Comparative Study on Antioxidative Activity of Glycyrrhiza uralensis and Glycyrrhiza glabra Extracts by Country of Origin (원산지별 감초 추출물의 항산화 활성 비교 연구)

  • In this work, comparative study on antioxidative activities of extracts from Glycyrrhiza uralensis (G. uralensis) produced in Korea and in China and Glycyrrhiza glabra (G. glabra) produced in Uzbekistan was conducted. Among three origins, 50% ethanol extracts (21.15 ${\mu}g/mL$), ethyl acetate fraction (29.15 ${\mu}g/mL$) and aglycone fraction (3.26 ${\mu}g/mL$) of G. uralensis from Korea showed the higher free radical (1,1-phenyl-2-picrylhydrazyl, DPPH) scavenging activity ($FSC_{50}$) than extracts from other origins. Reactive oxygen species (ROS) scavenging activities ($OSC_{50}$) of extracts from three origins on ROS generated in $Fe^{3+}-EDTA/H_2O_2$ system were investigated using luminol-dependent chemiluminescence assay 50% ethanol extract (1.00 ${\mu}g/mL$) and ethyl acetate fraction (0.34 ${\mu}g/mL$) of G. uralensis from China showed the most prominent ROS scavenging activity. The protective effects of extract/fractions of G. uralensis and G. glabra extracts on the rose-bengal sensitized photohemolysis of human erythrocytes were investigated. 50% ethanol extract and aglycone fraction of G. uralensis and G. glabra extracts from three origins showed cellular protective effects in a concentration dependent manner (5 ~ 50 ${\mu}g/mL$). Aglycone fraction of G. uralensis from Korea (${\tau}_{50}$ = 847.4 min)especially showed cellular protective effects four times higher than that from China (${\tau}_{50}$ = 194.3 min). These results indicate that G. uralensis and G. glabra extracts, which have been used as whitening agent, could be applicable to functional cosmetic ingredient as a natural antioxidant. Judging from the prominent cellular protecitve effects, it is concluded that G. uralensis and G. glabra extracts can protect the skin from $^1O_2$ and various ROS induced by UV.

  • Poultry are exposed to extremely high levels of oxidative stress as a consequence of the excessive production of reactive oxygen species (ROS) induced by endogenous and exogenous stressors, such as high-stocking densities, thermal stress, environmental and feed contamination, along with factors associated with intensive breeding systems. Oxidative stress promotes lipid peroxidation, DNA damage, and inflammation, which can have detrimental effects on the health of birds. During the course of evolution, birds have developed antioxidant defense mechanisms that contribute to maintaining homeostasis when exposed to endogenous and exogenous stressors. The primary antioxidant defense systems are enzymatic and non-enzymatic in nature and play roles in protecting cells from ROS attack. Recently, plant flavonoids, which have been established to reduce oxidative stress, have been attracting considerable attention as potential feed additives. Flavonoids are a group of polyphenolic compounds that can be stabilized by binding structural compounds with ROS, and can promote the elimination of ROS by inducing the expression of antioxidant enzymes. However, although flavonoids can contribute to reducing lipid peroxidation and thereby enhance the antioxidant capacity of birds, they have low solubility in the gastrointestinal tract, and consequently, it is necessary to develop a delivery technology that can facilitate the effect intestinal absorption of these compounds. Furthermore, it is important to determine the dietary levels of flavonoids by assessing the exact antioxidant effects in the gastrointestinal tract wherein the concentrations of dietary flavonoids are highest. It is also necessary to examine the expression of transcriptional factors and vitagenes associated with the efficient antioxidant effects induced by flavonoids. It is anticipated that the application of flavonoids as natural antioxidants will become a particularly important field in the poultry industry.

Antioxidant Activities of Gynura procumbens Extracts (명월초 추출물의 항산화 활성)

  • In this study, the methanol fraction and aglycone fraction were made from Gynura procumbens (G. procumbens) extracts and their antioxidative effects were investigated. The free radical scavenging activity (1,1-diphenyl-2-picrylhydrazyl, DPPH), total antioxidant capacity by luminol-dependent chemiluminescence assay, and the protective effects against reactive oxygen species (ROS) in erythrocytes were measured to evaluate the antioxidative activities of the extracts. Free radical scavenging activities ($FSC_{50}$) of the methanol fraction and aglycone fraction were 90.25 and $81.38{\mu}g/mL$, respectively. Total antioxidant capacities ($OSC_{50}$) of the methanol fraction and aglycone fraction were 16.96 and $12.30{\mu}g/mL$, respectively. The free radical scavenging activity and total antioxidant capacity of the aglycone fraction were greater than those of methanol fraction. The cellular protective effect on the $^1O_2$-induced cellular damage of human erythrocytes was confirmed by ${\tau}_{50}$ value. The ${\tau}_{50}$ value of the methanol fraction and aglycone fraction were 36.7 min and 76.1 min, respectively in $5{\mu}g/mL$, and the aglycone fraction showed about 2 times higher cellular protecive effect than (+)-${\alpha}$-tocopherol (35.4 min). These results indicate that the aglycone fraction of G. procumbens extracts has application possibility as antioxidant ingredient of cosmetic.

Effect of Ethanol Extracts of Goat's Beard on Streptozotocin Induced Diabetic Symptoms and Oxidative Stress in Rats (삼나물 에탄올 추출물이 Streptozotocin으로 유발한 흰쥐의 당뇨증상과 산화적 스트레스에 미치는 영향)

  • The effects of goat's beard (Aruncus dioicus var ramtschaticus Hara) ethanol extracts (AD) on the streptozotocin-induced diabetic symptoms and oxidative stress in rats were assessed. Experimental plots were divided into normal controls (NC), diabetes controls (DM), diet with 0.3% AD after diabetes induction (SA), and diet with 0.6% AD after diabetes induction (SB), then fed for 5 weeks. In the SA and SB groups as compared with the DM group, diabetic symptoms including weight loss, increase in feed and water intake, and enlargement of the liver and kidney were improved. The levels of blood glucose and serum fructosamine were reduced by $17.9{\sim}27.2%$ and $25.6{\sim}32.6%$, respectively. The activities of alanine aminotransferase, aspartate aminotransferase and the levels of triglycerides, total cholesterol, and LDL-cholesterol in the serum were reduced by $25.6{\sim}30.3$, $42.37{\sim}55.51$, $26.85{\sim}30.44$ and $37.29{\sim}39.11%$, respectively, whereas the level of HDL-cholesterol was increased by $37.29{\sim}39.11%$. Hepatic total (T) and O type (O) activities of xanthine oxidoreductase, O/T ratio(%) and level of lipid peroxide were significantly decreased, while the level of glutathione was increased, and also the activities of superoxide dismutase and glutathione S-transferase were increased by $56.84{\sim}94.90%$ and $57.14{\sim}68.92%$, respectively. In the above results, it was noted that AD has an antidiabetic effect which alleviated hyperglycemia and the AD reduced and/or prevented the tissue damage caused by diabetes yia the inhibition of reactive oxygen species (ROS) generating systems concurrent with an increase in ROS scavenging.

Quality Characteristics and Antioxidant Activity of Mulberry Leaf Tea Fermented by Monascus pilosus (Monascus pilosus로 발효시킨 뽕잎차의 품질특성과 항산화능)

  • This study was carried out to investigate the contents of monacolin K and citrinin, along with the sensory quality and antioxidant activity of mulberry leaf tea fermented by $Monascus$ $pilsous$ (FMM). Total monacolin K content of FMM was 0.058%, but citrinin was not detected. Redness of brewed FMM was remarkably higher than that of unfermented mulberry leaf tea (UFM). In sensory evaluation of brewed FMM, while astringent taste and savory taste were lower, flavor, color, and overall acceptability were significantly higher than those of UFM. Total polyphenol contents of UFM and FMM were 83.1 and 23.61 mg/g (dry basis), total flavonoid contents of UFM and FMM were 17.96 and 3.99 mg/g (dry basis), respectively. Xanthine oxidase (XO) inhibitory activity and superoxide dismutase (SOD)-like activity of FMM were lower than those of UFM. Electron-donating ability and ferric-reducing antioxidant power of FMM were slightly lower than those of UFM. However, the antioxidant activities of FMM per polyphenol content were markedly higher than those of UFM. These results suggest that FMM may scavenge excessive reactive oxygen spices (ROS) via inhibition of XO and SOD-like activity. Furthermore, FMM demonstrated relatively higher acceptability and antioxidant ability along with functionality of $Hongguk$ (red yeast rice), and therefore could be utilized to prevent various ROS-induced diseases.

Neuroprotective effects of Momordica charantia extract against hydrogen peroxide-induced cytotoxicity in human neuroblastoma SK-N-MC cells (산화적 스트레스에 대한 여주 (Momordica charantia) 추출물의 항산화 효과 및 세포사멸 억제 기전을 통한 신경세포보호효과)

  • Purpose: Many studies have suggested that neuronal cells protect against oxidative stress-induced apoptotic cell death by polyphenolic compounds. We investigated the neuroprotective effects and the mechanism of action of Momordica charantia ethanol extract (MCE) against $H_2O_2-induced$ cell death of human neuroblastoma SK-N-MC cells. Methods: The antioxidant activity of MCE was measured by the quantity of total phenolic acid compounds (TPC), quantity of total flavonoid compounds (TFC), and 2,2-Diphenyl-1-pycrylhydrazyl (DPPH) radical scavenging activity. Cytotoxicity and cell viability were determined by CCK-8 assay. The formation of reactive oxygen species (ROS) was measured using 2,7-dichlorofluorescein diacetate (DCF-DA) assay. Antioxidant enzyme (SOD-1,2 and GPx-1) expression was determined by real-time PCR. Mitogen-activated protein kinases (MAPK) pathway and apoptosis signal expression was measured by Western blotting. Results: The TPC and TFC quantities of MCE were 28.51 mg gallic acid equivalents/extract g and 3.95 mg catechin equivalents/extract g, respectively. The $IC_{50}$ value for DPPH radical scavenging activity was $506.95{\mu}g/ml$ for MCE. Pre-treatment with MCE showed protective effects against $H_2O_2-induced$ cell death and inhibited ROS generation by oxidative stress. SOD-1,2 and GPx-1 mRNA expression was recovered by pre-treatment with MCE compared with the presence of $H_2O_2$. Pre-treatment with MCE inhibited phosphorylation of p38 and the JNK pathway and down-regulated cleaved caspase-3 and cleaved PARP by $H_2O_2$. Conclusion: The neuroprotective effects of MCE in terms of recovery of antioxidant enzyme gene expression, down-regulation of MAPK pathways, and inhibition apoptosis is associated with reduced oxidative stress in SK-N-MC cells.

Usage- and daily intake-based cytotoxicity study of frequently used natural food additives in South Korea (국내 다빈도 사용 천연첨가물의 사용량 및 섭취량 기반 세포독성 연구)

  • Natural food additives have recently attracted attention as alternatives to synthetic additives. However, little information is available regarding their potential toxicity. In this study, we evaluated ten different natural food additives that are widely used in commercial foods in South Korea based on their actual usage level and daily intake. The results showed that none of the tested natural additives exhibited cytotoxicity in terms of inhibition of cell proliferation/viability and lactate dehydrogenase leakage. Additionally, the tested natural food additives did not generate intracellular reactive oxygen species (ROS), whereas they significantly decreased intracellular ROS levels produced by hydrogen peroxide. Moreover, none of the tested natural additives affected cell proliferation and viability in 2D and 3D intestinal epithelium models. Taken together, the ten natural food additives did not exhibit cytotoxicity in their actual usage levels. These findings can be used to further assess the toxicity of natural food additives.

Anti-oxidant and Anti-inflammatory Potentials of Sasa quelpaertensis Leaf Residue Extracts (제주조릿대 잎 잔사 추출물의 항산화 및 항염 활성)

  • Sasa quelpaertensis Nakai is a native Korean plant that grows only on Mt. Halla of Jeju Island. Its leaf is used for a popular bamboo tea due to its various health-promoting properties, and it has been increasingly used as food and cosmetic ingredients. To utilize the S. quelpaertensis leaf efficiently, the preparation method for phytochemical-rich extract (PRE) using the leaf's residue was previously reported, which was produced after hot water extraction. This study was undertaken to evaluate the anti-oxidant and anti-inflammatory potential of PRE and its solvent fractions. The ethyl acetate fraction of PRE (EPRE) showed higher DPPH, ABTS, and superoxide radical scavenging activities, and it effectively inhibited intracellular reactive oxygen species (ROS) and nitric oxide (NO) production in lipopolysaccharides (LPS)-stimulated RAW 264.7 cells. EPRE also induced the expression of heme oxygenase-1 (HO-1) by increasing the level of nuclear factor E2-related factor 2 (Nrf2) in a nuclear fraction. The inhibiting effect of EPRE on LPS-induced NO production was partially reversed by the HO-1 inhibitor (zinc protoporphyrin, ZPP), suggesting that HO-1 is involved in suppressing NO production. Taken together, the results suggest that EPRE has potential as a promising anti- oxidant and anti-inflammatory agent.

Effects of Fermented Kalopanax pictus on oxidative damage of neurofilament protein (신경세사 단백질의 산화적 손상에 엄나무 발효물이 미치는 영향)

  • This study was to investigate the effect of the extract(KP-HE) from Kalopanax pictus(KP) fermented with Hericium erinaceum(HE) mycelium on oxidative modification of neurofilament-L(NF-L) which is closely related to neurodegenerative disorders. The oxidative modification of NF-L was induced by AAPH producing peroxyl radicals in solution, and KP, HE, and KP-HE was investigated. KP and HE did not protect NF-L against peroxyl radical-mediated NF-L modification whereas KP-HE significantly prevented NF-L modification induced by peroxyl radical. KP-HE inhibited the formation of dityrosine in oxidative modification of NF-L and stimulated the peroxyl radical scavenging activity. The effects of KP, HE, and KP-HE on the modification of NF-L by tetrahydropapaveroline(THP), a neurotoxin found in patients with Parkinson's disease was investigated. KP-HE also prevented THP-mediated NF-L modification as compared to KP and HE. In addition, KP-HE significantly inhibited the formation of dityrosine in oxidative modified NF-L and enhanced the inhibition of reactive oxygen species(ROS) was generated by THP. The results suggested that KP-HE can contribute to protected cell from oxidative stress was induced by ROS and neurotoxin. Therefore, KP-HE could potentially be used as a valuable functional food ingredient to prevent neurodegenerative disorders.

Radical Scavenging Effect and Neuroprotective Activity from Oxidative Stress of Engelhardtia chrysolepis Leaf (Engelhardtia chrysolepis의 라디칼 소거능 및 신경세포의 산화 스트레스 보호효과)

  • In this study, the radical scavenging activity and protective effect of ethanol extract from leaf of Engelhardtia chrysolepis HANCE (ECE) against oxidative stress were investigated under in vitro and cellular system. ECE showed strong radical scavenging activities in 1,1-diphenyl-2-picrylhydrazyl, hydroxyl(${\cdot}OH$) and nitric oxide(NO) radical as a concentration-dependent manner. Particularly, strong scavenging activity against the ${\cdot}OH$ and NO radical were observed with the $IC_{50}$ value of $1.30{\mu}g/ml$ and $12.61{\mu}g/ml$, respectively. Furthermore, the cellular oxidative stress was induced by amyloid beta($A{\beta}_{25-35}$) in C6 glial cells. The treatment of $A{\beta}_{25-35}$ to C6 glial cells generated high levels of reactive oxygen species(ROS) and declined cell viability. However, production of ROS was decreased by the treatment of ECE. In addition, the cell viability was significantly increased at each concentration(10, 25, $50{\mu}g/ml$) as dose-dependent manner. The Alzheimer's disease-related protein expressions in $A{\beta}_{25-35}$-treated C6 glial cells were analyzed. The ECE treatment inhibited expression of amyloid precursor protein(APP), C-terminal fragment-${\beta}(CTF-{\beta})$, ${\beta}$-site APP cleaving enzyme(BACE), phosphorylated tau(p-tau) proteins in C6 glial cells induced by $A{\beta}_{25-35}$. The present study indicated that ECE has strong radical scavenging activity and neuroprotective effect through attenuating oxidative stress.
