Infectious Disease Prevention Act Written on Medical Books in Joseon Dynasty (조선시대 피역의서에 나타난 역병(疫病) 예방법)

  • There were many books on infectious disease prevention act, and still remained 5 books including Ganuibyeokonbang. Epidemics were seriously ill and widely contagious, so it was important to prevent them. Therefore, they wrote various preventive measures from epidemics on those books. They emphasized medication, and used not only compound prescriptions but also singular ones. They wrote 5 compound prescriptions including Sohaphyangwon and many singular ones on Ganuibyeokonbang, and they used folk medicine such as red-beans준 for practical use on that book. On Sinchanbyeokonbang, they emphasized Hyangsosan and presented many prescriptions to specialize in epidemics. Heojun presented various prescriptions for Dangdokyeok on Byeokyeoksinbang, and he excluded incantation methods to cope with epidemics medically. Since Ganuibyeokonbang they had tried to improve personal hygiene such as boiling clothes of patients.

Control Efficacy of Phloretin Isolated from Apple Fruits Against Several Plant Diseases

  • In the course of a searching natural antifungal compounds from plant sources, we found that the methanol extract ($3,000\;{\mu}g/ml$) of Malus domestica fruits had potential of control against rice blast (Magnaporthe grisea) and tomato late blight (Phytophthora infestans). Under bioassay-guided purification, we isolated phloretin, a phenolic compound, with in vivo antifungal activity against M. grisea. By 1-day protective application of phloretin ($500\;{\mu}g/ml$), the compound strongly inhibited the disease development of M. grisea and P. infestans on rice and tomato seedlings, respectively. And red pepper anthracnose caused by Colletotrichum coccodes also was moderately suppressed. However, rice sheath blight (Rhizoctonia solani AG1), and barley powdery mildew (Blumeria graminis f. sp. hordei) were hardly controlled. In addition, the compound showed in vitro antifungal activity against some plant pathogenic fungi including Phytophthora capsici, Alternaria panax, Sclerotinia sclerotiorum, R. solani AG4, and M. grisea. This is the first report on the antifungal activity of phloretin against plant pathogenic fungi.

An Antifungal Compound Against Phytophthora capsici Produced by Streptomyces sp. 3D3 (Streptomyces sp. 3D3 균주가 생산하는 항고추역병성 항생물질)

  • During the screening for the antifungal compounds against Phytophthora capsici causing phytophthora blight of red pepper, we isolated a strong active compound, bafilomycin $C_1$, produced by strain 3D3. The producing organism was identified as Streptomyces sp. based on taxonomic studies. The antifungal compound was purified from culture broth by HP-20 column chromatography, ethylacetate extraction, silica gel column chromatography and HPLC, and was identified as bafilomycin $C_1$ by color reaction, UV and $^{1}H$-NMR spectral data analysis. Bafilomycin $C_1$ showed strong antifungal activity against various phytopathogenic fungi.

Characteristics of Bacteriocin Produced by a Lactobacillus plantarum Strain Isolated from Kimchi (김치에서 분리한 Lactobacillus plantarum 균주가 생산하는 박테리오신의 특성)

  • Lactobacillus plantarum strain KK3 with tannase activity was isolated from Gochunipkimchi (red pepper leaf kimchi) and showed a high antagonistic activity against five kinds of food pathogens. Strain KK3 secreted antibacterial compound into culture medium and 24-h culture in MRS broth at $30^{\circ}C$ was enough for the antibacterial compound production. The crude antibacterial compound prepared from culture supernatant inhibited the growth of some Gram-negative bacteria and Bacillus cereus but not Listeria monocytogenes. The antibacterial activity was sensitive to proteinase K treatment, confirming its proteinaceous nature (bacteriocin). The crude bacteriocin was active in the pH range 3.5-8.5 and extremely stable after 15 min of heat treatment at $121^{\circ}C$. The strain KK3 produced equally active bacteriocin in Chinese cabbage juice as it produced in MRS broth.

  • Light fermentation has been conducted under different light conditions such as normal dark light, white light, and light emitting diodes (LEDs) various color (blue, green, red, white on blueberry powder with fermenting bacteria Bacillus subtilis (B2). The bacteria B2 was isolated and identified by 16S rRNA sequencing method. RYRP biologically converted to secondary metabolites through light fermentation in the presence of Bacillus subtilis, the bacteria actively involved in bioconversion process. LEDs fermentation to enhance the production of phenolic content while comparing to normal dark and white light. Among the different color LEDs, blue LEDs mediated fermentation showed higher amount of total phenolic and flavonoid content. Then blue LEDs mediated fermented compound were characterized by FTIR and GC-MS, subsequently the compound was analyzed antioxidant activity tests and the antioxidant activity exhibited higher. This is the first study to demonstrate that B. subtilis-LEDs mediated fermentation is useful for facilitating phenolic compound production and enhancing antioxidant activity, which may have greater application fermentation fields.

Experimental Study on the Anti-allergic Effects of Gamibojungikgi-tang (가미보중익기탕이 알레르기반응에 미치는 영향에 대한 실험적 연구)

  • This experimental research has been done to study the effects of Gmibojungikgi-tang(GBJIKT) on the anti-allergic reaction. We found the several important results from the research which has been performed by two experiments toward immediately type and delayed type in order to study the effects of GBJIKT on hypersensitivity response to mice. The results obtained from our research are as following: The survival rate of one group to which we injected only the compound 48/80 is almost 0% according to its density and timing test. In the other hand, the survival rates of the other group to which we injected both of the compound 48/80 and GBJIKT are 10%, 10%, 20%, 30%, 20%, and 40% according to 0.025, 0.05, 0.1, 0.25 0.5 and 1(mg/g) of compound 48/80. Time dependency test also shows the 0% survival rates in 5 and 10 minutes. GBJIKT revealed the significantly inhibitory effect on Compound 48/80 induced Mast cell degranulation. GBJIKT showed the significantly inhibitory effect in the delayed type hypersensitivity response to picry1 chloride GBJIKT showed the significantly inhibitory effect in the delayed type hypersensitivity response to sheep red blood cell. Our research provides the important evidence that GBJIKT is benificial to the prevention and treatment of allergy related diseases.

Identification of Irradiation -induced Volatile Marker Compounds in Irradiated Red Pepper Powder (방사선조사 고추가루로부터 휘발성 표지물질의 구명)

  • To develop a new detection method using irradiation-induced volatile marker compounds of red pepper powder (RP), the volatile compounds of irradiated RP (0, 1, 3, 5, and 10 kGy) were analyzed by purge and trap (P&T)/solid phase microextraction (SPME)/gas chromatography/mass spectrometry (GC/MS) methods. A total of 51 and 31 compounds were detected in IRP by SPME and P&T methods, respectively. Among these, 25 compounds, which were composed of 4 hydrocarbons, 7 aldehydes, 1 ketone, 3 alcohols, 4 aromatic compounds, 2 esters and 4 miscellaneous compounds, showed irradiation dependent manner with significant positive correlation (p<0.01 or p<0.05) between irradiation dose and relative concentration. However, all compounds except 1,3-bis(1,1-dimethylethyl)benzene were not suitable as marker compounds because of their low determination coefficients ($R^2$<0.80) between irradiation dose and their concentrations, and detectablilty in nonirradiated sample. Therefore, only one compound, 1,3-bis(1,1-dimethylethyl)benzene, was tentatively identified as a volatile marker compound to detect irradiated RP.

Response of the Growth Characteristics and Phytochemical Contents of Pepper (Capsicum annuum L.) Seedlings with Supplemental LED Light in Glass House (LED 보광처리가 고추(Capsicum annuum) 묘의 생장과 Phytochemical 함량에 미치는 영향)

  • This research was conducted to evaluate the effect of supplemental light-emitting diode (LED) light on growth characteristics and phytochemical content of pepper (Capsicum annuum L.) seedling using LED blue (470 nm, B), red (660 nm, R), blue + red (BR), far red (740 nm, FR) and UV-B (300 nm) light treatment, and without artificial light. Photon flux of LED light was 49, 16, 40, 5.0 and $0.82{\mu}mol\;m^{-2}s^{-1}$ for B, R, BR, FR, and UV-B light, respectively, during experiment. Supplemental LED light duration was $16hr\;day^{-1}$ and UV-B light duration was 10 min. per day after sunset up to 15 days (12 days after germination) of plants age. In our research, growth characteristics and phytochemical content of pepper seedlings were greatly influenced by supplemental LED light compare to control treatment. Red light increased the number of leaves, number of nodes, leaf width and plant fresh weight by 34%, 27%, 50% and 40%, respectively. Blue light increased the leaf length by 13%, and stem length and length of inter node were increased by 17% and 34%, respectively under grown far red light. After 15 days of light treatments phytochemical concentrations of pepper plants were significantly changed. Blue light enhanced the total anthocyanin and chlorophyll concentration by 6 times and 2 times, respectively. Red light increased the total phenolic compound at least two folds meanwhile far red light reduced the ascorbic acid and antioxidant activity 31% and 66%, respectively compared to control treatment.

Concanamycin B, Active substance Against Phytophthora capsici Produced by Streptomyces neyagawaensis 38D10 Strain (Streptomyces neyagawaensis 38D10 균주가 생산하는 concanamycin B의 항고추역병 활성)

  • During the screening of antifungal compounds from microbial secondary metabolites to control phytophthora blight of red pepper caused by Phytophthora capsici, a soil isolate, strain 38D10 was selected. Based on taxonomic studies, this strain was identified as Streptomyces neyagawaensis. The antifungal compound was purified from culture broth by HP-20 column chromatography, ethyl acetate extraction, silica gel column chromatography, HPLC and identified as concanamycin B by UV. $^1H$-NMR, $^{13}C$-NMR, SIMS analysis. Concanamycin B has strong antifungal activity against some phytopathogenic fungi but not antivacterial activity and preventive value were 50% and 100% at 125ppm and 250ppm in pot assay.
