In general, the precast columns can obtain its homogeneous quality as they are produced in a factory with a hollow concrete block type by using high strength concrete, so that they can generate the reduction of dead load. Such a method of precast hollow concrete columns is already implemented in USA and Japan and used for connecting between blocks which use PC tendons. However, it is inevitable to have uneconomical construction with excessive cost in early stage when PC tendons are used. This study aims to develop an economical precast column with high quality and constructability which consists of only splice sleeve and general reinforcing bar without using PC tendons in order to reduce the construction period and cost. To achieve this goal, this study tested the performance of total 5 minimized models in the experiment with the variables such as hollowness, diameter of main reinforcement bar and cross-sectional size for the cross section of precast column by using grouting type splice sleeve which is a new type joint rebar. And it also verified the performance of column in the experiment for a large-sized model in order to overview its applicability by excluding large scale effect.

To analysis effect of environment factors on productivity of sweet pepper according to greenhouse covering material (glass, plastic film), this was investigated. In glasshouse, outside light was positively correlated with yield as that $100MJ{\cdot}m^{-2}$ of outside light increased $300{\sim}500g{\cdot}m^{-2}$, also cumulative temperature was same tendency. On possibility of model development for yield estimate cumulative temperature was high than outside light. According to covering material, leaf photosynthesis, productivity per out-side light and term in glasshouse was more high 13%, 46%, and 47% compared with plastic film house, respectively. Result of analysis of effect of light, temperature, and $CO_2$ on yield, relative yield coefficient, yield increment coefficient, and yield reduction coefficient in glasshouse were more high 25%, 73%, and 34% compared with plastic film house, respectively. Hence, sweet pepper's growing in glasshouse compare with plastic film house had more productivity, but that had more sensitivity to charge of environment factors.
Rice is the most important crop as a model plant for functional genomics of monocotyledons. Rice is usually transformed using Agrobacterium tumefaciens. However, the transformation efficiency using previous method is still low. In this study, we established a new method by modifying the general Agrobacterium protocol especially in the inoculation and co-cultivation step. We directly inoculated Agrobacterium containing a CIPK15 gene under the control of CaMV 35S promoter and NOS terminator in the pCAM1300 vector into the pre-soaked seeds in N6D media for 24 hours. After 7 days of culture at $25^{\circ}C$, calli were formed on seeds cultured on the co-cultivation medium containing an antioxidant compound (1 mM dithiothreitol) and of Agrobacterium growth-inhibiting agent (3 mg/L silver nitrate). We obtained 35 and 22 transgenic plants in rice cultivars, Gopumbyeo and Ilpumbyeo, with increase of transformation efficiency by 30.4% and 22.6%, respectively compared to the general transformation method. The new method in this study would lead to reduction of substantial labor and time to generate transgenic plants.
Vacuolar protein sorting (VPS) 26b is a newly discovered member of the retromer complex; it is encoded by a single-copy gene located on mouse chromosome 9, and the complex has been reported as being composed of proteins VPS26, VPS29, and VPS35. We have previously shown that mice lacking VPS26b exhibited no significant body size or health issues. Although retromer components are widely expressed in mouse tissue, their roles have not yet been completely elucidated. The current study investigates whether the VPS26b-associated retromer complex can be used as a neurodegeneration model. Previously, we observed a significant reduction in VPS35 and VPS29 in the brain cells of in VPS26b-deficient mice as well as an absence of the VPS26b-VPS29-VPS35 retromer complex despite the normal presence of VPS26a-VPS29-VPS35. Recent studies have suggested that low levels of VPS35 can lead to Alzheimer's disease-like phenotypes including cognitive memory deficits. In this study, we successfully demonstrate an association between the absence of the VPS26b-VPS29-VPS35 retromer complex, reduced cell density in the CA3 region of the hippocampus, and learning disability in VPS26b knock-out mice. The results also indicate that the VPS26b-associated retromer complex affects neurodegenerative disorders and learning processes.

The objective of this study is to develop an automatic object changer unit to improve processing problems existed in the conventional horizontal machining center. To achieve this goal, this study designed a horizontal transfer as the second project continued to the first project that designed a upward and downward traverse unit. A horizontal traverse unit shows a symmetric structure and consists of frame, which consists of four unit tools, motor and reducer, which are fixed at a frame, operation unit with pinions, first traverse unit, and second traverse unit. Constraint conditions based on the operation mechanism with these elements were configured and obtained following results after modeling a model for a traverse motor. In the kinematic expression of sliding motion with one degree of freedom, the sliding motion is constrained. Also, the rack 3 installed at a frame is used to configure possible kinematic constraint conditions of the rack 2 according to the rolling motion of the pinion 2 in the first traverse unit. In addition, the moment of inertia that is a type of kinetic energy in a converted horizontal traverse unit in the side of the reducer can be applied to introduce the moment of inertia of a converted horizontal traverse unit in the side of the reducer by using the sum of kinetic energy in the rack and pinion, which is a part of the horizontal traverse unit. Also, the equation of motion of the converted upward and downward traverse unit in the side of the motor using the equation of motion of the motor. Furthermore, the horizontal traverse unit predetermines the mass of the first and second traverse unit and applied load including the radius and reduction ratio of the pitch circle in the pinion 1 and applied load to the rack 2. Then, a proper motor can be determined using several parameters in the upward and downward traverse unit in order to verify such predetermined specifications. In future studies later this study, a simulation that verifies the results of the previous two stages of studies using a finite element method.

In Korea, tuna purse seine fishery is the industry with the highest production volume in domestic deep-sea fishery. Research has been continuously carried out since the late 1980s, and research on modernization of technology has been actively conducted. However, while auxiliary works boats on the fishing vessels play an important role in the purse seine fishing, related research is not sufficient. In this study, we tried to rebuild the propulsion system of the Net-boat from the propeller to the waterjet type and to perform the hull form conversion to improve the resistance performance of the hull part and improve the motion performance. For this, we calculated the change of the fluid performance of the ship through computational numerical analysis. In addition, towing tank tests were carried out to verify the performance of the existing ship and the ship which changed the propulsion system. As a result, resistance performance of the waterjet type compared to the propeller type was improved by approximately 45 %. It was confirmed that this was due to reduction of the wetted surface area by removing net and improvements in hull form.
Obesity is an established risk factor for colorectal cancer. Pioglitazone is a peroxisome proliferator activated receptor$receptor{\gamma}$ ($PPAR{\gamma}$) agonist that induces differentiation in adipocytes and induces growth arrest and/or apoptosis in vitro in several cancer cell lines. In the present study, we investigated the effect of pioglitazone on the development of azoxymethane-induced colon aberrant crypt foci (ACF) in KK-$A^{\mathcal{Y}}$ obesity and diabetes model mice, and tried to clarify mechanisms by which the $PPAR{\gamma}$ ligand inhibits ACF development. Administration of 800 ppm pioglitazone reduced the number of colon ACF/mouse to 30% of those in untreated mice and improved hypertrophic changes of adipocytes in KK-$A^{\mathcal{Y}}$ mice with significant reduction of serum triglyceride and insulin levels. Moreover, mRNA levels of adipocytokines, such as leptin, monocyte chemoattractant protein-1 and plasminogen activator inhibitor-1, in the visceral fat were decreased. PCNA immunohistochemistry revealed that pioglitazone treatment suppressed cell proliferation in the colorectal epithelium with elevation of p27 and p53 gene expression. These results suggest that pioglitazone prevented obesity-associated colon carcinogenesis through improvement of dysregulated adipocytokine levels and high serum levels of triglyceride and insulin, and increase of p27 and p53 mRNA levels in the colorectal mucosa. These data indicate that pioglitazone warrants attention as a potential chemopreventive agent against obesity-associated colorectal cancer.

Objectives : Gal-Geun-Tang (GT) has been described from SANGHAN in Korean traditional medicine and known to act against cold, fever, hypertension, and nasal catarrh. However, little has yet been learned about the effect of GT on immune function. In the current study, in vitro and in vivo immunomodulatory activity of GT (water extract) was investigated.Methods : Water extract of GT induced in vitro proliferation of spleen cells and significantly increased their proliferative responses during anti-CD3 activation. Using purified splenic T and B cells, it was revealed that GT has a mitogenic activity to B cells and promotes their proliferation induced by lipopolysaccharide, whereas T cell proliferation was not triggered and GT was rather inhibitory to T cell activation caused by anti-CD3 antibody. In the presence of antigen presenting cells (APC), GT addition resulted in a significant increase of IFNγ and IL-4, but not IL-2, production. However, addition of high concentration (1,000㎍/㎖) of GT led to a marked reduction in T cell cytokine production and under such condition, GT facilitated apoptosis of T cells when examined by flow cytometry with propidium iodide staining.Results : In vivo immunomdulation of GT was also investigated using a mouse model. Following keyhole limpet hemocyanin (KLH) immunization, GT (1 ㎎/day) was orally administered for 9 days. Cell numbers in thymus, spleen and peripheral blood were not altered by GT administration, indicating that such dose is not immunotoxic. Cell numbers in draining lymph nodes (LN) and ex vivo Ag-specific proliferation of LN cells were significantly elevated by GT administration. However, any preferential stimulation of T or B and CD4+ or CD8+ T cell subpopulations was not observed in a flow cytometric analysis of LN cells. This result shows that GT does not promote in vivo B cell proliferation while GT enhances Ag-specific proliferation of LN cells, unlike what was observed in vitro.Conclusions : For a further understanding of in vivo immunomodulatory activity of GT, ex vivo cytokine production of LN cells obtained from KLH-immunized mice was evaluated. Ag-specific IFNγ production was significantly higher in GT-treated mice when compared to PBS-treated control mice. In contrast, IL-4 production in GT-treated group was comparable to control group unlike to in vitro data. In addition, GT administration did not result in any significant differences in serum levels of Ig (IgM, IgG1 and IgG2a) between GT-treated and control groups. Taken together, these data strongly support that GT promotes immune response, more profoundly type 1 helper T cell (Th1) activity and GT may be applicable for treatment of intracellular parasite infection such as viral diseases.
Ground magnetic surveys were conducted at four areas where the Yangsan fault, the most prominent lineament in the Kyeongsang basin, appears to be passed through. For data processing, IGRF correction, upward continuation and reduction-to-the-pole were performed. The automatic inversion by using a matrix computation method, which takes the depth to bottom layer of the horizontal two layer structure as the model parameter, has been attempted to delineate the subsurface structure. Upward continuation of the surface magnetic map to the same level of the aeromagnetic survey (KIER, 1989) resulted in very similiar patterns to those of aeromagnetic data. Subsurface modeling of eight profile data show that the strike and dip of the Yangsan fault in study areas are $N6^{\circ}-15^{\circ}E$, and near vertical to somewhat eastward, repectively, despite of the local lithological contrast of each study area. It seems that the magnetic effect of faulting in the study area 1, which locates in the most northern part of the survey areas, is disturbed by that of igneous intrusion. At study area 2, the possibility of volcanic or igneous intrusion, which is 200-300 meters wide along the fault plane was presented. At study area 3, unlike other study areas, distinct fracture zone of 500-700 meters in width was revealed along the surface fault line. The andesitic rocks of the study area 4 have very high susceptibilities and the fault line on surface of this area was shifted about 500 meter eastward, as compared with the inferred fault line by the previous study.

Influences of the magnetic field on 5, 10 and 15 wt% (1.5, 3 and 4.5 M) HCl solution behaviour, which has widespread applications in petroleum well acidizing, were investigated in various conditions. Differences in the pH of magnetized hydrochloric acid compared to that of normal hydrochloric acid were measured. Taguchi design of experimental (DoE) method were used to model effects of the magnetic field intensity, concentration, velocity and temperature of acid in addition to the elapsed time. The experimental results showed that the magnetic field decreases [$H^+$] concentration of hydrochloric acid up to 42% after magnetization. Increasing the magnetic field intensity (with 28% contribution), concentration (with 42% contribution), and velocity of acid increases the effect of magnetic treatment. The results also demonstrated that the acid magnetization was-not influenced by the fluid velocity and heating. It was also displayed that the acid preserves its magnetic memory during time. The optimum combination of factors with respect to the highest change of [$H^+$] concentration was obtained as an acid concentration of 10% and an applied magnetic field of 4,300 Gauss. Due to the reduction of HCl reaction rate under the magnetization process, it can be proposed that the magnetized HCl is a cost effective and reliable alternative retarder in the matrix acidizing of hydrocarbon (crude oil and natural gas) wells.
