Molecular Cloning and Characterization of Attacin from the Swallowtail Butterfly, Papilio xuthus

  • Attacin is an insect antibacterial protein that plays an important role in immune response to injury and infection. In this report, we have isolated and characterized of cDNA encoding for the attacin from the immunized larvae of swallowtail butterfly, $Papilio$ $xuthus$. A full length cDNA of $P.$ $xuthus$ attacin was obtained by employing annealing control primer (ACP)-based differential display PCR and 5' RACE. The complete $P.$ $xuthus$ attacin cDNA was comprised of 949 bp encoding a 250 amino acid precursor. It contains a putative 18 amino acid signal peptide sequence, a 42 amino acid propeptide sequence, and a 190 amino acid mature protein with a theoretical molecular mass of 19904.01 and a pI of 9.13. The putative mature protein of $P.$ $xuthus$ attacin showed 48-52% and 24-30% identity in amino acid sequences with that of lepidopteran and dipteran insects, respectively. Semiquantitive RT-PCR results revealed that the transcript of $P.$ $xuthus$ attacin gene was up-regulated at significant levels after injection with bacterial lipopolysaccharide (LPS). We sub-cloned cDNA fragment encoding mature $P.$ $xuthus$ attacin into the expression vector, highly expressed in $E.$ $coli$ BL21 cells, and its antibacterial activity was analyzed. Recombinant $P.$ $xuthus$ attacin evidenced considerably antibacterial activity against Gram-negative bacteria, $E.$ $coli$ ML 35 and $Klebsiella$ $pneumonia$.

Fabrication and Characteristics of HTS Field Winding of a 100 hp Synchronous Motor (100마력 동기전동기용 고온초전도 계자권선 제작과 특성)

  • To develop a 100 hp high temperature superconducting(HTS) motor with high efficiency first in Korea, we fabricated a HTS field winding and test. HTS field winding is composed of sixteen HTS race track shaped coils wound with stainless steel-reinforced Bi-2223 tape conductor by react and wind fabrication method. Nomex paper was used for electrical insulation. Each of four magnet pole assemblies was constructed with four double pancake sub-coils, mechanically stacked and electrically in series. Four magnet assemblies were fixed on an aluminum support structure to make effective heat transfer. The Critical current (Ic) was 41.5A at 77K and self field. However the lowest Ic value of sub-coils was 35A. Joule heat generated by each joints between sub-coils was lower than 1mW at 77K and 34A. And Joule heat generated by the joints between field coils was lower than 10mW at 77K and 34A. Joule heat of the whole field winding was 1W at 77K and 32A. And so, the lowest Ic value of sub-coils was more important than Joule heats generated by all joints. The operating current must be lower than the lowest Ic of all the sub-coils. In this paper, design, construction and testing of HTS field winding, Joule heat generated by the joints, and operating current were discussed.

Pathotype Classification of Plasmodiophora brassicae Isolates Using Clubroot-Resistant Cultivars of Chinese Cabbage

  • Clubroot disease caused by Plasmodiophora brassicae is one of the most serious diseases in Brassica crops worldwide. In this study, the pathotypes of 12 Korean P. brassicae field isolates were determined using various Chinese cabbage including 22 commercial cultivars from Korea, China, and Japan, and 15 inbred lines. All P. brassicae isolates exhibited the typical clubroot disease on non-clubroot resistant cultivar, indicating that the isolates were highly pathogenic. According to the reactions on the Williams' hosts, the 12 field isolates were initially classified into five races. However, when these isolates were inoculated onto clubroot-resistant (CR) cultivars of Chinese cabbage, several isolates led to different disease responses even though the isolates have been assigned to the same race by the Williams' host responses. Based on the pathogenicity results, the 12 field isolates were reclassified into four different groups: pathotype 1 (GN1, GN2, GS, JS, and HS), 2 (DJ and KS), 3 (HN1, PC, and YC), and 4 (HN2 and SS). In addition, the CR cultivars from Korea, China, and Japan exhibited distinguishable disease responses to the P. brassicae isolates, suggesting that the 22 cultivars used in this study, including the non-CR cultivars, are classified into four different host groups based on their disease resistance. Combining these findings, the four differential hosts of Chinese cabbage and four pathotype groups of P. brassicae might provide an efficient screening system for resistant cultivars and a new foundation of breeding strategies for CR Chinese cabbage.

Breeding of New Productive Bivoltine Hybrid, CSR12 $\times$ CSR6 of Silkworm Bombyx mori L.

  • With an objective of evolving quantitatively and qualitatively superior bivoltine breeds/hybrids of silkworm Bombyx mori L. for tropical conditions, breeding work was initiated in Central Sericultural Research and Training Institute, Mysore during 1992 by utilizing two Japanese hybrids namely BNl8$\times$BCS25 and Shunrei$\times$Shogetsu along with Indian evolved breed, KA. The breed CSR12 which is characterized with sex-limited larval marking and white oval cocoons was evolved from the Japanese hybrid BNl8 ${\times}$ BCS25 by crossing with KA, while the breed CSR6 which is characterized with normal marking (marked larvae) and white dumbbell cocoons was extracted from the Japanese commercial hybrid Shunrei${\times}$Shogetsu through continuous inbreeding coupled with selection. After fixation, these breeds along with other newly evolved breeds were subjected to hybrid study under optimum environmental conditions in the laboratory for expression of full potential of the genotypes. These hybrids were evaluated by Multiple Trait Evaluation Index (Mano et al., 1993). The hybrid CSR12${\times}$CSR6 was selected based multiple trait evaluation index value. The hybrid CSR12$\times$CSR6 recorded survival of 96.0%, shell weight of 50.0 cg, shell ratio of 24.3%, raw silk percentage of 19.6, filament length of 1,216 m, boil off loss of 22.4% and renditta of 5.1. On the other hand, the control hybrid (KA ${\times}$ NB4D2) has recorded survival of 90.6%, shell weight of 42.1 cg, shell ratio of 20.4%, raw silk percentage of 15.9, filament length of 999 m, boil off loss of 24.8% and renditta of 6.3. The hybrid CSR12$\times$CSR6 was authorized during 1997 by Central Silk Board, Government of India for commercial exploitation during favourable months based on national level race authorization test.

  • This research is conducted through a qualitative methodology, and it aimed for understanding of the generalized structure of getting-together experiences from South Korean who had conversations with North Korean refugees for a long term. As a result, the study, which includes the experiences along with consideration of both contexts and timeline, shows that 'One Korean but divided into South and North', 'Synthesize of water and oil', 'Different from us', 'Understand relativism and deal with it', 'Limitation as an one race: Living different ways on a peninsula' and 'Parallel horizons: Heading to vanishing point' are the key concepts. By understanding these keys, it was able to figure out how their consciousness and acts have been developed in a long term perspective, and thus, we suggest alternative solutions that can be practiced by social workers.

Spray Pattern Analysis for a Centrifugal Fertilizer Distributor with Two Shutter Holes (두 개의 셔터 구멍이 적용된 원심식 비료 살포기의 살포패턴 분석)

  • In this study, the spray pattern of a centrifugal fertilizer distributor with two shutter holes was analyzed and an effective driving width that satisfies proper spray uniformity was derived. The centrifugal fertilizer distributor was mounted on a tractor with a rated power of 23.7 kW and static and dynamic spray pattern tests were performed according to the standard procedure proposed by the American Society of Agricultural and Biological Engineers Standard ASAE S341.5. The height of the fertilizer distributor was 80 cm from the ground and the PTO (power take-off) shaft speed of the tractor was fixed at 540 rpm. The fertilizer scattered in space was collected using 275 evenly spaced collectors at shutter opening ratios of 25%, 50%, 75%, and 100%. The spray pattern was analyzed via the amount of sprayed fertilizer at each collector location and the coefficient of variation was used as an indicator of spray uniformity. Using the analyzed spray pattern, the effective driving width that satisfied less than 15% of the coefficient of variation was derived for different tractor driving patterns (race track mode, back and forth mode). From the results, spray uniformity increased as the shutter opening ratio decreased. The largest effective driving width was 8 m at a shutter opening ratio of 25% for both driving patterns.

Development of polymorphism genetic marker for identification of the silkworm races (누에 피브로인 유전자 다형성 마커 개발)

  • We have previously characterized the complete fibroin light chain gene from one of the silkworm race Baegokjam (Bombyx mori) and found two variable regions (FibL1, intron 2 ~ 3; FibL2, intron 6) with the primer sets designed to cover these variable regions. In this study, we tested the utility of these variable regions as genetic markers for classifying silkworm races. For the purpose, Europian races (Q, PK), Chinese races (C26, C31), Japanese races (N15, N9), and tropical races (SA2, SA5) were used in this experiment. The polymorphism of the FibL1 and FibL2 were divided into two and three types, respectively. The combination of the FibL1 and FibL2 polymorphisms were constant within the same races. The result suggest that the primer sets designed from two variable regions of fibroin light chain gene may be useful as the genetic markers for silkworm races.

Association with Genetic Polymorphism of rs117033348 and Allergic Disease in Korean Population

  • Allergy is an immune response that appears in certain people, and reactions such as coughing, shortness of breath, and hives occur. The immune system plays an important role in homeostasis and host defense, and allergies cause hypersensitivity reactions when an imbalance of immunity occurs. Mutations in the TLR genes are associated with autoimmune conditions such as allergies and asthma. It has been reported that a locus in the TLR1-TLR6-TLR10 region may be associated with atopic sensitization or allergy. Therefore, the purpose of this study was to select an allergy patient group and a healthy control group to determine how the genetic mutation of TLR1 affects the onset of disease. This study was conducted in 709 patients and 5,025 control groups out of 10,956 patients with data from KARE and HEXA cohorts. As a result of logistic regression analysis of 6 SNPs selected from the TLR1 gene, only rs117033348 showed a statistically significant correlation (P = 0.002356). The influence of rs117033348 was examined using PolyPhen-2, and a significant result was shown. Therefore, it can be predicted that the G base in rs117033348 will have an influence on the human body. In addition, Geography of Genetic Variants browser was used to confirm the geographical distribution of allele frequencies for the TLR1 gene. Although it was found that there was a large racial difference in the prevalence of TLR1 SNP, it could be confirmed that the polymorphism of rs117033348 conducted in this study was only specific in East Asia when compared with each race.

Perception of upper lip augmentation utilizing simulated photography

  • Background No head to head comparison is available between surgical lip lifting and upper lip filler injections to decide which technique yields the best results in patients. Despite the growing popularity of upper lip augmentation, its effect on societal perceptions of attractiveness, successfulness and overall health in woman is unknown. Methods Blinded casual observers viewed three versions of independent images of 15 unique patient lower faces for a total of 45 images. Observers rated the attractiveness, perceived success, and perceived overall health for each patient image. Facial perception questions were answered on a visual analog scale from 0 to 100, where higher scores corresponded to more positive responses. Results Two hundred and seventeen random observers with an average age of 47 years (standard deviation, 15.9) rated the images. The majority of observers were females (n=183, 84%) of white race (n=174, 80%) and had at least some college education (n=202, 93%). The marginal mean score for perceived attractiveness from the natural condition was 1.5 points (95% confidence interval [CI], 0.9-2.18) higher than perceived attractiveness from the simulated upper lip filler injection condition, and 2.6 points higher (95% CI, 1.95-3.24) than the simulated upper lip lift condition. There was a moderate to strong correlation between the scores of the same observer. Conclusions Simulated upper lip augmentation is amenable to social perception analysis. Scores of the same observer for attractiveness, successfulness, and overall health are strongly correlated. Overall, the natural condition had the highest scores in all categories, followed by simulated upper lip filler, and lastly simulated upper lip lift.

Necrotrophic Fungus Pyrenophora tritici-repentis Triggers Expression of Multiple Resistance Components in Resistant and Susceptible Wheat Cultivars

  • Tan spot of wheat, caused by Pyrenophora tritici-repentis (Ptr), results in a yield loss through chlorosis and necrosis of healthy leaf tissue. The major objective of this study was to compare gene expression in resistant and susceptible wheat cultivars after infection with Ptr ToxA-producing race 2 and direct infiltration with Ptr ToxA proteins. Greenhouse experiments included exposure of the wheat cultivars to pathogen inoculum or direct infiltration of leaf tissue with Ptr-ToxA protein isolate. Samples from the experiments were subjected to RNA sequencing. Results showed that ToxA RNA sequences were first detected in samples collected eight hours after treatments indicating that upon Ptr contact with wheat tissue, Ptr started expressing ToxA. The resistant wheat cultivar, in response to Ptr inoculum, expressed genes associated with plant resistance responses that were not expressed in the susceptible cultivar; genes of interest included five chitinases, eight transporters, five pathogen-detecting receptors, and multiple classes of signaling factors. Resistant and susceptible wheat cultivars therefore differed in their response in the expression of genes that encode chitinases, transporters, wall-associated kinases, permeases, and wound-induced proteins, among others. Plants exposed to Ptr inoculum expressed transcription factors, kinases, receptors, and peroxidases, which are not expressed as highly in the control samples or samples infiltrated with ToxA. Several of the differentially expressed genes between cultivars were found in the Ptr resistance QTLs on chromosomes 1A, 2D, 3B, and 5A. Future studies should elucidate the specific roles these genes play in the wheat response to Ptr.
