ATP가 마우스 담관세포의 세포크기 조절에 미치는 영향 (Effects of ATP on Regulatory Volume Decrease in Mouse Cholangiocytes)

  • ATP 분비 경로는 담관세포종 세포주를 포함한 일부 세포의 RVD 조절에 있어서 주요한 조절 역할을 수행할 것으로 생각되나, 정상 담관세포의 RVD에 있어서의 역할은 잘 알려져 있지 않다. 정상 담관세포의 RVD에 있어서 세포 외 ATP와 ATP 경로의 역할을 알아보기 위하여, 정상마우스에서 담관세포를 분리하여 연구에 이용하였다. BDCCs의 크기 변화는 정량측정 비디오현미경을 이용하여 횡단면을 간접 측정하였다. 정상마우스의 BDCCs의 횡단면은 저장액에서 노출한 후 10분에 $1.20{\pm}0.02$ (n=20)이었으나, 40분에는 $1.06{\pm}0.03$로 감소하였다. ATP와 ATP 가수분해 효소인 아피라아제 또는 P2 수용체 차단제인 슈라민 RVD 과정 중에 처리 하였으나 처리하지 않은 대조군과 중요한 차이를 보이지 않았다. 더 나아가, PKC 저해제인 비신돌아마이드 I 과 Ro 31-8220을 처리하여 RVD에 미치는 영향을 알아 보았으나 대조군과 중요한 차이를 보이지 않았다. 이러한 결과들에서 정상 마우스 담관세포는 담관세포종 세포주와 다른 결과를 보였으며, 정상 마우스의 RVD에 있어서 ATP는 중요한 역할을 하지 않음을 알 수 있었다.

Swelling-activated $Cl^-$ Channels in Human Salivary Gland Acinar Cells

  • The role of $Cl^-$ channels in regulatory volume decrease (RVD) in human salivary gland acinar cells was examined using a whole-cell patch clamp technique. Human tissues were obtained from healthy volunteers or from patients with oromaxillofacial tumors. During the measurements, $K^+$-free solutions were employed to eliminate contamination of whole-cell conductance by $K^+$ currents. When the cells were exposed to a 70% hypotonic solution, outward-rectifying currents, which were not observed in the resting state, were found to have significantly increased both in human labial and parotid gland acinar cells. The amplitudes of the currents were reduced in a low $Cl^-$ bath solution. Furthermore, the addition of $100{\mu}M$ 5-Nitro-2- (3-phenyl propylamino) benzoic acid (NPPB) or $100{\mu}M$ 4,4'-diisothio cyanatostilbene-2,2'-disulphonic acid (DIDS), known to partially block $Cl^-$ channels, significantly inhibited these currents. Its outward-rectifying current profile, shift in reversal potential in a low $Cl^-$ bath solution and pharmacological properties suggest that this is a $Ca^{2+}$-independent, volume activated $Cl^-$ current. We conclude therefore that volume activated $Cl^-$ channels play a putative role in RVD in human salivary gland acinar cells.

저장성 용액에 노출된 햄스터 난자에 관찰되는 이온전류의 변화 (Ionic currents elicited by the hypotonic solution in hamster eggs)

  • Cell volume regulatory mechanisms are usually disclosed by exposure of cell to anisotonic media. If a cell is suddenly exposed to hypotonic media, it swells initially like an osmometer but within minutes regains its original cell volume. This behavior has been labelled as regulatory cell volume decrease(RVD). RVD is believed to result from the loss of permeable ions through the membrane. In this study, we examined hypotonically induced changes in the membrance currents involved in RVD by using whole cell voltage clamp technique in the unfertilized hamster egg. At -40mV of the holding potential, the stationary current was maintained in the hamster egg exposed to isotonic solution composed of, mainly, 115mM NaCl and 40mM mannitol. Hypotonic solution was prepared by removing mannitol. Therefore, the concentrations of $Na^+$ and $Cl^-$ in this hypotonic media were the same as those in the isotonic solution. Following 30 to 60 sec after applying the hypotonic media to the egg, the inward current was evoked. This inward current was eliminated by $100{\mu}M$ 4-acetamido-4'-isothiocyanostil-bene-2,2'-disulfonic acid(SITS), an anion channel blocker, leaving the small outward current component. Further addition of 2mM $Ba^{2+}$, a broad $K^+$ channel blocker, completely abolished the small outward current left even in the presence of SITS during hypotonic stress. These results suggest that $K^+$ and $Cl^-$ move out of cells, resulting in RVD. To test the involvement of $Na^+$ in RVD, 20mM Na-isethionate was substituted for mannitol in isotonic media(135mM $Na^+$) and Na-isethionate (20mM) was freed the hypotonic solution. Only $Cl^-$ concentration in both isotonic and hypotonic media was kept constant at 115mM, whereas concentration of $Na^+$ was lowered in hypotonic solution to 115mM from 135mM in isotonic solution. Hypotonic medium induced the outward current in the egg equilibrated isotonically. This current was reduced by $100{\mu}M$ SITS but was augmented by 2 mM $Ba^{2+}$. In terms of RVD, these results imply that $Cl^-$ efflux is coupled with $K^+$, maybe for electroneutrality during hypotonic stress and/or with $Na^+$ via unknown transport mechanism(s). From the overall results, the hypotonic stress facilitates the movement of $Cl^-$ and $K^+$ out of the hamster egg to regain cellular volume with electroneutrality. If there exist a difference in $[Na^+]_0$ between isotonic and hypotonic solution, another transport mechanism concerned with $Na^+$ may, at least partly, participate in regulatory volume decrease.

플라즈마를 이용한 도시 쓰레기 소각재 용융처리 기술 (Slagging treatment of MSW incineration ash by plasma system)

  • A plasma melting system to vitrify ny ash from MSW(Municipal Solid Waste) incinerator has been operated in SHI(Samsung Heavy Industries) since 1996. Waste feeding rate was 200kg/hr. with maximum working power of 500㎾. Because of high melting temperature of fly ash, bottom ash was used as an additive to decrease melting temperature. Data analysis for discharged slag shows volume reduction up to 30% and no leaching of heavy metals such as Pb, Cd, Cr which were an obstacle for landfill and recycle. Atmospheric pollution gas like nitrogen oxides, carbon monoxide, and PCDD/PCDF were restrained below the regulatory limit.

Antitumor Effects of Fucoidan on Human Colon Cancer Cells via Activation of Akt Signaling

  • We identified a novel Akt signaling mechanism that mediates fucoidan-induced suppression of human colon cancer cell (HT29) proliferation and anticancer effects. Fucoidan treatment significantly inhibited growth, induced G1-phase-associated upregulation of p21WAF1 expression, and suppressed cyclin and cyclin-dependent kinase expression in HT29 colon cancer cells. Additionally, fucoidan treatment activated the Akt signaling pathway, which was inhibited by treatment with an Akt inhibitor. The inhibition of Akt activation reversed the fucoidan-induced decrease in cell proliferation, the induction of G1-phase-associated p21WAF1 expression, and the reduction in cell cycle regulatory protein expression. Intraperitoneal injection of fucoidan reduced tumor volume; this enhanced antitumor efficacy was associated with induction of apoptosis and decreased angiogenesis. These data suggest that the activation of Akt signaling is involved in the growth inhibition of colon cancer cells treated with fucoidan. Thus, fucoidan may serve as a potential therapeutic agent for colon cancer.

Ingestion Exposure to Nitrosamines in Chlorinated Drinking Water

  • Objectives: N-Nitrosodimethylamine (NDMA) is classified as a probable human carcinogen by the United States Environmental Protection Agency (US EPA) and is formed during the chlorination of municipal drinking water. In this study, selected nitrosamines were measured in chlorinated drinking water collected from Chuncheon, Kangwon-do, Republic of Korea, and a risk assessment for NDMA was conducted. Methods: Twelve water samples were collected from 2 treatment plants and 10 household taps. Samples were analyzed for 6 nitrosamines via solid-phase extraction cleanup followed by conversion to dansyl derivatives and high-performance liquid chromatography-fluorescence detection (HPLC-FLD). Considering the dietary patterns of Korean people and the concentration change of NDMA by boiling, a carcinogenic risk assessment from ingestion exposure was conducted following the US EPA guidelines. Results: NDMA concentrations ranged between 26.1 and 112.0 ng/L. NDMA in water was found to be thermally stable, and thus its concentration at the end of boiling was greater than before thermal treatment owing to the decrease in water volume. The estimated excess lifetime carcinogenic risk exceeded the regulatory baseline risk of $10^{-5}$. Conclusions: This result suggests that more extensive studies need to be conducted on nitrosamine concentration distributions over the country and the source of relatively high nitrosamine concentrations.

Humic Substances Suppresses the Proliferation of TC-1 Cells, the Lung Cancer Cell

  • In humic substances, fulvic acid (FA) is a subclass of diverse compounds known as humic substances, which are by-products of organic degradation from microorganisms. FA can suppress the proliferation of tumor cells. Despite numerous studies, the exact mechanism for the various effects of FA is not clearly understood. Based on results demonstrating anti-proliferation effects on human cancer, we investigated whether FA has similar effects on lung cancer in this study. Firstly, the anti-cancer effect of FA in pulmonary epithelial tumor cell lines (TC-1 cells) was examined by confirming its inhibitory effect on the cell proliferation of TC-1 cells. TC-1 cell proliferation was reduced by FA on a dose-dependent and time-dependent manner. After 24 hours of FA treatment, cell morphological changes such as cell volume decrease, non-adherence and increased number of apoptotic cells were clearly observed. In addition, FA induced a DNA ladder pattern by increased of DNA fragments in TC-1 cells. In the intracellular regulatory pathway by FA, we confirmed that FA induced the reduction of the anti-apoptotic protein, Bcl-2 protein levels. These results indicate that FA has anticancer effect by inducing intracellular apoptotic pathway. Further research on the mechanism of anticancer effects will be basic data for the development of potential anticancer drugs.

부동산정책이 부동산가격에 미치는 영향에 관한 연구: 조세정책과 금융정책 중심으로 (A Study on the Effect of Real Estate Policy on Real Estate Price: Focusing on Tax Policy and Financial Policy)

  • 본 연구에서는 부동산 정책, 조세정책, 금융정책, 규제지수에대한 이론적고찰과 선행연구를 살펴보고 전국의 2014년1월부터 2021년 12월 까지의 월별데이터를 이용하여 조세정책과 금융정책이 부동산가격에 미치는 영향을 분석하였다. 분석방법은 단위근 검정과 공적분 검정을 통해 VAR모형을 사용하여 분석하였으며 충격반응분석과 분산분해분석을 실시하였다. 본 연구의 분석결과는 다음과 같다. 첫째, 조세규제지수와 금융규제지수는 주택가격에 영향을 미치지 못하였다. 주택가격 상승기에 주택가격 안정을 위하여 규제 일변도의 정책은 효과가 없으며 오히려 거래량 감소 등으로 가격을 상승시키는 부작용이 생긴다. 둘째, 주택담보대출금리는 주택매매가격지수에 음(-)의 효과를 주었다. 즉, 이자율의 상승이 주택가격을 하락시키는 효과가 있다는 의미로 보여진다. 셋째, 양도차액의 상승 즉, 양도소득세의 과세는 주택가격에 양(+)의 효과를 준다. 이는 조세를 매수자에게 전가시키거나 조세부담으로 인해 매각을 보류하는 동결효과로 주택가격을 상승시키는 결과가 나왔다. 넷째, 취득세와 주택담보대출액은 주택가격에 별다른 영향을 미치지 못하였다.

휘발에 의한 가솔린 성분의 조성 및 용해도 변화특성 (Solubility Change of Gasoline Components Under Evaporation)

  • 회분식 실험을 통해 가솔린의 용해거동을 살펴보았다. 먼저 가솔린 중 상평형에 이르기까지 개별성분들의 용해 kinetics를 살펴보았으며 평형에 도달한 후 각각의 용해도를 Raoult의 법칙을 이용하여 설명하고자 하였다. 또한 서로 다른 정도로 휘발시켜 가솔린의 조성을 변화시킨 후 개별성분들과 TPH의 용해도를 측정하여 비교하였다. 정적인 상태에서 가솔린의 개별성분들의 용해 kinetics는 일정한 양상을 보였으나 다만 첨가제로 사용된 MTBE의 경우 다른 성분들에 비해 평형도달시간이 길었다. 가솔린과 물의 상평형 상태에서 가솔린 개별성분들의 분배계수는 각각의 단일물질 용해도와 log-log선형관계를 보였으나 가솔린을 이상용액으로 간주해서 Raoult의 법칙을 적용했을 경우보다는 약간 낮은 경향을 보였다. 가솔린에있는 개별물질들의 농도는 휘발정도가 증가함에 따라 증가하다가 감소하는 경향을 보였는데 물로의 용해도 변화도 이와 거의 일치하는 양상을 보였다. 휘발정도가 증가할수록 용출농도는 초기에 급격히 감소하다가 이후 점근적으로 감소하는 경향을 보였으며 BTEX등 독성이 높은 지하수오염 규제물질들의 용출농도는 더욱더 빠르게 감소하였다. 이러한 결과를 가솔린 누출로 인한 지하수 오염의 시나리오에 적용해 볼 때 휘발에 의한 가솔린 제거는 지하수 오염의 잠재성과 오염된 지하수의 독성을 효과적으로 저감시키며 그 정도는 가솔린 절대량의 제거효율보다도 훨씬 크다는 것을 알 수 있다.

연마재 워터젯을 활용한 처분터널 내 콘크리트 플러그 절삭 (Concrete plug cutting using abrasive waterjet in the disposal research tunnel)

  • 워터젯은 콘크리트와 암석 절삭에 적합하며 소음과 진동이 적어 도심지 시공에 널리 활용되는 기술이다. 최근 한국원자력연구원은 처분터널 내 공학적방벽의 열-수리-역학적 복합거동 현장시험의 콘크리트 플러그를 교란 및 손상 없이 해체하기 위해 연마재 워터젯 기술을 채택 및 적용하였다. 본 연구에서는 플러그 해체를 통한 연마재 워터젯의 터널 내 적용성을 평가하였고, 절삭 결과를 개발된 절삭 모델 결과와 비교하였다. 현장 적용성에 관해서는, 워터젯 활용은 경로선택이 용이하였으며 주변부의 추가적인 교란을 발생시키지 않았다. 펌프의 소음은 공회전 시 64.9 dB로 국내 생활소음·진동의 규제기준을 만족하였으나 공중 분사 및 절삭 시에는 터널 내에서 측정되어 그 기준을 상회한 것으로 나타났다. 절삭 모델 검증을 위해 연마재 워터젯의 반복 절삭, 유량, 연마재 투입량 및 이격거리를 주요 변수로 선정하였고 절삭 모델로 계산된 절삭 부피와 측정된 부피의 오차는 1회 절삭 시 12~13%, 2회 절삭 시 16%를 보였다. 절삭 깊이와 폭은 이격거리에 큰 영향을 받았으며, 이격거리가 작을수록 오차가 감소하는 경향을 보였다.
