Objective: Caseins and fatty acids of milk are synthesized and secreted by the epithelial cells of the mammary gland. All-trans retinoic acid (ATRA), an active metabolite of vitamin A, has been shown to promote mammary development. This study was conducted to determine the effect of ATRA on casein synthesis and fatty acid composition in MAC-T cells. Methods: MAC-T cells were allowed to differentiate for 4 d, treated with ATRA (0, 1.0, 1.5, and 2.0 μM), and incubated for 3 d. We analyzed the fatty acid composition, the mRNA expression of casein and fatty acid synthesis-related genes, and the phosphorylation of casein synthesis-related proteins of MAC-T cells by gas chromatography, quantitative polymerase chain reaction, and western blotting, respectively. Results: In MAC-T cells, ATRA increased the mRNA levels of αS1-casein and β-casein, janus kinase 2 (JAK2) and E74-like factor 5 of the signal transducer and activator of transcription 5 β (STAT5-β) pathway, ribosomal protein S6 kinase beta-1 (S6K1) and eukaryotic translation initiation factor 4E binding protein 1 of the mammalian target of rapamycin (mTOR) pathway, inhibited the mRNA expression of phosphoinositide 3-kinase and eukaryotic initiation factor 4E of the mTOR pathway, and promoted the phosphorylation of STAT5-β and S6K1 proteins. Additionally, ATRA increased the de novo synthesis of fatty acids, reduced the content of long-chain fatty acids, the ratio of monounsaturated fatty acids to saturated fatty acids (SFA), the ratio of polyunsaturated fatty acids (PUFA) to SFA, and the ratio of ω-6 to ω-3 PUFA. The mRNA levels of acetyl-CoA carboxylase 1, fatty acid synthase, lipoprotein lipase, stearoyl-CoA desaturase, peroxisome proliferator-activated receptor gamma, and sterol regulatory element-binding protein 1 (SREBP1) were enhanced by ATRA. Conclusion: ATRA promotes the synthesis of casein by regulating JAK2/STAT5 pathway and downstream mTOR signaling pathway, and it improves the fatty acid composition of MAC-T cells by regulating SREBP1-related genes.
세포 내 소기관들과 소포들은 세포 내에서 미세소관을 따라 적절한 구획으로 수송된다. 이러한 세포 내 수송과정은 분자 모터단백질인 kinesin과 dynein에 의하여 이루어진다. Kinesin 1은 오징어 축삭돌기 세포질로부터 처음 분리되었으며 2개의 중쇄단위체(KHCs, 또는 KIF5s) 및 이와 결합하는 경쇄단위체(KLCs)의 복합체를 형성한다. KIF5s는C-말단 고리 영역을 통해 많은 다양한 단백질과 결합하는데, 아직 그 결합단백질들은 충분히 밝혀지지 않았다. 본 연구에서는 KIF5A 결합단백질을 분리하기 위하여 효모 two-hybrid 탐색을 수행하여 스트레스 과립형성과 mRNP 위치결정에 관여하는 Ras-GTPase-activating protein (GAP) Src homology3 (SH3)-domain-binding protein 2 (G3BP2)를 분리하였다. G3BP2는 KIF5A의 C-말단 고리 영역에 존재하는 73개 아미노산을 포함하는 영역과 결합하였다. 그러나 G3BP2는 KIF5B, KIF5C, KLC1, KIF3A와는 결합하지 않았다. KIF5A는 G3BP2의 arginine-glycine-glycine(RGG)/Gly-rich 도메인과 결합하지만 G3BP1과는 결합하지 않았다. HEK-293T세포에 G3BP2와 KIF5A를 발현하여 면역침강한 결과 G3BP2와 KIF5A는 같이 침강하였다. 또한 HEK-293T 세포 내의 전체에서 두 단백질은 같은 부위에 존재하였다. 이러한 결과들은 세포 내에서 G3BP2는 KIF5A와 결합하는 결합단백질로 확인 되었다.
Subpopulations of nucleotides that bind specifically to a variety of proteins have been isolated from a population of random sequence RNA/DNA molecules. Roughly one in $10^{13}$ random sequence RNA/DNA molecules folds in such a way as to create a specific binding site for small ligands. Since the development of in vitro selection procedure, more than 50 nucleic acid ligands (aptamers) have been isolated. These molecules are very useful for the study of molecular recognition between nucleic acid and protein/organic compound. In addition to these basic studies this method gives us a dream to produce new drugs against several diseases. We focused on several aptamers which specifically binds to trypsin-like serine proteases (thrombin, human neutrophil elastase, activated protein C and NS3 protease of human hepatitis C virus) and want to introduce their structural characteristics and some functions.

Hub cells comprise a niche for germline stem cells and cyst stem cells in the Drosophila testis. Hub cells arise from common somatic gonadal precursors in embryos, but the mechanism of their specification is still poorly understood. Here we find that RNA binding proteins Lin28 and Imp mediate transcript stability of Bowl, a known hub specification factor; Bowl transcripts were reduced in the testis of Lin28 and Imp mutants, and also when RNA-mediated interference against Lin28 or Imp was expressed in hub cells. In tissue culture Luciferase assays involving the Bowl 3'UTR, stability of Luc reporter transcripts depended on the Bowl 3'UTR and required Lin28 and Imp. Our findings suggest that proper Bowl function during hub cell specification requires Lin28 and Imp in the testis hub cells.
Regulation of cell fate and lung cell differentiation is associated with Aminoacyl-tRNA synthetases (ARS)-interacting multifunctional protein 2 (AIMP2), which acts as a non-enzymatic component required for the multi-tRNA synthetase complex. In response to DNA damage, a component of AIMP2 separates from the multi-tRNA synthetase complex, binds to p53, and prevents its degradation by MDM2, inducing apoptosis. Additionally, AIMP2 reduces proliferation in TGF-β and Wnt pathways, while enhancing apoptotic signaling induced by tumor necrosis factor-α. Given the crucial role of these pathways in tumorigenesis, AIMP2 is expected to function as a broad-spectrum tumor suppressor. The full-length AIMP2 transcript consists of four exons, with a small section of the pre-mRNA undergoing alternative splicing to produce a variant (AIMP2-DX2) lacking the second exon. AIMP2-DX2 binds to FBP, TRAF2, and p53 similarly to AIMP2, but competes with AIMP2 for binding to these target proteins, thereby impairing its tumor-suppressive activity. AIMP2-DX2 is specifically expressed in a diverse range of cancer cells, including breast cancer, liver cancer, bone cancer, and stomach cancer. There is growing interest in AIMP2-DX2 as a promising biomarker for prognosis and diagnosis, with AIMP2-DX2 inhibition attracting significant interest as a potentially effective therapeutic approach for the treatment of lung, ovarian, prostate, and nasopharyngeal cancers.
eIF4E는 번역개시과정에서 중심조절자 역할을 한다. eIF4E와 eIF4G의 결합이 mRNA의 번역을 촉발하기 때문에, 여러 단백질들이 이 결합을 저해함으로써 번역과정을 조절한다. 인간 4E-T는 eIF4E 결합단백질 중의 하나로, 결합한 mRNA의 번역을 저해할 뿐 아니라, eIF4E를 processing body (P-body)로 이동시키는 기능을 가지고 있다. Clast4는 인간의 4E-T와 상동성을 가지는 생쥐 단백질로 번역 조절에 중요한 기능을 할 것으로 추측되지만, 그 특징은 아직 잘 알려져 있지 않다. 본 연구에서는 Clast4의 인산화된 상태와 eIF4E와의 결합력, Clast4 과발현시 세포증식의 변화에 대한 특징을 관찰하였다. Clast4는 PKA에 의해 in vivo에서 아미노말단의 몇몇 잔기가 인산화되는 것으로 확인되었다. 그러나 PKA에 의해 인산화된 Clast4는 eIF4E와의 결합력이나 Clast4의 세포 내 위치에는 큰 변화가 없었다. Clast4는 eIF4E1과 CPEB와 결합하며, Clast4의 보존된 eIF4E 결합 서열인 $YXXXXL_{\phi}$가 eIF4E1A와의 결합에서는 중요하지만 eIF4E1B와의 결합에서는 큰 영향이 없는 것으로 관찰되었다. 잘 알려져 있는 eIF4E 조절자인 4E-BP의 경우와 유사하게 Clast4를 과발현하였을 때 세포의 증식이 감소되었다. 이러한 결과는 Clast4가 세포 내에서 전반적인 번역 조절에 관여하고 있다는 것을 시사한다.
사람의 섬유아세포인 GM10을 사용하여 glucose의 배양조건에 다른 물질대사 및 IGFBP-3 발현을 살펴본 결과, glucose 농도에 따른 glucose 소비와 triglyceride 축적 수준은 고농도 glucose 배양 조건에서 증가한 반면, 총 단백질 함량은 고농도 glucose배양 조건에서 시간의 경과에 따라 감소하였다. 또한 고농도 glucose 배양 조건에서 5일 동안 배양한 세포 배양액내의 유리아미노산 함량은 저농도보다 고농도 glucose 배양 조건에서 높게 나타났다. IGFBP-3 단백질 수준과 mRNA수준은 저농도 glucose배양 조건에서 증가하였으나, IGFBP-3 단백질 분해효소에 따른 영향은 없었다. 이상의 결과에서 glucose 배양조건에 따른 물질대사는 부분적으로 세포를 이용한 당뇨병 모델 실험에서 in vivo와 같은 결과를 얻지 못하였지만, IGFs와 같은 세포 성장인자에 대한 연구를 통해 세포수준의 당뇨병 모델화가 가능하다고 여겨진다.
최근 시상하부에서 생성되는 nesfatin-1/NUCB2가 섭식과 에너지 대사를 조절한다는 사실이 새롭게 밝혀졌다. 본 연구에서는 이러한 단백질이 생쥐의 생식기관에서도 발현을 하는지, 그리고 그 수용체가 생식기관 내에 존재하는 지를 확인함으로써 nesfatin-1이 생식기능에 미칠 수 있는 가능성을 알아보고자 하였다. 암컷 생쥐에서 난소와 자궁을 획득하여 conventional PCR 방법으로 NUCB2 mRNA 발현을 조사하였고, real-time PCR 방법으로 상대적인 NUCB2 mRNA 발현량을 비교 분석하였다. 난소 내 nesfatin-1 단백질의 발현 위치를 조사하기 위하여 nesfatin-1 항체를 이용한 면역조직화학염색법을 수행하였으며, biotin conjugated nesfatin-1을 이용하여 nesfatin-1 결합 부위를 확인하였다. 또한 생식소 내 NUCB2 mRNA 발현이 성선자극호르몬에 의해 영향을 받는지 알아보기 위해 PMSG 투여 후 NUCB2 mRNA 발현량을 조사하였다. 실험 결과, 생쥐의 난소와 자궁에서 확인된 NUCB2 유전자가 시상하부에서 만큼이나 많은 양이 발현되고 있었다. 면역조직화학적 염색 결과, nesfatin-1 단백질은 협막세포와 대부분의 기질세포에서 발현되었고, 일부 황체세포에서도 발현이 확인되었다. 반면, 난포 내 과립세포에서는 발현되지 않았으나, 특정 난포 내 난자에서는 발현됨을 확인하였다. 한편, nesfatin-1 단백질의 결합 부위는 난소 백막 주위의 기질세포와 협막세포에서 관찰되었다. 또한 PMSG 투여 후 난소와 자궁에서 NUCB2 mRNA의 발현이 유의하게 증가함을 확인하였다. 이상의 결과에서 난소 내 nesfatin-1 단백질의 발현과 그 결합 부위의 존재는 nesfatin-1이 뇌에서 뿐만 아니라 생식기관에서도 국부조절인자로써 중요한 역할을 할 것으로 사료되며, 앞으로 생식기관에 미치는 nesfatin-1의 역할을규명하기위한더많은연구가필요하다고판단된다.
세포내 수송 기구는 세포의 작용과 생존에 필수적이다. 이러한 세포내 수송은 긴 미세소관을 따라서 운반체를 운반하는 미세소관 의존 분자 모터 단백질인 kinesin과 cytoplasmic dynein에 의하여 이루어진다. Kinesin은 ATP 의존적으로 미세소관의 plus-end방향으로 이동하는 모터 단백질로 세포내 소기관, 분비소포, RNA 복합체, 단백질 복합체들을 수송한다. Kinesins에 의한 다양한 운반체의 수송의 이상은 세포의 기능 이상과 연관된다. Kinesins에 의한 운반체 수송의 기본 단계는: 운반체 혹은 adaptor 단백질과의 결합, kinesin 기능 활성화와 미세소관을 따라서 이동, 그리고 올바른 위치에서 운반체와의 분리 단계로 나뉘어 진다. 최근의 연구결과들에서 kinesin 모터 기능 활성화, 운반체와의 결합, 운반체와의 해리 기전이 확인되고 있으며 세포내 운반체 수송은 kinesin과 운반체를 연결하는 adaptor 단백질에 의하여서도 조절된다. 단백질 인산화 효소, 탈 인산화 효소를 포함하는 kinesin 모터 활성 조절 단백질들은 kinesin의 인산화 혹은 탈 인산화를 통하여 직접적으로 세포내 수송을 조절하거나, c-Jun NH-terminal kinase-interacting proteins (JIPs)와 같은 adaptor 단백질들과 미세소관의 간접적 수식을 통하여 세포내 수송을 조절하기도 한다. 이러한 연구결과들은 세포의 기능과 형태 유지에 관여하는 kinesin에 의한 다양한 세포내 수송 조절 기전을 이해하는데 기초적인 토대가 된다. 또한 각각의 kinesin에 대한 조절 기전을 밝히는 것은 세포생물학과 신경생리학을 이해하는데 중요하므로 본 종설에서는 kinesin에 의한 세포내 수송을 조절하는 단백질과 kinesin과 수송체와의 결합이 어떻게 조절되는지를 고찰하고자 한다.
본 실험에서는 C형 간염바이러스 (HCV)의 외피 단백질인 E2 당단백질에 결합하는 세포단백질들을 클로닝하기 위해 간세포 cDNA를 phage 표면에 발현시킨 phage library를 제작하였고, 12-mer peptide library와 함께 E2 단백질에 대해 panning을 실시하였다. 검색결과 세포내 신호전달과 cytoskeleton 구성에 관여하는 tensin, membrane protein band 4.1 등 세포질내 단백질과 CCR7, CKR-L2, insulin-like growth factor-1 receptor 등 세포막 단백질 등이 확인되었다. 이들 단백질들을 발현하는 phage들은 수용성 E2단백질을 이용한 결합중화반응 결과 E2 단백질에 특이적으로 결합함이 확인되었다. 사람 T 세포에서 주로 발현되는 CCR7 유전자를 PHA로 활성화된 사람 T 세포의 total RNA를 이용하여 증폭하고 클로닝하였다. 293T 세포에 transfection시켜 단백질 발현양상을 flow cytometer로 분석하여 70% 이상의 세포들이 CCR7을 발현하고 있음을 관찰하였다. 수용성 E2 단백질을 CCR7이 transfection된 세포와 mock transfection된 대조군 세포에 각각 반응시킨 결과 dose-dependent 양상으로 CCR7에 결합하였다.
