A two-stage continuous stirred tank reactor (CSTR)/trickling biofilter reactor (TBR) system was developed for the degradation of gas-phase trichloroethlene (TCE) using Methylosinus trichoporium OB3b. Mrthylosinus trichosporium OB3b was immobilized on activated carbons in TBR and the microbial growth reactor of a CSTR was coupled for the reactivation of the deactivated cells during TCE degradation. The effect of operation variables on TCE conversion and degradation rate were studied. At inlet TCE concentrations ranging from 10 to 80 $\mu$mol/L, TCE degradation rate was increased up to 525 mg TCE/Lㆍday with 75% conversion. The TCE degradation rates were also increased with increse in broth recycle flow rate, gas flow rate and dilution rate. When the temperature of TBR was changed from 3$0^{\circ}C$ to 15$^{\circ}C$, TCE degradation rate and TCE conversion were increased due to the enhanced TCE transfer from gas-phase. The two-stage reactor system was found to be stable and has been operated for more than 270 days.
Hypersensitivity of cells lacking Brcal to DNA interstrand .ross-link (ICL) agents such as cisplatin and mitomycin C(MMC) implicates the important role of Brcal in cellular response following ICL treatment. Brca1 plays an essential role in DNA double-strand break (DSB) repair through homologous recombination (HR)-dependent and -independent process. Recently, our group has been reported that Brca1 involves in cellular ICL response through HR-dependent repair process (Yun J. et at., Oncogene 2005). In this report, the involvement of Brca1 protein in HR-independent repair process is examined using isogenic $p53^{-/-}\;and\;p53^{-/-}\;Brcal^{-/-}$ mouse embryonic fibroblast (MEF) and psoralen cross-linked reporter reactivation assay. Brcal-deficient MEFs showed significantly low HR-independent repair activity compare to Brca1-proficient MEFs. Hypersensitivity to MMC and ICL reporter repair activity were restored by the reconstitution of Brca1 expression. Interestingly, MEFs expressing exon 11-deleted isoform of Brca1 $(Brca1^{\Delta11/\Delta11})$ showed high resistance to MMC and ICL reporter repair activity comparable to Brca1-reconstituted MEFs. Taken together, these results suggest that Brca1 involves in ICL repair through not only HR-dependent process but also HR-independent process using N-terminal RINC finger domain or C-terminal BRCT domain rather than exon 11 region which mediate interaction with Rad50.
Luminescence dating of a coastal dune from the Shindu dunefield on the T'aean Peninsula shows that deposition of the dune structure began about 500-600 years ago. The lower section of the dune has remained stable since then but the upper part yields an age of about 30 years, suggesting reactivation or additional deposition since the 1970's. The two samples that were collected from the lower part of the dune at depths of 3.5 m and 2.0 m below the surface differ by an age interval of about 50-70 years. This indicates a net depositional rate of around 2.5 cm a year which is relatively slow for a coastal dune. Whilst only one dune structure has been dated for the time being and even though the dunefield was probably established much earlier in the Holocene, the OSL ages obtained demonstrate that some dunes in the area could be younger than 1000 years. Such chronologies point to a dynamic environment where the dune structures are not permanently fixed. Sedimentological properties of the dune sands are consistent with those of particles initially deposited under subaqueous conditions and then later transported by wind.
Raney nickel was used as catalyst in the hydrogen electrode for an alkaline fuel cell. The hydrogen electrode manufactured with the Raney nickel catalyst which was sintered at $700^{\circ}C$ was found to have the highest electrode performance. Using the Raney nickel powder of average particle size $90{\AA}$ for the electrode, the current density which had been measured was $450mA/cm^2$ at $80^{\circ}C$ using 6N KOH solution as an electrolyte. The effects of PTFE addition were investigated with CO-chemisorption, polarization curves and Tafel slope. CO-chemisorption had shown the optimum value when the Raney nickel was mixed with 5wt% of PTFE, but from the current density and Tafel slope at porous Raney nickel electrode, the appropriate value of PTFE addition was 10wt%. Recommendable Ni and Al portion for Raney nickel was 60 : 40 and loading amount was $0.25g/cm^2$. Also the influence of pressing pressure for manufacturing catalytic layer and for junction with gas diffusion layer was examined. The morphology of catalyst surface was investigated with SEM. The influence of reactivation time and heat-treatment temperature were also studied.
Since most large earthquakes occur by reactivation of preexisting active faults, it is important to understand the locations and characteristics of active faults in terms of earthquake hazard research and earthquake disaster prevention. Recently, several remote sensing techniques are broadly used for lineament analysis performed prior to field surveys in active fault surveys. The aim of this paper is introducing simple principles and application examples of each remote sensing technique (satellite remote sensing, airborne remote sensing, InSAR, LiDAR) widely used for active fault investigation. This paper also explains the analytical methods for the slope break generated by fault activity based on GIS and the horizontal displacement of the strike-slip fault. In discussion, we would like to discuss the problems and solutions on making DEM based on aerial photography, and a new developed technique (RRIM) to overcome the problems of DEM based on aerial LiDAR. Understanding remote sensing techniques used for active fault investigation and utilizing appropriate methods depending on the situation and limitations of each remote sensing technique are important for effective active fault investigation.
Jinsan gold deposit is a hydrothermal vein type deposit consisting of several fissure filling quartz veins developed within the Changri Formation of the Ogcheon Supergroup in Geumsan, Chungnam. This study is to provide an efficient exploration and development strategies based on the characteristics of the geology, geological structure, core logging, and ore vein occurrence and grade for the four pits (New pit, Main pit, Yanghapan pit and Teugho pit). Quartz veins are mostly developed with the strike of $N10^{\circ}-25^{\circ}W$ and $N5^{\circ}-20^{\circ}E$, and the thickness is in the range of 0.1~0.5 m, sometimes extending to over 1m. Although the quartz veins commonly form massive shape, they sometimes show zonal structure, comb structure as well as brecciated texture. Major ore minerals are pyrite and chalcopyrite, and pyrrhotite, sphalerite, galena, marcasite, electrum and chalcocite are also accompanied as minor phases. Gray and milky white quartz veins, which are occasionally crosscut by calcite vein, also include fluorite. Ore evaluations for the 22 samples revealed that the samples from the pits generally have very low Au contents, lower than 1 g/t, but some clay samples of drilled core show very high Au concentrations, up to 141 g/t, indicating that Au content is much higher within fault gouges rather than within fresh quartz veins. This may represent that gold might have been reworked and reprecipitated by hydrothermal fluids in association with reactivation of the faults, and thus suggest that ore occurrence in this deposit is very complex and irregular and therefore more precise and systematic exploration is required.
The Pungcheon-myeon, Andong, consists mainly of Precambrian metamorphic rocks, Jurassic igneous rocks, Cretaceous sedimentary rocks (Hasandong, Jinju and Iljik Formations) and Cretaceous igneous rocks (gabbroic rocks, dykes), in which several major faults are developed; Andong fault of ENE trend, which is the boundary fault of the Cretaceous Gyeongsang Basin and the Precambrian-Jurassic basement (Yeongnam Massif), Namhu fault parallel to it, Maebong fault of NNW direction, bow-shaped Gwangdeok fault of ENE direction which is convex toward SSE direction, and Hahoe fault of NNE direction. This paper is researched the geological structures around these major faults by means of the detailed geometric analysis on beddings, joints, faults and drag folds. As a result, a reverse slip faulting of top-to-the SSE movement accompanied with a regional drag folding is recognized from the arrangement of bedding poles measured around the Gwangdeok and Hahoe faults at its northeastern extension, and a zone of Gwangdeok drag fold of 150-300 m width, which is wider at the central and eastern parts of Gwangdeok fault and narrower at its western part and Hahoe fault, is also defined. It indicates that the Hahoe and Gwangdeok faults are a single fault and their movements are coeval unlike the results of earlier reasearchers. And, In this area are recognized two types of faults [(E)NE${\sim}$EW(fault I), WNW${\sim}$NNW (fault II), trending faults] and four types of joints [EW (I), (N)NW (II), NNE (III), NE (IV) trending joints]. These fractures were formed at least through four different events, named as Dn to Dn+3 phases. (1) Dn phase; the formation of joint (I) (Gwangdeok joint) and the intrusion of acidic dykes of EW trend under the compression of EW direction. (2) Dn+1 phase; the formations of joint (II) (Maebong joint), lens-shaped boudinage of acidic dykes, oblique-slip reverse fault (Fault I-Gwangdeok fault) under the compression of (N)NW direction, and the formation of regional zone of Gwangdeok drag fold accompanying the Gwangdeok faulting. (3) Dn+2 phase; those of joint (III), Fault II (Maebong fault) by dextral strike-slip movement of Maebong joint under the compression of NNE direction, and the extension cutting of Dn+1 structures due to the Maebong faulting. (4) Dn+3 phase; the jointing (IV) and the reactivation of Fault II as oblique-slip type with predominant dextral motion which took place under the compression of NE direction. It also suggests that the Maebong fault is not a tear fault deveolped during thrust tectonics of the Andong and Gwangdeok faults but is a post-fault during different tectonic event.
The Quaternary faults are extensively observed along major inherited fault zones (i.e. Yangsan Fault System, Ulsan Fault, Yeonil Tectonic Line, Ocheon Fault System) in SE Korea. Their geometry and kinematics provide a very useful piece of information about the Quaternary crustal deformation and stress field in and around Korean Peninsula. Using magnetic fabrics (AMS), we attempted to determine the slip senses of Jinti, Mohwa, Suseongji2, and Wangsan faults and then interpreted the fabric development process of fault gouge and the characteristics of stress field during the Quaternary. All the magnetic fabrics of the faults, except the Wangsan Fault, consistently indicate a dominant reverse-slip sense with weak strike-slip component. Most of the oblate fabrics are nearly parallel to the fault surface and the anisotropy degrees generally increase in proportion to the oblatenesses. These results suggest that the fabrics of the fault gouges resulted from a progressive deformation due to continuous simple shear during the last reactivation stage as reverse faulting. It is also interpreted that the pre-existing fabrics were overwhelmed and obliterated by the re-activated faulting. Paleostress field calculated from the fault slip data indicates an ENE-WNW compressive stress, which is in accord with those determined from previous fault tectonic analysis, focal mechanism solution, and hydraulic fracturing test in and around Korean Peninsula.
The p16 protein is a cyclin dependent kinase inhibitor that inhibits cell cycle progression from $G_1$ phase to S phase in cell cycle. Many p16 gene mutations have been noted in many cancer-cell lines and in some primary cancers, and alterations of p16 gene function by DNA methylation have been noticed in various kinds of cancer tissues and cell-lines. There have been a large body of literature has accumulated indicating that abnormal patterns of DNA methylation (both hypomethylation and hypermethylation) occur in a wide variety of human neoplasma and that these aberrations of DNA methylation may play an important epigenetic role in the development and progression of neoplasia. DNA methylation is a part of the inheritable epigenetic system that influences expression or silencing of genes necessary for normal differentiation and proliferation. Gene activity may be silenced by methylation of up steream regulatory regions. Reactivation is associated with demethylation. Although evidence or a high incidence of p16 alterations in a variety of cell lines and primary tumors has been reported, that has been contested by other investigators. The precise mechanisms by which abnormal methylation might contribute to carcinogenesis are still not fully elucidated, but conceivably could involve the modulation of oncogene and other important regulatory gene expression, in addition to creating areas of genetic instability, thus predisposing to mutational events causing neoplasia. There have been many variable results of studies of head and neck squamous cell carcinoma(HNSCC). This investigation was studied on 13 primary HNSCC for p16 gene status by protein expression in immunohistochemistry, and DNA genetic/epigenetic analyzed to determine the incidence, the mechanisms, and the potential biological significance of its Inactivation. As methylation detection method of p16 gene, the methylation specific PCR(MSP) is sensitive and specific for methylation of any block of CpG sites in a CpG islands using bisulfite-modified DNA. The genomic DNA is modified by treatment with sodium bisulfate, which converts all unmethylated cytosines to uracil(thymidine). The primers designed for MSP were chosen for regions containing frequent cytosines (to distinguish unmodified from modified DNA), and CpG pairs near the 5' end of the primers (to provide maximal discrimination in the PCR between methylated and unmethylated DNA). The two strands of DNA are no longer complementary after bisulfite treatment, primers can be designed for either modified strand. In this study, 13 paraffin embedded block tissues were used, so the fragment of DNA to be amplified was intentionally small, to allow the assessment of methylation pattern in a limited region and to facilitate the application of this technique to samlples. In this 13 primary HNSCC tissues, there was no methylation of p16 promoter gene (detected by MSP and automatic sequencing). The p16 protein-specific immunohistochemical staining was performed on 13 paraffin embedded primary HNSCC tissue samples. Twelve cases among the 13 showed altered expression of p16 proteins (negative expression). In this study, The author suggested that low expression of p16 protein may play an important role in human HNSCC, and this study suggested that many kinds of genetic mechanisms including DNA methylation may play the role in carcinogenesis.
The Gilan area in the central-northern part of Uiseong Block of Cretaceous Gyeongsang Basin is composed of Precambrian metamorphic rocks, Triassic Cheongsong granite, Early Cretaceous Hayans Group, and Late Cretaceous-Paleocene igneous rocks. In this area, the faults of various directions are developed: Oksan fault of $NS{\sim}NNW$ trend, Gilan fault of NW trend, Hwanghaksan fault of WNW trend, and Imbongsan fault of EW trend. Several fracture sets with various geometric indicators, which determine their relative timing (sequence and coexistence relationships) and shear sense, we well observed in the Cheongsong granite, the basement of Gyeongsang Basin. The aim of this study is to determine the development sequence of extension fractures and the movement sense of shear fractures in the Gitan area on the basis of detailed analysis of their geometric indicators (connection, termination, intersection patterns, and cross-cutting relations). This study suggests that the fracture system of the Gilan area was formed at least through seven different fracturing events, named as Pre-Dn to Dn +5 phases. The orientations of fracture sets show (W) NW, NNW, NNE, EW, NE in descending order of frequency. The orientation and frequency patterns are concordant with those of faults around and in the Gilan area on a geological map scale. The development sequence and movement sense of fracture sets are summarized as follows. (1) Pre-Dn phase: extension fracturing event of $NS{\sim}NNW$ and/or $WNW{\sim}ENE$ trend. The joint sets of $NS{\sim}NNW$ trend and of $WNW{\sim}ENE$ trend underwent the reactivation histories of sinistral ${\rightarrow}$dextral${\rightarrow}$sinistral shearing and of (dextral${\rightarrow}$) sinistral shearing with the change of stress field afterward, respectively. (2) Dn phase: that of NW trend. The joint set experienced the reactivations of sinistral${\rightarrow}$dextral shearing. (3) Dn + 1 phase: that of $NNE{\sim}NE$ trend. The joint set was reactivated as a sinistral shear fracture afterward. (4) Dn +2 phase: that of $ENE{\sim}EW$ trend. (5) Dn +3 phase: that of $WNW{\sim}NW$ trend. (6) Dn+4 phase: that of NNW trend. The joint set underwent a dextral shearing after this. (7) The last Dn +5 phase: that of NNE trend.
