The Effects of Bojungikgitang-gamibang Administration along with Mahwangshingungsan on the Rat Model with Allergic Rhinitis (보중익기탕가미방(補中益氣湯加味方)과 마황신궁산(麻黃辛芎散) 병용이 알레르기 비염 유발 흰쥐 모델에 미치는 영향)

  • Objectives : We aimed to investigate therapeutic effect of Bojungikgitang-gamibang(BI) and Mahwangshingungsan(MS) by observing changes in blood cells and the nasal mucosa of Sprague-Dawley(SD) rats with allergic rhinitis. Methods : Twenty-four SD rats were divided into three groups: normal, control, and sample group. Allergic rhinitis was induced in the control and sample group by intraperitoneal and intranasal sensitization with 0.1% and 0.4% Ovalbumin solution. Then BI was orally administered only to the sample group along with MS for 28days, while the rats in the control group was given normal saline. Results : BI and MS showed significantly decreased IgE level on the serum of the rat model, Bl and MS showed significantly decreased eosinophil level on the blood of the rat model. BI and MS inhibited the inflammatory reaction on the nasal mucosal tissue, according to nasal mucosal biopsy. Bl and MS had anti-allergic according to level, eosinophil level, nasal mucosal biopsy. BI and MS had no hepatoxicity, according to AST and ALT on the serum. Conclusion : According to the above results, it is considered that Bl and MS is helpful in treatment of allergic rhinitis.

Intestinal Absorption of Fibrinolytic and Proteolytic Lumbrokinase Extracted from Earthworm, Eisenia andrei

  • To investigate the intestinal absorption of a fibrinolytic and proteolytic lumbrokinase extracted from Eisenia andrei, we used rat everted gut sacs and an in situ closed-loop recirculation method. We extracted lumbrokinase from Eisenia andrei, and then raised polyclonal antibody against lumbrokinase. Fibrinolytic activity and proteolytic activity in the serosal side of rat everted gut sacs incubated with lumbrokinase showed dose- and time-dependent patterns. Immunological results obtained by western blotting serosal side solution using rat everted gut sacs method showed that lumbrokinase proteins between 33.6 and 54.7 kDa are absorbed mostly by the intestinal epithelium. Furthermore, MALDI- TOF mass spectrometric analysis of plasma fractions obtained by in situ recirculation method confirmed that lumbrokinase F1 is absorbed into blood. These results support the notion that lumbrokinase can be absorbed from mucosal lumen into blood by oral administration.

  • Dialysis and renal transplantation, the current therapies for end-stage renal disease (ESRD), have many limitations including severe complications, organ shortage, and graft failure. To overcome the limitations, the present study investigated the reconstruction of renal tissue in vivo by transplanting isolated fetal renal cells using fibrin gel to the kidney of renal failure rat model. After 4 weeks from the transplantation, blood urea nitrogen(BUN) and creatinine were examined from blood samples and histological examination of the implanted tissues revealed formation of renal-like structures and restoration of renal function.

  • Astrocyte has emerged as an active regulator of brain function, which connects between blood vessels and neurons as well as is a structural component of the blood-brain barrier, From its structural characteristics, astrocyte seems to sensitively respond to oxygen tension, and, in turn, generate diverse cellular cascades. Therefore, to reveal astrocytlc events by oxygen change, we screened genes whose expressions are upregulated under reoxygenation after hypoxic stress using cDNA representational difference analysis (RDA) technique. Meteorin that regulates glial differentiation was isolated from primary cultured rat astrocytes as a hypoxia/reoxygenation regulatory factor. We cloned rat version of Meteorin (rMe-teorin) and determined full-size sequences of rMeteorin. In addition, RT-PCR analysis revealed that Meteorin was increased under reoxygenation in astrocytes and highly expressed in the developing brain. Collectively, these results suggest that Meteorin may regulate astrocyte-mediated effects in response to the change of oxygen tension in the pathophysiological states.

Improving Effects of Brassica oleracea L. var. italica Sprout Extract on Alcoholic Liver Dysfunction

  • Alcoholic fatty liver disorder has become a frequent health concern worldwide. To investigate the effects of Brassica oleracea (B. oleracea) sprout extract (BOE), the present study was designed with alcoholic fatty liver in the rat. Initially, the effects of BOE on liver parameters were examined. Male rats were divided into five groups. The normal control group was fed the normal diet, and the BOE group was fed the high fat diet and ethanol with/without BOE for 4 weeks. After 4 weeks feeding period, rats were sacrificed and their livers and blood were used for fatty liver-related biomarkers analyses. As a result, BOE ameliorated fatty liver-related enzymes profiles in liver tissues and also reduced blood alcohol concentration in rat model. We demonstrated that BOE protected the high fat diet and alcohol-induced fatty liver in rat model. Furthermore, BOE increased detoxificative abilities against alcohol.

  • We have studied on the effects of Panax Ginseng on the carbohydrate metabolism of rat. Ginseng does not so much effect on the body weight of the nutrition diet groups, but effects much on the carbohydrate deficient diet groups; it remarkably inhibited the decrease of body weight. Liver glycogen, both in the nutrition and carbohycdrate deficient diet groups, is higher than that of the control. Particularly, the increase of the glycogen is eminent is the nutrition diet-ginseng administered group. In the nutrition diet group, the blood sugar level of ginseng-adminstered rats is the higher, while, in the carbohydrate deficient diet groups, the blood sugar level of ginseng-administered rats is decreased compared with that of control. the oxidation rate of glucose by rat liver slice indicates that only in the carbohydrate deficient diet groups, from the 6th to 15th hour, the oxidation rate of glucose in ginseng-administered group is increased, compared with that of the control. But there is no statistical significance between them. It is thought that Panax Ginseng would effect more on the enzyme system involved in gluconeogenesis and glycogenesis rather than that of carbohydrate oxidation.

  • Stroke occurs when local thrombosis, embolic particle or the rupture of blood vessele interrupts the blood floe to the brain. $\beta$-estradiol 17-valerate has been reported to exert neuroprotective effects when administered before an ischemic insult. Recently, the pathophysiology of cerebral ischemia has been studied extensively in rat with various methods. In the present study, we investigates whether $\beta$-estrodiol 17-valerate can protect against brain injury. RNA sample were extracted from the hippocampus of female rat, reverse-transcription in the presence of [$\alpha$32p] dATP. Differential gene express-ion profiles were revealed (Bone morphogenetic protein type 1A receptor, Protein disulphide isomerase, Leukemia inhibitor factor receptor, cytochrome bc- 1 complex-x core P, thiol-specific antioxidant protein). RT-PCR was used to validate the relative expression pattern obtained by the cDNA array. The precise relationship between the early expression of recovery genes and stroke is a matter of luther investigation.   • This experiment was undertaken to investigate the effect of splenectomy on the hematology and maintenance of pregnant rats. Those animals that had shown regular 4 or 5 day estrous cycles for more than two cycles were used. The day after mating ws designated Day 0 of pregnancy. Spleen was removed from Day 0(early), 6(middle) and 13(late) of pregnant rat, respectively. Blood was collected on Day 1, 7, 14 and 21 of the pregnancy. 1. The total serum protein was increased significantly (P<0.05) to the progress of pregnancy in control rat. All rats that splenectomized groups were decreased significantly(P<0.05) at Day 21 of pregnancy than control rats. 2. Albumin was not significantly changed. 3. Globulin was decreased significantly(P<0.05) between contol and middle treatment at Day 7, 14 of pregnancy. 4. Glucose was increased for the due to the pregress of pregnancy. It was no significance differences among the each groups. 5. Rate of abortion was increased in groups of early and middle of splenectomy compared with control. 6. Period of pregnancy was delayed in middle and late splenectomized gruops companed with control. 7. Litter size was lowed significantly (P<0.05) in early and middle splenectomized groups compared with control.

  • With the purpose of producing easily the basal ganglia infarction into Chen's, scerebral ischemic model which is almost cortical infarct made by the ligation of distal part of MCA and 1 hr obliteration of both common carotid arteries in rat, the MCA obstruction was extended between rhinal fissure and olfactory tract with electrocauterization in place of 10-0 silk suture ligation of distal part of MCA. Both original Chen's model and modified Chen's have shown the cortical infarction in dorsolateral & lateral frontoparietal cortex, but not any infarction in basal ganglia. However, the modified Chen's model have shown the effect of average 12% increase in cortical infarct than that of original Chen's model. This experimental results suggest the modified Chen's model can not reduce the blood flow of the lateral lenticulostriatal artery enough to make the basal ganglia infarction and that blood circulation of basal gagglia under its condition is probably being kept partly through the posterior cerebral artery via vertebral artery. Therefore, The follow-up observation on ischemic time lapse would be needed.

The Effect of Saponin Fraction of Panax ginseng C.A.Meyer on the Biosynthesis of Androgens in Rat Testis (인삼 사포닌이 쥐의 정소에서의 Androgen 생합성에 미치는 영향)

  • It was attempted to observe the effects of ginseng saponin, one of the major components of the roots of Panax ginseng, on androgen biosynthesis from cholesterol in vitro as well as in vivo in rat testis. Ginseng saponin was administered by stomach tubing prior to intraperitoneal injection of cholesterol containing (4-14C)-cholesteroll into adult male rats and the liver, testis and blood serum were analyzed. The first high radioactivity of the liver and blood serum of test animal was observed at 6 hours after radioactive cholesterol injection, while that of control appeared at 12 hours after the injection. In the case of testis, the first high radioactivity of test group appeared between 4 and 6 hours after the radioactive cholesterol injection, while that of control appeared at 10-14 hours. Analysis of radioactivity distribution of cholesterol, androstenedione and testosterone in the testis of rats fed with/without ginseng saponin piror to (4-14C)-cholesterol injection showed that the saponin stmulated the synthesis of androgens from cholesterol. This was confirmed again by in vitro experiment using testis homogenate as an enzyme source. From the above experimental results, it was suggested that the ginseng saponin stimulates both cholesterol transport and the biosynthesis of androgens from cholesterol in rat testis.
