501 preschool children from 4 to 6 years were examined for their salivary reductase activity and caries experience by Resazurin Disc Test and dental examination respectively. We asked the parents about their children's oral hygiene habits, between-meal eating habits, and physical exercise habits by the questionnaire. Toothbrushing frequency had negative relation to salivary reductase activity and caries experience. Caries experience was low when parents did toothbrushing for children, when teeth were brushed at bedtime, and when fluoride toothpastes were used. Salivary reductase activity and caries experience were high in bread & cookies group, chocolates & candies group, milk & soft drink group, and fruits & vegetables group in order. Caries experience was high in case of irregular between-meal eating. Sweet food eating frequency had positive relation to caries experience. Caries activity was low in case of eating homemade non-sweet between-meals. Salivary reductase activity and caries experience were low when gum-chewing frequency was high. Salivary reductase activity and caries experience were high when the amount of physical exercise was low.

The purpose of this study was to do an epidemiological survey of the salivary reductase activity and dental caries prevalence of the large group of preschool children and analyze the validity of the salivary reductase activity test as a caries activity test. One thousand and forty-four preschool children were examined for caries experience and salivary reductase activity by dental survey and the Resazurin Disc Test. 39.8% of children showed low salivary reductase activity, 39.4% showed middle, and 20.8% showed high. Salivary reductase activity increased as age increased. All indexes of caries experience were high when salivary reductase activity was high. There was positive relation between salivary reductase activity and caries experience. The salivary reductase activity of children with no caries experience was lower than that of children with caries experience. The salivary reductase activity of children with low caries experience was lower than that of children with high caries experience. It seemed that the salivary reductase activity test had basic validity as a caries activity test. However, the test's ability to select the small risk group of high caries susceptibility should be enhanced.

The purpose of this study was to compare the caries activity and cariogenic factors in children in different residence areas. The subjects of the study were 268 kindergarten children who were selected from the downtown area(D), apartment area(A), suburban area(S), and rural area(R) in and around the city of Iksan, Korea. Caries activity was measured as past caries experience (dft index and dfs index) and salivary reductase activity (Resazurin Disc Test scores). Cariogenic factors were investigated by a questionnaire. The purpose of this study was to investigate. the effects of the lactation of human or bovine milk and the method of using nursing bottles on the salivary invertase test scores and deciduous caries experience of children. 328 preschool children (boys 178, girls 150) in Iksan city were selected for this study. The parents of the children were asked to answer the questionnaire about the lactation of human or bovine milk and the time of using nursing bottles. The data were obtained from the salvary invertase activity score,(Resazurin Disc Test) deciduous caries experience and the questionnaire. The result were as follows. The invertase activity and caries experience of the human milk higher than that of the bovine milk group(P<0.05). There were significant positive relationships between the lactation period and the invertase activity and between the lactation period and the caries experience(P<0.05). Caries experience had a tendency to increase as the time of using nursing bottles increase (P>0.05).
Human carbonic anhydrase (CA) isozyme II has been used as protein target for disorder treatment including glaucoma. Current clinically used sulfonamide-based CA inhibitors can induce side effects, and so alternatives are required. This study aimed to investigate a natural CA inhibitor from Murraya paniculata. The previously developed yeast-based assay was used to screen 14 compounds isolated from M. paniculata and identified by NMR analysis for anti-human CA isozyme II (hCAII) activity. Cytotoxicity of the compounds was also tested using the same yeast-based assay but in a different cultivation condition. Two flavonoid candidate compounds, 5, 6, 7, 8, 3', 4', 5'-heptamethoxyflavone (4) and 3, 5, 7, 8, 3', 4', 5'-heptamethoxyflavone (9), showed potent inhibitory activity against hCAII with a minimal effective concentration of 10.8 and 21.5 μM, respectively, while they both exhibited no cytotoxic effect, even at the highest concentration tested (170 μM). The results from an in vitro esterase assay of the two candidates confirmed their hCAII inhibitory activity with IC50 values of 24.0 and 34.3 μM, respectively. To investigate the potential inhibition mechanism of compound 4, in silico molecular docking was performed using the FlexX and SwissDock software. This revealed that compound 4 coordinated with the Zn2+ ion in the hCAII active site through its methoxy oxygen at a distance of 1.60 Å (FlexX) or 2.29 Å (SwissDock). The interaction energy of compound 4 with hCAII was -13.36 kcal/mol. Thus, compound 4 is a potent novel flavonoid-based hCAII inhibitor and may be useful for further anti-CAII design and development.

Scutellaria pinnatifida A. Hamilt. (Lamiaceae) is an endemic Turkish herb. This plant is also endemic to Iran, and grows abundantly in other central and western Asian countries. Several species of the Scutellaria are known for their traditional uses in the treatment of hypertension, arteriosclerosis, inflammatory diseases, hepatitis, allergy, cancer and diarrhoea. Free-radical-scavenging property, antibacterial activity and brine shrimp toxicity of the n-hexane, dichloromethane (DCM) and methanol (MeOH) extracts of S. pinnatifida were assessed using the 2,2-diphenyl-1-picryl-hydrazyl (DPPH) assay, the resazurin microtitre plate based assay, and the brine shrimp lethality assay, respectively. The DCM and MeOH extracts exhibited free-radical-scavenging property, with the $RC_{50}$ values of 0.362 and 0.127 mg/ml, respectively. Among the solid-phase extraction fractions of the MeOH extract, the 50% aqueous-MeOH fraction showed the highest level of free-radicalscavenging activity ($RC_{50}$ = 0.039 mg/ml). While the DCM extract showed low level of antibacterial activity against Bacillus subtilis and ampicillin-resistant Escherichia coli, the MeOH extract was active against B. cereus, B. subtilis, E. coli and ampicillin-resistant E. coli. However, the minimum inhibitory concentrations (MIC) of the MeOH extract against these bacterial strains were >10 mg/ml. None of the extracts showed any significant toxicity towards brine shrimps ($LD_{50}$ = > 1.00 mg/ml).
PURPOSE. The aim of this in vitro study was to investigate the adhesion of initial colonizer, Streptococcus sanguis, on resin, titanium and zirconia under the same surface polishing condition. MATERIALS AND METHODS. Specimens were prepared from Z-250, cp-Ti and 3Y-TZP and polished with $1 {\mu}m$ diamond paste. After coating with saliva, each specimen was incubated with Streptococcus sanguis. Scanning electron microscope, crystal violet staining and measurement of fluorescence intensity resulting from resazurin reduction were performed for quantifying the bacterial adhesion. RESULTS. Surface of resin composite was significantly rougher than that of titanium and zirconia, although all tested specimens are classified as smooth. The resin specimens showed lower value of contact angle compared with titanium and zirconia specimens, and had hydrophilic surfaces. The result of scanning electron microscopy demonstrated that bound bacteria were more abundant on resin in comparison with titanium and zirconia. When total biofilm mass determined by crystal violet, absorbance value of resin was significantly higher than that of titanium or zirconia. The result of relative fluorescence intensities also demonstrated that the highest fluorescence intensity was found on the surface of resin. Absorbance value and fluorescence intensity on titanium was not significantly different from those on zirconia. CONCLUSION. Resin specimens showed the roughest surface and have a significantly higher susceptibility to adhere Streptococcus sanguis than titanium and zirconia when surfaces of each specimen were polished under same condition. There was no significant difference in bacteria adhesion between titanium and zirconia in vitro.
LIVE/DEAD$^{(R)}$ BacLight$^{TM}$ and alamarBlue$^{(R)}$ are fluorescent materials used for the enumeration of live and dead bacteria. LIVE/DEAD$^{(R)}$ BacLight$^{TM}$ is generally used for confocal microscopy applications to differentiate live from dead bacteria in a biofilm or planktonic state. AlamarBlue$^{(R)}$ has also been used widely to assay live and dead bacteria in a planktonic state. Whilst these materials are successfully utilized in experiments to discriminate live from dead bacteria for several species of bacteria, the application of these techniques to oral bacteria is limited to the use of LIVE/DEAD$^{(R)}$ BacLight$^{TM}$ in biofilm studies. In our present study, we assessed whether these two methods could enumerate live and dead oral bacterial species in a planktonic state. We tested the reagents on Streptococcus mutans, Streptococcus sobrinus, Porphyromonas gingivalis, Aggregatibacter actinomycetemcomitans and Enterococcus faecalis and found that only LIVE/DEAD$^{(R)}$ BacLight$^{TM}$ could differentiate live from dead cells for all five of these oral strains. AlamarBlue$^{(R)}$ was not effective in this regard for P. gingivalis or A. actinomycetemcomitans. In addition, the differentiation of live and dead bacterial cells by alamarBlue$^{(R)}$ could not be performed for concentrations lower than $2{\times}10^6$ cells/ml. Our data thus indicate that LIVE/DEAD$^{(R)}$ BacLight$^{TM}$ is a more effective reagent for this analysis.
To probe the subclinical mastitis in a herd of cows in Chonnam district, the electrical conductivity(EC) of 825 foremilk samples were measured for 2 years. Normal (n=487) and mastitic(n=110) foremilk samples were classified by the California mastitis test (CMT) and direct somatic cell count(DSCC) and investigated the relations between the changes of the EC value and the calving history, age, days of postpartum, estrus and causative organism isolated. Obtained results are summarized as follows. 1. In the normal foremilk samples, positive correlation, though not significant, was found between the EC value and calving history (r=0.573) and age (r=0.247). 2. In the normal foremilk samples, the EC values were lowered at 30~120 days of postpartum through the whole lactation period and revealed a tendencies to higher values following the day of postpartum increased untill to the drying off (r=0.823), and the days of postpartum was recognized as one of a influencing factor on the EC value(p<0.05). 3. In the mastitic foremilk samples, significant correlation between EC value and resazurin reduction test (RRT) were observed (r=0.904, p<0.05). 4. In the mastitic foremilk samples, EC values were obtained in the E. coli infection as 63.9mM-NaCl, in the Streptococcus spp. infection as 60.5mM-NaCl and in the Staphylococcus spp. infection as 57.0mM-NaCl. 5. At day 0 of estrus, the mean EC values of normal and mastitic foremilk samples were 41.2mM-NaCl and 68.3mM-NaCl respectively and the EC value of day 0 of estrus was higher than that of days before and after estrus.

Background: Mycobacterium tuberculosis (Mtb) is resistant to the β-lactam antibiotics due to a non-classical transpeptidase in the cell wall with β-lactamase activity. A recent study showed that meropenem combined with clavulanate, a β-lactamase inhibitor, was effective in multidrug-resistant (MDR) and extensively drug-resistant (XDR) tuberculosis (TB). However, in Korea, clavulanate can only be used as drugs containing amoxicillin. In this study, we investigated the susceptibility and genetic mutations of drug-resistant Mtb isolates to amoxicillin-clavulanate and meropenem-clavulanate to improve the diagnosis and treatment of drug-resistant TB patients. Methods: The minimum inhibitory concentration (MIC) of amoxicillin-clavulanate and meropenem-clavulanate was examined by resazurin microtiter assay. We used 82 MDR and 40 XDR strains isolated in Korea and two reference laboratory strains. Mutations of drug targets blaC, blaI, ldtA, ldtB, dacB2, and crfA were analyzed by polymerase chain reaction and DNA sequencing. Results: The MIC90 values of amoxicillin/clavulanate and meropenem/clavulanate in drug-resistant Mtb isolates were 64/2.5 and 16/2.5 mg/L, respectively. Gene mutations related to amoxicillin/clavulanate and meropenem/clavulanate resistance could not be identified, but T448G mutation was found in the blaC gene related to β-lactam antibiotics' high susceptibility. Conclusion: Our results provide clinical consideration of β-lactams in treating drug-resistant TB and potential molecular markers of amoxicillin-clavulanate and meropenem-clavulanate susceptibility.
