Error Analysis for Electromagnetic Surface Velocity and Discharge Measurement in Rapid Mountain Stream Flow (산지하천의 전자파 표면유속 측정에 기반한 유량 및 유속 관측 오차 분석)

  • Fixed Electromagnetic Wave Surface Velocimetry (Fixed EWSV) has been started to be used to measure flood discharge in the mountain stream, since it has various advantages such that it works well to continuously measure stream discharge even in the night time as well as very strong weather. On the contrary, the Fixed EWSV only measures single point surface velocity, thus it does not consider varying feature of the transverse velocity profile in the given stream cross-section. In addition, a conventional value of 0.85 was generally used as the ratio for converting the measured surface velocity into the depth-averaged velocity. These aspects could bring in error for accurately measuring the stream discharge. The capacity of the EWSV for capturing rapid flow velocity was also not properly validated. This study aims at conducting error analysis of using the EWSV by: 1) measuring transverse velocity at multiple points along the cross-section to assess an error driven by the single point measurement; 2) figuring out ratio between surface velocity and the depth-averaged velocity based on the concurrent ADCP measurements; 3) validating the capacity of the EWSV for capturing rapid flow velocity. As results, the velocity measured near the center by the fixed EWSV overestimated about 15% of the cross-sectional mean velocity. The converting ratio from the surface velocity to the depth-averaged velocity was 0.8 rather than 0.85 of a conventional ratio. Finally, the EWSV revealed unstable velocity output when the flow velocity was higher than 2 m/s.

Alveolar restoration following rapid maxillary expansion with and without corticotomy: A microcomputed tomography study in sheep

  • Objective: This study examined bone microstructure restoration after rapid maxillary expansion (RME) with and without corticotomy over multiple retention periods. Methods: Eighteen male Dorper sheep were randomly distributed into three groups (n = 6 each group): group 1, RME with corticotomy on the buccal and palatal sides; group 2, conventional RME treatment; and group 3, no treatment. Post-RME, trabecular bone microstructure and new bone formation were evaluated by using microcomputed tomography (microCT) and histomorphometry after a 4- or 12-week retention period. Intergroup differences in bone quality and bone remodeling were analyzed by using two-way analysis of variance with Bonferroni post-hoc test. Results: The bone volume fraction (bone volume [BV]/total volume [TV]) values relative to the control in groups 1 and 2 were 54.40% to 69.88% after the 4-week retention period and returned to approximately 80% after the 12-week retention period. The pooled BV/TV values of the banded teeth in groups 1 and 2 were significantly lower than those of the control after the 4-week retention period (p < 0.05). However, after the 12-week retention period, the pooled BV/TV values in group 2 were significantly lower than those in groups 1 and 3 (p < 0.05). Histomorphological analysis showed that the new bone formation area in group 1 was approximately two to three times of those in group 2 and control. Conclusions: Corticotomy significantly enhanced the restoration of bone quality after the retention periods for banded teeth. This benefit might result from the increased new bone formation after corticotomy.

  • Background and objective: If the Nelumbo nucifera spreads in a wetland at a high density, it can have considerable positive and negative ecological effects on habitats. For this reason, it is necessary to precisely investigate the impacts of its rapid proliferation. This study was conducted to propose the distribution and management of N. nucifera, which can cause the degradation of wildlife habitats due to the rapid spread of internal and external environmental factors that may affect the Junam wetland ecosystem. Methods: For the investigation and analysis of physical and ecological characteristics, factors of the abiotic environment such as general weather conditions, topography and water depth structure, and soil and water quality analysis, and bioenvironment characteristics such as changes in the N. nucifera community distribution were evaluated. To assess whether the differences in the soil depth and physicochemical characteristics between the N. nucifera community and the aquatic plant community are statistically significant, a One-way ANOVA was executed. Results: N. nucifera was presumably introduced in approximately 2007 and observed at a prevalence of only 0.8% in 2009, but had expanded to 11.1% in 2014. After that, the area was increased to 19.3% in 2015 and 40.0% in 2017, about twice that of the previous survey year. The rapid diffusion of an N. nucifera colony can have adverse effects on wildlife habitats and biodiversity at Junam Wetland. To solve these problems, four management methods can be proposed; water level management, mowing management, installation of posts and removal of lotus roots. Control of the N. nucifera community using these methods was judged to be suitable for cutting and water level management when considering expansion rate, water level variation, and wildlife habitat impacts. Conclusion: As the biotic and abiotic environmental factors are different for each wetland, it is necessary to determine the timing and method of management through a detailed investigation.

  • The runway occupancy time of landing aircraft is an important factor in determining runway capacity. The purpose of this study is to suggest improvement measures for runway occupancy time to improve the operation of existing airports. In order to derive improvement measures, a comparative analysis was conducted on the effectiveness of improvement using aircraft operation status data for specific days at the case airport. The FAA REDIM model was used to analyze the improvement plan, and the improvement application function of the model was used to confirm the effect of improving runway capacity by adding a rapid escape taxiway to an airport without a rapid escape taxiway. This study's approach can be applied to the derivation of runway improvement measures and preliminary prediction of effectiveness, and it presents cases that can be applied to future airport construction projects or airport improvement projects.

Development of a Rapid Method for the Screening of Conjugated Linoleic Acid (CLA)-Producing Strains of Bifidobacterium breve

  • This study was performed to isolate some strains of Bifidobacterium breve from fecal materials of neonates and to screen them for the biotransformation activity of converting linoleic acid into conjugated linoleic acid (CLA). Fecal samples were collected from twenty healthy neonates between 14 and 100 days old, and four hundred colonies were randomly selected from a Bifidobacterium selective transoligosaccharide medium. A duplex polymerase chain reaction technique was developed for the rapid and accurate molecular characterization of the B. breve strains that have been reported to show the species-specific characteristic of CLA production. They are identified by 16S ribosomal DNA, fructose-6-phosphate phosphoketolase encoding genes (xfp), and rapid pulsed field gel electrophoresis. Thirty-six isolates were identified as B. breve, and just two of the 12 neonates were harboring B. breve strains. Each isolate showed different CLA-producing ability in the spectrophotometric assay. All of the positive strains from the primary spectrophotometric assay were confirmed for their CLA-producing activities using gas-chromatographic analysis, and their conversion rates were different, depending on the strain isolated in this study. Some strains of B. breve were successfully isolated and characterized based on the CLA-producing activity, and further studies are necessary to characterize the enzyme and the gene responsible for the enzyme activity.

Development of a Novel Immunochromatographic Assay for Rapid Detection of OXA-23 β-lactamase-producing Acinetobacter baumannii

  • Among the several agents causing carbapenem resistance of Acinetobacter baumannii, the most common cause is OXA-23 ${\beta}$-lactamase, which is known to hydrolyze carbapenem. To effectively control dissemination of carbapenem-resistant Acinetobacter baumannii (CRAB), development of both rapid and easy-to-use detection methods are required. The aim of this study is to develop a novel immunochromatographic assay (ICA) for rapid detection of OXA-23 ${\beta}$-lactamase. Of the seven monoclonal antibodies (mAbs) screened by ELISA, four mAbs (4G6, 4H6, 6G4, 9A4) exhibited high reactivity. Of these four specific antibodies, the combination of 6G4/4G6 showed the greatest reactivity and this combination of mAbs (6G4/4G6 mAbs) was used to develop the OXA-23 ${\beta}$-lactamase ICA. Of 102 A. baumannii isolates tested, the OXA-23 ${\beta}$-lactamase ICA results were consistent with PCR analysis except one false positive and one false negative isolate. The overall sensitivity and specificity were 98.36% and 97.56%, respectively. In conclusion, to the best of our knowledge, we have developed the first specific antibody set to detect OXA-23 ${\beta}$-lactamase using an ICA kit. This novel ICA can be used as a reliable and easy-to-use immunological assay for detection of OXA-23 ${\beta}$-lactamase producing CRAB in clinical laboratories.

Rapid Evaluation of Chemical Components of Rice Grain Using Near Infrared Spectroscopy (근적외분광분석법에 의한 미질관련 성분 측정)

  • This study was conducted to establish the rapid evaluation method of chemical components of rice grain on the basis of non-destructive method. A near-infrared reflectance spectroscopic(NIRS) method was utilized, for the determination of amylose, protein, magnesium, and potassium content of rice. A multiple linear regression analysis for the data obtained by standard laboratory methods and NIRS method was carried out to make a calibration. The standard error of prediction for amylose, protein, magneisum and potassium content were 0.88%, 0.28%, 12.62mg and 10.79mg, respectively. It was concluded that the NlRS method can be useful the rapid determination of amylose, protein, magnesium and potassium content instead of the existing laboratory method.

Extraction of paddy field in Jaeryeong, North Korea by object-oriented classification with RapidEye NDVI imagery (RapidEye 위성영상의 시계열 NDVI 및 객체기반 분류를 이용한 북한 재령군의 논벼 재배지역 추출 기법 연구)

  • While utilizing high resolution satellite image for land use classification has been popularized, object-oriented classification has been adapted as an affordable classification method rather than conventional statistical classification. The aim of this study is to extract the paddy field area using object-oriented classification with time series NDVI from high-resolution satellite images, and the RapidEye satellite images of Jaeryung-gun in North Korea were used. For the implementation of object-oriented classification, creating objects by setting of scale and color factors was conducted, then 3 different land use categories including paddy field, forest and water bodies were extracted from the objects applying the variation of time-series NDVI. The unclassified objects which were not involved into the previous extraction classified into 6 categories using unsupervised classification by clustering analysis. Finally, the unsuitable paddy field area were assorted from the topographic factors such as elevation and slope. As the results, about 33.6 % of the total area (32313.1 ha) were classified to the paddy field (10847.9 ha) and 851.0 ha was classified to the unsuitable paddy field based on the topographic factors. The user accuracy of paddy field classification was calculated to 83.3 %, and among those, about 60.0 % of total paddy fields were classified from the time-series NDVI before the unsupervised classification. Other land covers were classified as to upland(5255.2 ha), forest (10961.0 ha), residential area and bare land (3309.6 ha), and lake and river (1784.4 ha) from this object-oriented classification.

Establishing of a rapid analytical method on uranium isotopic ratios for the environmental monitoring around nuclear facilities (원자력 시설 주변 환경 감시를 위한 토양 중 우라늄 동위원소 신속 분석법 확립)

  • The uranium isotopic ratio in environmental samples around nuclear facilities is important because it reveals information regarding illegal activities or anthropogenic pollution. Determination of uranium isotopes, however, is a challenging task requiring much labor and time because of the complex separation procedures and lengthy process. In this study, a rapid determination method for uranium isotopes in environmental samples was developed using. The sample was completely decomposed using the alkali fusion method. The separation procedure using extraction chromatography (UTEVA) was simplified in a single step without any further removal process for Si and major matrix elements. The established method can be completed within 3 h from sample dissolution to ICP-MS measurement. Most matrix elements and uranium isotopes in the soil samples were well separated and purified. Five types of were used to assess the method's accuracy and precision for a rapid uranium analysis method. The analytical accuracy for all CRM samples ranged from 95.1 % to 97.8 %, and the relative standard deviation was below 3.9 %. From the analytical results, one may draw conclusions that the evaluated method for uranium isotopes using alkali-fusion, the extraction chromatography process, and ICP-MS measurements is fast and fairly reliable owing to its recovering efficiencies. Thus, it is expected that the evaluated method can contribute to the improvement of environmental monitoring ability.

The study of manganese removal mechanism in aeration-sand filtration process for treating bank filtered water (강변여과수 처리를 위한 포기-모래여과공정에서 망간제거 기작에 관한 연구)

  • It is well known that manganese is hard to oxidize under neutral pH condition in the atmosphere while iron can be easily oxidized to insoluble iron oxide. The purpose of this study is to identify removal mechanism of manganese in the D water treatment plant where is treating bank filtered water in aeration and rapid sand filtration. Average concentration of iron and manganese in bank filtered water were 5.9 mg/L and 3.6 mg/L in 2008, respectively. However, their concentration in rapid sand filtrate were only 0.11 mg/L and 0.03 mg/L, respectively. Most of the sand was coated with black colored manganese oxide except surface layer. According to EDX analysis of sand which was collected in different depth of sand filter, the content of i ron in the upper part sand was relatively higher than that in the lower part. while manganese content increased with a depth. The presence of iron and manganese oxidizing bacteria have been identified in sand of rapid sand filtration. It is supposed that these bacteria contributed some to remove iron and manganese in rapid sand filter. In conclusion, manganese has been simultaneously removed by physicochemical reaction and biological reaction. However, it is considered that the former reaction is dominant than the latter. That is, Mn(II) ion is rapidly adsorbed on ${\gamma}$-FeOOH which is intermediate iron oxidant and then adsorbed Mn(II) ion is oxidized to insoluble manganese oxide. In addition, manganese oxidation is accelerated by autocatalytic reaction of manganese oxide. The iron and manganese oxides deposited on the surface of the sand and then are aged with coating sand surface.
