영년혼반초지에 있어서 예취빈도와 질소시비수준이 뿌리의 수량과 그 분포에 미치는 영향 (The Effect of Cutting Frequency and Nitrogen Fertilizing Level on the Root Production and its Distribution in the Pasture)

  • 혼파된 영년채초지에서의 예취빈도와 질소시비가 뿌리의 수량과 심층분포에 미치는 영향에 대하여 연구된 결과는 다음과 같다. 1. 조사된 식생구조에서의 뿌리의 분포는 모든 처리구에서 상층 20cm까지에 그 대부분이 존재하였다. 2. 뿌리의 양은 우점종에 관계없이 모두 높았지만 Arrhenatherum elatius 우점초지에서 가장 낮았고 Alopecurns pratensis우점초지에서 가장 높았다. 3.뿌리의 양은 예취빈도간에는 그 차이가 없었으나 적당한 N 시비수준(N-2)에서 가장 높았다. 4. 뿌리의 심층분포는 모든 처리구에서 1m 길이까지 존재하였다.

Analysis and Identification of Expressed Sequence Tags in Hairy Root Induced from Korean Ginseng (Panax ginseng C. A. Meyer)

  • Hairy roots were induced from Korean ginseng (Panax ginseng C. A. Meyer) root explants and studied for their gene expression. A total of 3,000 ESTs (expressed sequence tags) from ginseng hairy root were determined and about 2,700 ESTs have a length of readable sequence, which result in 1,352 unique ESTs sequences. The 879 ESTs showed significant similarities to known nucleotide or amino acid sequences in other plant species, which were divided into eleven categories depending upon gene function. The remaining 473 sequences showed no significant matches, which are likely to be transcripts or to be matched to other organisms. The results indicated that the analysis of the ginseng hairy root ESTs by partial sequencing of random cDNA clones may be an efficient approach to isolate genes that are functional in ginseng root in a large scale. Our extensive EST analysis of genes expressed in ginseng hairy root not only contributes to the understanding of the dynamics of genome expression patterns in root organ but also adds data to the repertoire of all genomic genes.

연근과 오미자를 이용하여 제조한 양갱의 특성 (Characteristics of Yanggaeng with Lotus Root and Omija)

  • In this study, lotus root(Nelumbo nucifera G.) and omija(Schizandrae fructus), which have been used in oriental medicine and folks remedy, were examined to apply to functional foods. We prepared yanggaeng with lotus root, sugar, oligosaccharide, agar, omija extract solution and analyzed the nutritional composition(moisture, protein, fat, ash, crude fiber, carbohydrate, free sugar and minerals). Also we investigated texture profile and evaluated sensory characteristics of developed yanggaengs and 8 commercial ones. Potassium and crude fiber contents of yanggaeng with lotus root were higher than the commercial yanggaengs. Cohesiveness, springiness and gumminess of lotus root yanggaeng were the same levels of commercial yanggaengs. Sensory evaluation with the ones, showed that the lotus yanggaeng was more desirable than the commercial ones. Yanggaeng with lotus had good scores in texture profile and sensory evaluation compared with commercial yanggaengs. These results demonstrated that lotus root hadsufficient values to use a foodstuff for yanggaeng.

작약 검은뿌리썩음병 발생실태와 방제 (Occurrence and Control of Black Root Rot of Peony (Paeonia lactiflora) on Continuous Cropping)

  • 작약검은뿌리썩음병은 연작을 할 경우 현저히 발생이 증가하였으며, 태백작약 품종에 비해 의성작약 품종에서 좀 더 심한 발병을 보였다. 종구로 사용하는 뇌두를 약제에 침지시 병원균은 검출되지 않았으나 물에만 침지한 경우 병원균이 검출되었다. 연작지의 토양개량시 검은뿌리썩음병의 방제는 효과가 없었으나 다조메입제로 토양을 훈증소독할 경우 토양훈증처리구에서 주근중과 근장이 단순연작구와 비교하여 50% 정도 증가하였으며 검은뿌리썩음병의 발병율은 무처리구의 경우 40.6% 정도 발병되었으나 토양훈증처리구는 4.7% 정도로 발병되어 아주 높은 방제효과를 보였으며 종구소독과 훈증처리를 병행시 발병율은 더욱 낮아졌다.

Effect of Trichokonins on the Growth Characteristics of Paeonia ostii 'Fengdan' Seedling Roots

  • The growth characteristics of Paeonia ostii 'Fengdan' seedlings roots in response to trichokonins-spray treatment were investigated in this study. One-year-old seedlings of P. ostii 'Fengdan' were potted in plastic cups containing garden mold and grown under field conditions. The results showed that application of trichokonins significantly promoted root growth in P. ostii 'Fengdan' seedlings. The total root projection area, total root surface area, total root volume, total root length, root number and root diameter of seedlings treated with 0.25 mg/L trichokonins were higher by 141.70, 116.59, 119.44, 55.97, 348.88 and 127.78%, respectively, than that of the control. Thus, the results supported the hypothesis that good growth condition for roots could directly improve their nutrient absorption and utilization efficiency, promoting plant growth and development.

Defoliation Effects on Root and Rhizome Development of Kura Clover

  • There is limited information on the relationship between defoliation and root and rhizome development of kura clover (Trifolium ambiguum M. Bieb.). To determine the effects of defoliation severity on root and rhizome growth of young kura clover plant (seedling about 8 wk old), this research was conducted in 2002 (Experiment 1) and 2003 (Experiment 2) in a glasshouse at the University of Wisconsin-Madison. Four kura clover entries were used in this experiment: two were started from seed materials (ARS-2678 and 'Rhizo') and two were clones from mature, field-grown Rhizo kura clover plant. Three defoliation frequencies (2-, 4- and 6-wk intervals) and two defoliation intensities (complete and partial defoliation) were imposed on each of the four kura clover entries. Root, rhizome, and leaf dry matter (DM) generally increased with less frequent defoliation, however, the increase in rhizome DM was not significant between 4- or 6-wk defoliation periods. The root and leaf DM under complete defoliation (CD) were significantly lower than under partial defoliation (PD). In Exp. 1, rhizome DM was not significantly different between CD and PD; it was significantly lower under CD in Exp. 2. ARS-2678 showed excellent root development characteristics, however, its rhizome DM was significantly lower than Rhizo clones. The rhizome development from Rhizo clones was greater than that from seed materials. If maximum root and rhizome growth are expected from young kura clover plant, the intensity and frequency of defoliation should be minimized or defoliation should be avoided.

Effects of localised liquid fertilization of N, P, K and Ca on root development in Zoysia matrella, Cynodon dactylon and Stenotaphrum secundatum

  • Turfgrass species were evaluated for their rooting and foliar characteristics, and their interaction with the soil. The rooting system was divided into three compartments, one above another, such that the top and bottom compartments of the root system could be supplied with a nutrient deprived solution. Exposure of parts of the roots to nitrate deprivation caused a localised retardation of root initiation and extension, compared with zones receiving the full supply of nutrients. This resulted in considerable modification to root form, coupled with a significant depression in foliar growth. The extension of roots was the least affected by the deprivation of potassium. Phosphate and calcium deprivations gave rise to similar responses in root and foliar formation. Results from this study showed that external concentrations of nitrogen, phosphorus, potassium and calcium are required by the root system in varying amounts for optimal growth of roots. Turfgrass coverage and turf quality ratings further reinforced these findings. No significant difference was observed between the different grasses examined here. All three species responded similarly to the deprivation of the various nutrients. Results from this study confirmed that targeted fertilization programs are beneficial and can help reduce cost, chemical usage and prevent leachate and contamination.

Efficacy of Pesticides and Growth Hormones against Root Disease Complex of Mulberry (Morus alba L.)

  • During mulberry cultivation, root disease complex caused by the association of root knot nematode (Meloidogyne incognita) with root rot pathogens like Fusarium solani and Botryodiplodia theobromae poses serious loss in leaf production. Therefore, an attempt was made to assess the efficacy of eight pesticides (Metayalaxyl+Mancozeb, Thiophanate methyl, Mancozeb, Bitertanol, Phenomiphos, Phorate, Thionazin & Carbofuran) and two growth hormones (Salicylic acid and Indole 3 acetic acid) at 0.1 and 0.2% concentrations under in vitro conditions against nematode (hatching of eggs and mortality of larvae) and root rot pathogens (poisoned food technique) for short listing the treatments to develop an IDM strategy. Results revealed that among the pesticides and growth hormones, Carbofuran followed by Salicylic acid were found to be effective at 0.2% concentration against both nematode and pathogenic fungi. Both the chemicals inhibited the hatching of nematode eggs by 83.5-78.9% and 80-76% larval mortality over the control and reduced the mycelial growth of both the pathogenic fungi to an extent of 75.5-77.8%. Though Mancozeb inhibited both the pathogenic fungi strongly (77-80%), it did not show any effectiveness against nematode. The rest of the chemicals were found either moderately or poorly effective in reducing the growth of pathogenic fungi, hatching of nematode eggs and enhancing the mortality of larvae. The two effective chemicals viz., Carbofuran and Salicylic acid, which rated as strong inhibitors against both nematode and pathogenic fungi, can be exploited in developing an IDM package as one of the component for better management of root disease complex in mulberry.

Water Extract of Kudzu Root (Pueraria radix) Decreases Apolipoprotein B100 and B48 Production in Vitro

  • We have previously demonstrated that kudzu root extracts have a hypocholesterolemic effect on rats fed diets high in fat and cholesterol. To further elucidate the mechanism involved, in this study we investigated the effect of water extracts of kudzu root, Pueraria radix, on the production of apolipoprotein B$_{100}$ (APo B$_{100}$) in HepG$_2$ liver cells and secretion of apolipoprotein B$_{48}$ (Apo B$_{48}$) in Caco$_2$ cells. Human cell lines, HepG$_2$ liver cells and Caco$_2$ intestinal epithelial cells, were grown with various concentrations (0%, 0.5%, 1.0%, 1.5%, 2.0%) of water extracts of kudzu root in the media. The kudzu root extract decreased Apo B$_{100}$ production and secretion. Treatment of HeP G$_2$ cells with the kudzu root extract also significantly decreased the intracellular total and free cholesterol concentration, and also decreased esterified cholesterol but was only significant at the highest dose of 2%. Apo B$_{48}$ production, but not secretion, from enterocytes was lowered by the kudzu root extracts. This research provided evidence that the hypocholesterolemic properties of kudzu root may be a consequence of decreased production and secretion of Apo B$_{100}$ in the liver and Apo B$_{48}$ in the intestine.

  • Er: YAG laser irradiation utilizing the newly developed RCLase side-firing spiral tip was used for the cleansing of root canals following their bio-mechanical preparation with ProTaper Ni-Ti files. The distal and palatal roots of 20 freshly extracted molar teeth were instrumented to size F3 with ProTaper files. In the experimental group (10 teeth) the pulp chamber and the root canals were filled with EDTA 17% and the root canals were lased for 30 s using the Er: YAG laser irradiation at 600 mJ per pulse and a frequency of 12 Hz. In the control group (10 teeth) the root canals were not lased. Scanning electron microscope analysis showed clean wall surfaces of the lased root canals with open dentinal tubules, free of smear layer and debris. In the scanning electron microscope photographs of the walls of the non-lased root canals a considerable amount of debris could be detected. It appears that an efficient cleansing of the root canal system can be achieved by using the Er: YAG laser irradiation with the RCLase Side-firing Spiral Tip following bio-mechanical preparation of the root canal with Ni-Ti Taper files.
