식물 생명공학 기술을 이용해 인간에게 유용한 치료단백질 및 백신을 생산하는 것은 최근에 각광받고 있는 연구 분야이다. 식물을 이용한 유용 단백질 생산은 다른 시스템에 비하여 경제적일 뿐만 아니라 병원성 인자에 대한 안전성이 있어서 유용하다고 할 수 있다. 암세포 표면에 특이적으로 발현하고 있는 분자 와 당 구조를 각각 인지할 수 있는 두 종류의 항체를 동시에 투여하는 면역 치료는 질병의 치료를 유도하는 데 있어서 효과적일 수 있다. 본 연구는 기존에 본 연구팀에서 확보하고 있었던 두 종류의 항체 단백질(mAb CO17-1A, mAb BR55) 생산 형질전환 식물체를 이용하여 상호교배를 통하여 한 식물에서 두 종류의 항체 단백질을 모두 생산하는 식물 발현 시스템 구축에 관한 연구이다. 각기 다른 유전자를 갖고 있는 식물체로부터 수분을 유도하여 씨앗을 얻고 이 씨앗을 배양하여 완벽한 식품 개체로 성장시켰으며, 그 식물체로부터 DNA, RNA, 단백질을 분리하여 형질전환 유전자를 포함하고 있는지 여부를 확인하였다. 그 결과, 개체에 차이는 있지만, 한 식물에서 두 항체 유전자를 갖고 있음을 확인할 수 있었고, 이 유전자는 식물체 내에서 안정적으로 transcription 되었음을 확인하였다. 또한, 두 종류의 항체를 동시 생산하는 식물체에서 분리한 단백질은 한 종류의 항체 단백질만 생산하는 식물체에 비하여 수용성 단백질 단위당 항체 발현률이 높게 나타나는 것을 확인하였다. 따라서 본 연구를 통하여 식물을 이 용한 유용 단백질 생산 효율을 높일 수 있는 시스템을 확립하였으며 앞으로 추가적으로 생산한 항체의 생물학적 활성 및 항암 효능, 당 구조 분석 등에 대한 연구용 수행한다면, 식물 생명공학적 방법을 통한 항체 생산에 대한 새로운 가능성을 제시할 수 있을 것으로 기대된다.
형질전환 동물에 있어서 외래 유전자의 조절되지 않은 과다 발현은 생리적인 부작용이나 독성을 나타내게 된다. 본 연구에서는 이러한 문제를 해결하기 위하여 외래 유전자 발현 조절 system인 tetracycline-inducible expression system(Tet system)을 도입하였다. 그러나 종래의 Tet system을 이용한 유전자 발현 조절은 system 자체의 구성 요소로 인한 미미한 leaky 현상 때문에 완벽하게 이루어지지 않는다. 본 연구에서는 보다 완벽한 외래 유전자의 발현 조절 system을 구축하기 위하여 rtTA 대신 진일보한 형태의 $rtTA2^SM2$를 도입하고 TRE 부분을 TRE-tight로 대체하였다. 확립된 retrovirus vector system을 이용하여 다양한 표적세포와 형질전환 닭으로부터 혈소판 생산의 일차적인 조절자이며, 조혈간세포의 생존과 증식에 있어서 매우 중요한 역할을 하는 human thrombopoietin(hTPO)를 생산하고자 하였다. In vitro 상의 연구에서, CEF 세포에서 발현되는 hTPO가 가장 높은 발현량과 발현 유도율을 나타내었으며, 상업적으로 판매되고 있는 hTPO나 다른 표적세포에서 생산된 hTPO에 비해 생물학적 활성이 가장 높은 것으로 확인되었다. 고농도로 농축한 재조합 retrovirus를 stage X단계의 계란의 배반엽 층에 미세주입하여 대리 난각 방법으로 배양한 결과, 미세주입한 138개의 계란 중 21일 후에 15개의 계란에서 병아리가 부화하였으며, 그 중 8마리가 형질전환 개체로 확인되었다. 이 형질전환 닭은 사료 1 g 당 0.5 mg의 doxycycline을 첨가하여 2주간 식이하였으며, 그 후 혈액을 채취하여 hTPO 농도를 측정한 결과 200ng/mL로 확인되었다. 또한 형질전환 개체 중 수컷의 정자에서 hTPO 유전자의 존재를 확인함으로써 germLine transmission의 가능성을 입증하였다. 이상의 연구 결과는 사람의 cytokine 단백질의 대량생산을 위한 생체반응기로서의 형질전환 닭의 생산 가능성을 제시한 데 의의가 있다.
본 연구에서는 CHO 세포를 이용해 세포를 별도의 적응기간 없이 무혈청 배지에서 배양했을 때 세포의 증식이 중단되는 원인을 찾고 배지 첨가 성분을 통해 이를 개선하고자 했다. 현재 개발된 무혈청 배지는 아직까지 혈청을 대체할 만한 성분을 포함하고 있지 않다. 때문에 무혈청 배지에 적응되지 않은 비적응 세포의 경우 계대 배양에 한계가 있다. 이런 한계가 나타나는 원인은 다양할 것으로 생각이 되지만 혈청의 부재로 인해 세포가 받게 되는 스트레스와 그로 인한 세포주기의 정지가 가장 근본적인 원인으로 생각된다. 무혈청 배지에서 세포가 받는 스트레스의 정도를 알아보고 배양 환경과 첨가물에 따른 ROS 농도의 변화를 측정하기 위해 배지와 세포의 ROS 농도를 측정하였다. ROS 농도를 측정한 결과 무혈청 상태에서 세포내 ROS가 엄청난 양으로 증가하는 것을 알 수 있었다. 이것은 혈청이 항산화능력을 갖고 있어서가 아니라 세포가 무혈청 환경에서 극심한 스트레스상태에 놓이기 때문인 것으로 생각된다. 이렇듯 증가한 ROS가 세포의 증식이 멈추게 되는 원인 중 하나로 생각되고, 항산화제를 첨가한 경우에도 증식력이나 ROS의 농도에 큰 차이가 없었던 것으로 미루어 보아 근본적으로 혈청과 같은 강력하게 증식을 촉진하는 성분을 배지에 첨가해야 할 것으로 여겨진다. ROS 이외에 세포의 증식이 멈추는 또 다른 원인으로 세포사멸의 여부를 확인했다. 무혈청 배지에서 배양한 적응세포와 비적응 세포 모두 특별한 세포사멸의 징후가 나타나지 않았다. 또한 무혈청 배지에서 증식이 멈춘 세포를 회수해 다시 혈청배지에서 배양한 경우 곧바로 증식력이 회복되기 때문에 대규모의 세포사멸은 발생하지 않는 것으로 생각된다. 위와 같은 현상들은 모두 혈청이 없기 때문에 발생하는 것으로 혈청을 대체할 수 있는 첨가물을 배지에 더해주면 세포의 증식이 개선될 것이다. 그래서 몇 가지 첨가물을 이용해 세포의 증식력에 변화가 나타나는지 알아보았다. 첨가물을 이용한 실험에서 IGF-I의 경우 장기간 배양에서 세포의 수를 안정적으로 유지하고 계대 횟수를 증가시키는 효과를 보였다. 이는 IGF-I이 어느정도 세포의 증식을 유지시켜주는 역할을 하기 때문인 것으로 생각된다. 무혈청 배지에서 비적응 CHO 세포의 계대 배양에 한계가 있는 것은 세포주기가 멈추기 때문인 것으로 생각된다. 세포주기가 멈추는 growth factor와 같이 세포의 증식을 지속적으로 유도할 수 있는 물질이 무혈청 배지에서는 부족하기 때문인 것으로 생각되고, IGF-I과 같은 첨가물을 통해 극복할 수 있는 문제라고 여겨진다.
Trichoplusia ni (T. ni) 세포는 사람 당 수송체를 이성질적으로 많은 양 생산하려 할 때 유용하게 사용되는, baculovirus 발현 시스템의 숙주세포로서 이용된다. 그러나 T. ni 세포에 존재하는 내재된 당 수송체의 높은 활동은, 발현된 외재적 당 수송체의 수송활성과 같은 직접적 증거 제시에 장애가 된다. 뿐만 아니라 곤충세포에 내재하는 당 수송체계의 특성에 대해서는 밝혀진 바가 거의 없다. 그래서 본 연구에서는 baculovirus 발현 시스템을 보다 잘 활용하기 위해 T. ni 세포의 2dGlc기질 수송에 D-fructose가 미치는 영향을 살펴 보았으며, T. ni 세포에 발현된 사람 당 수송체의 생물학적 활성을 보다 용이하게 검증하기 위해 발현된 수송체를 [$^3H$] cytochalasin B를 이용하여 photolabelling 하였다. 우선 감염되지 않은 세포와 recombinant AcMPV-GTL 감염시킨 T. ni 세포의 2dGlc uptake를 300 mM D-fructose가 있을 때와 없을 때, 그리고 $20{\mu}M$ cytochalasin B가 있을 때와 없을 때의 상황에서 살펴보았다. 감염되지 않은 세포에서의 육탄당 uptake는 D-fructose에 의해 강력하게 억제 되었으나 cytochalasin B에 의해서는 단지 미미한 억제 효과만을 보여주었다. 흥미롭게도 AcMPV-GTL 바이러스 감염된 T. ni 세포에서는 비록 2dGlc uptake율은 감염되지 않은 세포와 비교해 다소 낮았지만 육탄당 수송 억제 반응은 근본적으로 동일함을 보여 주었다. 또한 [$^3H$] cytochalasin B를 이용한 발현단백질 photolabelling에서는, L-glucose가 존재하는 상황 하에만 하나의 날카롭게 표지된 peak가, 바이러스 감염된 세포에서 관찰되었다. 감염되지 않은 세포에서는 이러한 peak는 관찰되지 않았다. 게다가 D-glucose 존재 하에서는 발현된 단백질의 photolabelling이 완전히 억제되어짐을 보여주어, labelling의 입체선택성(stereoselectivity)을 입증하였다.
TCTP는 다양한 진핵생물에서 풍부하게 존재하는 단백질 중에 하나이며, 암, 세포 증식, 유전자 조절 등과 관련된 세포의 생리학적 기작에서 중요한 역할을 담당하는 것으로 알려져 왔다. 더구나, TCTP는 dithiothreitol (DTT)나 hydrogen peroxide (H2O2)에 의해 유도되는 산화적 스트레스에 대한 저항성에 관여하는 중요한 단백질로 주목 받아 왔다. 한편, 극지역 서식 생물들은 강한 자외선에 의해 발생한 활성산소를 조절하기 위한 다양한 항산화 방어체계를 가지고 있다. 본 연구에서는 북극 동물플랑크톤 Calanus glacialis에서 분리된 TCTP가 산화적 스트레스 하에서 E. coli 세포의 저항성에 미치는 효과를 관찰하였다. C. glacialis에서 분리된 TCTP 유전자(ORF 522 bp) 서열을 분석하였고, 약 23 kDa의 재조합 단백질을 제작하였다. 관찰 결과, TCTP 재조합 단백질이 E. coli 세포에서 과발현 되었을 때, 세포들은 H2O2에 의해 유도된 산화적 스트레스에 대한 저항성이 증가하는 것을 확인하였다. 본 관찰을 통해, 북극 C. glacialis TCTP 단백질의 항산화 조절자로서의 역할에 대한 가능성을 처음으로 제시하였다.
In this study, we have cloned a novel cDNA encoding for a papain-family cysteine protease from the Uni-ZAP XR cDNA library of the polychaete, Periserrula leucophryna. This gene was expressed in Escherichia coli using the T7 promoter system, and the protease was characterized after partial purification. First, the partial DNA fragment (498 bp) was amplified from the total RNA via RT-PCR using degenerated primers derived from the conserved region of cysteine protease. The full-length cDNA of cysteine protease (PLCP) was prepared via the screening of the Uni-ZAP XR cDNA library using the $^{32}P-labeled$ partial DNA fragment. As a result, the PLCP gene was determined to consist of a 2591 bp nucleotide sequence (CDS: 173-1024 bp) which encodes for a 283-amino acid polypeptide, which is itself composed of an 59-residue signal sequence, a 6-residue propeptide, a 218-residue mature protein, and a long 3'-noncoding region encompassing 1564 bp. The predicted molecular weights of the preproprotein and the mature protein were calculated as 31.8 kDa and 25 kDa, respectively. The results of sequence analysis and alignment revealed a significant degree of sequence similarity with other eukaryotic cysteine proteases, including the conserved catalytic triad of the $Cys^{90},\;His^{226},\;and\;Asn^{250}$ residues which characterize the C1 family of papain-like cysteine protease. The nucleotide and amino acid sequences of the novel gene were deposited into the GenBank database under the accession numbers, AY390282 and AAR27011, respectively. The results of Northern blot analysis revealed the 2.5 kb size of the transcript and ubiquitous expression throughout the entirety of the body, head, gut, and skin, which suggested that the PLCP may be grouped within the cathepsin F-like proteases. The region encoding for the mature form of the protease was then subcloned into the pT7-7 expression vector following PCR amplification using the designed primers, including the initiation and termination codons. The recombinant cysteine proteases were generated in a range of 6.3 % to 12.5 % of the total cell proteins in the E. coli BL21(DE3) strain for 8 transformants. The results of SDS-PAGE and Western blot analysis indicated that a cysteine protease of approximately 25 kDa (mature form) was generated. The optimal pH and temperature of the enzyme were determined to be approximately 9.5 and $35^{\circ}C$, respectively, thereby indicating that the cysteine protease is a member of the alkaline protease group. The evaluation of substrate specificity indicated that the purified protease was more active towards Arg-X or Lys-X and did not efficiently cleave the substrates with non-polar amino acids at the P1 site. The PLCP evidenced fibrinolytic activity on the plasminogen-free fibrin plate test.
Background: Ginseng effectively reduces fatigue in both animal models and clinical trials. However, the mechanism of action is not completely understood, and its molecular targets remain largely unknown. Methods: By screening for proteins that interact with the primary components of ginseng (ginsenosides) in an affinity chromatography assay, we have identified muscle-type creatine kinase (CK-MM) as a potential target in skeletal muscle tissues. Results: Biolayer interferometry analysis showed that ginsenoside metabolites, instead of parent ginsenosides, had direct interaction with recombinant human CK-MM. Subsequently, 20(S)-protopanaxadiol (PPD), which is a ginsenoside metabolite and displayed the strongest interaction with CK-MM in the study, was selected as a representative to confirm direct binding and its biological importance. Biolayer interferometry kinetics analysis and isothermal titration calorimetry assay demonstrated that PPD specifically bound to human CK-MM. Moreover, the mutation of key amino acids predicted by molecular docking decreased the affinity between PPD and CK-MM. The direct binding activated CK-MM activity in vitro and in vivo, which increased the levels of tissue phosphocreatine and strengthened the function of the creatine kinase/phosphocreatine system in skeletal muscle, thus buffering cellular ATP, delaying exercise-induced lactate accumulation, and improving exercise performance in mice. Conclusion: Our results suggest a cellular target and an initiating molecular event by which ginseng reduces fatigue. All these findings indicate PPD as a small molecular activator of CK-MM, which can help in further developing better CK-MM activators based on the dammarane-type triterpenoid structure.

Recent studies showed that the PD-1/PD-L1 checkpoint blockade is a dramatic therapy for melanoma by enhancing antitumor immune activity. Currently, major strategies for the PD-1/PD-L1 blockade have mainly focused on the use of antibodies and compounds. Seeking an alternative approach, others employ endogenous proteins as blocking agents. The extracellular domain of PD-1 (ePD1) includes the binding site with PD-L1. Accordingly, we constructed a PD-1-based recombinantly tailored fusion protein (dFv-ePD1) that consists of bivalent variable fragments (dFv) of an MMP-2/9-targeted antibody and ePD1. The melanoma-binding intensity and antitumor activity were also investigated. We found the intense and selective binding capability of the protein dFv-ePD1 to human melanoma specimens was confirmed by a tissue microarray. In addition, dFv-ePD1 significantly suppressed the migration and invasion of mouse melanoma B16-F1 cells, and displayed cytotoxicity to cancer cells in vitro. Notably, dFv-ePD1 significantly inhibited the growth of mouse melanoma B16-F1 tumor cells in mice and in vivo fluorescence imaging showed that dFv-ePD was gradually accumulated into the B16-F1 tumor. Also the B16-F1 tumor fluorescence intensity at the tumor site was stronger than that of dFv. This study indicates that the recombinant protein dFv-ePD1 has an intensive melanoma-binding capability and exerts potent therapeutic efficacy against melanoma. The novel format of the PD-L1-blocked agent may play an active role in antitumor immunotherapy.
Edwardsiella tarda는 그람 음성의 장내세균과의 주요 어병세균으로 어류에 edwardsiellosis를 유발하는 전신감염성 병원체이다. 최근 병원성 세균의 외막 단백질들은 세균성 감염에 있어서 숙주와 반응하여 면역반응을 유도하는 것으로 여겨져 연구가 되고 있다. 일본의 연구팀은 어류에서 에드워드병의 원인체인 E. tarda의 37 kDa 단백질이 넙치에서 높은 항원성을 제시하는 것을 보고하였다. 또한 그 연구자들은 37 kDa 단백질의 N-말단 아미노산 서열이 GAPDH와 대응하는 것을 밝혔다. 본 연구에서는 다른 세균에서 알려진 N-말단 서열을 기반으로 primer를 제작하여 이에 상응하는 E. tarda DNA를 증폭하고 클로닝하였다. 이 DNA단편의 염기서열은 예상한 바와 같이 세균의 GAPDH유전자인 gapA와 높은 상동성이 있고, E. tarda GAPDH (etGAPDH)의 아미노산 서열은 다른 장내세균의 GAPDH와 70% 이상의 상동성을 보이는 것을 확인하였다. E. tarda의 외막단백질에 특이적으로 반응하는 항체를 이용하여 E. tarda의 GAPDH가 외막에 존재한다는 것을 증명하였고, gapA의 염기서열을 바탕으로하여 재조합 GAPDH를 과발현 시켰다. 과발현된 재조합단백질 GAPDH는 GAPDH 특이적인 항체를 제조하는데 사용되었고, 또한 넙치에 면역시켜 단일 단백질 백신으로서의 활용도를 모색하였다. 비록 재조합 GAPDH가 면역된 넙치에서 GAPDH에 특이적인 항체가 증가하였음에도 불구하고, E. tarda로 공격실험을 하였을 때 면역된 넙치의 생존율이 12.5%로 측정되어 면역된 그룹과 면역되지 않은 그룹간에 큰 차이가 없는 것이 확인되었다.
A commensal thermophile, Symbiobacterium toebii, isolated from hay compost (toebii) in Korea commensally interacted with a thermophilic Geobacillus toebii sp. nov., which was a new species within the genus Geobacillus on the basis of the phenotypic traits and molecular systematic data. S. toebii required the crude extracts and/or culture supernatant of the Geobacillus toebii for axenic growth and could grow on the temperature between 45 and $70^{\circ}C$ (optimum: $60^{\circ}C$; 2.4 h doubling time) and pH 6.0 and 9.0 (optimum: pH 7.5). The G+C content of the genomic DNA was $65 mol\%$, and the major quinones were MK-6 and MK-7. A phylogenetic analysis of its 16S rDNA sequence indicated that Symbiobacterium toebii was closely related with solely reported Symbiobacterium thermophilum. The presence of the commensal thermophile 16S rDNA and accumulation of indole in all the enriched cultures indicate that Symbiobacterium toebii is widely distributed in the various soils. The genome of S. toebii constituted a circular chromosome of 3,280,275 base pairs and there was not an extra-chromosomal element (ECE). It contained about 4,107 predicted coding sequences. Of these protein coding genes, about $45.6\%$ was encoded well-known proteins and annotated the functional assignment of 1,874 open reading frames (ORFs), and the rest predicted to have unknown functions. The genes encoding thermostable tyrosine phenol-lyase and tryptophan indole-lyase were cloned from the genomic DNA of S. toebii and the enzymatic production of L-tyrosine and L-tryptophan was carried out with two thermostable enzymes overexpressed in recombinant E. coli.

제 11 조 (회원의 의무)

제 4 장 서비스의 이용

① NDSL에서 제공하는 모든 저작물의 저작권은 원저작자에게 있으며, KISTI는 복제/배포/전송권을 확보하고
있습니다.
② NDSL에서 제공하는 콘텐츠를 상업적 및 기타 영리목적으로 복제/배포/전송할 경우 사전에 KISTI의 허락을
받아야 합니다.
③ NDSL에서 제공하는 콘텐츠를 보도, 비평, 교육, 연구 등을 위하여 정당한 범위 안에서 공정한 관행에
합치되게 인용할 수 있습니다.
④ NDSL에서 제공하는 콘텐츠를 무단 복제, 전송, 배포 기타 저작권법에 위반되는 방법으로 이용할 경우
저작권법 제136조에 따라 5년 이하의 징역 또는 5천만 원 이하의 벌금에 처해질 수 있습니다.
제 14 조 (유료서비스)
① 당 사이트 및 협력기관이 정한 유료서비스(원문복사 등)는 별도로 정해진 바에 따르며, 변경사항은 시행 전에
당 사이트 홈페이지를 통하여 회원에게 공지합니다.
② 유료서비스를 이용하려는 회원은 정해진 요금체계에 따라 요금을 납부해야 합니다.
제 5 장 계약 해지 및 이용 제한
제 15 조 (계약 해지)
회원이 이용계약을 해지하고자 하는 때에는 [가입해지] 메뉴를 이용해 직접 해지해야 합니다.
제 16 조 (서비스 이용제한)
① 당 사이트는 회원이 서비스 이용내용에 있어서 본 약관 제 11조 내용을 위반하거나, 다음 각 호에 해당하는
경우 서비스 이용을 제한할 수 있습니다.
- 2년 이상 서비스를 이용한 적이 없는 경우
- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
