In this study, we investigated the effects of linoleic acid (LA) treatment on Brucella abortus infection in professional phagocyte RAW264.7 cells, particularly during the pathogen's invasion and intracellular growth in these cells, as well as in murine model BALB/c mice focusing on bacterial splenic proliferation and immunoregulatory activities. LA inhibited the growth of Brucella in a dose- and time-dependent manner. The ability of the pathogen to enter the phagocytes was inhibited as was its survival within these cells. This was accompanied by increased nitrite accumulation in these cells at 24 h post-infection. The concentration of LA used in the present study did not affect the total body weight or liver function of the mice. During Brucella infection, the total splenic weight of these animals was not changed; rather, resistance to bacterial proliferation was enhanced in the spleen. Furthermore, mice treated with LA displayed elevated levels of IL-12 and IFN-γ but reduced levels of IL-10 during infection. The findings in this study showed the regulatory role of LA against B. abortus infection suggesting its potential use in designing intervention strategy for brucellosis.
QA 과정을 통해 30개의 정규화 보정값을 얻고, 1개의 기준 데이터 설정 후 29개의 정규화 보정값과 비교하였다. 또한 각 정규화 보정값을 PET raw data 에 적용시켜 재구성된 영상을 동일한 ROI를 그려 SUV 와 count 로 비교평가 하였다. 30개의 정규화 보정값은 차이가 발생하지 않았으며, 각각의 정규화 보정값을 적용시킨 PET 영상도 크게 차이가 없었다. 핵의학 영상은 정량분석을 위해 불감시간 보정 및 광자의 감쇠 및 산란 등 물리적 현상에 대한 보정이 필요하고, 정규화 보정 또한 무시할 수 없는 보정 과정이다. 따라서 이런 과정을 통해 PET 장비의 균일성이 유지되고 여러 디텍터의 효율도 균등해야 영상의 왜곡을 방지할 수 있고, 질 좋은 영상을 만드는데 꼭 필요한 과정이므로 매일 QA 과정이 필요하다고 사료된다.
Purpose: This study was performed to evaluate anti-oxidant activities and anti-inflammatory effects of Sungyoutanggagambang(SYTG). Methods: In the study of anti-oxidant activities. SYTG was investigated by DPPH radical scavenger activity. superoxide dismutase activity and superoxide anion radical scavenger activity. In the study of anti-inflammatory effects. SYTG was investigated using cultured cells and murine models. As for the parameters of inflammation. levels of several inflammatory cytokines and chemical mediators which are known to be related to inflammation were measured in mouse lung fibroblast cells(mLFCs) and RAW264.7 cells. Results: Prior to the experiment. we investigated the security of SYTG by measuring GOT and GPT in serum. 1. SYTG showed high antioxidant activity in a concentration-dependent degree by measured scavenging activity of DPPH free radical, superoxide dismutase and superoxide anion radical. 2. SYTG inhibited IL-1$\beta$, IL-6. TNF-$\alpha$, COX-2 and NOS-II mRNA expression as compared with the control group in a concentration-dependent degree in RAW264.7 cell line. 3. SYTG inhibited IL-1$\beta$, IL-6 production significantly at 100 ${\mu}g/ml$ and TNF-$\alpha$ production significantly at 50, 100 ${\mu}g/ml$ as compared with the control group in RA W264.7 cell line. 4. SYTG inhibited IL-1$\beta$, and IL-6 production significantly as compared with the control group in serum of acute inflammation-induced mice. and decreased IL-1$\beta$, IL-6 production in spleen tissue. and also decreased IL-1$\beta$, IL-6 production in liver tissue. Conclusion: These results suggest that SYTG can be useful in treating diverse female diseases caused by inflammation such as endometrosis, myoma, pelvic congestion. chronic cervicitis, chronic pelvic inflammatory disease and so on.
In this study, the anti-oxidant, anti-inflammatory, and anti-obesity activities of an ethanol extracts of Ulmus divididiana var. japonica (UDE) were investigated in vitro and in vivo. UDE anti-oxidant activity was evaluated with an Electron Spin Resonance (ESR) spectrometer, which measured 1, 1-diphenyl-2-picryl-hydrazyl (DPPH) radical scavenging activity. Cell viabilities were estimated using 3-[4,5-Dimethylthiazol-2-yl]-2,5-diphenyl-tetrazolium bromide (MTT) assay. LPS-stimulated BV-2 microglia were used to study the production of nitric oxide (NO). Cells stimulated with LPS produce more NO than normal control cells. However, cells treated with the UDE decreased this production in a concentration dependent manner (100, 250, 500, $1,000{\mu}g/mL$). Also, we investigated the anti-obese activity of UDE in SD rats. The SD rats were randomly divided into five groups: 10% low fat diet (N), 45% high fat diet (H), 45% high fat diet + garcinia extracts 200 mg/kg/day (HG200), high fat diet + UDE 200 mg/kg/day (HU200), high fat diet + UDE 400 mg/kg/day (HU400). UDE was found to lower whole body and abdominal and epididymal adipose tissue weights and lowered plasma levels of triglyceride (TG), compared to those in H group. Histological analyses of the liver and fat tissues of rat treated with UDE revealed significantly decreased number of lipid droplets and decreased size of adipocytes compared to the H group. These results suggest that UDE might be used to develop potent anti-oxidant, anti-inflammatory, and anti-obesity agents, and may be useful as ingredients for related new functional raw materials.

Background: Sulforaphane (SFN) is an isothiocyanate compound present in cruciferous vegetables. Although the anti-inflammatory effects of SFN have been reported, the precise mechanism related to the inflammatory genes is poorly understood. Objectives: This study examined the relationship between the anti-inflammatory effects of SFN and the differential gene expression pattern in SFN treated ob/ob mice. Methods: Nitric oxide (NO) level was measured using a Griess assay. The inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) expression levels were analyzed by Western blot analysis. Pro-inflammatory cytokines (tumor necrosis factor [TNF]-α, interleukin [IL]-1β, and IL-6) were measured by enzyme-linked immunosorbent assay (ELISA). RNA sequencing analysis was performed to evaluate the differential gene expression in the liver of ob/ob mice. Results: The SFN treatment significantly attenuated the iNOS and COX-2 expression levels and inhibited NO, TNF-α, IL-1β, and IL-6 production in lipopolysaccharide (LPS)-stimulated RAW 264.7 cells. RNA sequencing analysis showed that the expression levels of 28 genes related to inflammation were up-regulated (> 2-fold), and six genes were down-regulated (< 0.6-fold) in the control ob/ob mice compared to normal mice. In contrast, the gene expression levels were restored to the normal level by SFN. The protein-protein interaction (PPI) network showed that chemokine ligand (Cxcl14, Ccl1, Ccl3, Ccl4, Ccl17) and chemokine receptor (Ccr3, Cxcr1, Ccr10) were located in close proximity and formed a "functional cluster" in the middle of the network. Conclusions: The overall results suggest that SFN has a potent anti-inflammatory effect by normalizing the expression levels of the genes related to inflammation that were perturbed in ob/ob mice.
Objectives : In this study, it was investigated the anti-inflammatory, anticancer, and antioxidant effects of Jayangdaebo-tang (JDT) consisting of twelve herbs before and after fermentation. Methods : JDT extract was fermented using the Lactoplantibacillus plantanum (JDT-L), Bacillus subtilis (JDT-B), and L. plantanum plus B. subtilis (JDT-L+B). The effects of each extract were measured in LPS-stimulated RAW264.7 cells, MCF-7 breast cancer and A549 lung cancer cells, and H2O2-stimulated HepG2 cells. Results : The extracts of JDT-L, JDT-B and JDT-L+B at 1 ㎎/㎖ decreased significantly the levels of nitric oxide (NO) in LPS-treated RAW264.7 cells and also inhibited the expression of iNOS and COX-2, and the phosphorylation of ERK and NF-κB. The JDT-L+B extract decreased significantly the expression of apoptotic proteins, Bax, cleaved caspase-3, and PARP in MCF-7 and A549 cancer cells. The JDT-L, JDT-B and JDT-L+B extracts increased significantly the cell viability in H2O2-stimulated HepG2 cells and the JDT-L+B extract decreased significantly the expression of SOD, catalase, HO-1, and NRF-2. Among fermented JDT extracts, JDT-L+B was the best effective on response of macrophage inflammation, cancer cell apoptosis, and liver cell damage. Conclusions : Our results were suggested that the fermentation can be used as a useful way to enhance the biological activity of JDT.
An experiment was conducted to evaluate feeding values of food and other organic wastes, and to determine their dietary effects on performance and carcass yield in ducks. A total of 156 meat type ducklings at the age of 25 days were housed in 12 pens and assigned to 4 treatments, in which the birds were fed for 21 days. One of 4 diets, commercial duck feed, raw food waste (RFW), fermented food waste(FFW) and mixture of 50% RFW and 50% FFW (R+FFW). Feed consumption was significantly higher (p<0.05) in the RFW groups. Body weight gain showed no significant differences between the control and other treatments. The feed consumption ratio was significantly higher in the RFW groups (p<0.05) than that of the other groups. Carcass yield was significantly higher in the R+FFW groups (p<0.05) than the other groups. The weight of liver per live body weight showed significant differences among the treatment groups (p<0.05). The length of intestine was significantly different from those of the treated groups(p<0.05).
This study was performed to evaluate anti-thrombotic activities and anti-inflammatory effects of ChungyeolHaedogHwaeoTang water extract(CHHT). The results were summarized as follows. In experiment of anti-thrombotic effect; 1. CHHT inhibited human platelet aggregation induced by ADP and epinephrine as compared with the control group, and inhibited pulmonary embolism induced by collagen and epinephrine (inhibitory rate is 37.5%). 2. CHHT increased platelet number significantly, and also CHHT shortened PT and APTT significantly as compared with the control group in thrombus model induced by dextran. In experiment of anti-inflammatory effect; 3. CHHT inhibited $IL-1{\beta}$, IL-6, TNF-${\alpha}$, COX-2 and NOS-II mRNA expression as compared with the control group in a concentration-dependent degree, and inhibited NO production significantly at 50, 100 ${\mu}g/ml$, and also inhibited ROS production in a concentration-dependent degree as compared with the control of group in RAW 264.7 cell line. 4. CHHT inhibited $IL-1{\beta}$, IL-6 and TNF-${\alpha}$ production significantly in serum of acute inflammation-induced mice, and decreased $IL-1{\beta}$, IL-6 and TNF-${\alpha}$ production in spleen tissue, and also decreased $IL-1{\beta}$, and IL-6 production in liver tissue, but increased TNF-${\alpha}$ production in liver tissue of acute inflammation-induced Balb/c mice. 5. CHHT increased survival rate from the 3rd day in ICR mice with lethal endotoxemia induced by LPS. These results suggest that CHHT can be useful in treating diverse female diseases caused by thrombosis and inflammation such as menstrual pain, menstrual disorder, leukorrhea, vaginitis, cervicitis, pelvic inflammatory disease and so on.
This study was conducted to investigate the effects of Aralia elata, Acanthopanacis cortex and Ulmus davidiana water extracts on blood hemoglobin, $HbA_{1c}$, levels and biomarkers in the streptozotocin (STZ)-induced diabetic rats. Male Wistar rats divided into normal and diabetic groups. The diabetic groups subdivided into the control group (DM) and Araliaceae water extracts supplemented groups: Aralia elata (AE), Acanthopanacis cortex (AC) and Ulmus davidiana (UD). The extracts were supplemented in diet base on 11.42 g of raw Araliaceae/kg diet for 7 weeks. The diabetes was induced by injecting 572 (55 mg/kg B.W., i.p.) once 2 weeks before sacrifying. Relative weights of liver were significantly lowered in the DM group compared to the normal group, whereas those of kidney and heart were significantly increased in the DM group. Supplementation of the Araliaceae water extracts improved reduced liver weights in STZ-induced diabetic rats. Blood glucose level was significantly higher in the DM group than in the normal group, whereas insulin contents were significantly lowered in the DM groups. However, these parameters were normalized in the An, AC and UD supplemented groups, respectively. Blood hemoglobin and $HbA_{1c}$ levels were significantly higher in the DM group than in the normal group. When all of Araliaceae water extracts were supplemented to the diabetic rats lowered hemoglobin and $HbA_{1c}$ levels. Red blood cell, white blood cell and Lymphocyte were significantly higher in the DM group than in the normal group. The supplementation of Araliaceae family water extracts significantly lowered these parameters compared to the DM group. MCV, MCH contents were declined in the DM group, while the supplementation of Aralia elata, Acanthopanacis cortex and Ulmus davidiana water extracts elevated of these contents in STZ-induced diabetic rats. Accordingly, these results indicate that Aralia elata, Aeanthopanacis corex and Ulmus davidiana water extracts would seem to improve the blood biomarkers in STZ-induced diabetic rats.
Phongsaly Province, located in the northernmost area of Lao PDR, was previously suggested to be endemic for the liver fluke Opisthorchis viverrini infection. To confirm, or rule out, this suggestion, the Phonxay village in the Khoua District, Phongsaly Province, was selected for a survey. Ten volunteers (8 men and 2 women aged 31-57 years) who consumed raw freshwater fish and had gastrointestinal troubles were treated with a single dose of praziquantel (40 mg/kg) and pyrantel pamoate (10 mglkg) and purged with magnesium sulfate to recover any worm parasites. Eight of the 10 volunteers expelled 1 or more species of trematodes, nematodes, or cestodes (worm positive rate; 80%). The worms were morphologically identified as H. taichui (861 worms from 8 people), H. yokogawai (59 from 6 people), Phaneropsolus bonnei (1 from 1 person), Trichostrongylus sp. (2 from 2 people), Ascaris lumbricoides (2 from 1 person), Enterobius vermicularis (11 from 3 people), and Taenia saginata (1 strobila with scolex from 1 person). The results indicate that the mountainous area of Phongsaly Province, Lao PDR, is not endemic for the liver fluke but endemic for intestinal flukes, in particular, Haplorchis taichuiand H. yokogawai.
