Interspecies somatic cell nuclear transfer (iSCNT) is a useful method to preserve endangered species and to study the reprogramming event of a nuclear donor cell by the oocyte. Although several studies of iSCNT using murine cells and bovine oocytes have been reported, the development of murine-bovine iSCNT embryos beyond the 8-cell stage has not been successful. In this paper, we examined the developmental potential of embryos reconstructed with a murine embryonic fibroblast as the nuclear donor and a bovine oocyte as the cytoplasm recipient. The reconstructed embryos were cultured in CZB (murine medium) or CR1aa (bovine medium). In addition, for the development of a murine-bovine iSCNT blastocyst, the antioxidant ${\beta}$-mercaptoethanol (${\beta}ME$) was supplemented to CR1aa medium. Furthermore, to verify the mouse genome activation in murine-bovine iSCNT embryos, RT-PCR analysis of murine Xist was performed. The development of the murine-bovine iSCNT embryos cultured in CR1aa was significantly higher than that in CZB (p<0.05). With respect to the effect of BME on the development of the murine-bovine iSCNT blastocyst, addition of BME produced a significant increase in blastocyst development (p<0.05). Karyotype analysis confirmed that the reconstructed embryos were derived from murine cells (40XX). The Xist gene was gradually increased from the 8-cell stage to the blastocyst stage. This is the first report of blastocyst development of iSCNT embryos derived from murine somatic cells and bovine oocytes. These results demonstrate that bovine cytoplasm can support the development of later stages of a preimplantation embryo from murine-bovine iSCNT.

Because of the properties of software such as invisibility, complexity, and changeability, software configuration management (SCM) for software artifacts generated during software life-cycle has been used for guarantee of the quality of the software. However, the existing SCM system has only focused on code artifacts and software development document artifacts such as Software Requirements Specification (SRS), Software Design Description (SDD), and Software Test Description (STD). Moreover, software research-oriented project comes out late the code artifacts and the software development document artifacts. Therefore, there is a need for trace and management of software research document artifacts composed of highly abstracted non-functional requirements like 'the purpose of the project', 'the objectives', and 'the progress' before generation of the code artifacts and the software development document artifacts for a long time. Nevertheless, the existing SCM system cannot trace and manage them. In this paper, we propose content-based configuration management system comprised of the relevance link generation phase and content-based testing phase to trace and manage them. The preliminary application results show applicability and feasibility of the proposed system.

Medicinal plants resources are becoming important assets since their usages have been expanded to the development of functional foods for human health, natural cosmetics, and pharmaceutical industries. However, names are different from each country and their phylogenetic origins are not clear. These lead consumers to be confused. In particular, when they are morphologically similar and distributed as dried roots, it is extremely difficult to differentiate their origins even by specialists. Recently, molecular markers have been extensively applied to identify the origin of many crops. In this review, we tried to overview the current research achievements for the development of suitable 'origin identification' regarding to the differentiation of Angelica species. Furthermore, more advanced techniques including amplification genome based marker analyses are also discussed for their practical applications in the authentication of particular medicinal plant in Angelica species.

Dissolved oxygen is one of the most important factors controlling growth in aquatic organisms. Hypoxia is generally defined as dissolved oxygen less than 2.8 mg $O_2/L$ (equivalent to 2 mL $O_2/L$ or 91.4 mM). Therefore, hypoxia zone can cause a serious problem in marine ecosystem. In this study, to investigate embryotoxic (fertilization and embryo development rates) effects of hypoxia on sea urchin Strongylocentrotus nudus were exposed to dissolved oxygen levels of 7.6 mg $O_2/L$ (normoxia) and 1.8 mg $O_2/L$ (hypoxia) for 2 days at $15^{\circ}C$ and 33 ‰. Also, Expression levels of stress related gene (HSP70) and antioxidant related gene (glutathione reductase) in the sea urchins exposed to hypoxia were confirmed by Immunoblotting and RT-PCR analysis. In results, we showed that developmental rates were dramatically reduced in hypoxia condition. Molecular analysis demonstrated that higher HSP70 (5.5 fold) and glutathione reductase gene (2.79 fold) were present in the sea urchin exposed to hypoxia. Our results suggested that hypoxia can cause the abnormal development and elicits a stress and antioxidant response on sea urchin.
The bio-molecules involved in defense mechanisms can be used as efficient biomarkers for physiological changes in organisms caused by both of internal and external stress. Thus, the expression level of genes which encoding such molecules serve as critical 'early warning system' for environmental assessment as well as health diagnosis of biological organisms. In this study, Cytochrome P450, Heat shock protein 90, Ubiquitin and ${\beta}$-actin gene were isolated for the first time from a red alga Gracilaria textorii. The quantitative differential gene expression analyses of three genes, GteCYP1A, GteHsp90 and Gte-UB, were carried out in G. textorii sporophytes collected from two different localities, polluted Sujeong (Masan, Korea) and potentially unpolluted Danggeum (Daemaemuldo Is., Korea). The transcripts of all three tested genes were highly expressed in the Sujeong population. The results suggest: 1) the Sujeong site was more polluted than the Danggeum site; 2) G. textorii could be applicable to marine environment monitoring in coastal regions.

The use of groundwater in Korea has increased in recent years to the point where its extraction is restricted in times of drought. This work models the groundwater pumping field as a confined aquifer in a simplified simulation of groundwater flow. It proposes a genetic algorithm to maximize groundwater development using a conceptual model of a steady-state confined aquifer. Solving the groundwater flow equation numerically calculates the hydraulic head along the domain of the problem; the algorithm subsequently offers optimized pumping strategies. The algorithm proposed here is designed to improve a prior initial groundwater management model. The best solution is obtained after 200 iterations. The results compare the computing time for five simulation cases. This study shows that the proposed algorithm can facilitate better groundwater development compared with a basic genetic algorithm.

Shigella sonnei is a causal agent of fever, nausea, stomach cramps, vomiting, and diarrheal disease. The present study describes a quantitative polymerase chain reaction (qPCR) assay for the specific detection of S. sonnei using a primer pair based on the methylase gene for the amplification of a 325 bp DNA fragment. The qPCR primer set for the accurate diagnosis of Shigella sonnei was developed from publically available genome sequences. This quantitative PCR-based method will potentially simplify and facilitate the diagnosis of this pathogen and guide disease management.
The blood-epididymis barrier (BEB) forms a unique microenvironment that is crucial for the maturation, protection, transport, and storage of spermatozoa in the epididymis. To characterize the function of tight junctions (TJs), which are constitutive components of the BEB, we determined the expression and localization of TJ proteins such as zonula occludens (ZO)-1, 2, and 3, occludin, and claudin3 (Cldn3) during postnatal development in the goat epididymis. To assess the expression patterns of TJ proteins in immature (3 months of age) and mature (14 months of age) goat epididymides, two different experimental methods were used including immunofluorescence labeling and western blotting. We show that, ZO-1, 2, and 3, and occludin, were strictly expressed and localized to the TJs of the goat epididymis, whereas Cldn3 was present in basolateral membranes as well as TJs. All TJ proteins examined were more highly expressed in the immature epididymis compared to levels in mature tissue. In conclusion, our study indicates that at least five TJ proteins, namely ZO-1, ZO-2, ZO-3, occludin, and Cldn3, are present in TJs, and the expression strength and pattern of TJ proteins tend to be age dependent in the goat epididymis. Together, these data suggest that the distinct expression patterns of TJ proteins are essential for regulating components of the luminal contents in the epididymal epithelium and for forming adequate luminal conditions that are necessary for the maturation, protection, transport, and storage of spermatozoa in the goat epididymis.

There are the national science and technology standard classification system used in Korea, the classification according to the standard classification system for educational organization units, and the Korean standard education classification by the National Statistical Office. It is not suitable for calculation or evaluation, and classification is still mixed depending on the purpose of use. Therefore, in this study, the current status of academic standard classification, issues related to the standard classification system such as research foundation associations and research institutes, and issues related to the academic standard classification through the analysis of existing prior research issues, etc. As a result of the research, first, it is necessary to maintain and strengthen the linkage of the academic classification system, such as maintaining the linkage between the relevant departmental classification systems and strengthening the linkage with the relevant classification system, as a result of analysis of major issues in the academic standard classification system, and the systematic improvement cycle of the revision process and management system and settings are required.
