  • As a result of studing on the "Joa-gui Em", we could reach conclusions as follows. 1. "Joa-gui Em" shows in vivo effect of that it supplement Kidney-Um essence and nomalize a physiological function of Kidney-Qi. 2. "Joa-gui Em" supplement Kidney-Um essence, it does not use in The Rise of Insubstantial Fire(음허화왕) body condition. 3. "Joa-gui Em" use in many ways women disease, in case of menstruation and meonpause disorder. 4. "Joa-gui Em" and "Joa-gui Hwan" are modification prescription of "Uk-mi-ji-hwang-hwan. 5. "Joa-gui Em" is a pure prescription in supplement Kidney-Um essence, Roast Glycyrrhizae Radix of prescription has effect of supplement Sam-cho Qi. 6. "Joa-gui Hwan" is similarly "Joa-gui Em." Colla Cornus Cervi and a Colla Cornus Glemmys of prescription have effect of supplement DUMAI and RENMAI CHONGMAI.

Dyeing and Functional Property of Cotton Fabrics dyed with Glycyrrhizae Radix Extract(I) - The Color Depth of Cotton Fabrics by Mercerization & Tannic Acid Treatment - (감초추출물에 의한 면직물의 염색성과 기능성(I) - 머서화, 탄닌산처리에 의한 면직물의 농색화 -)

  • The purpose of this research was to raise K/S value by mercerization and tannic acid treatment in cotton fabrics. K/S value was 2.84-3.56 in mercerized none-mordant fabrics and it is 5.12-5.36 in tannic acid treatment. As the result, it showed higher in processed cotton fabrics than in unprocessed ones. Processed cotton fabrics had high K/S value in pH 3, pre-mordant. Antibacterial activity showed 99.9% in tannic acid and Cu mordanted fabrics. Deodorization and UV-cut effect were showed a higher effect in dyed fabrics than in undyed ones.

Herb medicine Bo-du-san induces caspase dependent apoptosis and cell cycle arrest human gastric cancer cells, SNU-1 (보두산(寶豆散)에 의한 SNU-1 세포의 Apoptosis 유도와 Cell cycle arrest)

  • Objectives : The purpose of this study was to investigate the effect of Bo-du-san (BOS) on apoptosis in human gastric cancer cells, SNU-l cells. BOS, a drug preparation consisting of two herbs, that is, Crotonis Fructus (Strychni ignatii Semen, bodu in Korean) and Glycyrrhizae Radix (Glycyrrhizae uralensis FISCH, Gamcho in Korean). Methodss : In this study, methanol extract of BOS was examined for cytotoxic activity on human gastric cancer cells, SNU-1 cells, using XTT assay, with an IC50 value was 0.7 mg/ml and 0.3 mg/ml at 24 hrs and 48 hrs, respectively. Apoptosis induction by BDS in SNU-l cells was verified by the induction of DNA fragmentation, cleavage of poly ADP-ribose polymerase (PARP), and activation of caspase-3, -8 and -9. Inhibitors of caspase-3, -8 and -9 (Ac-DEVD-CHO, Z-IETD-FMK and Z-LEHD-FMK) efficiently blocked BOS-induced cell death of SNU-l. Resultss : BOS-induced cell death was via caspase dependent apoptosis. Moreover, treatment of BOS result in the decrease the G1/S cycle regulation proteins (cyclin D1 and E) expression and increase CDK inhibitor proteins (p21 and p27) expression, and increase apoptotic protein, p53 expression. Thus, BOS induces apoptosis in SNU-1 cells via cell cycle arrested in G1 phase. Conclusions : These results indicated that BOS has some potential for use as an anti-cancer agent.

The Enhancing Effect of Atopic Dermatitis by cosmetics containing estern medical herbs (한방소재 화장품의 아토피 피부염 개선 효과)

  • Sixteen moderate atopic dermatitis patients were treated with applying toner and cream using estern medical herbs[Estern medical complex(Radix Glycyrrhizae, Radix Angelicae Dahuricae, Rhizoma Ligustici Chuanxiong, Fructus ponciri Seu Aurantii Immaturus, Radix Rehmanniae Praeparata, Radix Puerariae, Rhizoma Corydalis), Cypress, Green tea, Radix Astragali, Red ginseng, Syrup ferment, Apple ferment filtrate] on the atopic area 2-3 times per day for 6 weeks. After 6 weeks of study, the result was a decrease in SCORAD index($26.9{\pm}11.8$ to $14.9{\pm}9.7$(p<0.05)), IgE($641.9{\pm}1294.6$ to $565.8{\pm}1076.8$), Eosinophil count($246.7{\pm}203.5$ to $203.3{\pm}130.7$(p<0.05)) and TEWL($16.7{\pm}5.1$ to $15.4{\pm}8.7$(p<0.05)). And skin hydration was increased($36.6{\pm}10.0$ to $44.0{\pm}10.3$(p<0.01)) maintaining skin pH level. The patient;s and physician's global assessment also improved. Therefore, estern medical herbs may play a role in treatment of atopic dermatitis.

Effects of Bojungikgi-tang on Trimellitic Anhydride-induced Contact Hypersensitivity in BALB/c Mice (보중익기탕(補中益氣湯)이 접촉성 과민반응에 미치는 영향(影響))

  • Objective : The water extract of Bojungikgi-tang (BTE) composed with Ginseng Radix, Astragali Radix, Angelicae gigantis Radix, Atractylodes Rhizoma alba, Aurantii nobilis Pericarpium, Glycyrrhizae Radix, Cimicifuga Rhizoma, and Bupleuri Radix and it has been traditionally used for chronic diseases or weakness after illness in oriental countries. However, little is known about the effects of aqueous extract of BTE on trimellitic anhydride (TMA)-induced contact hypersensitivity (CHS). Therefore, the aim of this study was to investigate the effects of BTE on TMA-induced CHS in BALB/c mice.Methods : In this study, the extract of BTE was prepared by extracting with distilled water at 100℃ for 2.5 h. The extract was freeze-dried following filtration through 0.45 μm filter. Mice were orally administrated with or without BTE extract of different doses (25-200 ㎎/㎏/day) for 28 days. During the challenge period, mice were externally applied with different doses of BTE extract one time per day 30 min before TMA treatment. I examined the effects of BTE on the serum levels of IgE, IgG1, IgG2a, and IgG2b, nitric oxide (NO) and prostaglandin E2 (PGE2) production in isolated peritoneal macrophages, Th2 cytokine production in isolated spleen cells, ear swelling responses, and the leukocyte infiltration induced by TMA.Results : The orally and externally administration of BTE dose-dependently reduced the serum levels of hapten-specific immunoglobulin E (IgE), IgG1 titer, and PGE2 production as well as ear swelling responses and leukocyte infiltration in TMA-induced BALB/c mice. The levels of NO and PGE2 production from peritoneal macrophages stimulated with TMA-BSA were markedly suppressed by pretreatment with BTE in a concentration dependent manner. Furthermore, the levels of Th2 cytokines (IL-4, IL-5, IL-13) production from spleen cells stimulated with ConA were markedly suppressed by BTE treatment in a concentration dependent manner.Conclusions : These results suggest that BTE treatment suppresses chronic contact hypersensitivity, and it can be assumed that the suppression of ear swelling, serum IgE, NO and PGE2 levels, leukocyte infiltration, and Th2 cytokines in an animal model. Therefore, these properties may contribute to the strong anti-CHS response effect of BTE.

Compatibility Analysis Through the System of Chief, Deputy, Assistant, and Envoy for Socheongnyong-tang (소청룡탕(小靑龍湯)의 군신좌사(君臣佐使) 배오(配伍) 분석)

  • Objectives : We analyzed the principle of compatibility of socheongnyong-tang and expanded the range of application in clinical practice. Methods : Socheongnyong-tang was analyzed by the compatibility principle through the system of chief, deputy, assistant, and envoy, but the basic formulas of "Discussion of Cold Damage" and "Synopsis of the Golden Chamber" was used. Results : Socheongnyong-tang treats exterior wind-cold and interior water accumulation. Exterior wind-cold is treated mainly by exterior-releasing herbs constituting mahwang-tang and gyeji-tang in "Discussion of Cold Damage", and interior water accumulation is treated mainly by retained fluid-resolving herbs constituting yeonggamomigangsinha-tang, yeonggamomigagangsinbanhahaengin-tang and yeonggamomigagangsinbanhaengdaehwang-tang in "Synopsis of the Golden Chamber". Depending on the weight of exterior wind-cold and interior water accumulation, the disease condition is classified as a case where exterior wind-cold is heavier than interior water accumulation, a case where exterior wind-cold is lighter than interior water accumulation, and a case where it is equivalent. When exterior wind-cold is heavier than interior water accumulation, chief herbs are Ephedrae Herba and Cinnamomi Ramulus of exterior-releasing herbs series and deputy herbs are Zingiberis Rhizoma and Asari Radix et Rhizoma of retained fluid-resolving herbs series. When exterior wind-cold is equal to the disease condition of interior water accumulation, chief herbs are Ephedrae Herba of exterior-releasing herbs series and Zingiberis Rhizoma of retained fluid-resolving herbs series, and deputy herbs are Cinnamomi Ramulus of the exterior-releasing herbs series and Asari Radix et Rhizoma of retained fluid-resolving herbs series. When exterior wind-cold is lighter than interior water accumulation, chief herbs are Zingiberis Rhizoma and Asari Radix et Rhizoma of retained fluid-resolving herbs series and deputy herbs are Ephedrae Herba and Cinnamomi Ramulus of exterior-releasing herbs series. In any case, assistant herbs are Pinelliae Rhizoma, Paeoniae Radix alba, and Schisandrae Fructus, and envoy herb is Glycyrrhizae Radix preparata. Conclusions : In conclusion, socheongnyong-tang must differently formulate the system of chief, deputy, assistant, and envoy according to the grade of exterior wind-cold and interior water accumulation. These results suggest that socheongnyong-tang can be applied flexibly when applied in clinical practice to enhance the therapeutic effect.

The Growth Promoting Effect of Bifidobacterium bifidum by Combination of Natural Products Bearing Antioxidative Capacity (항산화 천연산물의 조합구성에 의한 Bifidobacterium bifidum의 성장 촉진 효과)

  • The growth of B. bifidum was promoted by natural products bearing antioxidative capacity and mixed combined two, three and four kinds of them. B. bifidum was expressed a good growth by Nelumbo nuclfera gaertner, Corni fructus, Beiamcanda chinensis, alone, and two mixed combinations were composed of Nelumbo nuclfera gaertner and Corni fructus, Nelumbo nuclfera gaertner and Beiamcanda chinensis, Nefumbo nuclfera gaertner and Theae folium, three mixed combinations were oraganized with Nelumbo nuclfera gaertner, Corni fructus and Theae folium, Nelumbo nuclfera gaertner, Corni fructus and Beiamcanda chinensis, Nelumbo nuclfera gaertner, Theae folium and Beiamcanda chinensis, and four mixed combinations were formed with Nelumbo nuclfera gaertner, Theae folium, Beiamcanda chinensis aud Corni fructus, Theae folium, Beiamcanda chinensis, Corni fructus and Glycyrrhizae radix, Nelumbo nuclfera gaertner, Corni fructus, Teae folium and Glycyrrhizae radix, and Nelumbo nuclfera gaertner, Corni fructus, Beiamcanda chinensis and Glycyrrhizae radix. These few mixed combinations promoted cell growth by 2.6 times than that of control, and its antioxidative capacity was also 5.6 times higher, and the ratio of elimination of hydroxyl radical was more than 80% in each dilution rate. As these combinations of natural products could activate some parts of body, they might be applied to pharmaceuitcal applications, functional foods, also expected to multifunctional fermentative beverages.

Analysis of Liquiritigenin, an Aglycone of Liquiritin in Licorice by High Performance Liquid Chromatography (감초 중 리퀴리티게닌 분석법 개발 및 함량분석)

  • Licorice(Glycyrrhizae Radix et Rhizoma) is recorded as the root of Glycyrrhiza uralensis Fischer or Glycyrrhiza glabra Linne or Glycyrrhiza inflata Bat.(Leguminosae) in Korean Pharmacopoeia $9^{th}$ edition (KP9) and Chinese Pharmacopoeia 2005(CP2005), Glycyrrhiza uralensis Fischer or Glycyrrhiza glabra Linne in Japanese Pharmacopoeia 2005(JP2005). It is established the content standard of Glycyrrhizin 2.5 % and liquiritin 1% in KP9 and CP2005. But, according to the reports, all Licorice species were not sufficient for content standard of liquiritin 1.0% for licorice in KP9 and CP2005. It shows different content of liquiritin among G. uralensis, G. glabra and G. inflata. Also, it was reported liquiritin, liquiritin apioside are transformed into liquiritigenin by human internal flora. Therefore, we have studied for the pre-treatment condition and analytical method of liquiritigenin; It was good efficinet in 2M HCl reflux(1 hr) for hydrolysis and in methylene chloride for solvent fractionation. And 1% acetic acid in DW(A) and acetonitrile(B) with gradient condition as a mobile phase was most effective in HPLC analytical condition. According to these experimental methods, we have anlayzed content of liquiritigenin about 77 Licorice sample. In this research, it was also examined the content of liquiritin and liquiritigenin for Glycyrrhizae Radix related growing area. According to the results, we suggested the content standard of glycyrrhizin more than 2.5%, liquiritigenin more than 0.7%(after hydrolysis) of licorice.

Anti-inflammatory Effect of Yukil-san Water Extract on LPS-induced RAW 264.7 Cells (LPS로 활성화된 RAW 264.7 cell에서 NF-𝜅B억제를 통한 육일산(六一散) 물추출물의 염증억제효과)

  • Objectives : Yukil-san (YIS, 六一散; Liu yi san) is composed of Talcum and Glycyrrhizae Radix, the name is said to be derived from the proportion of the two herbal components of the formula. The YIS originated from 'Formulas from the discussion illuminating the Yellow Emperor's Basic Question'(黃帝素問宣明論方; Huang di su wen xuan ming lun fang) written by Liu Wan-Su (劉完素). YIS could clear summerheat, resolve dampness, and augment the qi. This formula may be used to treat the common cold, influenza, acute gastroenteritis, cystitis, urethritis and bacillary dysentery. But, there is insufficient of study about the effects of YIS on the anti-inflammatory activities. The present study evaluated the anti-inflammatory effects of YIS on lipopolysaccharide (LPS)-activated RAW 264.7 cells. Methods : Cell viability was assessed by MTT assay and nitric oxide (NO) was evaluated by measuring the nitrite content in culture medium. Pro-inflammatory cytokines such as tumor necrosis factor-α, interleukin-1β and IL-6 were quantified by ELISA kit. The expression of proteins related with nuclear factor-κB (NF-κB) pathway and inducible NO synthase (iNOS) were assessed by western blot analysis. Results : YIS significantly inhibited the expression of iNOS increased by LPS, and thus significantly inhibited the production of NO. In addition, YIS significantly inhibited pro-inflammatory cytokines. In the regulation of inflammation, NF-κB pathway plays a crucial role. YIS inhibited the expression of p-IκBα and thus inhibited the translocation of NF-κB to the nucleus. Conclusions : These results suggest that YIS ameliorates inflammatory response in LPS-activated RAW 264.7 cells through the inhibition of inflammatory mediators, via suppression of the NF-κB pathway. Therefore, this study provides objective evidence for the anti-inflammatory effect of YIS including the underlying mechanisms.

Effects of Herbal Medicine on Breastfeeding - Analysis of Marker Substances in Saenghwa-tang by HPLC and LC/MS/MS - (산모의 한약 복용이 모유에 미치는 영향 - HPLC와 LC/MS/MS를 이용한 생화탕 지표성분 분석 -)

  • Objectives: We took breast milk samples and analyzed them using HPLC and LC/MS/MS, to evaluate the effects of taking Saenghwa-tang during breastfeeding on breast milk. Methods: The study participants were 20 lactating women who admitted in Korean medical postpartum care center. Breast milks were collected from paticipants who have been administrated Saenghwa-tang for more than 3 days. We used HPLC and LC/MS/MS for the determinations of amygdalin, liquiritins, 6-gingerol, decursin and decursinol angelate in Saenghwa-tang. Results: 1. Participants' $Mean{\pm}S.D$ (standard deviation) of age is $31.05{\pm}1.96$, and 15 participants had normal delivery and 5 participants had cesarean delivery. 12 participants were primipara and 8 participants were multipara. $Mean{\pm}S.D$ of lactating date is $9.4{\pm}0.94$. 2. Using HPLC, we learned LOQ level peak that matches the peak retention time of standard components of Saenghwa-tang was not detected from 20 breast milk samples. 3. Using LC/MS/MS, decursin of Angelicae Gigantis Radix was detected from HMSP 02, HMSP 04, HMSP 06, HMSP 11, and the each concentrations are 16, 2, 64, 11 ppb. Liquiritin of Glycyrrhizae Radix was not detected from HMSP 13~HMSP 18. Conclusions: Data obtained by this approach shows that this method is reliable and suitable for determining the safety of taking Saenghwa-tang during breastfeeding.
