Effect of Ginseng on the Intestinal Absorption of Ethanol (인삼 성분이 Ethanol의 장관내 흡수에 미치는 영향)

  • The effect of ginseng butanol fraction (total sponin) on the absorption rate of ethanol in rat intestine was examined. Ginseng butanol fraction showed inhibitory effect on the intestinal absorption of ethanol in situ as well as in vitro test. Ginseng butanol fraction markedly decreased the ethanol blood level, delayed onset time of ethanol effect and shortened sleeping time when it was adminstered orally together with ethanol. These results suggest that ginseng may alter the ethanol blood level by decreasing the intestinal absorption of ethanol.

  • Several epidemiological studies suggested that arsenic exposure was strongly correlated with the development of cardiovascular disease such as hypertension. In order to examine whether arsenic affects vasomotor tone in blood vessels, we investigated the effect of arsenic on agonist-induced vasorelaxation using the isolated rat aortic rings in in vitro organ bath system.   • Counts of RBC and proportion of packed cell volume(PCV) showdc no difference among treatment groups. the values of hemoglobin in rat showed a tendeuy to increase in herbal-acupuncture group, however the values of hemoglobin in guineapig and rabbit showed no significant difference in the treatnent groups. the count of WBC showed high in the chishiI(BL52) herbal-acupuncture group. The proprtion of neutrophils showed no significant difference in rabbit, however the values in guinea-pig and rat showed a tendency to increased in herbal-acupuncture groups. The proportion of lymphocytes in rabbit and guinea-pig showed a tendency to decreast in herbal-acupuncture groups, however the values in rat showed no significant difference. The proportion of Eosinophils and monocytes showed no significant difference in all the treated groups. Results from this study indicated that shinsu(BL23) and chishil(BL52) herbal-acupuncture with palmijihwangtang elivate the biglogical function in Rabbit Guinea-pig and Rat.

Effect of Ginseng on the Alcohol Metabolism in Alcohol Treated Rat (알톨 대사에 미치는 인삼의 영향)

  • After pretreatment with ginseng followed by induction of acute intoxication of alcohol, the activities of alcohol dehydrogenase (ADH), microsomal ethanol-oxidizing system (MEOS) and aldehyde dehydrogenase(Ald DH) increased respectively compared to the groups treated with alcohol alone. In case that ginseng was given to rats fed with 5% alcohol instead of water for 60 days, the activities of ADH and MEOS increased compared to the groups treated. On the contrary, the activity of Ald DH in mitochondrial fraction decreased to an extent of about 35% in chronic alcoholism, but after pretreatment of ginseng the activity was restored to the control level. On the other hand, the catalase activity was not significantly affected by either treatment. Ginseng butanol fraction significantly increased the serum isocitrate dehydrogenase activity which is inhibited by alcohol-treated in rat. Alcohol-induced lactate dehydrogenase activity was decreased to control level in liver by ginseng treatment. And the serum level of lactic acid also decreased by ginseng treatment in alcohol-intoxicated rat. Ginseng butanol fraction markedly decreased the xanthine oxidase activity in the ethanol-treated rat liver. It was also observed that ginseng reduced the blood concentration of uric acid on experimentally reduced hyperuricemia by alcohol treatment. Uricase activity was not affected by either treatment. Ginseng butanol fraction decreased the hepatic aniline hydroxylase activity which was induced by alcohol-treated rat. These results suggest that the treatment with ginseng can be promoted the recovery from alcohol intoxication and some therapeutic effect on alcoholinduced metabolic disease.

  • In the present study, we employed the confocal laser scanning microscopy (CLSM) system to visualize the blood flow field with $1{\times}1{\mu}m^2$ spatial resolution. Based on the confocal microscopic image of red blood cells (RBCs), we performed the velocity vector field measurement and evaluated characteristics of cell migration from the cell depleted layer thickness calculation. The rat and mouse's blood were supplied into a micro glass tubes in vitro. The line scanning rate of confocal microscopy was 15 kHz for a $500{\times}500$ pixels image. As a result, the red blood cell itself can be used as a tracer directly without any kind of invasive tracer particle to get the velocity vector field of blood flow by performing particle image velocimetry (PIV) technique.

Effect of Arsenic on Heat Shock Protein and Vascular Contractility of Rat Aorta (횐쥐 대동맥의 수축반응과 열충격단백질에 대한 비소의 영향)

  • In order to examine if arsenic, one of environmental stresses, contributes to hypertension as one of cardiovas cular pathological factors, this study was perfarmed in vivo and in vitro, using intacted or pithed rats and aorta ring preparation, respectively. And also the relationship between expression of heat shock protein (HSP) 90 and vasoactives-induced contractile response was elucidated. To measure blood pressure, the carotid arterial pressure was recorded on physiograph(Grass Co. 79E) connected to strain gauge. On the other hand, contractile response of vascular ring preparation isolated from rat was determined in organ bath and was recorded on physiograph connected to isometric transducer. And HSP was detacted by Western blotting whole cell Iysis. Preganglionic nerve stimulation was increased by 26.0% in arterial pressure of rat treated with arsenic. Vascular contractile response was monitored and HSP were measured by Western blotting of whole Iysis prepared from samples exposed with 0, 0.5, 1, 2 and 4 mM of arsenic for 8 hours. The dose-vascular responses of potassium chloride were augmented by increasing dose of arsenic in the strips exposed to arsenic for 8 hours, and were not augmented for 1, 3, 5 hours. And the response of relaxation of rat aorta induced by histamine was not influenced by arsenic stress. The increase of HSP 90 expression in rat aorta was pronounced at 8 hours after 4 mM of arsenic treatment, but HSP 60 expression was not. Arsenic stress not only increased the expression of HSP 90 in the rat aorta, but also augmented contractions to potassium chloride. These results indicated that arsenic stress was sufficient to induce heat shock protein 90, resulting in increased vascular contractility in rat aorta.

  • Using aged Wistar rat living body change by aging CheongSimYeonJaTang was each orally administrated and achieved research about aging control. In Wistar rat 10, 30, 50 week and 40 week CheongSimYeonJaTang between 10 weeks form condition change of weight, change of intestine weight, hematology, blood chemistry, research result about serum content following conclusion get. 1. Observed gain in weight than control group form of CheongSimYeonJaTang to aged Wistar rat. 2. Is thought to promote activation of living body action gaining intestine weight along with gain in weight. 3. Displayed decrease of MDA's content of serum than control group form of CheongSimYeonJaTang to aged Wistar rat. 4. Change that is Wistar rat's hematological value by aging according to 10, 30, 50 week WBC, RBC, Hgb, monocytes, eosinophil etc. increase, and HCT, PLT etc. showed tendency that decrease according to old-week, and observed improvement that is hematological value than control group form of CheongSimYeonJaTang. 5. Change that is Wistar rat's biochemical value by aging was measured highest in 50 week because ALT, AST, BUN, CRN, T-bili., T-chol, TG, TP, ALB, A/G, P etc. increase according to 10, 30, 50 week, and observed improvement that is biochemical value than control group form of CheongSimYeonJaTang. Is considered by being effect that CheongSimYeonJaTang living body function decline by aging by this result.

Effects of Oyaksoonkisan and Kamioyaksoonkisan on Hypertension and Pulse Rate (오약순기산과 가미오약순기산이 고혈압과 심박동수에 미치는 영향)

  • Experiments were performed to determine the effects of the Oyacksunkisan(烏藥順氣散) and Kamioyacksunkisan(加味烏藥順氣散) liquid extract on the hypertension and the pulse rate in Sprague-Dawley rat(SDR) and Spontaneous Hypertensive rats(SHR). The results were obtained as follows ; 1. Blood pressure was significantly decreased firstday and 11th day after administration of Oyacksunkisan extract and pretreatment of phentolamine. 2. Blood pressure and pulse rate were regulated first day after administration of Kamioyacksunkisan and blood pressure was significantly decreased after pretreatment of clonidine and phentolamine. 3. Oyacksunkisan and Kamioyacksunkisan didn't show any significant changes of blood pressure and pulse rate after pretreatment of prdpranolol, hydralazine and verapamil From the above result, it was concluded that Oyacksunkisan(烏藥順氣散) and Kamioyacksunkisan(加味烏藥順氣散) could be applied effectively to the hypertension.

In vivo evidence for brain-to-blood efflux transport of taurine and regulation of this transport by tumor necrosis factor-$\alpha$ at the blood-brain barrier

  • The purpose of this study is to examine whether the efflux system for taurine from brain to blood is present on the blood-brain barrier (BBB) using the brain efflux index (BEl) method and taurine transport system is regulated by CNS cell damage with oxidative stress agent such as diethyl maleate (DEM) or tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$) in vivo. [$^3$H]Taurine was microinjected into parietal cortex area 2 (Par2) of the rat brain, and was eliminated from the brain with efflux transport rate of 1.22 10$\^$-2//min, and the process is saturable with a $K_{m}$ of 43.5 ${\mu}$M.
