Photosynthesis and Respiration of Ginseng Leaf and Root in Relation to Senescence of Aerial Part (지상부 노화별 인삼의 엽과 근의 광합성 및 호흡)

  • Photosynthesis and respiration of leaf and root of field grown Panax ginseng were investigated according to aerial part sensecence. No apparent photosynthesis activity was detected in senescenced leaf(less than 0.7mg total chlorophyll/g FW) and leaf dark respiration was consistent relation with senescence. Leaf respiratory Q$_{10}$ consistently increased with senscence. Root respiration and Q$_{10}$ tended to decrease with aerial part senescence only in the range of optimum temperature of leaf growth. Apparent photosynthesis or respiration of leaf was negatively or positively correlated, respectively with the increase of air temperature. Root respiration with temperature was well accordance with Arrhenius plot which was not consistent with aerial part senescence. Accelerated senescence may be recommendable for better root yield unless any reserve in stem or leaves contributes to root through translocation.

  • Identification of the age of ginseng root is very important in commercial market as well as in research field. However, any criterion abut it has not been clearly established yet. We studied to find a clear morphological key for identification of ginseng root\\`s age using the histochemical staining method. Fresh sections of 3, 4, 5, and 6 year old roots were stained with 1% nile blue, observed under the light microscopy, and compared each other. The number of secretory duct layers(SDL) is a useful key to confirm the age of ginseng root as follow; three-year-old root has two, four-year-old one has three, fie-year-old one has four, and six-year-old one has five resin duct layers on each cortical region of roots. Secretory ducts are thought to be formed by the vascular cambium every year. Unlike the surrounding parenchyma cells, secretory epithelial cells lack starch grains in the cytoplasm.

  • Maxillary lateral incisors usually exhibit a single root with a single canal. However, maxillary lateral incisor teeth with unusual morphology of root canal system are frequently reported. These cases of variable root canal anatomy can be treated well by nonsurgical endodontic methods. A detailed description of root canal morphology is fundamental for successful endodontic treatment. Treatment using an operating microscope, radiographs from different angles, and cone-beam computerized tomography (CBCT) can produce more predictable endodontic outcomes.

  • Objectives: Management of a horizontal root fracture of an anterior teeth is challenging and often requires multiple approaches for improving the functional and esthetic outcomes. This case report describes the treatment and 2-yr follow up of 3 maxillary incisors with horizontal root fracture. Two maxillary central incisors were treated with Mineral Trioxide Aggregate (ProRoot MTA, Dentsply, Tulsa, OK, USA). Left maxillary lateral incisors were treated with endodontic treatment and submerged. During 2-yr of follow-up evaluation, the root-fractured teeth of the present patients were well retained in the arch, showing periodontal healing even after endodontic treatment.

  • In Japan, propagation of cyclamen is mainly from seedlings. However, seeds are expensive and germination is slow and non..uniform. Therefore, to achieve genetically uniform propagation, multiplication must be vegetative. The rooted tuberous stems without shoots as sources of explants were cultured on the media containing BA and sucrose. After 30 days cultivation, tuberous roots were produced from the root ends attached to a tuberous stem and its capability was dependent on the type of media. The highest percentage of tuberous root formation was observed in Culture on the medium of 1/3 MS containing 0.05mgL$^{-1}$ NAA, 0.5mg L$^{-1}$ BA and 5% sucrose. Growth rates of the tuberous roots were greatly influenced by the cutting positions of a root in explants. The highest growth of was observed if small amount of root end was cut at initiation of tissue culture.

Effects of Polyamine on Growth and Anthocyanin Contents of Carrot Hairy Root (당근 Hairy root의 성장 및 Anthocyanin 함량에 미치는 Polyamime의 영향)

  • The effects or polyamines on growth, anthocyanin contents and $\beta$-glucan synthetase(GSII) activity in carrot hairy root were studied. Growth of hairy root was stimulated somewhat when each polyamine concentration was treated, especially addition of 1mM spermidine resulted in about 20% increase. On the whole, the axial diameter of hairy root was increased in response to increase in concentration of polyamine. On the other hand, GSII activity was stimulated in response to increase in concentration of polyamine, especially addition of 1mM spermine resulted in about 100% increase of activity. Therefore increased activity of GSII stimulated growth and thickness of hairy root. Anthocyanin contents were not affected by the polyammine.

  • Production of new flavanol derivatives with cytotoxic activity, gericudranin A and B, was studied by using hairy root cultures of Cudrania tricuspidata. Schenk and Hildebrandt (SH) medium was chosen for root growth and gericudranin production. After 35 days culture in a half-strength liquid SH medium containing $30g^{glucose}$/l, hairy root growth reached $138g^{FW}$/I and gericudranin A and B were produced at concentrations of 27mg/l and 21 mg/l, respectively. It was also observed that the contents of gericudranin A and B in hairy root were eight and six times higher than those of cudraniae radix, respectively.

  • Fifty two mandibular permanent canines were chosen to study the anatomy of the root canal. The experimental teeth were injected with China ink, decalcified, cleared and observed to investigate the number of root canals, frequency and location of lateral canals, the location of apical foramens, the frequency of apical deltas, and the curvature of root canals. The results were as follows: 1) all of the experimental teeth demonstrated single canal. 2) of the 52 canals, 9.6% of the canals were found to have lateral canals and the locations of the lateral canals were in the apical third of the roots 3) Thirty three apical foramens were located laterally on the root apices and nineteen foramens were located on the centers of the root apices. 4) 37 canals showed straight curvature, 12 canals distal curvature. 3 canals labial curvature.

  • The existing evaluation of root barrier resistance takes at least two years as an evaluation based on plant growth, and it is difficult to determine early performance in the process of quality control in the field. In this study, thus, the direction of the root barrier resistance evaluation system was presented by considering the construction specifications and design standards for root barrier design. As a result, it was necessary to establish a test method for reproducing simple and consistent test results for evaluating the durability of root barrier materials.

  • This study was carried out to compare the root yields, root characters, saponin and ginsenosides contents of 6-year-old p. ginseng and p. quinquefolium. These two ginseng species showed difference in the diameter and ten비h of mainroot. The main root length or p. quinquefolium was shorter than that of p. ginseng, whereas Jakyung-jong and Hwangsook-jong of P. ginseng showed similar root length. Proximate composition were similar between the two species, however, crude fibercontent was significantly higher in main and lateral root of Jakyung-jong and Hwang sook-jong of P ginseng than P quinquefolium. In regard to mineral contents of root, P ginseng contained more Ca and Mn and less Fe and Al than P. quinquefolium. P. quinquefolium contained more of Rbl and Rd of protopanaxadiol saponin, and less or Re, $Rg_1$ and $Rg_2$ of protopanaxatriol saponin than P ginseng. However, no Rf was detected in the p. quinquefolium. Key words Panax ginseng, Panax quinquefolium, ginseng character, ginsenoside.
