Stability-enhanced mutants, H44, 11-94, 5A2-84, and F8, of L-threonine aldolase(L-TA) from Streptomyces coelicolor A3(2)(SCO1085) were isolated by an error-prone PCR followed by a high-throughput screening. Each of these mutant, had a single amino acid substitution: H177Y in the H44 mutant, A169T in the 11-94 mutant, D104N in the 5A2-84 mutant and F18I in the F8 mutant. The residual L-TA activity of the wild-type L-TA after a heat treatment for 20 min at $60^{\circ}C$ was only 10.6%. However, those in the stability-enhanced mutants were 85.7% for the H44 mutant, 58.6% for the F8 mutant, 62.1% for the 5A2-84 mutant, and 67.6% for the 11-94 mutant. Although the half-life of the wild-type L-TA at $63^{\circ}C$ was 1.3 min, those of the mutant L-TAs were longer: 14.6 min for the H44 mutant, 3.7 min for the 11-94 mutant, 5.8 min for the 5A2-84 mutant, and 5.0 min for the F8 mutant. The specific activity did not change in most of the mutants, but it was decreased by 45% in the case of mutant F8. When the aldol condensation of glycine and 3,4-dihydroxybenzaldehyde was studied by using whole cells of Escherichia coli containing the wild-type L-TA gene, L-threo-3,4-dihydroxyphenylserine(L-threo-DOPS) was successfully synthesized with a yield of 2.0 mg/ml after 20 repeated batch reactions for 100 h. However, the L-threo-DOPS synthesizing activity of the enzyme decreased with increased cycles of the batch reactions. Compared with the wild-type L-TA, H44 L-TA kept its L-threo-DOPS synthesizing activity almost constant during the 20 repeated batch reactions for 100 h, yielding 4.0 mg/ml of L-threo-DOPS. This result showed that H44 L-TA is more effective than the wild-type L-TA for the mass production of L-threo-DOPS.
The purpose of this study was to estimate genetic parameters for productive traits in Duroc breed. In this study, 40,657 records for productive traits and the pedigree data of 47,974 families were collected from 41 farms registered at the Korean Animal Improvement Association (KAIA) from 2004 to 2011. The REMLf90 program was used to analyze a multiple traits animal model with fixed effects of sex, contemporary group, parity and age at the end of the test as covariate and random effects of animal and residual error. The heritabilities of days to 90 kg (D90KG), average daily gain (ADG), backfat thickness (BF) and eye muscle areas (EMA) were estimated to be 0.334, 0.340, 0.335, and 0.200, respectively. The genetic correlation coefficients were -0.992 between D90KG and ADG, -0.142 between ADG and BF, -0.361 between ADG and EMA, and -0.243 between BF and EMA. Conversely, positive genetic correlations for D90KG with BF and EMA were 0.13 and 0.36, respectively.
Recently, non-destructive stress measurement method using magnetic anisotropy sensor has been applied to the construction site such as steel bridges and steel pipes. In addition, steel rib used in the tunnel construction site was found to be possible to measure the stress by non-destructive method. In this study, steel loading experiments using magnetic anisotropy sensor developed in Japan and strain gauges were conducted to derive stress sensitivity curve for domestic steel SS400. Also, additional steel loading experiments and numerical analysis were performed for evaluation of applicability for non-destructive stress measurement method using magnetic anisotropy sensor. As a result of this study, stress sensitivity curves for domestic steel SS400 were derived using output voltage measured by magnetic anisotropy sensor and average of stress measured by strain gauges depending on the measurement location. And as a result of comparing additional steel loading experiments with the numerical analysis, error level of magnetic anisotropy sensor is around 20MPa. When considering the level of the yield stress(245MPa) of steel, in case of using magnetic anisotropy sensor in order to determine the stress status of steel, it has sufficient accuracy in engineering. Especially, magnetic anisotropy sensor can easily identify the current state of stress which considers residual stress at steel structure that stress measurement sensor is not installed, so we found that magnetic anisotropy sensor can be applied at maintenance of steel structure conveniently.

Two crystal structures of fully dehydrated silver and potassium exchanged zeolite A, stoichiometries of $Ag_{9.3}K_{{2.7}^-}A$ (${\alpha}$ = 12.282(2) ${\AA}$) and $Ag_{10.7}K_{{1.3}^-}{\AA}$ (${\alpha}$ = 12.287(2) A) per unit cell, have been determined from 3-dimensional x-ray diffraction data gathered by counter methods. All structures were solved and refined in the cubic space group Pm3m at 21(1)$^{\circ}C$ . The crystals of $Ag_{9.3}K_{{2.7}^-}A$ and $Ag_{10.7}K_{{1.3}^-}A$ were prepared by flow method using exchange solutions in which mole ratios of $AgNO_3$ and $KNO_3$ were 1:10 and 1:5, respectively, with total concentration of 0.05M. The structures of the dehydrated $Ag_{9.3}K_{{2.7}^-}A$ and $Ag_{10.7}K_{{1.3}^-}A$ were refined to yield the final error indices $R_1$ = 0.037 and $R_2$ = 0.040 with 321 reflections, and $R_1$ = 0.042 and $R_2$ = 0.043 with 371 reflections, repectively, for which I > 3${\sigma}$(I). In both structures, eight $Ag^+$ ions are found nearly at 6-ring centers and each $Ag^+$ ion is nearly in the (1 1 1) plane at its O(3) ligands. The 8-ring sites are preferentially occupied by $K^+$ ions in both structures. 1.3 and 1.7 reduced silver atoms per unit cell were found inside of sodalite units of $Ag_{9.3}K_{{2.7}^-}A$ and that of $Ag_{10.7}K_{{1.3}^-}A$, respectively. These reduced silver species were presumably formed from the reduction of $Ag^+$ ions by oxide ions of residual water molecule or of the zeolite framework. These two crystals may be presented as hexasilver cluster in 21.7% and 28.3% of sodalite unit cells for $Ag_{9.3}K_{{2.7}^-}A$ and $Ag_{10.7}K_{{1.3}^-}A$, repectively.
The purpose of this study is to suggest how to investigate the reliability of the assessment, which consists of items generated by automatic item generation using empirical example data. To achieve this, we analyzed the illustrative assessment data by applying the multivariate generalizability theory, which can reflect the design of responding to different items for each student and multiple error sources in the assessment score. The result of the G-study showed that, in most designs, the student effect corresponding to the true score of the classical test theory was relatively large after residual effects. In addition, in the design where the content domain was fixed, the ranking of students did not change depending on the item types or items. Similarly, in the design where the item format was fixed, the difficulty showed little variation depending on the content domains. The result of the D-study indicated that the original assessment data achieved a sufficient level of reliability. It was also found that higher reliability than the original assessment data could be obtained by reducing the number of items in the content domains of operation, geometry, and probability and statistics, or by assigning higher weights to the domains of letters and formulas, and function. The efficient measurement conditions presented in this study are limited to the illustrative assessment data. However, the method applied in this study can be utilized to determine the reliability and to find efficient measurement conditions for the various assessment situations using automatic item generation based on measurement traits.
Partitioning tracer method is a useful tool to characterize large domains of the aquifers contaminated with light nonaqueous phase liquids (LNAPLs). Sorption of the partitioning tracers to the organic matter content of soil can potentially influence the efficacy of partitioning tracer method. LNAPL-water partitioning coefficients of tracers ($K_{nw}$), measured by static method, showed linear relationship. Sorption isotherm tests were conducted to evaluate the sorption capacity of the soils packed in the columns and the results were appropriately represented by Freundlich sorption isotherm. The sorption of tracers proportionally increased with the increase of the organic matter content of the soil. Laboratory experiments were conducted in four columns each packed with soils of different organic matter contents to determine the potential interference effects of sorption to soil organic matter content and correction factors for the errors in estimation of LNAPLs by partitioning tracer method. Though there were no contaminants added, breakthrough curves from columns packed with mixture of Jumunjin standard sand and organic matter showed separation of tracers. Columns were then contaminated to residual saturation with kerosene and breakthrough curves were obtained. The results show that sorption of tracers to soil organic matter leads to an increase in the retardation factor (R) and hence, to an overestimation of the saturation of LNAPLs. A relation between the percentage of organic matter content and the corresponding percentage error in the estimation of NAPLs has been developed.
The aim of this study is to present a graphical method in order to evaluate stages in shrinkage cracking. Firstly, the distribution of crack openings is established by sorting the openings of individual cracks in the soil cracking system. Secondly, it is normalized in a range of 0 to 1 to obtain the normalized crack opening distribution. Thirdly, three S-shape curve models introduced by Brooks and Corey(1964), Fredlund and Xing(1994) and van Genuchten(1980) are chosen to fit the normalized crack opening distribution using a curve fitting method. The accuracy of fitting which is described through fitting parameters by the van Genuchten equation is much higher than that by the Brooks and Corey equation and slightly higher than that by the Fredlund and Xing equation; thus the van Genuchten model is used. Finally, the stages of shrinkage cracking are graphically evaluated by drawing three separate straight lines corresponding to three linear parts of the fitted normalized crack opening distribution. The proposed method is tested with different sample thicknesses. The measured data are fitted by the selected model with the fairly high regression coefficient and small root mean square error. The results show graphically that shrinkage cracking comprises three stages; namely, primary, secondary and residual stages. Subsequently, the ranges of evaluated crack opening for each of these stages are presented.

Objective: This study was aimed to estimate the genetic parameters, including genetic and phenotypic correlations, of milk yield, lactation curve traits and milk composition of Thai dairy cattle from three government research farms. Methods: The data of 25,789 test-day milk yield and milk composition records of 1,468 cattle from lactation 1 to 3 of Holstein Friesian (HF) and crossbred HF dairy cattle calved between 1990 and 2015 from three government research farms in Thailand were analysed. 305-day milk yield was estimated by the Wood model and a test interval method. The Wood model was used for estimating cumulative 305-day milk yield, peak milk yield, days to peak milk yield and persistency. Genetic parameters were estimated using linear mixed models with herd, breed group, year and season of calving as fixed effects, and animals linked to a pedigree as random effects, together with a residual error. Univariate models were used to estimate variance components, heritability, estimated breeding values (EBVs) and repeatability of each trait, while pairwise bivariate models were used to estimate covariance components and correlations between traits in the same lactation and in the same trait across lactations. Results: The heritability of 305-day milk yield, peak milk yield and protein percentage have moderate to high estimates ranging from 0.19 to 0.45 while days to peak milk yield, persistency and fat percentage have low heritability ranging from 0.08 to 0.14 in lactation 1 cows. Further, heritability of most traits considered was higher in lactation 1 compared with lactations 2 and 3. For cows in lactation 1, high genetic correlations were found between 305-day milk yield and peak milk yield (0.86±0.07) and days to peak milk yield and persistency (0.99±0.02) while estimates of genetic correlations between the remaining traits were imprecise due to the high standard errors. The genetic correlations within the traits across lactation were high. There was no consistent trend of EBVs for most traits in the first lactation over the study period. Conclusion: Both the Wood model and test interval method can be used for milk yield estimates in these herds. However, the Wood model has advantages over the test interval method as it can be fitted using fewer test-day records and the estimated model parameters can be used to derive estimates of other lactation curve parameters. Milk yield, peak milk yield and protein percentage can be improved by a selection and mating program while days to peak milk yield, persistency and fat percentage can be improved by including into a selection index.
We investigated the effect of spectral fitting wavelength interval variations and selection of absorption cross-section on the sulfur dioxide slant column density (SCD) retrievals from the scattered sunlight observation using a UV-Vis hyperspectral instrument. The sulfur dioxide slant column densities were retrieved from the combinations of multiple spectral fitting intervals and absorption cross-sections. The observation was carried out at the site 0.53 km away from a combustion site located in Gimhae from December 1, 2023, to January 23, 2024. The radiances were obtained on the line of measurement sight toward the stack of the combustion facility. The best spectral fitting intervals were found to be from 305.7 to 321.1 nm. In terms of the absorption cross-section dependency, the SO2 (293 K), O3 (223 K, 243 K) show the best spectral fitting for the observed radiances with both the smallest fitting residual and SCD error. The effects of the fitting interval and cross sections found in this study can be useful information for improving SO2 retrievals based on UV hyperspectral measurements.

This study was conducted to derive the optimal estimation equations for deriving the green and dry weights of Larix kaempferi (Japanese larch) and Pinus rigida (Rigida pine), which are major coniferous tree species in South Korea. The equations were then used to develop weight tables. Table development began with the sampling of 150 L. kaempferi and 90 P. rigida trees distributed throughout the national scale, after which green weights were measured on-site. Samples from each stand were then collected, and their dry weights were measured in a laboratory. The equation used to calculate green and dry weights was divided into a one-variable formula that uses only the diameter at breast height (DBH) and a two-variable equation that employs DBH and height. The equations used to estimate the green and dry weights of logs were divided into one- and two-variable equations using DBH. Statistical data, such as the fitness index (FI), root mean square error, standard error of estimation, and residual diagram, were used to verify the suitability of the estimation equations. Applicability was examined by calculating weights using the derived optimal equations. The equation W = bD+cD2 was used in measurements involving only DBH, whereas the equation W = aDbHc was employed in cases involving both diameter and height at breast height. The FI of W = bD+cD2 was 0.91, while that of W = aDbHc was 0.95, both of which are high values. With these estimation formulas, weight tables for the green and dry weights of L. kaempferi and P. rigida were prepared and compared with weight tables created 20 years ago. The green and dry weight tables of both species were larger.
