조사속도 및 조사탕온도의 고저가 자동조사성적에 미치는 영향 (Effect of Reeling Velocity and Temperature on the Automatic Silk Reeling Process)

  • 본 시험은 우리나라의 원료견질에 적합한 자동조사의 기술체계를 확립하기 위한 기초연구로서 다음 결과를 얻었다. 1. 생사량비율 및 해서률은 조사속도의 상승 및 조사탕온도의 저하에 따라서 감소하는 경향이다. 2. 조사장력은 조사속도의 상승 및 조사탕속도의 저하에 따라서 증대하는 경향이다. 3. 사조고장 회수는 조사속도 및 조사탕온도의 상승에 따라서 증가하는 경향이다. 4. 정섬도 자동조사에 있어서 조사속도 90~120m/min 및 조사탕온도 35~45$^{\circ}C$ 범위내에서는 생사품위의 차이를 인정할 수 없다. 5. 이상의 결과를 종합하여 볼 때 우리나라의 원료견질에 적합한 자동조사의 자동속도는 120m/min 내외로 다조조사의 약 2배이고 조사탕온도는 4$0^{\circ}C$내외라고 본다.

식혜의 이소말토올리고당에 관한 연구 -4보 찹쌀식혜- (A Study on Sugars in Korean Sweet Rice Drink "Sikhye" -4. Glutinous Rice Sikhye-)

  • 찹쌀 20%, 엿기름 4%를 가하여 7시간 동안 당화시켜 제조한 식혜는 말통스 10.1%, 한계덱스트린 7.3%, 말토트링스 1.3%, 글루코오스 0.18%, 밥알 1.75%를 나타냈다. 알코올 침전, Biogel P-2의 겔 크로마토그래피로 식혜의 한계덱스트린을 정제하여 1H-NMR 분석한 결과 한계덱스트린은 $\alpha$-1,4-글루코시드 결합과 $\alpha$-1,6-글루코시드 결합이 5:1로 이루어졌고, pullulanase 처리한 결과 말토오스와 말토헥사오스까지의 분포를 나타내어 멥쌀식혜에서 얻은 한계덱스트린가 구조가 같은 결과이다. 밥알의 당함량은 26.4%, 단백질 함량은 41.6%를 나타냈다. 참쌀식혜의 한계덱스트린과 밥알에 30unit/ml의 $\alpha$-아밀라아제, 글\ulcorner아밀라아제, $\alpha$-글루코시다아제, $\beta$-아밀라아제를 작용시킨 결과 글루코아밀라아제 외에는 일부밖에 가수분해하지 at하였다. 인체의 효소인 $\alpha$-아밀라아제와 $\alpha$-글루코시다아제를 함께 작용시킨 결과 한계덱스트린의 가수분해율은 18%, 밥알의 가수분해율은 26%를 나타냈다.

Screening of Immunostimulatory Probiotic Lactic Acid Bacteria from Chicken Feces as Animal Probiotics

  • The principal objective of this study was to screen and select acid-tolerant Lactobacillus strains from chicken feces, feeds, and other sources. Fourty six strains evidencing acid tolerance (pH 3.5) were isolated in this study. Among them, nine strains exhibited marked immunostimulatory effects. Therefore, nine candidate strains were characterized for probiotic use. In order to evaluate macrophage activation, NO production was measured using RAW 264.7 cells. In particular, three strains (FC812, FC222, and FC113) evidenced the highest levels of NO production measured at $38.39{\pm}20.01,\;35.06{\pm}27.73$, and $33.88{\pm}15.99{\mu}M$, respectively, at a concentration of $10^{8}CFU/mL$. The majority of strains, with the exception of strain FC322, evidenced marked resistance to artificial gastric juice (pH 2.5 with 1%(w/v) pepsin). Additionally, strains FC222, FC421, FC511, and FC721 were highly resistant to artificial bile acid (0.1%(w/v) oxgall), whereas strains FC113, FC322, FC422, FC621, and FC812 were the least resistant to bile. All nine strains exerted antimicrobial effects against chickenrelated pathogens. Additionally, all nine strains were found to be resistant to several antibiotics. The isolated strains, except for strain FC322, were tentatively identified as Lactobacillus salivarius, using an API 50 CHL kit. These results demonstrate that some probiotic organisms may potentially probiotic properties, and thus may serve as an effective alternative to antibiotics in animal applications.

발아현미 첨가에 따른 국수의 제조 특성과 GABA 함량 및 항산화 활성 변화 (Quality Characteristics and Changes in GABA Content and Antioxidant Activity of Noodle Prepared with Germinated Brown Rice)

  • 발아 전과 후의 현미와 미강을 첨가한 밀가루 복합분으로 만들어진 국수의 품질 특성과 GABA 함량 및 항산화 활성의 변화를 조사하였다. 발아 전후의 현미와 미강을 각각 5, 10, 15% 첨가하여 국수를 제조하였다. 이렇게 제조된 국수는 조리 및 제면특성(cooking loss, swelling index, water holding capacity), 조리에 따른 GABA 함량과 항산화 성분(polyphenol) 및 활성(ABTS와 DPPH의 radical scavenging activities) 변화를 통해 기능성 국수로의 이용가능성을 알아보고자 하였다. 실험결과, 발아 후 rice bran의 첨가에 따른 cooking loss가 증가하는 것으로 관찰되었으며 반면에 swelling index에서는 차이를 나타내지 않았다. 발아 후 현미와 미강을 첨가한 조리 전후 국수에서 발아 전 현미와 미강을 첨가한 국수보다 높은 GABA 함량을 확인하였으며 발아에 따른 polyphenol 함량과 항산화 활성에서는 큰 변화가 없음을 확인할 수 있었다. 따라서 발아현미가 첨가된 국수는 밀가루 국수의 영양학적 가치와 항산화 기능을 향상시킬 수 있는 소재로써 이용 가능할 것으로 생각된다.

온라인 유통중인 과메기·야채세트의 미생물학적 안전성 평가 (Microbial Contamination in Cololabis saira and Vegetables Distributed through Online Markets)

  • The consumption of Gwamegi, a semi-dried saury Cololabis saira, and its vegetable sides has increased owing to its availability online. Therefore, this study investigated the microbial contamination levels in Gwamegi and its accompanying vegetable sides bought online by measuring total viable bacteria, coliforms, Escherichia coli, Staphylococcus aureus and fungi. The total viable bacteria ranged from 3-5 log CFU/g. The fungi in Gwamegi and garlic were 3.4 and 3.9 log CFU/g, respectively. The positive rate of bacterial contamination was 100% (2-3 log CFU/g) in Gwamegi, cabbage Brassica rapa subsp. pekinensis, and green chili Capsicum annuum, whereas the contamination positive rate was 80% and 60% (< 2 log CFU/g) in chives Allium ascalonicum L. and garlic A. sativum L., respectively. The positive rates of E. coli were 0%, 20%, 60%, and 40% in Gwamegi, green chili, cabbage, and chives, respectively. The contamination levels of E. coli were 1-2 log CFU/g. S. aureus was detected at < 1 log CFU/g in all raw materials. The data on microbial contamination levels may be used for microbial risk assessment of Gwamegi and vegetables for controlling the level of microbial contamination and securing microbiological safety.

Characterization and functional inferences of a genome-wide DNA methylation profile in the loin (longissimus dorsi) muscle of swine

  • Objective: DNA methylation plays a major role in regulating the expression of genes related to traits of economic interest (e.g., weight gain) in livestock animals. This study characterized and investigated the functional inferences of genome-wide DNA methylome in the loin (longissimus dorsi) muscle (LDM) of swine. Methods: A total of 8.99 Gb methylated DNA immunoprecipitation sequence data were obtained from LDM samples of eight Duroc pigs (four pairs of littermates). The reference pig genome was annotated with 78.5% of the raw reads. A total of 33,506 putative methylated regions (PMR) were identified from methylated regions that overlapped at least two samples. Results: Of these, only 3.1% were commonly observed in all eight samples. DNA methylation patterns between two littermates were as diverse as between unrelated individuals (p = 0.47), indicating that maternal genetic effects have little influence on the variation in DNA methylation of porcine LDM. The highest density of PMR was observed on chromosome 10. A major proportion (47.7%) of PMR was present in the repeat regions, followed by introns (21.5%). The highest conservation of PMR was found in CpG islands (12.1%). These results show an important role for DNA methylation in species- and tissue-specific regulation of gene expression. PMR were also significantly related to muscular cell development, cell-cell communication, cellular integrity and transport, and nutrient metabolism. Conclusion: This study indicated the biased distribution and functional role of DNA methylation in gene expression of porcine LDM. DNA methylation was related to cell development, cell-cell communication, cellular integrity and transport, and nutrient metabolism (e.g., insulin signaling pathways). Nutritional and environmental management may have a significant impact on the variation in DNA methylation of porcine LDM.

Network RTK 품질 분석 방법론별 성능 지표와 사용자 항법 정확도의 상관성 (Correlation between the Position Accuracy of the Network RTK Rover and Quality Indicator of Various Performance Analysis Method)

  • 정지한 객체의 측위에 사용되던 Network RTK (real time kinematics) 기술을 이동형 항체의 항법에 적용하기 위해서는 보정정보와 함께 사용자의 성능을 대표할 수 있는 지표가 함께 제공되어야 한다. 이를 위하여 본 논문에서는 I95 (ionospheric index 95) / G95 (geodetic index 95), SBI (semivariance based index), RIU (residual interpolation uncertainty) 등의 지표 도출 알고리즘을 분석하고 이를 국토지리정보원의 기준국 원시 데이터와 VRS (virtual reference station) 사용자에 적용함으로써 정밀 항법 성능 지표로의 활용 가능성을 타진하였다. 24시간 데이터를 처리한 결과 보정정보의 비선형성을 나타낼 수 있는 RIU 지표와 Network RTK 사용자의 위치 정확도와의 상관성이 0.52로 타 지표에 비해 훨씬 높은 것으로 나타났으므로 향후 이동 항체의 항법 성능 지표로 사용이 가능할 것으로 예상된다.

우유첨가 취반미의 물성에 관한 연구 (Rheological Evaluation of Cooked Rice with Milk)

  • This study was attempted to enhance nutritional value of cooked rice by adding milk in cooking water. Cooked and soaked rice with five different levels of milk in cooking water (0%, A: 30%, B: 50%, C: 70%, D: 100%, E) was tested for rheological parameters, fine structural changes, sensory evaluation. 1. Water absorbance of raw rice in cooking water with varying amountes of milk, was tested at $5^{\circ}C$ and $15^{\circ}C$ for 2 hours. Water absorption ratio was decreased as milk content was increased and soaking temperature was low. Time for maximum water absorption of sample A was 40 min at 1$15^{\circ}C$ but for sample B to D, it was not reached until 120 min. 2. Electronmicroscopic observation revealed that starch granules of rice lost their regular forms by soaking for 90 minutes, but recovered most of initial regularity after 24 hours. Increase in milk content of soaking water decreased marginal sharpness of the starch granule, presumably due to reduced swelling of the granule. 3. Degree of gelatinization of cooked rice was highest in sample A and progressively decreased as milk content was increased. It was, however, increased in all samples when the cooking water to rice ratio was raised from 160% to 180%. During 4 hour storage, rates of retrogradation were not different between A and B samples, but those of C, D and E were about 2.5 times higher than A and B under the optimal condition of 170% cooking water to rice ratio. This was in the order reverse to hardness order of AC>A, D>E at cooking water to rice ratios of 160% and 170%, above which A sample surpassed the rest of samples. 4. Sensory evaluation conducted by fifteen university students as panelists showed that there were more significant differences among five samples in flavour, texture than appearance and a notable preference for b and C over A, D and E.

  • 왜문어 Octopus vulgaris의 천일건조 및 건조 후 3개월간 저장 중의 핵산관련물질의 변화를 이온교환수지 column chromatography 법으로 실험하여 다음과 같은 결과를 얻었다. 살아있는 왜문어에서 절단한 생시료(다리) 중에는 inosine이 $9.4\;{\mu}mole/g{\cdot}dry\;wt.$로서 가장 함량이 많았고, 어류와는 달리 IMP는 존재하지 않았다. 건조중 ATP, ADT 및 inosine은 감소하고, AMP 및 hypoxanthine은 증가하였다. 특히 inosine은 생시료때 $9.4\;{\mu}mole/g\;dry\;wt.$이던것이 $3.0\;{\mu}mole/g\;dry\;wt.$로 감소하는 반면, hypoxanthine은 $5.1\;{\mu}mole/g\;dry\;wt.$에서 $17.6\;{\mu}mole/g\;dry\;wt.$로 증가하였다. 건조후 3개월간 저장 중에는 ADP, AMP 및 inosine은 감소하고 hypoxanthine은 약 2배로 증가하였다.

비만 여성의 체형 특성 분석을 위한 비만 판정 지수의 비교 (A Comparative Study on Obesity Judgment Indices for Body Characteristics Analysis in Korean Obese Women)

  • As a preliminary research of body characteristics analysis of Korean obese woman, this study aims to select optimal obesity judgment tools for reliable sampling of obese subjects from 2,425 female measurement data out of 2004 Size Korea project's raw data. From previous researches related to obesity, 7 obesity judgment tools were chosen. 2007 obesity rate(26.3%) of Korean female adults was refered in selecting optimal obesity judgment criteria in the study. The results are as follows. Firstly, it was verified that BMI was the most suitable in judging and sampling the obese subjects by the percentile analysis. R$\ddot{o}$hrer index was also reliable in grouping the obese subjects from a population. Secondly, it was concluded that the obesity ratios of relative weight 120 and higher group, R$\ddot{o}$hrer index 1.6 and higher group and waist girth 80cm and higher group were the most similar to obesity rate of Korean female adults by 2007 National Health & Nutrition Survey. Thirdly, 30 direct measurements, age, 2 drop values and 6 ratios of 7 groups by the obesity judgment tools showed the significance each other at p<0.001 level. On the other hand, "bust point to bust point" and "waist to hip length" measurements didn't show the significant differences among 7 groups. Conclusively, 4 to 5 satisfactions out of 7 obesity judgment criteria were adequate and sufficient in sampling the obese subjects. If it is needed the strict criteria for judging the obesity, 5 satisfactions and higher group will be the best choice as the obese subjects. However 4 satisfactions and higher group generally, will be adequate for sampling of the obese subjects.
