MicroRNAs are a class of endogenous small RNAs that play important roles in post-transcriptional gene regulation by directing degradation of mRNAs or facilitating repression of target gene translation. In this study, three small RNA cDNA libraries from the mammary gland tissues of Laoshan dairy goats (Capra hircus) were constructed and sequenced, individually. Through Solexa high-throughput sequencing and bioinformatics analysis, we obtained 50 presumptive novel miRNAs candidates, and 55,448 putative target genes were predicted. GO annotations and KEGG pathway analyses showed the majority of target genes were involved in various biological processes and metabolic pathways. Our results discovered more information about the regulation network between miRNAs and mRNAs and paved a foundation for the molecular genetics of mammary gland development in goats.
The objectives of this study were to identify the histamine-forming bacteria and bacteriocin- producing lactic acid bacteria (LAB) isolated from Myeolchi-jeot according to sequence analysis of the 16S rRNA gene, to evaluate the inhibitory effects of the bacteriocin on the growth and histamine accumulation of histamine-forming bacteria, and to assess the physico-chemical properties of the bacteriocin. Based on 16S rRNA gene sequences, histamine-forming bacteria were identified as Bacillus licheniformis MCH01, Serratia marcescens MCH02, Staphylococcus xylosus MCH03, Aeromonas hydrophila MCH04, and Morganella morganii MCH05. The five LAB strains identified as Pediococcus acidilactici MCL11, Leuconostoc mesenteroides MCL12, Enterococcus faecium MCL13, Lactobacillus sakei MCL14, and Lactobacillus acidophilus MCL15 were found to produce an antibacterial compound with inhibitory activity against the tested histamine-producing bacteria. The inhibitory activity of these bacteriocins obtained from the five LAB remained stable after incubation at pH 4.0-8.0 and heating for 10 min at $80^{\circ}C$; however, the bacteriocin activity was destroyed after treatment with papain, pepsin, proteinase K, ${\alpha}$-chymotrypsin, or trypsin. Meanwhile, these bacteriocins produced by the tested LAB strains also exhibited histamine-degradation ability. Therefore, these antimicrobial substances may play a role in inhibiting histamine formation in the fermented fish products and preventing seafood-related food-borne disease caused by bacterially generated histamine.
한국의 여러 지역에서 채취한 토양으로부터 셀룰로오스를 유일한 탄소원으로 이용하여 자라는 591균주의 점액 세균을 순수 분리하였다. 세포, 집락, 그리고 자실체의 모양과 같은 형태적인 특징과 16S rRNA 염기서열은 이들 균주들이 모두 Sorangium cellulosum임을 나타내었다. 그리고 생리활성조사는 114균주 중 최소한 20균주가 Candida albicans의 성장을 저해하는 항진균물질을 생산함을 보여주었다.
MicroRNAs (miRs) are a family of small noncoding RNAs that regulate gene expression by targeting mRNAs in a sequence specific manner, inducing translational repression or mRNA degradation. In this paper we have first analyzed by microarray the miR-profile in erythroid precursor cells from one normal and two thalassemic patients expressing different levels of fetal hemoglobin (one of them displaying HPFH phenotype). The microarray data were confirmed by RT-PCR analysis, and allowed us to identify miR-210 as an highly expressed miR in the erythroid precursor cells from the HPFH patient. When RT-PCR was performed on mithramycin-induced K562 cells and erythroid precursor cells, miR-210 was found to be induced in time-dependent and dose-dependent fashion, together with increased expression of the fetal $\gamma$-globin genes. Altogether, the data suggest that miR-210 might be involved in increased expression of $\gamma$-globin genes in differentiating erythroid cells.
Effects of allopurinol (2mM), a specific inhibitor of xanthine oxidase, on the growth and metabolism of llantoin in dark grown Chinese cabbage (Brassica campestris L.) seedlings were investigated. Allopurinol treatment maintained the fresh and dry weights of cotyledons at higher levels, but inhibited the elongation of hypocotyls and roots of the seedlings. Total nitrogen content in the cotyledons decreased at slower rate by allopurinol. Accordingly, the levels of total nitrogen contents in the hypocotyls and roots, were depressed by the inhibitor. In the cotyledons, allopurinol began to elevate RNA levels after day 3, which it did not affect DNA level throughout the experiment. Activities of xanthine oxidase (XO:EC 1.2.3.2), uricase (UO:EC 1.7.3.3) and allantoinase (AL:EC 3.5.2.5) in the cotyledons were examined. The activity of XO was not detected, but the accumulation of xanthine by allopurinol treatment presented an indirect evidence of the existence of XO in the organ. Allopurinol kept UO activity high up to day 2 after sowing and depressed AL activity throughout the experiment. By allopurinol treatment, allantoin content was kept high over the control both in cotyledons and roots, but it was kept low in hypocotyls. The level of allantoic acid in the 3 organs were shown to be depressed by allopurinol. These results suggest that allantoin and allantoic acid produced by the degradation of stored and newly synthesized RNA are transported from the storage tissue to hypocotyls and roots as important nitrogen sources for the development of Chinese cabbage seedlings.
This study examined the anti-osteoclastogenic effects of baicalin on receptor activator of NF-${\kappa}$B ligand (RANKL)-induced RAW264.7 cells. Baicalin is a flavonoid that is produced by Scutellaria baicalensis and is known to have multiple biological properties, including antibacterial, anti-inflammatory and analgesic effects. The effects of baicalin on osteoclasts were examined by measuring 1) cell viability; 2) the formation of tartrate-resistant acid phosphatase (TRAP) (+) multinucleated cells; 3) RANK/RANKL signaling pathways and 4) mRNA levels of osteoclast-associated genes. Baicalin inhibited the formation of RANKL-stimulated TRAP (+) multinucleated cells and also suppressed the RANKL-stimulated activation of p-38, ERK, cSrc and AKT signaling. Baicalin also inhibited the RANKL-stimulated degradation of $I{\kappa}B$ in RAW264.7 cells. In addition, the RANKL-stimulated induction of NFATc1 transcription factors was found to be abrogated by this flavonoid. Baicalin was further found to decrease the mRNA expression of osteoclast-associated genes, including carbonic anhydrase II, TRAP and cathepsin K in the RAW264.7 cells. Our data thus demonstrate that baicalin inhibits osteoclastogenesis by inhibiting the RANKL-induced activation of signaling molecules and transcription factors in osteoclast precursors.
이 연구에서는 PacBio RS II 플랫폼을 사용하여 동해에서 분리된 해양 미생물 Pseudoalteromonas donghaensis $HJ51^T$의 전체 유전체 서열화를 수행하였다. 그 결과, 크기 3,646,857 bp, G + C 함량 41.8%인 염색체와 함께 크기 842,855 bp, G + C 함량 41.3% 및 크기 244,204 bp, G + C 함량 40.4%인 두 개의 플라스미드로 구성된 3개의 유전체 서열을 획득하였다. 이 유전체는 4,083개의 단백질 암호화 유전자와 127개의 RNA 유전자를 포함하고 있다. 다양한 생체 고분자 분해효소의 유전자원과 대장균과 유사한 세포외 단백질 분비 숙주로서의 개발에 이용될 수 있을 것으로 생각된다.

In this study, we evaluated the effect of Rodgersia podophylla leave extracts (RPL) on ${\beta}$-catenin level in human cancer cells. RPL dose-dependently inhibited cell proliferation in SW480, A549, MDA-MB-231, PC-3 and AsPC-1 cells. RPL dramatically decreased ${\beta}$-catenin protein level in all cancer cells. However, decreased level of ${\beta}$-catenin mRNA expression was observed in A549 and AsPC-1 cells. In addition, RPL dramatically attenuated cyclin D1 mRNA expression in all cancer cells. MG132 decreased the downregulation of ${\beta}$-catenin protein level induced by RPL in all cancer cells, while RPL-induced downregulation of ${\beta}$-catenin was inhibited by the inhibition of $GSK-3{\beta}$ by LiCl in MDA-MB-231 cells. RPL phosphorylated ${\beta}$-catenin and $GSK-3{\beta}$. In addition, the inhibition of $GSK-3{\beta}$ by LiCl attenuated RPL-induced ${\beta}$-catenin phosphorylation. Based on these findings, RPL may be a potential candidate for the development of chemopreventive or therapeutic agents for human cancer.

Ziziphus jujuba Mill. (ZJM), a traditional folk medicine and functional food in South Korea and China, has been reported to having pharmacological activities against anti-cancer, anti-oxidative, and anti-obesity. However, the effect of ZJM related to myoblast differentiation has not been known. In this study, we investigated the effects and mechanism of ZJM on myogenic differentiation of C2C12 cells. ZJM promotes myogenic differentiation and elevates the formation of multinucleated myotube compared to the control group. ZJM significantly increased the mRNA and protein expression of MyHC1, myogenin and MyoD in dose- and time-dependent manner. Interestingly, ZJM significantly inhibited the mRNA and protein expression of protein degradation markers, atrogin-1 and MuRF-1, in dose- and time-dependent manner. Taken together, our data suggest that ZJM is a potential functional candidate for muscle growth and strength by promoting myogenic differentiation.
Compost is widely used as an organic additive to improve the bioremediation of diesel-contaminated soil. In this study, the effects of compost amendment on the remediation performance, functional genes, and bacterial community are evaluated during the bioremediation of diesel-contaminated soils with various ratios of compost (0-20%, w/w). The study reveals that the diesel removal efficiency, soil enzyme (dehydrogenase and urease) activity, soil CH4 oxidation potential, and soil N2O reduction potential have a positive correlation with the compost amendment (p < 0.05). The ratios of denitrifying genes (nosZI, cnorB and qnorB) to 16S rRNA genes each show a positive correlation with compost amendment, whereas the ratio of the CH4-oxidizing gene (pmoA) to the 16S rRNA genes shows a negative correlation. Interestingly, the genera Acidibacter, Blastochloris, Erythrobacter, Hyphomicrobium, Marinobacter, Parvibaculum, Pseudoxanthomonas, and Terrimonas are strongly associated with diesel degradation, and have a strong positive correlation with soil CH4 oxidation potential. Meanwhile, the genera Atopostipes, Bacillus, Halomonas, Oblitimonas, Pusillimonas, Truepera, and Wenahouziangella are found to be strongly associated with soil N2O reduction potential. These results provide useful data for developing technologies that improve diesel removal efficiency while minimizing greenhouse gas emissions in the bioremediation process of diesel-contaminated soil.
