  • This experiment was carried out to investigate the effect of planting density on thegrowth and yield of Rehmannia glutinosa. A local variety was planted on the 100cm row, with different planting density of 20, 30 and 40 plants per $m^2$. Nitrogen, phosporus and potassium fertilizers were applied as 12, 12, and 16 kg /10a, respectively with compost of 1,000kg /10a. Plant height became shorter with increase in the planting density from 20 to 40 $plants\;/\;m^2$. But emergence date, leaf length and width were not significantly changed with planting density. As the planting density was increased, tuber diameter got slender and tuber number was decreased, but tuber length was not affected. The tuber yield was 793kg /10a at the planting density of 20 $plants\;/\;m^2$ and it increased 40% at 30 $plants\;/\;m^2$ and 45% at 40 $plants\;/\;m^2$ Regarding on the farmer's income, optimum planting density was estimated 30 $plants\;/\;m^2$.

  • This study was conducted to evaluate the appropriateness cultivation of R. glutinosa by plug seedling. Cultivation by direct seeding (DS) and plug seedling (PS) of R. glutinosa was conducted in each of the Pyeongchang and Eumseong areas. As a result, locally, for the Eumseong area, aerial parts in leaf length, leaf width and number of leaves per plant were better than for the Pyeongchang area. In the cultivation method, leaf width and number of leaves per plant were better with PS cultivation than with DS cultivation. Locally, rhizome yield of underground parts in the Eumseong area was better than that in the Pyeongchang area, but the rhizome yield with DS cultivation in the Eumseong area and PS cultivation in the Pyeongchang area were statistically the same. The fresh rhizome yield for DS in Eumseong, PS in Eumseong, DS in Pyeongchang and PS in Pyeongchang was 1,253 kg/10a, 1,376 kg/10a, 923 kg/10a and 1,256 kg/10a, respectively. Finally, the catalpol content for DS in Eumseong, PS in Eumseong, DS in Pyeongchang and PS in Pyeongchang was 3.67%, 2.03%, 2.96% and 1.60%, respectively. From these results, it can be seen that the R. glutinosa PS cultivation method can increase the rhizome yield.

  • The effects of soil properties and N, P and K application rates on the contents of inorganic constituents in the rhizomas of Ji-whang were investigated to know the condition for producing the better quality rhizoma of Ji-whang. Most of soil texture for Ji-whang cultivating belongs to sandy clay loam and sandy loam soils. The nitrogen contents in the rhizomas of Ji-whang was negatively correlated with the contents of organic matter and total nitrogen in surface soils, whereas positively correlated with the sand contents in subsoils. The calcium contents in the rhizomas of Ji-whang were negatively correlated with the sand contents, however, positively correlated with the contents of clay and organic matter in soils. The iron contents was positively correlated with the contents of clay, available phosphorus and exchangeable potassium in surface soils. Much more contents of the inorganic constituents in the leaf of Ji-whang was generally detected than that in the rhizoma of Ji-whang. The contents of iron and copper in the leaf was determined to be more than that in the rhizomas by ten times. In the results of field experiments, it is apparent that the nitrogen contents in the rhizoma and leaf increased with increasing N application rate more than 20kg/10a and 10kg/10a, respectively.

  • The current study was conducted to evaluate beneficial effects of a new formula (CWC-9) using four traditional Oriental medicinal herbs, Cynanchum wilfordii, Rehmannia glutinosa, Polygala tenuifolia, and Acorus gramineus, on hippocampal cells and memory function. To examine the neuroprotective effects of a new four-herb extract, cell viability, cytotoxicity, and reactive oxygen species (ROS) assays were performed in HT22 cells and behavioral tests (Morris water maze and passive avoidance retention), Western blot, and immunohistochemistry were performed in a mouse model of focal cerebral ischemia. In HT22 hippocampal cells, pretreatment with CWC-9 resulted in significantly reduced glutamate-induced cell death with suppression of ROS accumulation caused by glutamate. In a mouse model of focal cerebral ischemia, we observed significant improvement of spatial and short-term memory function by treatment with CWC-9. Phosphorylated p38 mitogen-activated protein kinases (MAPK) in hippocampus of ischemic mice were decreased by treatment with CWC-9, but phosphorylated phosphatidylinositol-3 kinase (PI3K) and cAMP response element binding protein (CREB) were significantly enhanced. By immunohistochemical analysis, we confirmed higher expression of phosphorylation of CREB in the hippocampal neurons of CWC-9 treated mice. These results suggest that new multi-herb formula CWC-9 mainly exerted beneficial effects on cognitive function through regulation of neuro-protective signaling pathways associated with CREB.

  • Objective : The 5-HMF was not index material suitable to do the quality control of Rehmanniae Radix Preparata. In this study, We estimated the changes of oligosaccharide contents in Rehmanniae Radix Preparata using high-performance anion-exchange chromatography with pulsed amperometric detection(HPAEC-PAD). Methods : The analysis of oligosaccharide was conducted by HPAEC-PAD with Carbopac PA1, $250{\times}4mm$, 5um, and Carbopac PA1 guard column. Column temperature was kept at $30^{\circ}C$. Elution was carried out at 1000 ${\mu}l/min$ with 70mM NaOH and the injection volume was $10{\mu}l$. Each component was detected by PAD. Results : Nine constituents were found from merchandising Rehmanniae Radix Preparata(MR), while seven constituents were found in various processed Rehmanniae Radix Preparata. Not all constituents were defined but stachyose and raffinose were found in all cases. And The most common constituents of Rehmanniae radix was stachyose. In the course of processing, most of stachyose and raffinose were decreased. Stachyose was decreased slowly in the course of processing with rice wine(RR), amomi and rice wine(AR), and crataegi and rice wine(CR). However stachyose was decreased rapidly in the course of processing with fresh rehmannia juice(FR). The method with crataegi and rice wine(CR) showed the smallest decrease of stachyose. And processing method with crataegi and rice wine(CR) showed the most abundant amount for stachyose after the nineth processing. Conclusion : The changes of oligosaccharides in the course of processing were a very important direct barometers to do the quality control and set up a standard of Rehmanniae Radix Preparata.

  • Multiple shoots obtained in MS medium suppler with 5.0 mg/L BA though shoot-tip culture. The frequency of vitrified shoot was lower on Bacto-agar medium than on Gelrite as gelling agent. Addition of activated charcoal at concentrations of 0.1~0.3% reduced vitrification and markedly increased shoot growth, and formation and growth of roots, but significantly reduced the number of shoots formed. The ratio of fresh weight to dry weight was decreased by increasing light intensity and agar concentration. Eight-tenths times of macroelement of MS medium was observed to be effective for shoot formation. Addition of IAA effectively promoted shoot formation in both shoot tip and node-bud explants. Supplement of 5.0 mg/L BA, 0.3 mg/L IAA to MS medium was most effective in shoot proliferation on shoot tip and node-bud explants.Multiple shoots obtained in MS medium suppler with 5.0 mg/L BA though shoot-tip culture. • Background: Allergic diseases like such as atopic dermatitis, asthma, and rhinitis have recently increased both domestically and globally. The present study was undertaken to select candidates with anti-allergic activity from plant resources. Methods and Results: Fifty-six plant extracts at $20{\mu}g/m{\ell}$ were screened against ${\beta}$-hexosaminidase production and interleukin (IL)-4 release in degranulated rat basophilic leukemia (RBL)-2H3 cells. The anti-allergy activity of three plant extracts selected from the preliminary screening experiment, Polygonatum sibiricum F. Delaroche (root), Pyrus pytifolia var. culta (Makino) Nakai (leaf), and Rehmannia glutinosa (Gaertn.) Libosch. ex Steud (root) were measured at concentrations of $2-250{\mu}g/m{\ell}$ in three cell lines as RBL-2H3, HaCaT and Jurkcat T cells. The assay showed the root extract of R. glutinosa to have an inhibitory activity of 4.2% - 28.6% on ${\beta}$-hexosaminidase production from IgE-sensitized RBL-2H3 cells. Each extract of P. sibiricum and R. glutinosa reduced IL-4 release in IgE-sensitized RBL-2H3 cells, respectively. The leaf extract of P. pyrifolia var. culta showed a significantly potent suppressive effect of 10.2% - 74.7% on the mRNA expression of tumor necrosis factor (TNF)-${\alpha}$ in HaCaT cells sensitized with TNF-a and INF-g, and showed inhibitory effect of -8.6% - 90.9% on the mRNA expression of IL-2 in Jurkat T cells sensitized with PMA and A23187. Conclusions: The results showed that the root of R. glutinosa and leaf of P. pyrifolia var. culta could be useful candidates as antiallergy materials.

  • Medicinal herbs have long been used as a remedy for diverse diseases in Asia owing to their various pharmacological effect. In this study, the immuno-enhancing activity of medicinal herbs was investigated using macrophage cell lines. Specifically, we examined the effects of extracts of twelve medicinal herbs on nitric oxide (NO) production in RAW 264.7 cells, and selected five that were highly effective (Glycyrrhiza glabra, Rehmannia glutinosa, Angelica gigas, Platycodon grandflorum, and Actinidia polygama) for further immune related studies. The effects of extracts from five theses medicinal herbs, which were mainly composed of polysaccharides and proteins on the production of immune-related cytokines in the RAW 264.7 macrophage cell line and the Molt-4 T cell line were investigated. The extracts of all investigated medicinal herbs increased the production of NO and cytokines such as tumor necrosis factor-alpha (TNF-${\alpha}$), interleukin-1beta (IL-$1{\beta}$), interleukin-6 (IL-6) and interleukin-10 (IL-10). Additionally, they slightly increased the proliferation of T-cells when compared to the control. Overall, the result of this study suggests that the five medicinal herb extracts investigated herein are useful natural immune enhancing agents.
