Identification of Actinobacillus pleuropneumoniae Genes Preferentially Expressed During Infection Using In Vivo-Induced Antigen Technology (IVIAT)

  • Porcine pleuropneumonia is an infectious disease caused by Actinobacillus pleuropneumoniae. The identification of A. pleuropneumoniae genes, specially expressed in vivo, is a useful tool to reveal the mechanism of infection. IVIAT was used in this work to identify antigens expressed in vivo during A. pleuropneumoniae infection, using sera from individuals with chronic porcine pleuropneumonia. Sequencing of DNA inserts from positive clones showed 11 open reading frames with high homology to A. pleuropneumoniae genes. Based on sequence analysis, proteins encoded by these genes were involved in metabolism, replication, transcription regulation, and signal transduction. Moreover, three function-unknown proteins were also indentified in this work. Expression analysis using quantitative real-time PCR showed that most of the genes tested were up-regulated in vivo relative to their expression levels in vitro. IVI (in vivo-induced) genes that were amplified by PCR in different A. pleuropneumoniae strains showed that these genes could be detected in almost all of the strains. It is demonstrated that the identified IVI antigen may have important roles in the infection of A. pleuropneumoniae.

  • I will introduce robotics education, Novel Engineering, STOMP have hosted in CEEO at Tufts University. CEEO is a leading center for Robotics Education in all over the USA. So far lots of universities and centers have adopted these programs. CEEO have hosted many workshops for students and teachers every summer vacation. And Simple Robotics course is required for every Engineering department freshmen. Novel Engineering(NE) is not familiar to us. I think that NE will be a good tool for integrating reading literacy and engineering. When teachers design a engineering class, members of STOMP help teachers achieve the goals of class. Software Education has been widely spreaded for last several years in South Korea. If our country adopts these programs government-widely. I think that It will be able to contribute to foster SW-talented persons.

A Gene-Tagging System for Monitoring of Xanthomonas Species

  • A novel chromosomal gene tagging technique using a specific fragment of the fatty acid desaturase-like open reading frame (des-like ORF) from the tox-argK gene cluster of Pseudomonas syringae pv. phaseolicola was developed to identify Xanthomonas spp.released into the environment as biocontrol agents. X. campestris pv. convolvuli FB-635, a pathogen of Convolvulus arvensis L., (bindweed), was chosen as the organism in which to develop and test the system. A 0.52 kb DES fragment amplified from P. syringae pv. phaseolicola C-199 was inserted into pGX15, a cosmid clone containing a 10.3 kb Eco RI-HindIII fragment derived from the xanthomonadin biosynthetic gene cluster contained in plasmid pIG102, to create a pigG::DES insertion. The 10.8 kb EcoRI-BamHI fragment carrying the pigG:: DES insertion was cloned into pLAFR3 to generate pLXP22. pLXP22 was then conjugated into X. campestris pv. convolvuli FB-635 and the pigG::DES insertion integrated into the bacterial chromosome by marker exchange. Rifampicin resistant, tetracycline sensitive, starch hydrolyzing, white colonies were used to differentiate the marked strain from yellow pigmented wild-type ones. PCR primers specific for the unique DES fragment were used for direct detection of the marked strain. Result showed the marked strain could be detected at very low levels even in the presence of high levels of other closely related or competitive bacteria. This PCR-based DES-tagging system provides a rapid and specific tool for directly monitoring the dispersal and persistence of Xanthomonas spp.released into the environment.

Computer-Aided Diagnosis for Pulmonary Tuberculosis using Texture Features Analysis in Digital Chest Radiography (질감분석을 이용한 폐결핵의 자동진단)

  • There is no exact standard of detecting pulmonary tuberculosis(TB) in digital image of simple chest radiography. In this study, I experimented on the principal components analysis(PCA) algorithm in the past and suggested six other parameters as identification of TB lesions. The purpose of this study was to develop and test computer aided diagnosis(detection) method for the detection and measurement of pulmonary abnormalities on digital chest radiography. It showed comparatively low recognition diagnosis rate using PCA method, however, six kinds of texture features parameters algorithm showed similar or higher diagnosis rates of pulmonary disease than that of the clinical radiologists. Proposed algorithms using computer-aided of texture analysis can distinguish between areas of abnormality in the chest digital images, differentiate lesions having pulmonary disease. The method could be useful tool for classifying and measuring chest lesions, it would play a major role in radiologist's diagnosis of disease so as to help in pre-reading diagnosis and prevention of pulmonary tuberculosis.

Deleuze and Guattari's Death, the Minor Literature and the Minor Philosophy of Education (소수자 문학과 문학교육의 과제)

  • This essay examines the relationship between death and the minor literature and the minor philosophy of education. Deleuze and Guattari conceive death as the source of question and the problem. Death overcomes Freud's death-drive and is conceived as the universal event. Then, death is understood as the essential problem, the condition of thought about life, and the paradox of actuality. On the other hand, their minor literature is able to figure in Kafka's works, Metamorphosis and so on, excellently. Deleuze and Guattari argue that Kafka's works push ahead to the edge of the deterritorization. Their argument on the minor literature has the following three characteristics: first, the territorization of the language; second, the directional connection between the individual and the social; third, the collective arrangement of the statement. Death and the minor literature share the "intensity" as the theoretical tool and thus presuppose each other. From this, we are able to draw the possibility of the minor philosophy of education. It makes a crack on the major philosophy of education, exposing the limitation of the doctrines. In other words, the major philosophy of education enforces the reading of grand philosophers by the defined ways only. Instead, Deleuze and Guattari show the creative way through their research of the history of philosophy and extend the field of thought. In following this way, we can repeat the novel in the philosophy of education also. In this essay, we examine their "affect" for the possibility.

  • In an advanced computer and information network technologies, users are rapidly approaching environment in which information will be created, delivered, managed and stored, using vastly different methods to traditional pap r based systems. Information seeking behavior contains not only physical access, but also cognitive process in which reading, viewing, analyzing, reasoning etc. The most Important factor determining the usability of electronic documents is cognitive process. But, currently Navigation tools for using electronic documents depend on physical searching without considering cognitive process. Therefore, this study examined feasibility of using overal cognitive process in developing navigation tools. Six cognitive process style have been analyzed to find common skill of information seeking process. Also this study suggests skills that needs to introduce to functions of navigation tool, such as searching, filtering, visualization, traversal, content structure.

  • This study aims to investigate the effects of literary texts as a teaching tool to enhance college students' English communicative competence both in linguistic aspects and affective ones. The control group used only the course book as study material, whereas the target group read four short stories along with it and engaged in a series of follow-up tasks. To measure their English competence, the researcher had both groups take a pre-test and a post-test, compared the results, and analyzed the data using SPSS. The study indicates that though the target students' post-test scores increased, the result failed in reaching a significant level. Nevertheless, reading and discussing literature facilitated the target students' affective aspects of communication. This article points out some other limitations of utilizing literary texts in language teaching and suggests the need for further research to deal with the issues.

GENOME STRUCTURE OF Bombyx mori NUCLEOPOLYHEDROVIRUS

  • Baculoviruses are characterized by large double-stranded circular DNA genomes and rod-shaped enveloped virions. Bombyx mori nucleopolyhedrovirus(BmNPV) is a major pathogen, which causes severe damage in sericulture. Currently, BmNPV is recogtnized as an improtant tool in molecular biology, especially for expression of useful genes in B.mori cells and silkworm larvae. Our laboratories have focused on the studies of the molecular mechanisms of BmNPV replication and the application of BmNPV to agriculture and medicine. The entire nucleotide sequence of the BmNPV genome has recently determined. The BmNPV genome possessed 135 putative genes and 7 homologous repeated sequence (hrs) regions. Relatively little space, a few to a few hundred base-pairs, was observed between the open reading frames and hrs. Termination codons often overlapped. These results showed a compactly packde BmNPV genome. Based on comparative sequence analyses, we speculated that the ancestor of BmNPV was a baculovirus similar to Autographa californica NPV(AcNPV). The function of the BmNPV genes were characterized by gene deletion analysis; p35 was found to be involved in blocking apoptosis and cysteine proteinase was found to be involved in horizontal virus transmission by degrading viral-infected larval host. By AcNPV and BmNPV coinfection experiments, we identified a BmNPV gene involved in expanding host specificity of AcNPV. The identified gene was likely encoded a DNA helicase based on the amino acid sequence analysis; a few amino acid substitutions in the putative DNA helicase gene resulted in the expansion of host range of AcNPV. These findings indicate that BmNPV evolved within a short period from an AcNPV-like ancestral virus due to rapid evolution including specific amino acid substitutions and gene deletions/insertions.
