Effects of Origins of Soybean Meal on Growth Performance, Nutrient Retention and Excreta Microflora of Broilers

  • An experiment was conducted to determine the effects of origins of soybean meal (SBM) on growth performance, nutrients and energy retention and fecal microflora in broilers. The SBM originating from Korea, Brazil and India were used. A total of 480 broiler chicks (average initial BW, 41.8 g) were randomly allotted to 6 treatments. Each treatment had 4 replicate pens with 20 chicks per pen. Birds were fed diets containing SBM originated from Korea (domestic SBM), Brazil or India (imported SBM) and the diets were based on the total amino acid (TAA) or true digestible amino acid (TDAA). Experimental diets were fed in two phases, starter (d 0~21) and finisher (d 22~35). The overall weight gain, feed intake and FCR were better (P<0.05) in birds fed Korean SBM. During starter and finisher periods, birds fed diets containing Korean SBM had greater (P<0.05) retention of GE, DM, and CP (P<0.05) as compared with Birds fed diets containing SBM from India. In addition, diets formulated on TDAA basis had grater (P<0.05) CP retention during starter period and DM, GE and CP retention during finisher period. There were no differences (P>0.05) in the excreta microflora and overall feed cost per kg body weight gain in birds fed SBM from different origins; however, diets formulated on TDAA basis had lesser (P<0.05) feed cost per kg body weight gain in birds when compared with diets formulated on TAA basis. These results indicate that Korean SBM has better nutrients digestibility than SMB originated from Brazil and India, which contributes to the improved performance of broilers. In addition, better performance was obtained when diets were formulated on TIDAA basis.

  • The ability of neurotropic alpha herpesviruses to replicate within synaptically linked neurons has made these pathogens valuable tools for transneuronal analysis. Recent studies suggest that unique gene products expressed by genetically engineered strains of virus may permit the use of multiple strains in complex tracing paradigms. In the present study we have examined the invasiveness of two genetically engineered strains of the swine pathogen known as pseudorabies virus(PRV). The two strains were isogenic with the attenuated Bartha strain of PRV; in one strain a lacZ reporter gene was inserted into the gC locus (PRV-BaBlu; $4.75\times10^8pfu/ml$) contrained a PRV envelope glycoprotein gene that was absent in PRV-BaBlu. Simultaneous or temporally separated sequential injection of $4\mu\ell$ of each strain into the ventral wall of the stomach produced a predictale course of retrograde synaptic infection. The results were as follows: 1. PRV-BaBlu and PRV-D infected the dorsal motor nucleus of vagus nerve(DMV) and paraventricular nucleus(PVN). 2. Invasion and replication of PRV-D occured at a faster rate than the parental strain or PRV-BaBlu. 3. PRV-D was much more virulent than PRV-BaBlu or the parental strain. 4. Co-injection of PRV-D and PRV-BaBlu produced an infection that was more virulent than that produced by the parental strain (PRV-Bartha), 5. Neurons in DMV were permissive to co-infection with PRV-D and PRV-BaBlu when they were injected simultaneously into the same site. 6. Replication of PRV-BaBlu was compromised by prior infection of the same circuit with PRV-D. 7. Prior infection of neurons with PRV-D maked them resistant to infection with PRV-BaBlu.

해남 플럭스 타워 지점에서의 Advanced Microwave Scanning Radiometer E 토양수분자료의 검증 (Advanced Microwave Scanning Radiometer E Soil Moisture Evaluation for Haenam Flux Monitoring Network Site)

  • 본 연구에서는 원격탐사자료인 Advanced Microwave Scanning Radiometer E (AMSR-E) 토양수분자료의 시간적 변통성 검증을 위해 해남 플럭스 지점에서 관측된 2년간의 (2004년과 2006년) 토양수분자료를 이용하였다. 지상관측 토양수분자료에 비해 상대적으로 큰 공간해상도를 가진 원격탐사자료(AMSR-E)는 2년간의 비교/검증에서 지상관측토양수분자료와 시간적 상관성이 있는 것으로 파악되었으며, 차후에 보다 다양한 식생조건과 현장조건에서 심도 있는 검증연구가 이루어져야 할 것으로 판단된다. 이를 위해서는 보다 다양한 지역에서 현장측정이 이루어져야할 것으로 사료된다. 이를 통하여 보다 정확한 비교/검증 연구가 수행된다면, 원격탐사 토양수분자료의 활용성을 기대할 수 있으며 또한 수문기상학적 관점에서 지표면 대기의 상호작용에 대한 올바른 이해를 향상시킬 수 있을 것으로 판단된다.

Construction of a Transposon-mediated Baculovirus Vector Hanpvid and a New Cell Line for Expressing Barnase

  • In this study we developed the transposon-mediated shuttle vector 'Hanpvid', which composed of HaNPV (Heliothis armigera nuclear polyhedrosis virus) genomic DNA and a transposon cassette from Bacmid of Bac-to-Bac system. Hanpvid replicates in E. coli in the same way as Bacmid and retains infective function in cotton bollworm cells (Hz-AM1). Using Hanpvid we constructed a recombinant virus, which could infect Hz-AM1 cells and generate recombinant HaNPV (rHa-Bar) containing the barnase gene, a ribonuclease gene from Bacillus amyloliquefaciens. Since the expression vector carrying barnase gene cannot replicate in the absence of barstar, a specific inhibitor of barnase, we constructed a new cotton bollworm cell line (AM1-NB) using the marker rescue method. In AM1-NB barstar was integrated into the cellular chromosome to sustain the replication of rHa-Bar. To screen out recombinant HaNPV for potential use as biopesticide, Hz-AM1 and AM1-NB cell lines were infected with rHa-Bar, respectively. The results obtained indicate that Viral progenies in AM1-NB were 23 and 160 times greater than those in Hz-AM1 48 h and 72 h after infection, respectively. With additional insertion of the polyhedron gene from AcNPV (Autographa californica nuclear polyhedrosis virus) into the Hanpvid genome, rHa-Bar regained the polyhedron phenotype and its pest-killing rate greatly improved. Toxic analysis showed that the lethal dosages ($LD_{50}$) and the lethal time(s) ($LT_{50}$) of rHa-Bar were reduced by 20% and 30%, respectively, compared to wt-HaNPV in the third instar larvae of cotton bollworm. This study shows that in AM1-NB barnase can be effectively produced and used as pest-killing agent for the biological control of cotton pests.

Nucleotide Analysis of Phaffia rhodozyma DNA Fragment That Functions as ARS in Saccharomyces cerevisiae

  • The chromosomal DNA fragment from Phaffia rhodozyma CBS 6938 which is able to autonomously replicate in the yeast Saccharomyces cerevisiae was cloned on an integrative URA3 plasmid. Its minimal fragment exhibiting autonomously replicating activiy in the S. cerevisiae gave a higher frequency transformation efficiency than that found for centromere-based plasmid, and enabled extrachromosoma1ly stable transmission of the plasmids in one copy per yeast cell under non-selective culture condition. The 836-bp DNA element lacked an ORF and did not contain any acceptable match to an ARS core consensus. Sequence analysis, however, displayed a cluster of three hairpin-Ioop-sequences with individual $\triangle {G_{25}}^{\circ}C$ free energy value of -10.0, -17.5, and -17.0 kcal. $mor^{-l}$as well as a 9-bp sequence with two base pair mismatches to the S. cerevisiae/E. coli gyrase-binding site. This 836-bp sequence also included one 7-bp sequence analogous to the core consensus of centromeric DNA element III (CDEIII) of S. cerevisiae, but CDEIII-like 7 bp sequence alone did not give a replicative function in this yeast.

Improving Pneumovirus Isolation Using a Centrifugation and AZD1480 Combined Method

  • The isolation of respiratory viruses, especially from clinical specimens, often shows poor efficiency with classical cell culture methods. The lack of suitable methods to generate virus particles inhibits the development of diagnostic assays, treatments, and vaccines. We compared three inoculation methods, classical cell culture, the addition of a JAK2 inhibitor AZD1480, and centrifugation-enhanced inoculation (CEI), to replicate human respiratory syncytial virus (HRSV) and human metapneumovirus (HMPV). In addition, a combined method using AZD1480 treatment and CEI was used on throat swabs to verify that this method could increase virus isolation efficiency from human clinical specimens. Both CEI and AZD1480 treatment increased HRSV and HMPV genome replication. Also, the combined method using CEI and AZD1480 treatment enhanced virus proliferation synergistically. The combined method is particularly suited for the isolation of interferon-sensitive or slowly growing viruses from human clinical specimens.

Kinetic behavior of sophoricoside by gas chromatography/mass spectrometry in rats

  • Sophoricoside was isolated as the inhibitor of IL-5 bioactivity from Sophora japonica (Leguminosae). To develope as novel anti-allergic drug, kinetic study was performed in rats. Serum concentration of sophoricoside was measured by gas chromatography-mass spectrometry (GC/MS) in male Sprague-Dawley rat (250${\pm}$10g, n=5) after oral administration of sophoricoside (100mg/kg). The recovery of sophoricoside after extraction and concentration was above 95 % from rat serum. Between-day precision(relative standard deviation 2.2-2.8%) and within-day precision(2.0-12.1%) were determined from replicate analysis of a spiked control and incurred serum sample. The detection limits of sophoricoside in this serum was approximately 0.1 ng/mL. The Pharmacokinetic parameters were derived from the noncompartmental analysis. The C$\_$max/(3.56${\pm}$0.34 $\mu\textrm{g}$/mL) value for sophoricoside in male rat was observed at 7.6 h. The elimination half-life(t$\_$1/2/) of sophoricoside was approximately 4.47 h, the mean residence time (MRT) averaged 10.75 h, the total body clearance (Cl) averaged 0.0042 mL/min/kg. and the area under the serum concentration-time curve (AUC$\_$0-$\infty$/) was 24.93 $\mu\textrm{g}$$.$hr/mL.

Uncertainties in blast simulations evaluated with Smoothed Particle Hydrodynamics method

  • The paper provides an inside look into experimental measurements, followed by numerical simulations and their related uncertainties. The goal of the paper is to present findings related to blast loading and the handling of defects that are inherent in experiments. Very often it might seem that experiments are simplified reflections of real-life conditions. In most cases this is true, but there is a good reason for that. The more complex an experiment is, the larger the amount of uncertainties that can be expected. This especially applies when the blast loading of concrete is the subject of research. When simulations fail to reproduce the results of experimental measurements, it does not necessarily mean there is something wrong with the numerical model. The problem could be missing information. Put differently, the numerical simulation may lack information that seemed irrelevant with regard to the experiment. In the presented case, a reference simulation with a proven material model unexpectedly failed to replicate the results of an experiment where concrete slabs were exposed to blast loading. This resulted in a search for possible unknowns. When all of the uncertainties were examined, the missing information turned out to be the orientation of the charge to the concrete slab. Since the experiment was burdened with error, a sensitivity study had to take place so the influence of this factor could be better understood. The findings point to the fact that even the smallest defect during experiments must somehow be taken into account when designing numerical simulations. Otherwise, the simulations are not correlated to the experiments, but merely to some expectations.

Replacement of Yellow Maize with Pearl Millet (Pennisetum typhoides), Foxtail Millet (Setaria italica) or Finger Millet (Eleusine coracana) in Broiler Chicken Diets Containing Supplemental Enzymes

  • An experiment was conducted to study the performance of broilers chicks (2 to 42 d of age) fed diets containing pearl millet (PM, Pennisetum typhoides), foxtail millet (FOM, Setaria italica) or finger millet (FIM, Elusine coracana) totally replacing (w/w) yellow maize (YM) with and with out supplementing non-starch polysaccharide (NSP) hydrolysing enzymes at the rate of 0.5 g/kg diet. Enzyme preparation contained amylase 2,400 units, hemi-cellulase 5,400 units, cellulase 12,000 units, protease 2,400 units and beta-glucanase 106 units/g. Each diet was fed to eight replicates (five female Vencob broilers/replicate) housed in stainless steel battery brooders. The estimated metabolizable energy (ME) contents of YM, PM, FOM and FIM were FM (PM) were about 3,389, 2,736, 3,303 and 2,846 kcal/kg, respectively. Total replacement of YM with FOM did not influence the body weight gain, ready to cook yield, relative weights of giblet, liver, intestine, lymphoid organs (bursa and spleen) and length of intestine, antibody titers and livability at 42 d of age. But the food efficiency decreased significantly in FOM fed broilers compared those fed YM. Further, the fat content in thigh muscle reduced with FOM fed groups compared to those fed YM. The performance of broilers decreased significantly in PM and FIM fed broilers compared to those fed YM. The relative weights of giblet, gizzard and liver increased in FIM fed groups compared to those fed YM as the principal source of energy in broilers. Incorporation of NSP hydrolysing enzymes in commercial broiler diets improved the efficiency of feed utilization during starter phase but not at 42 d of age. The results thus indicate that yellow maize can be replaced in toto on weight basis in commercial broiler diets without affecting the performance. Supplementation of NSP hydrolysing enzymes was beneficial in enhancing feed utilization during the starter phase.

Effects of Supplemental Synthetic Amino Acids to the Low Protein Diets on the Performance of Growing Pigs

  • A total of 120 pigs $(L\;{\times}\;LW\;{\times}\;D)$ averaged 14.16 kg of body weight were reared under six dietary treatments to evaluate the effects of amino acid supplementation on their performances. Treatments were 1) 18% CP diet (control); 2) 15% CP+0.28% Lys (B, 15L); 3) B+0.06% Met (15LM); 4) B+0.13% Thr (15LT) ; 5) B+0.06% Met+0.13% Thr (15LMT); 6) B+0.06% Met+0.13% Thr+0.05% Trp (15LMTT). Each treatment had 4 replicates with 5 pigs per replicate. The daily weight gains were statistically similar in 15LMT, 15LMTT and control groups. Threonine and methionine supplementation resulted in improved growth performance and nutrient digestibilities, while tryptophan supplementation had little beneficial effect. However, the best feed conversion was found in the control group. Dry. matter and CP digestibilities were improved in the 15LMT and 15LMTT groups. Gross energy, crude fat and phosphorus digestibilities were not affected by the treatment. Among the 15% CP groups, nitrogen digestibility showed the tendency to be increased as the more synthetic amino acids were added. Dry matter and nitrogen excretions were significantly reduced by feeding low protein, amino acid fortified diets, while phosphorus excretion was not influenced. Essential amino acids digestibility was higher in 15LT, 15LMT and 15LMTT groups and lower in the control. The 15LT group showed the best lysine digestibility, and methionine digestibilities were higher in all treated groups than control one. Threonine digestibilities higher in 15LT 15LMT, and 15LMTT than the control. The result shows that threonine and methionine should be added to the diet containing 15% CP diet, fortified with lysine to get same performance of 18% CP diet.
