Real-Time RT-PCR for Validation of Reovirus Type 3 Safety During the Manufacture of Mammalian Cell Culture-Derived Biopharmaceuticals (세포배양 유래 생물의약품 생산 공정에서 Reovirus Type 3 안전성 검증을 위한 Real-Time RT-PCR)

  • Validation of viral safety is essential in ensuring the safety of mammalian cell culture-derived biopharmaceuticals, because numerous adventitious viruses have been contaminated during the manufacture of the products. Mammalian cells are highly susceptible to Reovirus type 3 (Reo-3), and there are several reports of Reo-3 contamination during the manufacture of biopharmaceuticals. In order to establish the validation system for the Reo-3 safety, a real-time RT-PCR method was developed for quantitative detection of Reo-3 in cell lines, raw materials, manufacturing processes, and final products as well as Reo-3 clearance validation. Specific primers for amplification of Reo-3 RNA was selected, and Reo-3 RNA was quantified by use of SYBR Green I. The sensitivity of the assay was calculated to be $3.2{\times}10^0\;TCID_{50}/ml$. The real-time RT-PCR method was proven to be reproducible and very specific to Reo-3. The established real-time RT-PCR assay was successfully applied to the validation of Chinese hamster ovary (CHO) cell artificially infected with Reo-3. Reo-3 RNA could be quantified in CHO cell as well as culture supernatant. When the real-time RT-PCR assay was applied to the validation of virus removal during a virus filtration process, the result was similar to that of virus infectivity assay. Therefore, it was concluded that this rapid, specific, sensitive, and robust assay could replace infectivity assay for detection and clearance validation of Reo-3.

Impact of Corporate Entrepreneurship, Human Resource Innovation on the Firms' Innovation Activities and Nonfinancial Performance: A Exploratory Research of KOSDAQ Companies (사내기업가정신, 인적자원혁신성이 기업혁신활동과 비재무적 성과에 미치는 영향에 관한 탐색적 연구)

  • New business management methods different from the past are necessary because of the rapid changes of the corporates' environment. KOSDAQ(Korean Securities Dealers Automated Quotation) companies should be expected the more affirmative business performance of companies by listing, but it is a well-known that they have problems of low business performance mostly. This paper aims to investigate the influential factors on enhancing corporate innovation and nonfinantial business performance, and to clarify practical measures and present a solution of KOSDAQ companies' problems through analysis of previous researches and an empirical research. This research present corporate entrepreneurship and human resources innovation as impact factors on the business performance to apply finely the path of technological innovation for the solution of the relevance investigation limit between the complexity of corporates' innovation paths and the firms' performance. And also knowledge management activities and external networks management or the firms have been adopted as a corporate innovation activities for free from quantitative measures, such as conventional research and development(R&D) activities by considering recent corporates' knowledge business operations. The results of the empirical analysis shows that significant impact factors on corporate innovation activities are the firms' propensities of competitive advantage initiative, risk taking and chief executive officer's innovation. These can be interpreted that the CEOs' innovation propensity should be enhanced for stimulating corporate's innovaton activities, which include the CEOs' interest in the development of new technology, the exploiting new businesses and their support of the innovation discipline for employees. In addition, it can be said that it is necessary to intensify more initiatives within those enterprise for enhancing the competitive advantage in the identical industry. The significant impact factors of corporate entrepreneurship and human resource innovation on the non-financial performance are resulted as the propensities of firms' competitive advantage initiative, CEOs' innovation and employees' innovaton. This shows that the higher propensities of firms' competitive advantage initiative, CEOs' innovation and employees' innovaton, the higher the cognitive degrees of business performance within each corporate, which include the members' awareness about firms' sales growth, market share growth, profit ratio growth, customers' preference and corporates' awareness.

Estimation on the Distribution Function for Coastal Air Temperature Data in Korean Coasts (한반도 연안 기온자료의 분포함수 추정)

  • Water temperature due to climate change can be estimated using the air temperature because the air and water temperatures are closely related and the water temperatures have been widely used as the indicators of the environmental and ecological changes. It is highly necessary to estimate the frequency distribution of the air and water temperatures, for the climate change derives the change of the coastal water temperatures. In this study, the distribution function of the air temperatures is estimated by using the long-term coastal air temperature data sets in Korea. The candidate distribution function is the bi-modal distribution function used in the previous studies, such as Cho et al.(2003) on tidal elevation data and Jeong et al.(2013) on the coastal water temperature data. The parameters of the function are optimally estimated based on the least square method. It shows that the optimal parameters are highly correlated to the basic statistical informations, such as mean, standard deviation, and skewness coefficient. The RMS error of the parameter estimation using statistical information ranges is about 5 %. In addition, the bimodal distribution fits good to the overall frequency pattern of the air temperature. However, it can be regarded as the limitations that the distribution shows some mismatch with the rapid decreasing pattern in the high-temperature region and the some small peaks.

  • Korea is far behind other OECD countries in economy-wise productivity: Korea's labor productivity in terms of GDP per hour worked is the lowest among OECD countries. Against the existing productivity gap, there is a worrying sign in Korea's investment trend - rapid fall in machinery and equipment investment with slow increase in R&D investment. The challenge facing Korea is how to transform her economy from catching-up model to a knowledge-based one. The paper shows that, in tandem with the structural changes that today's Korean industries are experiencing, industry's innovation system is also changing. Innovation networks are emerging as the result of economy-wise restructuring since the financial crisis of 1997 and, though still not a dominant force, the newly emerging innovation networks will be the main threads of industry's innovation activities in the future. The changes in industrial innovation system would positively contribute in raising the productivity of the Korean economy. The paper contains a case study on Korea's automobile industry in order to highlight some of main characteristics of the structural changes, in addition to a chapter that gives an overview of the evolutionary paths of the Korea's industrial innovation. The paper assesses that changes can be considered as a positive sign of future growth perspective; but there are further challenges to make the Korea's industrial innovation system effective. The list of such challenges includes strengthening upstream sectors of currently leading industries, expanding the innovation base to SME and promoting technological co-operation between domestic firms and foreign firms.

The Production Objectives and Optimal Standard of Density Control Using Stand Density Management Diagram for Pinus densiflora Forests in Korea (임분밀도관리도를 이용한 소나무림의 적정 임분밀도 관리 기준 및 수확목표)

  • This study has utilized the stand density management diagram to devise an efficient management standard for the stand density for Pinus densiflora that secures the health of the stands and predicted the harvest goals. The appropriate stand control level was estimated by modeling the relationship of the relative yield index (Ry) to the ratio of slender trees within the stand through an exponential function; the coefficient of determination ($R^2$) was found to be 0.424 according to the estimation. The ratio of slender trees within the stand showed a tendency of rapid increase at a certain relative yield index; with this relational function, the appropriate Ry value of 0.84 was obtained. By estimating the curve of the Ry value 0.84, which was the appropriate stand density management level, as well as the height of dominant trees in the central region of Korea, the production objective for each site index was set. Assuming that the final age by the site indices ranged from 10 to 16 for the P. densiflora in central region of Korea, the number of production was estimated to be between 426 to 1,311 trees per ha. It was predicted that the production of medium-diameter logs larger than 30 cm in diameter is possible for the target DBH at a site index of more than 16; small-diameter logs larger than 20 cm in diameter for site indices 12 and 14 enabled, and small-diameter logs of less than 20 cm for site index 10.

  • Combustion characteristics for co-firing of biomass (Walnut Shell) as blending fuel in coal fired boiler have investigated using thermogravimetric analyser (TGA) and drop tube reactor (DTR). The results show that devolatilization and char combustion for WS occurs at lower temperature than those of existing coals and has lower activation energy value, which is resulting in higher reactivity. When the WS is blended with coal, TGA results show linear profiles depending on blending ratio for each fuel. However, DTR results exist the non-additive phenomena for blending of WS. As blending ratio of WS increase, the UBC decrease at BBR 5%, but the UBC rather increase from BBR 10% due to oxygen deficiency formed from rapid combustion of WS. This paper propose that fuel lean condition by oxygen rich lead to higher blending ratio of biomass by solving the oxygen deficiency condition.

  • Solid phase enzyme linked immunosorbent assay (ELISA) was developed to detect the whole cells of Edwardsiella tarda from infected tissues of flounder. Cross-reaction test was performed by ELISA against fish pathogens such as A. hydrophila ATCC7966. V. anguillarum HYUFP5001, Y, ruckeri 11-4, E. ictaluri and Streptococcus sp. NG8206. Rabbit anti-E, tarda Edk-2 sera highly cross-reacted with A. hydrophila ATCC7966 and V. anguillarum HUFP5001. However, the cross-reaction was removed by using the anti-serum pre-adsorbed with A, hydrophila ATCC7966 FKC. The intra-species cross-reaction among E. tarda isolates was very high. ELISA with the whole cell antigens present in tissue homogenate appeared with highly decreased sensitivity, presumably by the co-coating of lipid or proteins in tissues. Thus, it would be necessary to use the infected tissue homogenates diluted more than 100 times with PBS for diagnosis. Interestingly, compared with the using of FKC antigen, the direct detection of viable cells in tissue homogenate showed more sensitive results with detection limit of $1{\times}10^3$ cells/ml in buffer or diluted tissue homogenate. Consequently, the ELISA method developed in this study was specific, rapid and sensitive for diagnosing edwardsiellosis.

Surface Checking Reduction Effect of Preservative-treated Korean Larch Round-woods with Various Physical Treatments (물리적 처리에 따른 낙엽송 방부원주목의 표면할렬 방지효과)

  • In this study, the surface check reduction effect of preservative-treated Korean larch round-wood was investigated by applying three physical treatments, such as incising, kerfing, and kiln-drying. Moreover, the possibility of long-term service life was also checked by comparing the depth of surface check and the penetration depth of preservative. A rapid gradient of wood moisture content between surface and center causes a surface check. Thus, the effect of reducing surface check was carried out at the lowest equilibrium moisture content (EMC) condition, temperature ($2^{\circ}C$), humidity (44.6%), in outdoor locations in Korea until the test specimens reach to the 8.6% EMC. As a result, the preservative-treated specimens without incising, kerfing, kiln-drying (Type C) could not ensure the long-term service life due to a large surface check. Because the surface check depth of all specimens was deeper than the penetration depth of preservative. In case of the incising treated specimens (Type I), 80% of them, the depth of surface check was not deeper than the penetration depth of preservative. However, when a kerfing was additionally treated (Type B), a possibility of happening deeper surface check than penetration depth of preservative was increased rather than Type I.

  • Three edible species, e.g. Lyophyllum sp. and Armillaria mellea introduced from Japan in 1979 and Auriaularia auricula-judae from Taiwan in 1978 were selected for artifcial cultivation. The media, MSDS for Lyophyllum sp, MHS and PDS for A. auricula-judae, and MSDS and MHS for A. mellea were selected. The range of optimum pH of Lyophyllum sp. was 6.0 and pH 8.0 for A. auricula-judae and pH 6.0 to 7.0 for A. mellea. The range of optimum temperature for the mycelial growth of Lyophyllum sp. was 25 to $30^{\circ}C,\;30^{\circ}C$ for A. auricula-judae and $25^{\circ}C$ for A. mellea. When the moisture content 75 to 80% (w/w) in the substrates, the mycelial growth of Lyophyllum sp. and A. auricula-judae, was rapid. 65% (w/w) for A. mellea. As the spawning substrates, wheat for Lyophyllum sp. oak sawdust for A. auricula-judae and poplar sawdust for A. mellea were selected, respectively. In this experiment, yields of Lyophyllum sp. and A. auricula-judae were high on the poplar sawdust added 20% (w/w) of rice bran. Lyophyllum sp. and A. auricula-judae for sources of new edible mushrooms seems to be possible.

Effect of Transplanting Dates on the Occurrence of Rice Stripe and Black-Streaked Dwarf Virus Diseases in Yeongnam District (영남지방에서 벼 이앙시기가 줄무늬잎마름병과 검은줄무늬오갈병의 발병에 미치는 영향)

  • The experiments were conducted to clarify the influence of transplanting date on the occurrence of rice virus in field condition of 1984. The rate of RBSDV (rice black-slreaked dwarf virus) viruliferous vector, smaller brown planthopper (Laodelphax striatellus Fallen), was shown to be $13.6\%$ at the 2nd adult and that of rice stripe virus (RSV) viruliferous was $6.7\%$ at the 2nd adult. The vector in the field was begun to come from May 29, the maximum densities were 19.6 insects per hill on June 13 in cultivar Chucheongbyeo, 19.3 in Nagdongbyeo, 7.4 in Cheongcheongbyeo and 4.9 in Samgangoyes. The number of vectors per hill was inclined to increase by early transplanting. Although the infection of rice virus in nursery bed was not recognized until May 30 transplanting, the nursery infection could be seen from June 10 transplanting. The highest rate of nursery infection with RSV was $4.1\%$ at June 10 transplanting plot, and that of RBSDV was $14.2\%$at June 20 trans planting plot. The infection of rice virus in paddy field was the highest at May 20 transplanting plot, the lowest at July 10 plot. The earlier transplanting, the more severe occurrence of rice viruses. Occurrence of infected plants with RBSDV was shown to increase more rapidly at May 20 and May 30 planting plot than May 10 plots. However, the occurrence of infected plant with RSV was more rapid at May 10 transplanting plot than May 20 and 30.
