통도산(通導散)이 실험동물(實驗動物)의 심혈관계(心血管系)에 미치는 영향(影響) (Effects of Tongdosan on the Cardiovascular System in the Experimental Animals)

  • The present experiment was desinged to investigate the effects of Tongdosan water extracts on the Cardiovascular System in the Experimental Animals. Thus, the changes of blood pressure and heart rate were measured after oral admini- stration. Measurment of Mortality rate was observed for measuring the effect of Tongdosan water extract. Tongdosan water extract against pulmonary thrombo- embolism induced by collagen the mixture(0.1ml/10g, 2mg/kg B.W) plus serotonin(5mg/kg B.W) in mouse. The effect of Tongdosan water extract was examined by observing the change of collagen-induced platelet aggregation, coagulation activity, ex vivo and in vitro fibrinolytic activity of euglobulin fraction in rats. The results were summarized as followings. 1. Tongdosan dropped the blood pressure in spontaneous hypertensive rat. 2. The drug increased the auricular blood flow in rabbit. 3. The drug relaxed the artery contraction by pretreated norepinephrine in rat. 4. The drug inhibited the death rate of mouse which was led to thromboembo- lism by serotonin and collagen. 5. The drug inhibited the platelet aggregation in rat. 6. The drug prolonged the prothrombin time and activated partial thromboplastin time on the test of plasma coagulation factor activity in rat, but was not valuable. 7. The drug reduced the fibrinogen lyses time of rat ex vivo assay and lyses area was increased. 8. Tongdosan reduced fibrinogen lyses time of rat in vitro assay. According to the above mentioned results, Tongdosan increased the blood flow and dropped the blood pressure by the dilation of blood vessel. And the drug presented the antithrombin acivity, inhibited the platelet aggregation.

  • 랫드 꼬리는 연속적인 채혈을 위해서 주로 이용되는 채혈부위로 꼬리 절개법, 꼬리 절단법, 꼬리 천자 등의 방법을 통해 채혈이 이루어지고 있다. 하지만, 랫드 꼬리에 대한 혈관 구조는 아직 잘 밝혀져 있지 않다. 그래서 본 연구에서는 독성학이나 약리학에서 가장 많이 이용되는 폐쇄군인 Sprague-Dawley 랫드 꼬리의 혈관구조 규명하기 위해 혈관조형술과 면역조직학적인 방법을 실시하였다. 그 결과 SD 랫드 꼬리는 한 개의 등쪽 정맥과 두개의 외측정맥, 한쌍의 배쪽 정맥과 동맥이 존재했다. 그러므로, 본 연구는 SD 랫드 꼬리를 이용해서 채혈을 하고자 하는 연구자들이 꼬리의 혈관구조를 이해하고, 그 연구 목적에 맞게 채혈부위와 방법을 선택하는데 도움이 될 것으로 생각된다.

흰쥐의 대퇴동맥 혈전 모델에서의 스트렙토키나제-덱스트란 포합체의 혈전용해효과의 평가 (Evaluation of Thrombolytic Effect of Streptokinase-Dextran Conjugate in a Rat Model of Arterial Thrombosis)

  • To evaluate the thrombolytic activity of streptokinase-dextran conjugate, a rat model of arterial thrombosis was used. Briefly, the femoral artery was exposed and a filter paper saturated with 70% $FeCl_3$ solution was placed around the femoral artery in order to stop the blood flow. Six minutes after the stop of the blood flow in the femoral artery, streptokinase $(10000{\sim}30000\;units\;per\;rat)$ or streptokinase-dextran conjugate $(5000{\sim}17000\;units\;per\;rat)$ was administered by i.v. bolus injection through the femoral vein. Then the blood flow in the femoral artery was monitored using a Doppler laser flow meter. The i.v. bolus administration of streptokinase could not restore the blood flow in the femoral artery in the dose range of $10000{\sim}30000$ units per rat. The i.v. bolus administration of streptokinase-dextran conjugate could restore the blood flow in the femoral artery in the dose range of $5000{\sim}17000$ units per rat. A good correlation between the dose of streptokinase-dextran conjugate and the total thrombolytic effect was observed. In addition, the lag time between the injection of streptokinase-dextran conjugate and the restoring of the blood flow was decreased as the i.v. dose of streptokinase dextran conjugate increased. These results show the superior beneficial effect of streptokinase-dextran conjugate compared with the unconjugated streptokinase with respect to the elongation of thrombolytic activity, the administration method (single injection versus continuous infusion), and the reduced dose necessary for a equivalent thrombolytic effect.

Streptozotocin-Induced Pro-Inflammatory Response in Rat Blood and its Attenuation by External Albumin Administration

  • Previous report has suggested that the albumin levels were reduced in the peripheral blood mononuclear cells (PBMCs) and consequently oxidative stress was elevated in streptozotocin (STZ)-induced diabetic rats as albumin is the predominant antioxidant in plasma. In this study, we suggest that the levels of pro-inflammatory cytokine such as interleukin-6 (IL-6) and tumor necrosis factor-${\alpha}$ ($TNF-{\alpha}$) were increased by approximately 3.4- and 2.9-fold, respectively, in the serum of STZ-diabetic rats, compared to those of normal rat. In addition to the cytokines, the levels of C-reactive protein (CRP) were also about 3.6-fold higher, indicating that STZ induced a pro-inflammatory response in rat blood. However, when purified rat albumin was externally co-administrated with STZ through the tail vein, the serum levels of IL-6, $TNF-{\alpha}$, and CRP were markedly reduced, although the values were still higher than those of normal (non-diabetic) rats. Albumin administration also decreased STZ-induced oxidative stress in serum and PBMCs. Moreover, the decrease in cytokine and CRP levels was dependent on the dose of injected albumin. These results suggest that STZ-induced pro-inflammation and oxidative stress in rat blood might be attenuated by treatment with exogenous albumin.

불포화도가 다른 유지가 단식과 정상식이 흰쥐의 체중 및 간대사에 미치는 영향 (Effect of Various Fat and Oil Types on Weight Gain and Liver Metabolism in Fasting or Normal Diet Rats)

  • 단식 또는 기초사료를 투여한 성장기 rat 에 우지 또는 들깨유를 매일 일정량 투여하여 2일 간격으로 체중변화와 장기중량, 혈액의 생화학적검사 간조직검사를하여 다음의 결론을 얻었다. 완전 단식rat군(F)과 단식중 우지 (FT)와 들깨유(FP)를 투여한 rat군은 1일 평균 -6.2~-4.0%인 체중감소를 보이고 기초식이군 (B) 또는 기초식이에 우지 (BT)나 들깨유(BP)를 투여한 rat군은 1일 2.3~4.0%의 체중증가를 보였다. F군은 4일만에 모두 아사하였고, FT와 FP군은 6~10일 사이에 모두 아사하였다. 단식에 의한 영향으로 아사직전 이들 rat는 간중량(3.6~4.0g) serum cholesterol(70~73.2mg/㎗), serum triglyceride (6.5~7..5mg/㎗) blood glucose (76-91mg/㎗)의 현저한 감소와 GPT (117.5-186.5u), blood urea nitrogen (38~65mg/㎗)을 대폭 증가하는 특징을 가졌다. 유지시료 불포화도에 따른 영향에서 FP군은 FT군에 비해 약 2일 정도 생존일수의 감축과 serum cholesterol, serum triglyceride, blood urea nitrogen 감소를 보이나 GPT, blood glucose를 약간 증가하였다. BP군은 BT군에 비해 serum cholesterol, triglyceride를 상당히 감소하고 blood urea nitrogen의 근소한 감소와 GPT는 대폭 증가하였다. Blood glucose는 거의 차이가 없었다. 간조직 검사 결과 F군은 아사직전 간실질내 염증 세포 침윤을 보이고 FT, FP군은 유지 시료의 불포화도 차이에 관계없이 아사직전 일부 rat가 염증세포 침윤, 간실질의 염증, 간세포의 손상된 모양이 관찰되었다. 따라서 어떤 목적으로 단식 또는 유지만을 공급하는 무단백질 제한식이를 할 때 극심한 체중감소를 보이고 혈중 cholesterol과 trig1yceride는 크게 낮아지며 간을 포함한 모든 조직의 붕괴의 조짐으로 blood urea nitrogen이 크게 증가되고 그에 따른 간기능저하로 GPT도 크게 상승하고 rat의 경우 4~6일만에 사망하였다. 단식중이거나 혹은 정상식이내 고도 불포화 유지 (들깨유)의 섭취는 포화지방 (우지)에비해 혈중 cholesterol 과 triglyceride를 감소시키는 효과는 있지만 GPT가 유의적으로 증가됨을 볼때 장기간 고도 불포화 유지를 다량 섭취함은 간기능을 저하 시킬 수 있음이 나타났다.

$\imath$--Muscone의 실험관계에 관한 약리연구 (Pharmacological Actions of $\imath$--Muscone on Cardiovascular System)

  • In order to investigate the pharmacological properties of ι-muscone, effects of ι-muscone and musk were studied on the cardiovascular system with various experimental models. In isolated rat aorta, ι-muscone and musk made the relaxation of blood vessels in maximum contractile response to phenylephrine (10$^{-6}$ M) in endothelium-containing rings of the rat aorta, but not in endothelium-denuded rings. However, ι-muscone and musk in the presence of the inhibitor of NO synthase and guanylate cyclase did not make the relaxation of blood vessels. In spontaneously hypertensive rats (SHRs), ι-muscone and musk slightly reduced blood pressure but significantly decreased heart rate. In the isolated perfused rat hearts, ι-muscone and musk did not affect significantly on LVDP, contractile force, coronary flow and (-dp/dt)/(+dp/dt). These results suggest that ι-muscone and musk have weak cardiovascular effects with relaxation of blood vessel and decrease of heart rate, but without significant cardiac functions.

휜쥐의 심혈관 기능에 대한 carbofuran독성작용 (carbofuran Induces Cardiovascular Dysfunction in Rat)

  • Of all pesticides, carbamates are known to be most common, since alternatives such as organophosphates have long lifetime and are extremely toxic to produce a delayed neurotoxic effect. Although a number of studies about toxicity of carbofuran, a most widely used carbamate, have been reported, its cardiovascular toxicity has not yet been studied. In the present study, we investigated its cardiovascular toxic effect in anesthetized rat in vivo and in isolated Langendorff rat heart, In anesthetized rat model, carbofuran (10 mg/kg) significantly reduced heart rate, and transiently increased blood pressure. In isolated rat heart, carbofuran (10${\mu}{\textrm}{m}$) caused a significant depression in the left ventricular developed pressure (LVDP), indicating contractile dysfunction by carbofuran. Carbofuran (10${\mu}{\textrm}{m}$) also decreased coronary flow rate (CFR) in isolated heart, indicating carbofuran-induced coronary dysfunction. These results suggest that carbofuran can cause cardiac dysfunction in rat in vivo and vitro.

티타늄 금속나노입자의 혈액응고작용 검색 (Screening Test of Titanium Dioxide Nanoparticles on Blood Coagulation and Platelet Aggregation)

  • Effects of titanium dioxide nanoparticles on blood coagulation and platelet aggregation were investigated using diluted whole blood and platelet rich plasma (PRP) solution prepared from human and rat blood, respectively. Blood coagulation was monitored by using a whole blood impedance aggregometer and titanium dioxide nanoparticle (10, 20, and 40 ppm) did not show any effect on the coagulation both in human and in rat blood. When platelet aggregation was measured by turbidometric method after addition of titanium dioxide nanoparticles to PRP solution with final concentrations of 1, 5 and 10 ppm. no aggregation was observed.
