Easy Red Ginseng Production Using Household Microwave Ovens (가정용 전자레인지를 이용한 간편 홍삼 제조)

  • The study was about to produce red ginsengs easily, using a household microwave oven to promote the consumption of fresh ginsengs in the home. Producing red ginsengs with a household microwave oven 'defrost function' takes 13 minutes (A), 'cook function' 6 minutes (B), and finally, 'defrost function' 44 minutes (C). For characteristics of microwave-produced red ginsengs, total saponin loss, color of powder, polyphenol content and saponin composition were compared with common red ginsengs. The color test for red ginseng powder showed that the color of household microwave-produced 6-minute cooked red ginseng (B) or 44-minute defrosted red ginseng (C) was closer to that of the common red ginsengs (E). The total saponin content in water eluted during red ginseng production showed that the saponin loss in microwave red ginseng was negligible compared to the common red ginsengs. Microwave red ginsengs showed no difference in phenol content that of the and higher total ginsenoside content than common red ginsengs. The ginsenoside $Rg_1$, Re, Rf, $Rg_2+Rh_1$, $Rb_1$, Rc, $Rb_2$, $Rb_3$, Rd and $Rg_3$ contents of microwave red ginsengs (A, B) were higher compared to that of the common red ginsengs; the ginsenoside Re, Rc, $Rb_2$, $Rb_3$, Rd and $Rg_3$ contents of 44-minute defrosted red ginseng (C) were higher compared to the common red ginsengs. It is considered that red ginseng production, using microwave oven at home, can be a fast and convenient way to produce highly functional red ginsengs with high ginsenoside content.

Effects of Panax ginseng on Type I Hypersensitivity (제1형 과민 반응에 미치는 고려인삼의 영향)

  • Effects of Panax ginseng on allergic reactions were studied uslng various in vivo and in vitro experimental models such as 48-hr passive cutaneous anaphylaxis, mediators-induced skin reactions, histamine release from rat peritoneal mast cells, hexosaminidase release from RBL-2H3 cells, and lipoxygenase assay . In all of anti-allergic experiments we conducted, ginseng components (50% ethanol extract or ginseng total saponin or ginsenosides) extracted from Korean red ginseng, did not show significant anti-allergic actions. In 48-hr passive cutaneous anaphylaxis and mediators-induced skin reactions, 50% ethanol extract did not suppress hypersensitivity reactions. Total saponin, 50% ethanol extract, and 8 major ginsenosides did not show inhibitory effects on lipoxygeanse activity. Ginseng total saponin did not inhibit histamine release from rat peritoneal mast cells. All of the ginseng components mentioned above were also tested on RBL-2H3 cells, but none of them inhibited hexosaminidase release from this cell line. These results suggest that Panax ginseng does not have effects on allergic reactions at the level of 50% ethanol extract or total saponin used. All of 8 major saponin components tested ($Rb_1$, $Rb_2$, Rc, Rd, Re, Rf, $Rg_1$, $Rg_2$), did not inhibit lipoxygenase activity and degranulation events.

The Effect of Korean Red Ginseng Saponins on the Recombinant Serotonin Type 3 Receptor Expressed in Xenopus Oocytes (Xenopus oocytes에서 발현된 유전자재조합 세로토닌 제3형 수용체에 대한 한국산 홍삼 사포닌의 효과)

  • The effect of Korean Ginseng saponins (total saponin, PD saponin and PT saponin) on the serotonin type 3 receptor, which is known to be involved in nausea and vomiting following anticancer chemotherapy or the general anesthesia, was investigated. after in vitro transcribed recombinant serotonin type 3 receptor in the Xenopus laevis oocyte, classic two electrodes voltage clamp technique was used. All of ginseng saponins inhibited the response of the agonist, serotonin, on the serotonin type 3 receptor in a dose-dependent manner. PT saponin showed to have the inhibitory effect more than 2 times as potent as PD saponin. Total saponin shifted the serotonin dose response plot to the right (EC$\_$50/, 0.70$\pm$0.17 $\mu$M into 3.57$\pm$1.42 $\mu$M, and Hill coefficient, 2.14$\pm$0.60 into 1.52$\pm$1.00). Ginseng saponin did not change the reversal potential (∼0 mV) of serotonin type 3 receptor. These results suggest that Korean ginseng saponin may have the inhibitory effect on serotonin type 3 receptor.

  • The present study was undertaken to elucidate the behavioral characteristics of nootropic candidates, entrophenoxine, N-methyl-D-glucamine, piracetam and red ginseng saponin components on stereotyped sniffing behavior induced by apomorphine in rats. Apomorphine, a direct dopaminergic receptor agonist, induced stereotyped behaviors including sniffing licking growing and biting in a dosedependent manner, and that behaviors were completely inhibited when measured at 1 week after 6-ydroxydopamine(6-HDA) treatment. Centrophenoxine, N-methyl-D-glucamine, red ginseng total saponin(TS), panaxatriol (PT), and Rg1 enhanced but panaxadiol (PD) inhibited, whereas piracetam and Rb1 were not effective of the sterotyped sniffing behavior induced by apomorphine(1mg/kg). The enhanced stereotyped behavior by centrophenoxine, N-methyl-D-glucamine, red ginseng total saponin, panaxatriol(PT), and Rg1 seems to have a similarity to entrophenoxine, N-methyl-D-glucamate in modulating of dopaminergic neuroal activity and also my be useful for the nootropic candidates.

  • Total saponins and ether extracts of red and white ginseng were obtained and their effects on blood pressure in cat and their histamine liberating activities in rabbits were measured. 1) Ether extract of red ginseng showed a transient hypotensive effect and subsequently showed a remarkable and persistent hypotensive effect, whereas other three fractions, such as saponin fractions of red and white ginseng and ether extract of white ginseng showed only a initial transient hypotensive effects. 2) Histamine levels liberated into blood after administration of each fractions measured by the bioassay with guinea pig ileum. Ether extract of red ginseng immediately increased histamine contents in plasma but the histamine levels decreased to normal level within 10min in spite of decreased blood pressure was sustained. Although white ginseng saponin lowered blood pressure immediately when it is administered, histamine release was observed after 10min. The results suggest that hypotensive effects of ginseng seems to have no correlation with the histamine liberating activity. Ginseng appears to show hypotensive effect via some other mechanisms.

  • This study was conducted to elucidate the effects of light intensity using polyethylene (p.E) net shading on the specific gravity, red ginseng quality, sugar and saponin contents of ginseng root. The specific gravity significantly increased in the ginseng roots grown under the P.E net shading as compared with that of common straw shading. The red ginseng quality under the P.E net shading was improved in order of 10, 5, 15, 20, 30% of light intensity and the inside cavity and inside white part decreased remarkably as compared with those of common straw shading. The ginseng roots grown under the P.E net shading at 10% and 15% light intensity showed a significant increase in the total sugar content but a significant decrease in the reducing sugar content at 15% light intensity as compared with those of common straw shading. The amount of total saponin of ginseng roots was increased under the P.E net shading at high light intensity as compared with that of common straw shading and the ginseng roots grown under the P.E net shading at 10% light intensity showed an increase in the diol group saponin but the ratio of PT/PD was decreased. Extract contents of ginseng root under the P.E net shading was higher than those of common straw shading and the roots grown under the P.E net shading at 15% and 20% light intensity resulted in a remarkable increase in extract contents.

  • In order to enhance the components of bioactive ginsenosides and the manufacturing process of red ginseng, we developed the simplified method for red ginseng production. The red ginseng extract was prepared from red ginseng produced with the simplified method, and the production rate of extract ($62^{\circ}$ brix) was more than 60%. The ginsenosides of red ginseng were purified and analyzed by HPLC using ELSD. Ginsenoside-$Rg_3,\;Rh_2$ and $Rh_1$, specific artifacts found only in red ginseng, were detected by HPLC. Especially, contents of ginsenoside-$Rg_3$ and Rh1 were detected high than two times in red ginseng produced the simplified method compared to commercial products.

Extracting Conditions for Promoting Ginsenoside Contents and Taste of Red Ginseng Water Extract (홍삼 물 추출액의 사포닌 함량 및 맛의 증진을 위한 추출 조건)

  • In this study, red ginseng extract solutions were analyzed to set up the functional saponin content and quality optimization condition. The highest saponin content among the total red ginseng extracts was 64.6 mg / 100 ml which was extracted at $75^{\circ}C$ for 18 hours. In addition, the saponin content decreased according to the increased extraction temperature and time. The highest total content of $Rb_2$ and Re was 11.8 mg / 100 ml at $75^{\circ}C$ for 12 hours which decreased according to the increased extraction temperature and time. The prosapogenin content of red ginseng extract was increased at $75^{\circ}C$ and $85^{\circ}C$ while the content decreased at $95^{\circ}C$, in which the highest prosapogenin content was 34.9 mg / 100 ml at $85^{\circ}C$ for 24 hours. The total sugar content and cloudness were increased according to the increased extraction time at $95^{\circ}C$, but pH and hue value were decreased according to the increased extracted time. The highest sweetness content was 4.0% which was found at $95^{\circ}C$ for 24 hours extract. Therefore, the most appropriate red ginseng extracting method was lower the temperature for saponin content at first time in combination with raise the temperature for taste at second time.

Effect of Tota1 Saponin from Red Ginseng on Acvtivities of Antioxidant Enzymes in Pregnant Rats (홍삼 사포닌이 수태중인 흰쥐의 항산화 효소활성에 미치는 영향)

  • Pregnancy is a physiological state accompained by a high energy demand of many bodily functions and an increased oxygen requirement. Because of the increased intake and utilization of oxygen, increased levels of oxidative stress would be expected. So we observed the activities of the hepatic antioxidant enzymes from rat treated with total saponin from the red ginseng against free raicals produced in pregnant rats. The activity of superoxide dismutase (SOD) in the control group was slightly decreased during pregnancy, and SOD activity in total saponin treated group was not observed any siginificant change compared with the control group. The activities of glutathione peroxidase (GPX), glutathione reductase (GRD) and catalase in the control group have shown the decreasing tendency during pregnancy, whereas the activities of GRD and catalase in total saponin treated group showed significant increased tendency compared with the control group. GPX activity in total saponin treated group was slightly decreased tendnency compared with the control group. The activity of glutathione-S-transferase (GST) in the control group was increased to keep the state of homaeostasis tendency in pregnant rats. On the other hand, the activity of GST after total saponin treatment was increased than control group. Activity of all enzymes in the control group and total saponin treated group recovered the normal level after delivery of rats. In spite of the physiological changes in vivo, the inflaunce of total saponin on activaties of hepatic antioxidant enzyme in pregnant rats seems to be regulated the biological homeostatic adaptation mechanism which protects the maternal liver aganist oxygen induced toxicity

Effects of Extrusion Conditions on the Physicochemical Properties of Extruded Red Ginseng

  • The effects of variable moisture content, screw speed and barrel temperature on the physicochemical properties of red ginseng powder extrudates were investigated. The raw red ginseng powders were processed in a co-rotating intermeshing twin-screw extruder. Primary extrusion variables were feed moisture content (20 and 30%), screw speed (200 and 250 rpm) and barrel temperature (115 and $130^{\circ}C$). Extruded red ginseng showed higher crude saponin contents (6.72~7.18%) than raw red ginseng (5.50%). Tested extrusion conditions did not significantly affect the crude saponin content of extrudates. Increased feed moisture content resulted in increased bulk density, specific length, water absorption index (WAI), breaking strength, elastic modulus and crude protein content and decreased water solubility index (WSI) and expansion (p<0.05). Increased barrel temperature resulted in increased total sugar content, but decreased reducing sugar content in the extrudate (p<0.05). Furthermore, increased barrel temperature resulted in increased amino acid content and specific length and decreased expansion and bulk density of extrudates only at a higher feed moisture content. The physicochemical properties of extrudates were mainly dependent on the feed moisture content and barrel temperature, whereas the screw speed showed a lesser effect. These results will be used to help define optimized process conditions for controlling and predicting qualities and characteristics of extruded red ginseng.
