Antimicrobial Activity of the Solvent Extract from Different Parts of Orostachys japonicus (와송 부위별 추출물의 항균활성)

  • This study was to determine the inhibitory effect against food borne pathogens of ethanol and water extracts from leaf, stem and root of Orostachys japonicus. On the paper disc assay, no detectable bactericidal activity in the water extracts from leaf, stem and root of Orostachys japonicus and ethanol extracts form stem and root of Orostachys japonicus was shown. However, ethanol extract of Orostachys japonicus leaf showed the highest antimicrobial activity. Minimum inhibitory concentration (MIC) of ethanol extracts was determined to range from 0.05 to 0.1% in leaf of Orostachys japonicus against gram positive bacteria and yeast. Antimicrobial activity of ethanol extracts was stable by heating at $121^{\circ}C$ for 15 min, and not affected by pH $2{\sim}10$ except for B. subtilis. These findings suggest ethanol extract from leaf of Orostachys japonicus may be useful as natural preservative.

A Clinical Study on Relevance between Classified Dampness-Phlegm Pattern of Metabolic Syndrome and Heart Rate Variability (대사증후군의 습담 변증군과 심박변이도의 관련성에 관한 고찰)

  • Objectives : This study was performed to investigate characteristic indice of heart rate variability (HRV) according to presence of metabolic syndrome (MS), pattern identification. Methods : We analyzed 167 subjects who participated in a stroke check-up. We classified the subjects into two groups as MS (n=48) and non-MS (n=119) based on NCEP ATP III. First, we investigated pattern identification of all subjects based on Korean Standard Pattern Identifications for Stroke-III. Second, we surveyed significance of HRV indices between MS and non-MS groups. Third, we investigated significance of HRV indices between classified DP of MS and classified DP of non-MS. Fourth, we investigated significance of HRV indices between classified DP of MS and non-MS. Fifth, we investigated significance between components of MS and HRV indices. Results : 1. SDNN, RMS-SD, HF, Ln(HF), and HF (NORM) were significantly lower in the MS than non-MS group. LN (TP), LF/HF were significantly higher in the MS than non-MS. 2. Ln (HF), Ln (TP), and LF/HF were statistically significant in the classified DP of MS group. 3. RMS-SD, and Ln (HF) were significantly lower in the abdominal obesity group than in the non-abdominal obesity group. SDNN, RMS-SD, TP, Ln (TP), VLF, Ln (VLF), and Ln (LF) were significantly lower in the hypertension group than in the non-hypertension group. RMS-SD was significantly lower in the diabetes group than in the non-diabetes group. Mean HRT,LF/HF were significantly higher in the hypertriglyceridemia than non-hypertriglyceridemia group, but Ln (HF), RMS-SD, and HF (NORM) were significantly lower in the hypertriglyceridemia than non-hypertriglyceridemia group. Conclusions : We found a few significant relationships between classified DP of MS and HRV indices. The resluts demonstrate that MS has potentially negative effects on the cardiovascular system, and these effects could be detected by HRV.

Antioxidative and Anticancer Activities of Ethanol Extract of Millettia erythrocalyx (Millettia erythrocalyx 에탄올 추출물의 항산화 활성 및 항암 활성에 관한 연구)

  • Millettia erythrocalyx, a species of plant in the Fabaceae family, is widely distributed in the tropical and subtropical regions of the world, such as the Indies, China, and Thailand. The antiviral activity of flavonoids from M. erythrocalyx has been reported; however, the antioxidative and anticancer activities of M. erythrocalyx remain unclear. In this study, we evaluated the antioxidative and anticancer effects of ethanol extract of M. erythrocalyx (EEME) and the molecular mechanism of its anticancer activity in human hepatocellular carcinoma HepG2 cells. EEME exhibited significant antioxidative effects, with a concentration at 50% inhibition ($IC_{50}$) value of $2.74{\mu}g/ml$, as measured by 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging assay; moreover, it inhibited cell proliferation in a dose-dependent manner in HepG2 cells. Cell cycle analyses showed that EEME induced HepG2 cell accumulation in the subG1 phase in a dose-dependent manner. EEME also induced apoptosis of HepG2 cells, with increases in apoptotic cells and apoptotic bodies, as detected by Annexin V and 4,6-diamidino-2-phenylindole (DAPI) staining, respectively. Treatment with EEME resulted in increased expression of First apoptosis signal (Fas), a death receptor, and Bcl-2-associated X protein (Bax), a proapoptotic protein, and the activation of caspase-3, 8, and 9, resulting in the cleavage of poly (Adenosine diphosphate-ribose) polymerase (PARP). Collectively, these results suggest that EEME may exert an anticancer effect in HepG2 cells by inducing apoptosis via both the intrinsic and extrinsic pathways.

Tag-SNP selection and online database construction for haplotype-based marker development in tomato (유전자 단위 haplotype을 대변하는 토마토 Tag-SNP 선발 및 웹 데이터베이스 구축)

  • This report describes methods for selecting informative single nucleotide polymorphisms (SNPs), and the development of an online Solanaceae genome database, using 234 tomato resequencing data entries deposited in the NCBI SRA database. The 126 accessions of Solanum lycopersicum, 68 accessions of Solanum lycopersicum var. cerasiforme, and 33 accessions of Solanum pimpinellifolium, which are frequently used for breeding, and some wild-species tomato accessions were included in the analysis. To select tag-SNPs, we identified 29,504,960 SNPs in 234 tomatoes and then separated the SNPs in the genic and intergenic regions according to gene annotation. All tag-SNP were selected from non-synonymous SNPs among the SNPs present in the gene region and, as a result, we obtained tag-SNP from 13,845 genes. When there were no non-synonymous SNPs in the gene, the genes were selected from synonymous SNPs. The total number of tag-SNPs selected was 27,539. To increase the usefulness of the information, a Solanaceae genome database website, TGsol (http://tgsol. seeders.co.kr/), was constructed to allow users to search for detailed information on resources, SNPs, haplotype, and tag-SNPs. The user can search the tag-SNP and flanking sequences for each gene by searching for a gene name or gene position through the genome browser. This website can be used to efficiently search for genes related to traits or to develop molecular markers.

Analysis of current nursing homes in Korea and to assess the role and prospect of dentists in comparison to Japanese long term care insurance system (한국노인요양시설 구강 위생 현황과 일본 노인요양보험법 비교를 통한 치과의사의 역할과 전망)

  • Statement of problem: The introduction of "The Long-term Care Insurance System" (a public nursing care insurance scheme) is scheduled from July in 2008. Lately, the importance of oral health care had increased. Introduction and establishment of the methodology (nursing care procedure) based on professional dental knowledge is inevitable. Purpose: The purpose of this study was to rouse recognition of the importance of dental care in the long term elderly care in nursing homes, with implementation of the new insurance law. Material and methods: Visited two institutions for the elderly, Yudang Village and Sungjiwon located in Suwon city to investigate the present conditions in terms of (1) the detailsof the institution, (2) the activities concerning with dental care in the institution, and (3) the consciousness and recognition regarding dental care of the staffs. Results: In two institutions, under the present conditions, oral cleaning (including the cleaning of denture) for residents was operated with no professional advices and limited professional dental care. It was found that there was very little awareness of aspiration pneumonia. The members of staff however, did recognizethe necessity of professional maintenance and management of oral care in daily nursing care, and that many residents hold dental problems. They were very eager in introducing the methodology (nursing care procedure) in dental care in nursing homes.

Anti-Visceral Obesity Effect of Apios americana Medikus in Diet-Induced Obese Mice (식이로 유도한 비만마우스에서 아피오스의 내장지방 감소 효과)

  • This study examined the effects of apios (Apios americana Medikus) supplementation on visceral obesity in high-fat diet-induced obese mice. C57BL/6N mice were fed a high-fat diet (40% calories from fat) with or without apios powder (10%, w/w) for 12 weeks. At the end of the experiment, apios supplementation reduced visceral fat mass significantly by 14.3% compared to the control group. Apios decreased significantly the atherogenic index, serum leptin level, hepatic lipid (free fatty acid and triglyceride) content, and lipid droplets, whereas it increased the serum high density lipoprotein-cholesterol/total cholesterol ratio. Hepatic lipogenic gene expression of peroxisome proliferator activated receptor gamma, fatty acid synthase, and diacylglycerol O-acyltransferase 2 was down-regulated by apios supplementation. These results suggest that apios is a healthy food for preventing high-fat diet-induced visceral obesity and fatty liver.

  • The status of the forest owners and the ownership arrangement has been surveyed in the joint ownership forest of the total 250.59 ha, 17 plots including plot No. 2, in Cha-ri, Duseo-myeon, Ulju-gun, Kyongnam. It has been surveyed that the total number of the practical owners relevant to the ownership is 133 persons. However, the number of the owners is recorded as 55 person in the official documents and there has been a big difference between the number of the practical owners and that of the practical owners and that of the owners in the official documents. As 79 forestowners, 59% of the total forestowners, can not be certainly guaranteed in the execution of their legal rights to the ownership, it could be one of the factors which will hinder the promotion of the investment willingness to forest works. In order to go through with the formality of the ownership registration correspond to reality, some necessary works such as the boundary survey should be followed. Therefore it is required that any governmental or public organizations should take part in technical and financial aids to solve these matters.

A study on obesity pattern and related factors of the doctors (의사를 대상으로한 비만양상과 그 관련요인에 관한 연구)

  • The author surveyed overall obesity indicies and factors concerned with obesity such as dietary intake, physical activity, stress and life style with the subject of doctors. The number of subjects was total 508 with 396 men and 112 women. They were subgrouped into surgical part, medical part and service and basic part by speciality. And also subgrouped into intern and resident, pay doctor, and practitioner by working type. The results were as follows. 1) Obesity indices: BMI of total doctor was $23.1{\pm}2.8$, and WHR was $0.87{\pm}0.08$ and overweight prevalence(BMI>25.0) was 23.6%. It was within normal limit but slightly over the Korean standard. The degree of obesity indices of subgroups by speciality was 'surgical part > medical part > service and basic part', and by working type was 'practitioner > pay doctor > intern and resident'. 2) Dietary intake and Physical activity: Average dietary intake was $2148{\pm}451kcal/day$. The degree of dietary intake by speciality was 'surgical part > medical part > service and basic part'. By working type it was 'practitioner > pay doctor > intern and resident'. Average physical activity was $29{\pm}5$ METs/day. The degree of physical activity also showed similiar pattern. But there was no significant difference among each groups. 3) Comparision between over-weight and non-over-weight group: The items that showed significant difference between two groups were dietary intake, skip breakfast, regular exercise, smoking, heavy drinking, chronic disease etc.

Suppression of Nitrate Accumulation in Vegetables by Foliar Application of Micronutrients (미량원소 엽면 처리에 의한 엽채류의 질산태 질소 축적 억제)

  • Suppression of nitrate accumulation in vegetables through foliar application of micronutrients was investigated. Spinach and lettuce were grown in pots under greenhouse condition. Micronutrient solutions containing Cu, Mn, Mo, and Zn were used; chitosan was added into one and the other contained chitosan oligomers. The micronutrient solutions were sprayed on the leaves at 3 and 4 weeks after transplanting of 20-day-old seedlings. Plants were harvested at 5-weeks after transplanting. Yield, contents of chlorophyll, Brix value, micronutrient, and nitrate, and nitrate reductase activity were measured. Fresh weights of lettuce and spinach were significantly increased by the foliar application of micronutrients. Contents of chlorophyll and micronutrients were higher in micronutrient-treated plants, while those of nitrate were reduced by about 10 and 14-23% in lettuce and spinach, respectively. Compared to the control plants, nitrate reductase activity was higher in plants treated with micronutrients. Results of this study indicate the effect of micronutrients on the suppression of nitrate accumulation was relatively small in comparison to the contents of nitrate in leaves of spinach and lettuce. To maximize the effect, nutrient composition in solution, application time, and frequency should be further examined, taking into consideration nitrogen level in soil and other environmental factors including light condition.

Rare Metal Occurrences within the Anorthosite in the Hadong-Sanchong area, Kyungnam Province, Korea (하동-산청지역 회장암에 배태된 희유금속자원에 관한 연구)

  • Allanite crystals rich in rare-earth elements(REE) occur in soil developed on top of anorthositic rocks in the Jungsu-ri area of Okjong-myun, Hadong-run, where large Ti orebodies are embedded in the bed rock. In this study allanite is investigated mainly by transmitted light microscopy, electron microprobe analysis, atomic absoption spectrophotometry, X-ray diffraction, infrared spectrocopy. In addition, its specific gravity and micro=indentation hardness value are measured. Allanite occurs with max. dimension of $3cm{\times}6cm$ and coexists with quartz, epidote, zircon, biotite and muscovite. It shows nearly nonmetamict crystallinity, although ${\alpha}$-particles bombardment from the disintegration of the radioactive element Th is detected by an autoradiography. The allanite is particularly enriched in REE(19.88-23.99 wt.%), but is deficient in CaO(8.35-10.29wt.%). Genesis of the allanite in this area is not understood yet. It is, however, assumed to have been formed from magmatic fluid rich in REE and Ti, based on the facts that it ocexists with zircon and that it has high $TiO_2$(0.89-1.13 wt.%) whose concentration is significant in the country rocks.
