흰쥐 혀침샘의 점액질에 미치는 N-Nitrosodimethylamine 독성에 대한 녹차의 완화 효과 (The Alleviating Effects of Green Tea on N-Nitrosodimethylamine Toxicity to Mucins in the Rat Lingual Salivary Glands)

  • 흰쥐의 혀침샘인 혀 von Ebner샘과 혀 점액샘의 점액질에 영향을 미치는 N-nitrosodimethylamine(NDMA)의 독성에 대한 녹차의 완화 효과를 조사하기 위해 Sprague- Dawley계 수컷 흰쥐를 대조군, NDMA 1주 투여군(NDMA 군), NDMA 1주 투여 후 녹차 1주 투여군(NDMAGT군), 녹차와 NDMA 1주 투여군(GTNDMA군) 및 녹차 1주 사육 후 녹차와 NDMA 투여군(GTGTNDMA군)으로 나누어 점액질의 조직화학적 성상을 PAS 반응, AB pH 2.5 염색, AB pH 2.5-PAS 염색, AF pH 1.7-AB pH 2.5 염색 및 HID-AB pH 2.5 염색법으로 관찰하였다. NDMA군에서 혀 von Ebner 샘의 형태적인 주된 변화는 장액샘꽈리의 위축 및 파괴, 장액세포의 세포질 과립의 감소 및 소실, 세포질의 공포변성 그리고 도관상피세포의 점액화였으며 혀 점액샘의 주된 형태적 변화는 점액샘꽈리의 확장, 융합, 파괴 및 점액세포의 세포질의 과립 소실 및 공포변성, 도관상피의 점액화였다. NDMAGT군에서는 회복되는 경향이 나타났고 GTNDMA 군에 비해 GTGTNDMA군에서 대조군으로 현저히 회복되었음이 관찰되었다. 혀 von Ebner샘의 중성점액질 양은 대조군에 비해 NDMA군에서 감소되었으나 NDMAGT군, GTNDMA군에서 회복되는 경향을 나타났으며 GTGTNDMA 군에서는 대조군에 비해 중성점액질의 염색성이 더 강했다. NDMA군에서 혀 점액샘의 중성점액질과 산성점액질의 양은 대조군에 비해 감소하였는데, 산성점액질 중 강 sulfomucin을 분비하는 점액샘꽈리는 대조군에 비해 감소하고 non-sulfomucin(sialomucin)을 분비하는 점액샘꽈리는 증가하는 경향을 나타내었다. 녹차를 투여한 모든 실험군들에서 는 회복되는 경향이 나타났는데 NDMAGT군보다 GTNDMA 군에서 더 많은 회복을 보였으며, GTGTNDMA군에서는 대조군과 거의 유사한 수준으로의 회복이 관찰되었다.

  • 발암물질인 NDMA 17m/kg을 경구 투여했을 때 흰쥐설 von Ebner선과 설 점액선의 glycoconjugates에 미치는 영향을 prelectin 조직화학법인 PAS반응, AB pH 2.5 염색법, AB pH 0.4 염색법, AB pH 2.5 -PAS 염색법, AF pH 1.7-AB pH 2.5 염색법 및 HID-AB pH 2.5 염색법으로 연구하였다. NDMA 투여군의 설 von Ebner선의 형태적인 주된 변화는 장액선포의 위축 및 파괴, 장액세포의 세포질 과립의 감소 및 소실, 세포질의 공포변성 그리고 도관상피세포의 점액화였으며 NDMA 투여후 12시간군부터 72시간군까지 현저하였다. NDMA 투여군의 설점액선의 주된 형태적 변화는 점액선포의 확장, 융합, 파괴 및 점액세포의 세포질의 과립 소실 및 공포변성, 도관상피의 점액화 등이었으며 NDMA 투여후 12시간군부터 72시간군까지 현저하였다. 설 von Ebner선의 neutral glycoconjugates가 대조군에 비해 NDMA 투여후 12시간군과 72시간군에서 현저히 감소되었으며 NDMA 투여후 96시간군후에 회복되는 경향을 나타내었다. 대조군에서 나타나지 않던 acidic glycoconju-gates가 NDMA 투여후 6시간군부터 48시간군, 그리고 120시간군에서 약간의 장액세포들에서 나타났다. 설 점액선의 neutral glycoconjugates와 acidic glycocon-jugates가 대조군에 비해 NDMA 투여후 3시간군, 24시간군, 48시간군에서 다같이 현저히 감소되었으며, 후자는 NDMA 투여후 72시간군에 아주 현저히 감소하였다. Aci-dic glycoconjugates중 sulfated glycoconjugates는 대조군에 비해 NDMA 투여후 감소하는 경향을 나타내며 NDMA 투여후 72시간군에 가장 현저한 감소를 나타내는데 비해 sialic glycoconjugates는 NDMA 투여후 3시간군, 12시간 군 및 48시간군에서 증가하는 경향을 나타내었다.

Altered Expression of Aquaporins in Rat Submandibular Glands after Parasympathetic Denervation

  • The salivary glands produce 1.5L of fluid per day. As in other exocrine organs, the general mechanism in the salivary glands is that water movement occurs secondary to osmotic driving forces created by active salt transport. Therefore, high water permeability in the salivary glands is expected to have a variety of aquaporin (AQP), a water channel. Although some AQPs have been known to be present in the salivary glands, roles of parasympathetic nerve in AQP expression have not yet been examined. This study was designed to examine the changes of AQPs and extracellular signal-regulated kinase (ERK) in the submandibular glands after parasympathetic denervation. Right chorda-lingual nerve was cut, and each right (experiment) and left (control) submandibular gland was excised at 1, 3, 7, 14, 30 days after denervation. The denervated right submandibular glands were resulted in weight loss and morphologic changes, including cell loss and atrophy, as the time elapsed after parasympathetic denervation increased, whereas there were no histologic alteration in control side. AQP5 which is known to reside in apical membrane and secretory caraliculi of the submandibular acini were gradually underexpressed according, as the time after denervation increased. Expression of AQP4 in submandibular ductal epithelium was down-regulated after denervation. Besides, AQP3 and 8, which is known to be present in basolateral membrane of the glandular acini, were gradually underexpressed after denervation, similar to the pattern of other types. Expression of ERK, a mitogen-activated protein kinase, was downregulated after parasympathetic denervation in the submandibular gland. These results suggest that parasympathetic nervous system regulates the expression of AQPs in salivary glands, and is in part mediated by ERK pathway.

각종 가축 설근부의 조직학적 구조에 대한 비교 연구 (Comparative Study of Histological Structure in the Tongue Root of Domestic Animals)

  • In order to know the comparative histological structure of the tongue of some domestic and laboratory animals, the root of the tongue has been investigated by light microscopy. The filiform papillae were mainly distributed in the root of the tongue of most animals and many foliate papillae were also found in the dog and rabbit. On the other hand, only filiform papillae were found in the fowl. The very well-developed mucosa was found in the cattle and in the dog, fowl, rat and mouse was fair. The lamina propria was well developed in the cattle, dog and pig, The muscle layer was well developed in the cattle. The adipose tissue which distributed between the muscle fibers was mainly found in the dog. The lingual glands were well developed in the pig, fowl and rabbit. The properties of these glands were shown strung acid mucopolysaccharide in the dog and pig, weak acid mucopolysaccharide in the fowl, rabbit and rat and neutral or acid mucopolysaccharide in the mouse. The species differences on the histological structure of the tongue were also ascertained by light microscopical observations.

CsA Affects the Rat Submandibular Glands via Regulating the CypA Expression

  • The effects of the an immunosuppressive drug cyclosporine A (CsA), on the salivary gland are largely unknown, even though clinical trials for the stimulation of salivation using CsA have been attempted. Cyclophilin A (CypA) is known to be a binding protein for CsA. CypA has cell proliferation and tissue matrix change activities. In our present study, the presence of CypA in the gland and effects of CsA on CypA expression were investigated by immunohistochemistry, immunoblotting and RT-PCR analyses. CypA was immunohistochemically detected in various kinds of ducts in the submandibular glands of Sprague Dawley rats. The CypA mRNA level was highest at postnatal day 1 and gradually decreased in a time-dependent manner up to adulthood. The expression of CypA increased after a 10 day subcutaneous administration of CsA in postnatal day 1 rats. Surgical sections of the chorda-lingual nerve with impaired salivation showed no changes in CypA expression. A cell proliferation assay using PCNA anti-serum showed increased cell division following CsA treatment. These results suggest that CsA and CypA may act on ductal cells to regulate saliva composition rather than salivation levels.
