Molecular Cloning of Two Genes Encoding Cinnamate 4-Hydroxylase (C4H) from Oilseed Rape (Brassica napus)

  • Cinnamate 4-hydroxylase (C4H) is a key enzyme of phenylpropanoid pathway, which synthesizes numerous secondary metabolites to participate in development and adaption. Two C4H isoforms, the 2192-bp BnC4H-1 and 2108-bp BnC4H-2, were cloned from oilseed rape (Brassica napus). They both have two introns and a 1518-bp open reading frame encoding a 505-amino-acid polypeptide. BnC4H-1 is 57.73 kDa with an isoelectric point of 9.11, while 57.75 kDa and 9.13 for BnC4H-2. They share only 80.6% identities on nucleotide level but 96.6% identities and 98.4% positives on protein level. Showing highest homologies to Arabidopsis thaliana C4H, they possess a conserved p450 domain and all P450-featured motifs, and are identical to typical C4Hs at substrate-recognition sites and active site residues. They are most probably associated with endoplasmic reticulum by one or both of the N- and C-terminal transmembrane helices. Phosphorylation may be a necessary post-translational modification. Their secondary structures are dominated by alpha helices and random coils. Most helices locate in the central region, while extended strands mainly distribute before and after this region. Southern blot indicated about 9 or more C4H paralogs in B. napus. In hypocotyl, cotyledon, stem, flower, bud, young- and middle-stage seed, they are co-dominantly expressed. In root and old seed, BnC4H-2 is dominant over BnC4H-1, with a reverse trend in leaf and pericarp. Paralogous C4H numbers in Brassicaceae genomes and possible roles of conserved motifs in 5' UTR and the 2nd intron are discussed.

Detection Characteristics of Gamma-Irradiated Seeds by using PSL, TL, ESR and GC/MS (PSL, TL, ESR 및 GC/MS 분석을 통한 감마선 조사된 유지종실류의 검지 특성 연구)

  • In this study, we investigated the applicability of the photostimulated luminescence (PSL), thermoluminescence (TL), electron spin resonance (ESR) and gas chromatography/mass spectrometry (GC/MS) methods for 5 seeds which are not allowed to be irradiated in Korea. All 5 seeds including evening primrose seed, safflower seed, rape seed, sunflower seed and flax seed were analyzed. Samples were irradiated at 1~10 kGy using a $^{60}Co$ gamma-ray irradiator. In PSL study, the photon counts of all the unirradiated samples showed negative (lower than 700). The photon counts of irradiated (1, 5, 10 kGy) samples showed positive (higher than 5,000). In TL analysis, results showed that it is possible to apply TL method to all foods containing minerals. In ESR measurements, the ESR signal (single-line) intensity of irradiated foods was higher than non-irradiated foods. The hydrocarbons 1,7-hexadecadiene ($C_{16:2}$) and 8-heptadecene ($C_{17:1}$) from oleic acid were detected only in the irradiated samples before and after the treatment at doses ${\geq}$ 1 kGy, but they were not detected in non-irradiated samples before and after treatment. These two hydrocarbons could be used as markers to identify irradiated safflower seed, rape seed, Sunflower seed and flax seed. And then, the hydrocarbons 1,7,10-hexadecatriene ($C_{16:3}$) and 6,9-heptadecadiene ($C_{17:2}$) from linoleic acid were detected in the evening primrose seed, safflower seed and sunflower seed. According to the results, PSL, TL and GC/MS methods were successfully applied to detect the irradiated foods. It is concluded that PSL, TL and GC/MS methods are suitable for detection of irradiated samples and a combined method is recommendable for enhancing the reliability of detection results.

Studies on the Sterol of Sesame Oil Sold In Markets and Used in Restaurants (시중(市中) 참기름의 Sterol에 관(關)한 연구(硏究))

  • Methods for the determination of sterols in sesame oils were studied. The sesame oils were saponified and the sterols isolated from the unsaponifiable matter by Florisil column chromatography, and the individual components were determined by means of gas chromatography. Campesterol, ${\beta}-sitosterol$, stigmasterol were found in sesame oil including unknown Ⅰ and Ⅱ. The use of SE-30 gas chromatographic column allows the slow elution, duplication of peaks and relatively low reproducibility, therefore, 3% OV-17 was suitable for the sterol analysis. The result of this study showed that contents of sterols in sesame oil were campesterol 8.4%, stigmasterol 4.5%, ${\beta}-sitosterol$ 33.9% and others 53.0% involving 8.8% of unknown I and 44.3% of unknown Ⅱ. There has been no specific test available for identifying the sesame oil among common edible oils. But the ratio of sterols in sesame oils allowed the estimation of genuiness. The ratio of sterols vs. campesterol in genuine sesame oils were stigmasterol 0.3- 0.6, ${\beta}-sitosterol$ 3.0-3.8 and unknown Ⅱ 3.0, respectively. The 65 samples were composed of genuine sesame oil 40%, mixed rape seed oil 3%, cotton seed oil 1. 5% others were reused soybean oil or re-extracted oil.

  • Soybean and rape seed are common feedstocks for biodiesel product ion in USA and Europe, respectively. On the other hand, South Eastern countries like Malaysia and Indonesia have surplus palm crops. However due to substantial amount of saturated fats in palm, the palm biodiesel has poor low temperature properties. To improve the low temperature flow properties as biodiesel, the dependence of the cold filter plugging point (CFPP) on the fatty acid compositions was examined. Two different kinds of biodiesels, palm and soybean biodiesels, were blended with the different volume ratios. And the low temperature flow properties of 0.5%, 1%, and 5% biodiesel in diesel blend fuels was tested. The decrease of CFPP was not observed for BD1 with Palm BD. Also, WDI test didn't exceed in the range of 4oC by the mixing of Palm BD upto 5% in commercial diesels.

  • Performance tests of small diesel engine were carried out, using the blend fuel oil as the substitute fuel oil and the following results were obtained. (1) The character at the blend oil as substitute fuel for small diesel engine was examined. (2) In the case of operating small diesel engine with blend oil, the exhaust gas volume increases at 8% in comparison with diesel oil. (3) In the case of operating small diesel engine with blend oil, the fuel consumption Increases at 3% in comparison with diesel oil. (4) In the brake thermal efficiency, blend oil is similar to diesel oil.

  • Compared with ordinary varieties, the cold resistant lines were heavy in fresh weight of seedling, low in stalk elongation. Their leaf-types were complete runner types, and they were high in wintering rate, and were abundant in the branch number and in pod numbers per branch. They were high in the seed yield.

Influence of Vegetable oils on the Serum Cholesterol of Rabbit (가돈의 혈청 Cholesterol에 미치는 식물유의 영향 (1))

  • The level of serum cholesterol was successively determined for 4 weeks after the basic diet feeding for 8 weeks in the rabbits. It was observed that the individual serum cholesterol levels were highly elevated by the oral administration of cholesterol for 18 days, and then the rabbit were fed by sesame and rape seed oils for 11 weeks to survey the effect of the oils which was playing a role of decreasing factor for serum cholesterol values. The results were obtained as follows ; 1. The rabbit's serum cholesterol level was 128±29.0mg/dl in average, it was higher in female (162±50.6) than male(95±17.6). 2. It can be possible elevating experimentally the level of serum cholesterol by the cholesterol administration per os. 3. There were significant effect of the vegetable oils to decrease the experimental hypercholesteremia in the rabbits.

  • Effect pf rape seed oil cake compast (OM) active carbon(C) compound fertilizer (NPK) and magnesium sulfate (Mg) application on Zoysia koreana growth by lysimeter of 50cm soil column filled with a low fertility loam. Effect on growth was in order of OM>OM . NPK interaction> NPK . active carbon and Mg were not effective OM . NPK interaction effect was negative on leaf width and root dry matter yield. OM and OM . NPK interaction were most effective on rhizome growth. Their effect did not change the ratio of growth rates between aerial part and rhizome but change that between aerial part and root. Leaf width was most sensitive to the treatments.

Studies on the Contents of Special Components and Estimation of Purity of Sesame Oil II (참기름의 특이 성분 함량과 순도 결정에 관한 연구(제2보))

  • The verification of genuine sesame oil can be examined by determination of the ratio of fatty acid. Fatty acids were extracted from the saponifiable substance of sesame oils. Fatty acids were methylated with the 14% boron trifluoride methanol solution and injected into a gas chromatograph with Unisole 3000 column and finally determined the molecular weight by mass spectrometry. The fatty acids in laboratory prepared sesame oils were composed mainly of oleic acid 36.7-42.8% and linoleic acid 39.0-46.6%, including palmitic acid 7.9-9.l%, stearic acid 4.1-5.6%, linoleic acid 0.1-3.0%, arachidic acid 0.5-1.0% and eicosenoic acid 0.1-0.5%. The above results allow the estimation of genuine sesame oil, mixed with rape seed oil, soybean oil, perilla oil, etc. In 53 samples, 14 samples were estimated as genuine and it was found that erucic acid was contained in 31 samples, linoeic acid was highly contained in 14, high quantity of linolenic acid was in 7 and palmitic and oleic acid were highly involved in 3.
