyclic loading test was conducted to a maximum drift ratio of 2.3%. Subsequently, the PC beam was replaced by a new PC beam, and the repaired PC connection was loaded to a maximum drift ratio of 5.1%. The structural performance of the repaired PC connection was then compared with that of the original PC connection. The difference in the load at the drift ratio of 2.3% between the original and the repaired PC specimens was only 0.2%. The residual drift ratio in the repaired PC specimen did not exceed 1.0% at the 2.0 % drift ratio cycles, which satisfies the life safety performance level specified in ACI 374.2R-13. When the developed PC connection system is used, structural performance can be restored by rapidly replacing the damaged elements.

Advanced Oxidation Processes (AOP) have been interested for removing micropollutants in water. Most of water treatment plants (WTPs) located along the lower part of Nakdong River have adopted the $O_3/BAC$ process and have interesting in peroxone process a kind of AOP. This study evaluated the removal characteristics of residual hydrogen peroxide ($H_2O_2$) combining with the biofiltration process in the next BAC process when the hydrogen peroxide is applied for the WTP operating $O_3/BAC$ process. In the experiment, changing the temperature and the concentration of $H_2O_2$ of influent, the biofiltration process showed rapidly dropped the biodegradability when the $H_2O_2$ concentration was increased and lowered water temperature while BAC process maintained relatively stable efficiency. The influent fixed at $20^{\circ}C$ and the concentration of $H_2O_2$ at 300 mg/L was continuously input for 78 hours. Most of the $H_2O_2$ in the influent did not remove at the biofiltration process controlled 5 to 15 minutes EBCT condition after 24~71 hours operating time while BAC process controlled 5 to 15 minutes EBCT showed 38~91% removal efficiency condition after 78 hours operating time. Besides, after 78 hours continuously input experiment, the biomass and activity of attached bacterial on the biofilter and BAC were $6.0{\times}10^4CFU/g$, $0.54mg{\cdot}C/m^3{\cdot}hr$ and $0.4{\times}10^8CFU/g$, $1.42mg{\cdot}C/m^3{\cdot}hr$ respectively. These biomass and activity values were decreased 99% and 72% in biofilter and 68% and 53% in BAC compared with initial condition. The biodegradation rate constant ($k_{bio}$) and half-life ($t_{1/2}$) in BAC were decreased from $1.173min^{-1}$ to $0.183min^{-1}$ and 0.591 min to 3.787 min respectively according to increasing the $H_2O_2$ concentration from 10 mg/L to 300 mg/L at $5^{\circ}C$ water temperature and the $k_{bio}$ and $t_{1/2}$ were $1.510min^{-1}$ to $0.498min^{-1}$ and 0.459 min to 1.392 min at $25^{\circ}C$ water temperature. By increasing the water temperature from $5^{\circ}C$ to $15^{\circ}C$ or $25^{\circ}C$, the $k_{bio}$ were increased 1.1~2.1 times and 1.3~4.4 times. If a water treatment plant operating $O_3/BAC$ process is considering the hydrogen peroxide for the peroxone process, post BAC could effectively decrease the residual $H_2O_2$, moreover, in case of spilling the $H_2O_2$ into the water process line, these spilled $H_2O_2$ concentration can be able to decrease by increasing the EBCT at the BAC process.
Responses of real-time control parameters, such as ORP, DO and pH, to the conditions of biological animal wastewater treatment process were examined to evaluate the stability of real-time control using each parameter. Also an optimum index for supplemental carbon source addition based on NOx-N level was determined under a consideration of denitrification rate by endogenous respiration of microorganism and residual organic matter in liquor. Experiment was performed with lab-scale sequencing batch reactor(SBR) and working volume of the process was 45L. The distinctive nitrogen break point(NBP) on ORP-and DO-time profiles, which mean the termination of nitrification, started disappearing with the maintenance of low NH4-N loading rate. Also the NBP on ORP-and DO-time profiles was no longer observed when high NOx-N was loaded into the reactor, and the sensitivity of ORP became dull with the increase of NOx-N level. However, the distinctive NBP was constantly occurred on pH(mV)-time profile, maintaining unique profile patterns. This stable occurrence of NBP on pH(mV)-time profile was lasted even at very high NOx-N:NH4-N ratio(over 80:1) in reactor, and the specific point could be easily detected by tracking moving slope change(MSC) of the curve. Revelation of NBP on pH(mV)-time profile and recognition of the realtime control point using MSC were stable at a condition of over 300mg/L NOx-N level in reactor. The occurrence of distinctive NBP was persistent on pH(mV)-time profile even at a level of 10,000mg/L STOC(soluble total organic carbon) and the recognition of NBP was feasible by tracing MSC, but that point on ORP and DO-time profiles began to disappear with the increase of STOC level in reactor. The denitrfication rate by endogenous respiration and residual organic matter was about 0.4mg/L.hr., and it was found that 0.83 would be accepted as an index for supplemental carbon source addition when 0.1 of safety factor was applied.
Polychlorinated dibenzo-p-dioxins (PCDDs), polychlorinated dibenzofurans (PCDFs) and dioxin-like polychlorinated biphenyls (dl-PCBs) are bioaccumulative chemicals that are considered to be toxic contaminants based on several epidemiological studies. These chemicals in colostrum were investigated and estimated for their residual consistencies by maternal characteristics like parity and maternal ages. Test subjects were healthy primipara and multipara mothers with a mean age of 31.5 (S.D=3.6) in 2007. Seven isomers of PCDDs, 10 of PCDFs, 4 of non-orthopolychlorinated biphenyls(non-ortho PCBs) and 8 of mono-orthochlorinated polychlorinated biphenyls (mono-ortho PCBs) were analyzed by HRGC/HRMS. From the analyzed data, the mean level of total WHO-TEQs was 9.41 pg TEQ/g lipid, which is significantly lower than the level found in individuals from other countries. The main contributors to the total WHO-TEQs with increasing percentages were 2,3,4,7,8-PeCDF, 1,2,3,7,8-PeCDD and 3,3',4,4',5-PCB (#126), and they accounted for more than 60% of the total WHO-TEQs. PCDFs concentrations and total WHO-TEQs were negatively associated with parity (p<0.05), and maternal age was positively associated with total WHO-TEQs (p<0.01). However, the associations with body mass index (BMI) and fish intake during pregnancy were not significant. These results were suggested that parity and maternal age are an important factor affecting the concentrations of PCDD/DFs and dl-PCBs in these specimens.
The initiation and growth processes of cyclic ice body in porous systems are affected by the thermo-physical and mass transport properties, as well as gradients of temperature and chemical potentials. Furthermore, the diffusivity of deicing chemicals shows significantly higher value under cyclic freeze-thaw conditions. Consequently, the disintegration of concrete structures is aggravated at marine environments, higher altitudes, and northern areas. However, the properties of cyclic freeze-thaw with crack growth and the deterioration by the accumulated damages are hard to identify in tests. In order to predict the accumulated damages by cyclic freeze-thaw, a regression analysis by the response surface method (RSM) is used. The important parameters for cyclic freeze-thawdeterioration of concrete structures, such as water to cement ratio, entrained air pores, and the number of cycles of freezing and thawing, are used to compose the limit state function. The regression equation fitted to the important deterioration criteria, such as accumulated plastic deformation, relative dynamic modulus, or equivalent plastic deformations, were used as the probabilistic evaluations of performance for the degraded structural resistance. The predicted results of relative dynamic modulus and residual strains after 300 cycles of freeze-thaw show very good agreements with the experimental results. The RSM result can be used to predict the probability of occurrence for designer specified critical values. Therefore, it is possible to evaluate the life cycle management of concrete structures considering the accumulated damages due to the cyclic freeze-thaw using the proposed prediction method.
This study was conducted to evaluate the substitution effect of red beet powder on sodium nitrite in emulsion-type pork sausages, and to investigate the effect of the addition of red beet powder on the physicochemical characteristics of emulsion-type pork sausages at 10℃ for 6 weeks. The treatments were divided into five groups: Control (sodium nitrite 0.01%), T1 (sodium nitrite 0.005% + red beet powder 0.5%), T2 (sodium nitrite 0.005% + red beet powder 0.25%), T3 (red beet powder 0.5%), and T4 (red beet powder 0.25%). The redness value was higher in T1, whereas the yellowness value was higher in the T3 and T4 groups compared with the others (p<0.05). The pH values of emulsion-type pork sausages were significantly decreased in the T3 and T4 groups by the addition of red beet powder (p<0.05). However, the DPPH radical-scavenging activity was higher in the T1 and T3 groups than in the others (p<0.05). The residual nitrite ion was the highest in the control group (p<0.05). Therefore, it is determined that red beet powder can substitute for nitrite as a natural colorant, and it has a slightly antioxidant effect in emulsion-type pork sausages.
There has been an increasing public concern about environmental safety and human health of field crops while customer consumes more and more field crops, to which overdose and residual chemicals were applied. As a solution for the problems, construction of sustainable agroecosystem is spreading out to pursuit the economic value of agricultural management as well as to meet environment concern. Public has extended their understanding on the preservation of environment and safeness of agricultural products, and governmental policy toward sustainable agriculture drives rapid increase of the production of sustainable agricultural products. Under this circumstances, it is time to encourage more consumption and to activate market system for the sustainable agricultural products. This study was initiated to diagnose the problems and future direction of domestic sustainable agriculture by analyzing the overall opinions of consumer on the sustainable agriculture. The results are as follows: The pursuit of low input management is a prerequisite to creat high valued agricultural products, and serious consideration should be taken to produce clean crops using natural products. Consummers are willing to pay 10∼50% more prices if the products get certified by official eco-label programs. It is believed that practice of sustainable agriculture using biopesticide and natural pesticide would accelerate the rapid extension of this pro-environmental agricultural management. To activate production and consumption of sustainable agricultural products correct informations on safety should be addressed to customer, and confidence has to be brought about from customer. This could be done by obtaining various and efficient distribution route, product competency for quality, upgraded sales strategy, maximum utilization of certificate system, and practice of recall system, so on.
Enzymatic activities and fluorescence spectroscopic properties of the double mutant proteins P96L/F139W, P96L/F258W and a triple mutant protein P96L/F139W/F258W of tryptophan synthase $\alpha$ subunit from Escherichia coli was examined to study tertiary and local structure changes around the tryptophan residues. The enzymatic activities of P96l./F139W and P96L/F258W were similar, but P96L/F139W/F258W had lower activity, as compared to wild type. The fluorescence intensities of double mutant, P96L/F139W and P96L/F258W, were decreased but that of a triple mutant, P96L/F139W/F258W, was increased when compared to wild type. The sum of the maximum fluorescence intensity (fluorescence intensity at the λ$_{max}$) for the double mutant proteins was not equal to the intensity seen in the triple mutant protein. The enzymatic activity and fluorescence data indicate that the replacement of Pro$^{96}$ longrightarrowLeu might affect on the stability of helix 8 and the loop located between strand 4 and helix4. The result suggests that the tertiary structure of triple mutant (P96L/F139W/F258W), being different from wild type, might have more compact residual structure at the vicinity of 139 and 258.8.
Food irradiation is an emerging technology which offers many advantages such as reduction of microorganisms, extension of shelf-life of foods, reduction in the use of post-harvest chemicals, and destruction of insects and parasites. The commercial utilization of food irradiation, however, has been restricted because of the uncertainty of consumers' responses to it. Because success of food irradiation in the marketplace will depend upon their acceptability by consumers, this study focused on the consumers' perception and acceptance toward food irradiation in order to get basic data for commercial utilization of food irradiation and give information to consumers to help rational consumption behavior. The survey with 411 respondents living in Youngnam area was conducted during the spring of 1997 by the questionnaires. The results and implications from this study are as follows. First, consumers' knowledge about food irradiation is scanty. Two-thirds of respondents in the survey had not heard of irradiated foods and many people confused irradiation with radioactivity. In the willingness to accept food irradiation, one-third of respondents showed a wait-and-see attitude. This result indicated consumers had insufficient information about the irradiation process and nationwide education of food irradiation technology should be undertaken. Second, although the purchase and use of food are very important consumption behaviors, consumer education by mass communication has been rarely done. For the successful commercialization of food irradiation, the information provision by mass communication for the consumers should be made. Third, consumers generally worried about residual pesticide and intended to purchase irradiated foods if radioactivity was not retained in the foods. Therefore, food irradiation could be an alternative method to the use of pesticide Fourth, consumers pointed out that they wanted to extend shelf-life of milk and dairy foods, fish and seafood and to irradiate these foods. Therefore, research for the safety of irradiated foods should be continually conducted. finally, labeling for irradiated foods is needed to provide the information and to further increase public understanding. Especially, the labeling should show the definite reason why irradiation is being used. In conclusion, recently, under the circumstances that the commercial utilization of food irradiation and irradiation for the import and export products have been increased in many countries, many efforts are needed to improve the quality of irradiated foods, and prove the safety of them in Korea. In addition, consumer education for food irradiation should be given to help consumers to make decision for food purchase and use.
This study was investigated on properties and thermostability of gelatin-degrading proteinases in the fruit of Actinidia chinensis (kiwifruit) for the industrial application. Three gelatin-degrading proteinases (PI, PII and PIII) were detected from the pulp of fruits. The molecular weights of these proteinases, PI, PII and PIII, were approximately 220 kD, 51 kD, and 26 kD respectively, on the basis of gelatin-containing SDS-PACE. The optimum pH of these proteinases ranged from 2.0 to 5.0 with a maximal activity at pH 4.0. These proteinases had a high sensitivity to E-64 and iodoacetate which are cysteine protease inhibitors, and required DTT, cysteine, and $\beta$-mercaptoethanol for their activities which are stimulators for cysteine proteases. These results indicate that these proteinases are cysteine proteinases and the proteinase PIII is actinidin (EC 3.4.22.14), based on the molecular weight and/or susceptibility against proteinase inhibitors. These proteinases were strongly activated by $Ca^{2+}$, $Mg^{2+}$ and $Mn^{2+}$, whereas strongly inhibited by Zn$^{2+}$ and Hg$^{2+}$. However, these proteinases have slightly different susceptibility against other cations ($Ca^{2+}$, $Cu^{2+}$, $Al^{3+}$, $Ca^{3+}$. The temperature stability of proteinase PIII was more stable than proteinases PI and PII. Moreover, proteinase PIII remained stable below $50^{\circ}C$ for 48hr, showing the residual activity above 75% of the enzyme activity.
